AIM:To investigate the mRNA expression of cyclooxygensae-2(COX-2)in benign and malignant ascites,and to explore the difference in COX-2 mRNA expression among different diseases.METHODS:A total of 36 samples were collected from the Fifth Affiliated Hospital of Sun Yat-Sen University and divided into two experimental groups:benign ascites(n=21)and malignant ascites(n=15).Benign ascites included cirrhotic ascites(n=10)and tuberculous ascites(n=5).Malignant ascites included oophoroma(n=7),cancer of colon(n=5),cancer of the liver(n=6),gastric cancer(n=2),and bladder carcinoma(n=1).The mRNA expression of COX-2 in ascites was examined with reverse transcriptase polymerase chain reaction(RT-PCR)technology,and the positive rate of COX-2 mRNA was compared between different diseases.RESULTS:The positive rate of COX-2 mRNA in malignant ascites was 42.9%(9/21),which was significantly higher than in benign ascites,6.7%(1/15),difference being significant between these two groups(χ2=4.051,P=0.044).The proportion of the positive rate in the malignant ascites was as follows:ovarian cancers57.1%(4/7),colon cancer 40.0%(2/5),liver cancer33.3%(2/6),gastric cancer 50.0%(1/2),and bladder cancer 0.00%(0/1).However,there was no significant difference in COX-2 mRNA expression among various tumors with malignant ascites(χ2=1.614,P=0.806).Among the benign ascites,COX-2 mRNA levels were different between the tuberculous ascites(0/5)and cirrhotic ascites(1/10),but there was no significant difference(P=1.000).CONCLUSION:COX-2 mRNA,detected by RT-PCR,is useful in the differential diagnosis of benign and malignant ascites,which also has potential value in the clinical diagnosis of tumors. 展开更多

In order to investigate the expression of cyclooxygenase-2 (COX-2) in human lower segments of myometrium obtained from women in labor and those not in labor and identify the splicing variant of COX-2, reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of COX-2. The primers were designed and synthesized according to the sequence of rat COX-2 splice variant which was discovered firstly by us. Then the splicing variant of COX-2 in human myometrium from woman in labor was identified, cloned into vector and sequenced. The results showed that the expression of COX-2 mRNA was lower in human myometrium obtained from women who were not in labor than that in labor women and a new band of COX-2 was obtained in myometrium from labor woman. The fragment included an unspliced intron, which pitched between exons 7 and 8. It was suggested that COX-2 gene was not only expressed highly in human myometrium from woman in labor, but also produced splicing variant by alternative splicing. 展开更多

Walnut dreg protein hydrolysates(WDPHs)exhibit a variety of biological activities,however,the cyclooxygenase-2(COX-2)inhibitory peptide of WDPHs remain unclear.The aim of this study was to rapidly screen for such peptides in WDPHs through a combination of in silico and in vitro analysis.In total,1262 peptide sequences were observed by nano liquid chromatography/tandem mass spectrometry(nano LC-MS/MS)and 4 novel COX-2 inhibitory peptides(AGFP,FPGA,LFPD,and VGFP)were identified.Enzyme kinetic data indicated that AGFP,FPGA,and LFPD displayed mixed-type COX-2 inhibition,whereas VGFP was a non-competitive inhibitor.This is mainly because the peptides form hydrogen bonds and hydrophobic interactions with residues in the COX-2 active site.These results demonstrate that computer analysis combined with in vitro evaluation allows for rapid screening of COX-2 inhibitory peptides in walnut protein dregs. 展开更多

