Fosmid library construction and screening for the maize mutant gene Vestigial glume 1

The maize mutant gene Vestigial glume 1(Vg1) has been fine-mapped to a narrow region by map-based cloning and the candidate gene for Vg1 spanned 19.5 kb. Here we report Vg1 genomic fosmid library construction and screening. The fosmid library of Vg1 consisted of574,000 clones with an average insert size of 36.4 kb, representing 7.9-fold coverage of the maize genome. Fosmid stability assays indicated that clones were stable during propagation in the fosmid system. Using Vg1 candidate gene-specific primers, a positive clone was successfully identified. This discovery will pave the way for identifying the function of Vg1 in maize development.

Construction and characterization of a cDNA library from human liver tissue with chronic hepatitis B

Objective: To construct a cDNA library from human liver tissue with chronic hepatitis B and check its quality for investigating the expression level of liver tissue infected by hepatitis B virus. This will then be used to find the relevant genes and interesting proteins associated with the development of hepatitis B. Methods: The total RNA from liver tissue with chronic hepa- titis B was extracted and the mRNA was purified using TRIZOL method. Switching mechanism at 5′ end of the RNA transcript (SMART) technique and CDS III/3′ primer were used for first-strand cDNA synthesis. Long distance polymerase chain reaction (LD PCR) was then used to synthesize the double-strand cDNA that was then digested by Sfi I and fractionated by CHROMA SPIN-400 column. The longer than 0.4 kb cDNAs were collected and ligated to λTriplEx2 vector. Then λ phage packaging reaction and library amplification were performed. The qualities of both unamplified and amplified cDNA libraries were strictly checked by conventional titer determination. Fourteen plaques were randomly picked and tested using PCR with universal primers derived from the sequence flanking the vector. Results: The titers of unamplifed and amplified libraries were 1.94×106 pfu/ml and 1.49×109 pfu/ml respectively. The percentages of recombinants from both libraries were 98.15% in unamplified library and 98.76% in amplified library. The lengths of the inserts were 1.23 kb in average, 1?2 kb in 64.29%, and 0.5?1.0 kb in 35.71%. Conclusion: A high quality cDNA library from human liver tissue with chronic hepatitis B was successfully constructed.

Construction of human Fab library and screening of a single-domain antibody of amyloid-beta 42 oligomers

Screening humanized antibodies from a human Fab phage display library is an effective and quick method to obtain beta-amyloid oligomers. Thus, the present study prepared amyloid-beta 42 oli- gomers and constructed a have human Fab phage display library based on blood samples from six healthy people. After three rounds of biopanning in vitro, a human single-domain antibody that spe- cifically recognized amyloid-beta 42 oligomers was identified. Western blot and enzyme-linked im- munosorbent assay demonstrated this antibody bound specifically to human amyloid-beta 42 tetramer and nonamer, but not the monomer or high molecular weight oligomers. This study suc- cessfully constructed a human phage display library and screened a single-domain antibody that specifically recognized amyloid-beta 42 oligomers.

An Examination on the Support of Library Information to the Construction of National Research Organizations

The country’s economic development determines the political and cultural status.At the present time,China’s rapid economic development has greatly improved the country’s international prominence.Although China has high economic and political achievements,China’s cultural soft power is still in a weak position.Cultural soft power is of great significance in improving the country’s overall national strength and international competitiveness.Research organizations are an important part of the country’s soft power and are of great significance to the strengthening of national soft power.Research organizations are the“peripheral brain”in the decision-making process of the government.National government agencies need democratic science in making decisions,and the adopted policy methods must have positive and effective outcomes on the people.Therefore,the national government agencies cannot achieve this without the help of research organizations.This resulted in the development and rising construction of research organizations.With the comprehensive development and construction of research organizations,whilst making progress,has also exposed several shortcomings.The construction of China’s research organizations requires various supports;especially the intelligence agencies and access to library information,which has a unique advantage in promoting the construction of research organizations[1].This paper first introduces the definition,classification and characteristics of a research organization.It will also provide a preliminary understanding of research organizations.The paper will then highlight and analyze the problems encountered by the research organization.Finally,through the grasp of the advantages of the library and intelligence agencies,a solution is proposed.

