These sponges harbor diverse bacteria with metabolic potentiality. From biodiversity to biotechnological prospecting, scientific investigations related to sponge associated microorganisms have expanded, but remain rather limited to few geographic locations. In this study, culturable bacteria associated with two demosponges viz Dysidea granulosa, Sigmadocia fibulata and the ambient water were screened for commercially important enzymes such as amylase, protease, gelatinase, lipase, deoxyribonucleic, phosphatase and urease. Amylase and phosphatase were the predominant enzymes produced by >80% of sponge-associated bacteria compared to the ambient water. Nearly 50% of the sponge-associated bacteria expressed multiple enzymatic activities (> 4) with variation in the percentage of expression of individual enzymes. More than 65% of the culturable heterotrophic bacteria associated with sponges were Gammaproteobacteria. The order Vibrionales was the main source for multiple enzyme production. Sponge associated bacteria formed more closely related clusters than the water isolates based upon their activity pattern. High recovery of sponge-associated bacteria with multiple enzymatic activities suggest that these versatile bacteria are yet unexploited potential for bioprospecting.展开更多

By plate assay, lipase, protease, amylase,...The potential of 324 bacteria isolated from different habitats in polar oceans to produce a variety of extracellular enzymatic activities at low temperature was investigated. By plate assay, lipase, protease, amylase, gelatinase, agarase, chitinase or cellulase were detected. Lipases were generally present by bacteria living in polar oceans. Protease-producing bacteria held the second highest proportion in culturable isolates. Strains producing amylase kept a relative stable proportion of around 30% in different polar marine habitats. All 50 Arctic sea-ice bacteria producing proteases were cold-adapted strains, however, only 20% were psychrophilic. 98% of them could grow at 3% NaCl, and 56% could grow without NaCl. On the other hand, 98% of these sea-ice bacteria produced extracellular proteases with optimum temperature at or higher than 35℃, well above the upper temperature limit of cell growth. Extracellular enzymes including amylase, agarase, cellulase and lipase released by bacteria from seawater or sediment in polar oceans, most expressed maximum activities between 25 and 35℃. Among extracellular enzymes released by bacterial strain BSw20308, protease expressed maximum activity at 40℃, higher than 35℃ of polysaccharide hydrolases and 25℃ of lipase.展开更多
Endophytic fungi are being investigated for their ability to create industrially relevant secondary metabolites.In recent years,there has been a surge in interest in these fungi as a source of novel enzymes,particular...Endophytic fungi are being investigated for their ability to create industrially relevant secondary metabolites.In recent years,there has been a surge in interest in these fungi as a source of novel enzymes,particularly hydrolytic enzymes.The present study investigated the effect of different carbon,organic and inorganic nitrogens on the growth and ligninolytic enzyme production by the endophytic fungus Diaporthe phaeolorum.The fungus was isolated from the leaves of Dillenia indica and analyzed by morpho-molecular basis.The fungus showed promising results for in vitro production of ligninolytic enzymes.Sucrose was the most favorable carbon compound for growth among all the carbon compounds tested.It displayed maximum lignin peroxidase(Lip)activity in fructose(3.5 U/ml),followed by pectin(2.60 U/ml)and glucose(2.53 U/ml).Glucose gave the highest manganese peroxidase(MnP)activity i.e.,6.88 U/ml followed by starch,sucrose and raffinose.Similarly,the maximum laccase activity was 44.5 U/ml in pectin.Potassium nitrate and L-asparagine were the best inorganic and organic nitrogen for growth.In the case of ligninolytic enzyme production,ammonium acetate and ammonium phosphate were the best media for LiP and MnP,whereas laccase production was highest in ammonium nitrate supplemented medium.In organic nitrogen sources,medium supplemented with DL-tryptophan gave the highest Lip production,whereas MnP and laccase production was observed in the medium containing L-tyrosine and L-asparagine.To the best of our knowledge,this is the first report related to the growth and ligninolytic enzyme production by Diaporthe phaseolorum.The findings from the study will assist researchers in improving the production of ligninolytic enzymes by this fungus under in vitro conditions on an industrial scale.展开更多