目的:探究胃癌(GC)组织中长链非编码RNA HOXC反义RNA1(lncRNA HOXC-AS1)、胰岛素样生长因子2 mRNA结合蛋白2(IGF2BP2)mRNA表达水平与患者临床病理特征及术后5年内生存的关系。方法:选取我院2015年5月至2018年4月收治的GC患者139例,手术... 目的:探究胃癌(GC)组织中长链非编码RNA HOXC反义RNA1(lncRNA HOXC-AS1)、胰岛素样生长因子2 mRNA结合蛋白2(IGF2BP2)mRNA表达水平与患者临床病理特征及术后5年内生存的关系。方法:选取我院2015年5月至2018年4月收治的GC患者139例,手术收集GC组织及癌旁组织。荧光定量PCR法检测lncRNA HOXC-AS1和IGF2BP2 mRNA表达。对GC患者进行术后随访5年,记录GC患者生存情况,分为生存组88例,死亡组51例。比较GC组织和癌旁组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平、不同临床病理特征患者GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达情况、生存组和死亡组临床病理特征和GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平。分析GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平的相关性、GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达情况与术后5年生存的关系、影响GC患者术后5年内生存的危险因素。结果:与癌旁组织相比,GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平均显著升高(P<0.05);GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平呈正相关(P<0.05);GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平与淋巴结转移、TNM分期、肿瘤分化程度相关(P<0.05);lncRNA HOXC-AS1高表达组、IGF2BP2 mRNA高表达组患者术后5年总生存率分别显著低于lncRNA HOXC-AS1低表达组、IGF2BP2 mRNA低表达组(P<0.05);生存组和死亡组淋巴结转移、TNM分期、肿瘤分化程度比较差异有统计学意义(P<0.05);死亡组患者GC组织中lncRNA HOXC-AS1和IGF2BP2 mRNA表达水平显著高于生存组(P<0.05);TNM分期为III期、肿瘤低分化、lncRNA HOXC-AS1高表达、IGF2BP2 mRNA高表达是影响GC患者术后5年内生存的独立危险因素(P<0.05)。结论:GC患者癌组织中lncRNA HOXC-AS1及IGF2BP2 mRNA表达水平均显著升高,且术后5年内死亡的GC患者较存活患者更高,二者与淋巴结转移、TNM分期、肿瘤分化程度密切相关,二者高表达是影响GC患者术后5年内生存的独立危险因素。 展开更多

目的 探究宫颈脱落细胞中mybl2 mRNA的表达在宫颈低级别鳞状上皮内病变(LSIL)患者中的分流价值。方法 180例宫颈病变患者,根据病理结果分为宫颈炎(NC)组(40例)、LSIL组(100例)及宫颈高级别鳞状上皮内病变(HSIL)组(40例)。比较三组的宫... 目的 探究宫颈脱落细胞中mybl2 mRNA的表达在宫颈低级别鳞状上皮内病变(LSIL)患者中的分流价值。方法 180例宫颈病变患者,根据病理结果分为宫颈炎(NC)组(40例)、LSIL组(100例)及宫颈高级别鳞状上皮内病变(HSIL)组(40例)。比较三组的宫颈脱落细胞中mybl2 mRNA及人乳头瘤病毒(HPV) E6/E7 mRNA表达量、阳性率;比较LSIL组中不同转归者的宫颈脱落细胞中mybl2m RNA及HPV E6/E7 mRNA表达量、阳性率;分析LSIL患者宫颈脱落细胞中mybl2 mRNA与HPV E6/E7 mRNA的相关性。结果 NC组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA表达量分别为(1.23±0.16)、(1.76±0.32), LSIL组分别为(3.39±0.59)、(5.01±1.32), HSIL组分别为(6.56±0.93)、(8.10±1.69)。三组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA表达量比较,差异有统计学意义(P<0.05);HSIL组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA表达量显著高于NC组、LSIL组, LSIL组显著高于NC组,差异有统计学意义(P<0.05)。NC组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA阳性率分别为25.00%、27.50%, LSIL组分别为76.00%、71.00%, HSIL组分别为95.00%、100.00%。三组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA阳性率比较,差异有统计学意义(P<0.05);HSIL组的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA阳性率显著高于NC组、LSIL组,LSIL组显著高于NC组,差异有统计学意义(P<0.05)。LSIL组中不同转归者的宫颈脱落细胞中mybl2mRNA及HPV E6/E7 mRNA表达量、阳性率比较,差异有统计学意义(P<0.05);进展者的宫颈脱落细胞中mybl2 mRNA及HPV E6/E7 mRNA表达量、阳性率显著高于持续者和消退者,持续者显著高于消退者,差异有统计学意义(P<0.05)。Pearson相关性分析显示, LSIL患者宫颈脱落细胞中mybl2 mRNA与HPV E6/E7 mRNA呈正相关(r=0.816, P<0.05)。结论 LSIL患者宫颈脱落细胞中mybl2 mRNA的表达与HPV E6/E7 mRNA密切相关,其在LSIL患者中的分流价值较高。 展开更多