Research on the Construction of Chinese Universities from Intelligent Library to Smart Library

The study aims to discuss the construction and development of university library,and transform the management and operation mode of university library,thereby realizing the transformation and upgrading to smart library.Taking Anhui University of Finance and Economics as the research object,first,based on the literature and data combined with questionnaire design and expert interview,data statistics was made on the development of smart library.Second,analytical hierarchy process(AHP)and big data analysis were applied to complete the construction of the smart library system.Third,based on the data analysis method,the problems in the construction of the smart library were discussed.The results showed that the Chinese smart library started late and developed rapidly after 2012;from information perception to smart service,the construction of smart library system had realized the transformation and upgrading of library construction;the key to solve the construction of smart library was to standardize library construction,realize resource co-construction and sharing,promote technological innovation,and strengthen the construction of smart librarians.To sum up,the construction of smart library is expected to make the library and librarians become the cooperative objects of teaching and scientific research in colleges and universities.

Development and construction of China's higher education libraries

Libraries in China's higher education institutions have been developing in keeping pace with the flourishing development of China's higher education. This article aims to make an introduction to the construction of China's higher education libraries, especially the recent three decades' achievements since China's reform and opening-up in 1978. In this article, the authors draw a general picture of the development of libraries in China's higher education institutions, covering such eight aspects as management, types and positioning,organizational structure and personnel, expenditure and buildings, reader service, building and sharing of resources as well as automation system.

Construction and screening of suppression subtractive hybridization library of renal cell carcinoma

Objective To construct and screen the suppression subtractive hybridization (SSH) library of human renal cell carcinoma (RCC). Methods Poly A+ RNA was isolated from RCC lines 786-O(tester) and renal cell(RC) lines HK-2 ( driver), respectiely. SSH procedure was performed according to the protocol of the PCR-Select cDNA Subtraction Kit ( Clontech), and PCR products were cloned into pT-Adv vector and transformed E. coli TOP10F’. All positive clones picked out were digested and some of which were sequenced. Results The SSH library contained 362 clones with SSH cDNA fragments distributed mainly from 0.3 to 0.9 kb. Among 50 clones sequenced randomly,2 represented unknown genes and the other 48 derived from 36 known genes. Conclusion The quality of the SSH library of human RCC is reliable and is construction is the basis for further screening differentially expressed genes of RCC. 6 refs,4 figs, 1 tab.

[目的/意义]智慧图书馆已经成为当今图书馆学界和业界最热门的主题之一。智慧图书馆建设的理论研究与实践工作已经取得了初步成效,生成式AI也在蓬勃发展的同时参与到各行各业的建设中。因此有必要立足当下发展,放眼远景目标,确定生成式AI参与智慧图书馆建设的策略。[方法/过程]本文从智慧图书馆空间升级、服务打造、业务重组、人才培养、体系建设5个方面阐明了生成式AI视域下智慧图书馆建设的关键路径。[结果/结论]关键路径研究将为生成式AI引入智慧图书馆下一阶段的建设工作提供借鉴与依据。

数字时代图书馆法治建设高质量发展的五维审思

图书馆是文化资源的储存库和提取库,担负着凝聚民族亲和力与团结力的历史使命。数字时代《公共图书馆法》及其相关法律为数字化法治化图书馆的设立、运行及服务等提供了规范依据,呈现出布局时空、内驱外引、业态迭新、为民服务的发展优势。2023年2月中共中央、国务院相继印发《质量强国建设纲要》,指出要大力推动图书馆、博物馆等公共文化场馆数字化发展,加快线上线下服务融合。图书馆所蕴含的文化、教育、开放、共享等价值理念成为人民实现和平与发展的精神品格,有助于保障人民公共利益权利,有助于满足人民群众对文化资源获取的美好期待,有助于团结全体人民智慧共建人类文明共同体。

真实工作场景下“搜索即学习”任务集的构建与分析

文章采用日记法收集来自16个国民经济行业的真实工作场景下具有学习特征的搜索任务实例,结合任务概念框架对搜索任务进行表征,构建面向“搜索即学习”研究的多维任务集,并进一步剖析了真实工作场景下搜索任务的属性特征,对任务相关特征、参与者认知变化、行为及关系进行探讨,根据参与者的学历层次、完成任务的认知层次,以及在完成任务时所处的时间压力进行分组比较三组变量间的关系。研究发现:参与者的任务执行熟悉度与任务感兴趣程度,所需信息类型的数量显著影响认知变化,任务执行者特征对认知变化与行为的影响在不同情境下均存在显著差异。本研究开发的任务集可作为“搜索即学习”研究的公共研究资源,以及评估生成式人工智能技术复杂任务处理能力的评测集,为开发面向真实工作任务情境与高效的信息系统提供依据。