目的分析宫颈高级别鳞状上皮内病变(HSIL)冷刀锥切术后复发的影响因素。方法收集唐山市工人医院和唐山市妇幼保健院2015年1月—2020年6月368例宫颈HSIL患者的临床病理及随访资料,所有患者均行冷刀锥切术,术前均行人乳头瘤病毒(HPV)E6/E7... 目的分析宫颈高级别鳞状上皮内病变(HSIL)冷刀锥切术后复发的影响因素。方法收集唐山市工人医院和唐山市妇幼保健院2015年1月—2020年6月368例宫颈HSIL患者的临床病理及随访资料,所有患者均行冷刀锥切术,术前均行人乳头瘤病毒(HPV)E6/E7 mRNA检测并定期复查,应用免疫组织化学方法检测宫颈组织ⅡA型拓扑异构酶(TOP2A)和p16表达情况,分析术后复发的影响因素。结果HPV E6/E7 mRNA持续感染组复发率(25.4%)显著高于未持续感染组(6.3%),TOP2A高表达组复发率(12.5%)显著高于TOP2A低表达组(2.9%),差异均有统计学意义(P<0.05)。年龄、象限受累范围、腺体受累、组织学分级、p16表达与复发无关(P>0.05)。Logistic回归分析显示HPV E6/E7 mRNA持续感染和TOP2A高表达是术后复发的独立危险因素(OR=4.989,OR=3.883;均P<0.05)。不同型别HPV持续感染复发率的差异无统计学意义(P>0.05)。HPV E6/E7 mRNA持续感染与TOP2A的高表达呈正相关(P<0.05)。结论HPV E6/E7 mRNA持续感染与TOP2A高表达是宫颈HSIL冷刀锥切术后复发的高危因素,二者可能在复发机制上存在内在联系,检测HPV E6/E7 mRNA持续感染情况及TOP2A表达对宫颈HSIL的术后临床管理具有指导意义。 展开更多

目的探讨染色体凝缩蛋白复合物I(NCAP)D2、Krüppel样因子(KLF)5蛋白及跨膜受体蛋白Notch1 mRNA对老年结肠癌患者预后的预测价值。方法选取老年结肠癌患者85例,术中采集肿瘤组织和正常组织(距离肿瘤组织5 cm以上),Western印迹检测... 目的探讨染色体凝缩蛋白复合物I(NCAP)D2、Krüppel样因子(KLF)5蛋白及跨膜受体蛋白Notch1 mRNA对老年结肠癌患者预后的预测价值。方法选取老年结肠癌患者85例,术中采集肿瘤组织和正常组织(距离肿瘤组织5 cm以上),Western印迹检测组织标本中NCAPD2、KLF5蛋白表达,反转录-聚合酶链反应(RT-PCR)检测组织标本中Notch1 mRNA表达。收集老年结肠癌患者的人口学特征和临床病理资料;以患者出院时日期为随访起点,进行3年随访,记录患者生存情况,分为预后良好组(生存),不良组(死亡)。结果老年结肠癌患者结肠癌组织中NCAPD2、KLF5蛋白及Notch1 mRNA表达水平明显高于正常组织(P<0.01)。NCAPD2、KLF5蛋白及Notch1 mRNA表达水平在不同病理分期、分化程度、淋巴结转移情况、远端转移情况的结肠癌患者差异均有统计学意义(P<0.01);其中,浸润程度T3~T4、有淋巴结转移、有远端转移的结肠癌患者NCAPD2、KLF5蛋白及Notch1 mRNA表达水平明显高于浸润程度T1~T2、无淋巴结转移、无远端转移的结肠癌患者(P<0.01);随着分化程度的降低,结肠癌患者NCAPD2、KLF5蛋白及Notch1 mRNA表达水平逐渐升高(P<0.01)。不良组NCAPD2、KLF5蛋白及Notch1 mRNA表达水平明显高于预后良好组(P<0.01)。构建受试者工作特征(ROC)曲线分析结果显示,NCAPD2、KLF5蛋白及Notch1 mRNA表达水平对老年结肠癌患者预后均具有较高的预测价值(P<0.01);各指标联合检测对老年结肠癌患者预后的预测价值最高。结论老年结肠癌患者肿瘤组织中NCAPD2、KLF5蛋白及Notch1 mRNA高表达,且与浸润程度、分化程度及转移情况密切相关,联合检测可用于患者预后评估。 展开更多

目的应用全转录组测序与实时聚合酶链锁反应(Real-time polymerase chain reaction,RT-qPCR)技术,探索参芪复方调控糖尿病GK大鼠肝脏信使RNA(messenger RNA,mRNA)、长链非编码RNA(long non-coding RNA,lncRNA)表达谱的机制研究。方法采... 目的应用全转录组测序与实时聚合酶链锁反应(Real-time polymerase chain reaction,RT-qPCR)技术,探索参芪复方调控糖尿病GK大鼠肝脏信使RNA(messenger RNA,mRNA)、长链非编码RNA(long non-coding RNA,lncRNA)表达谱的机制研究。方法采用高脂高糖饲料建立2型糖尿病模型,将GK大鼠随机分为模型组、参芪复方组和西药组。另将10只Wistar大鼠设为空白组,予普通饲料喂养。参芪复方组大鼠予参芪复方浸膏灌胃,西药组予西格列汀混悬液灌胃,模型组及空白组灌服生理盐水。干预12周,每周检测空腹血糖(fasting blood glucose,FBG)。运用HE染色法观察大鼠肝脏组织病理形态,在空白组、模型组和参芪复方组中每组随机选取4只大鼠进行全转录组测序,构建mRNA、lncRNA差异表达谱,分析差异基因生物学功能,并进行RT-qPCR验证。结果与空白组相比,糖尿病GK大鼠FBG显著升高(P<0.05),模型组大鼠肝脏可见肝细胞排列紊乱,边界模糊,出现弥漫空泡状改变,呈中度脂肪变性,伴见炎性浸润、弥漫水肿;与模型组相比,参芪复方组大鼠FBG在12周时明显降低(P<0.05),肝脏组织排列较整齐,脂质沉积、细胞水肿及炎细胞浸润明显减轻。全转录组结果显示,与空白组相比,模型组大鼠mRNA、lncRNA表达谱存在显著差异,决定了糖尿病大鼠生理病理上的差异表现。参芪复方可广泛调控mRNA、lncRNA的差异表达,富集分析显示参芪复方通过调控多条内分泌系统及代谢过程相关信号通路改善糖尿病,包括胰岛素分泌、非酒精性脂肪性肝病、胆固醇代谢、鞘脂代谢、胰岛素抵抗等信号通路。RT-qPCR结果显示,基因Irf1、Trim30、Tmcc3、Insig1与转录组结果一致,转录组准确性较高。结论参芪复方发挥调节血糖及改善肝脏脂质沉积、水肿的作用,可能与广泛调控mRNA、lncRNA的差异表达有关。 展开更多

目的探讨宫颈脱落细胞mybl2 mRNA表达水平在绝经后细胞学检查为低级别鳞状上皮内病变(LSIL)或无明确诊断意义的不典型鳞状细胞(ASC-US)且合并高危型HPV(HR-HPV)感染患者中的诊断分流价值。方法收集宫颈细胞学筛查为LSIL或ASC-US且合并HR... 目的探讨宫颈脱落细胞mybl2 mRNA表达水平在绝经后细胞学检查为低级别鳞状上皮内病变(LSIL)或无明确诊断意义的不典型鳞状细胞(ASC-US)且合并高危型HPV(HR-HPV)感染患者中的诊断分流价值。方法收集宫颈细胞学筛查为LSIL或ASC-US且合并HR-HPV感染绝经后患者的宫颈脱落细胞共120例,用qPCR检测宫颈脱落细胞的mybl2 mRNA表达水平。所有患者均经阴道镜下宫颈活检术行病理检查,根据病理结果分为宫颈癌(SCC)组(n=4),高级别鳞状上皮内病变(HSIL)组(n=18),LSIL组(n=38),宫颈炎症(NC)组(n=60),比较组间的mybl2 mRNA表达水平;根据HPV感染类型分为16/18型感染组(A组,n=35),16/18型合并其他HR-HPV感染组(B组,n=38),其余HR-HPV感染组(C组,n=47),比较组间的mybl2 mRNA水平。结果SCC组、HSIL组、LSIL组mybl2 mRNA表达水平均明显高于NC组(P<0.05);SCC组、HSIL组mybl2 mRNA表达水平均明显高于LSIL组(P<0.05);SCC组mybl2 mRNA表达水平明显高于HSIL组(P<0.05)。不同HPV感染组间宫颈脱落细胞mybl2 mRNA表达水平比较,A组、B组mybl2 mRNA表达水平明显高于C组(P<0.05),而A组和B组mybl2 mRNA表达水平差异无统计学意义(P>0.05)。结论宫颈脱落细胞的mybl2 mRNA表达水平随宫颈病变程度的加重而升高,且16/18型HPV患者的mybl2 mRNA表达水平明显更高,提示mybl2 mRNA可能作为细胞学为LSIL或ASC-US且合并HR-HPV感染绝经后患者的诊断分流指标,以提高宫颈癌筛查准确率并减少医疗资源浪费。 展开更多

血清ST2、ST2mRNA水平与脑出血患者脑损伤严重程度及预后的关系

目的 探讨脑出血患者血清肿瘤发生抑制蛋白2(ST2)、ST2mRNA表达水平与脑损伤严重程度及预后的关系。方法 选取我院收治的106例脑出血患者为研究组,根据格拉斯哥昏迷(GCS)评分将其分为中轻度组、重度组;另选同期50例健康体检者作为健康... 目的 探讨脑出血患者血清肿瘤发生抑制蛋白2(ST2)、ST2mRNA表达水平与脑损伤严重程度及预后的关系。方法 选取我院收治的106例脑出血患者为研究组,根据格拉斯哥昏迷(GCS)评分将其分为中轻度组、重度组;另选同期50例健康体检者作为健康对照组。检测两组血清的ST2及ST2mRNA表达水平,对研究组出院后进行半年随访,根据格拉斯哥预后(GOS)评分分成预后不良组和预后良好组。分析不同脑损伤严重程度、预后患者的血清ST2及ST2mRNA水平差异,并评估其在脑损伤及预后方面的价值。结果 研究组血清ST2及ST2mRNA表达水平明显高于对照组,重度组高于中轻度组,预后不良组高于预后良好组(P<0.05)。ROC曲线分析结果显示,血清ST2及ST2mRNA评估脑损伤严重程度的AUC分别为0.826、0.776;血清ST2及ST2mRNA评估脑出血患者预后的AUC分别为0.724、0.664,证明其在脑损伤严重程度和预后评估方面的价值更高。结论 脑出血患者脑出血后血清ST2及ST2mRNA水平明显升高,且不同脑损伤和预后患者存在明显差异,其可较好评估脑损伤严重程度和预后。 展开更多

目的探究胰岛素样生长因子2 mRNA结合蛋白2(insulin like growth factor 2 mRNA binding protein 2,IGF2BP2)协助甲基转移酶样3(methytransferase like 3,METTL3)通过N6-甲基腺苷(N6-methyladenosine,m6A)修饰调控HBV复制的分子机制。... 目的探究胰岛素样生长因子2 mRNA结合蛋白2(insulin like growth factor 2 mRNA binding protein 2,IGF2BP2)协助甲基转移酶样3(methytransferase like 3,METTL3)通过N6-甲基腺苷(N6-methyladenosine,m6A)修饰调控HBV复制的分子机制。方法以HBV稳定复制细胞系HepG2.2.15及其亲本细胞HepG2为模型,斑点杂交分析m6A修饰水平,RT-qPCR和Western blot检测METTL3、IGF2BP2表达。生物信息学及免疫共沉淀分析METTL3与m6A阅读蛋白及其相互作用。将METTL3质粒、METTL3 siRNA和(或)IGF2BP2 siRNA转染至HepG2.2.15细胞,分别记为OE-METTL3、si-METTL3、si-IGF2BP2、OE-METTL3+si-IGF2BP2、si-METTL3+si-IGF2BP2、Control组;qPCR检测HBV DNA、HBV rcDNA、HBV cccDNA拷贝数,RT-qPCR或Western blot检测HBV pgRNA、METTL3、IGF2BP2表达。结果与HepG2细胞相比,在HepG2.2.15细胞中m6A修饰富集,METTL3、IGF2BP2表达水平升高(P<0.05)。与Control组相比,在OE-METTL3组中m6A修饰富集增强,IGF2BP2表达水平升高(P<0.05),HBV复制相关指标(HBV DNA、HBV rcDNA、HBV cccDNA、HBV pgRNA)升高(P<0.01);在si-METTL3组中则相反。生物信息学分析及免疫共沉淀显示METTL3与IGF2BP2为互作蛋白。与Control组相比,在si-IGF2BP2组中m6A修饰富集减弱,METTL3表达水平降低(P<0.01),HBV复制相关指标降低(P<0.01)。在OE-METTL3+si-IGF2BP2组中,HBV复制相关指标较OE-METTL3组降低(P<0.001)。在si-METTL3+si-IGF2BP2组中,HBV复制相关指标较si-METTL3组、si-IGF2BP2组降低(P<0.05)。结论METTL3依赖IGF2BP2富集m6A通过增强HBV rcDNA转化为cccDNA,进而增强pgRNA逆转录复制病毒。 展开更多

目的初步探讨肝衰竭患者外周血单个核细胞人瑞粒酶逆转录酶(hTERT)mRNA及血清血管生成素-2(Ang-2)和细胞间黏附分子-1(ICAM-1)水平变化的临床意义。方法2019年1月~2021年5月我院诊治的74例肝衰竭患者【亚急性肝衰竭(SALF)13例,慢加急性... 目的初步探讨肝衰竭患者外周血单个核细胞人瑞粒酶逆转录酶(hTERT)mRNA及血清血管生成素-2(Ang-2)和细胞间黏附分子-1(ICAM-1)水平变化的临床意义。方法2019年1月~2021年5月我院诊治的74例肝衰竭患者【亚急性肝衰竭(SALF)13例,慢加急性肝衰竭(ACLF)39例和慢性肝衰竭(CLF)22例】和同期健康体检者30例,分离并检测外周血单个核细胞(PBMC)hTERT mRNA水平,采用ELISA法检测血清Ang-2和ICAM-1水平。结果肝衰竭组PBMC hTERT mRNA、血清Ang-2和ICAM-1水平分别为(0.9±0.3)、(1344.6±55.3)ng/L和(540.2±22.4)ng/ml,显著高于健康人组【分别为(0.4±0.1)、(1062.5±36.2)ng/L和(167.3±15.6)ng/ml,P<0.05】;SALF患者PBMC hTERT mRNA、血清Ang-2和ICAM-1水平分别为(1.5±0.6)、(1507.8±38.2)ng/L和(647.3±26.2)ng/ml,显著高于ACLF患者【分别为(1.2±0.4)、(1398.6±35.8)ng/L和(582.7±24.1)ng/ml,P<0.05】或CLF患者【分别为(0.7±0.2)、(1280.5±46.3)ng/L和(468.9±20.3)ng/m,P<0.05】;经过1~3 m治疗,SALF组死亡3例,ACLF组死亡5例,CLF组死亡4例。结论肝衰竭患者PBMCs hTERT mRNA及血清Ang-2和ICAM-1水平显著升高,它们可能为判断肝损伤程度提供客观依据,值得进一步研究。 展开更多

BACKGROUND Inflammatory bowel diseases(IBD)are a worldwide health problem and mainly affect young people,consequently affecting the workforce.Available treatments are often associated with side effects,and new therapeutic options are needed.For centuries,plants have represented important substrates in the field of drug development.Lafoensia pacari(L.pacari)is a plant whose pharmaceutical potential has been described,and may have biological activity relevant to the treatment of IBD symptoms.AIM To investigate the activity of keto-alcoholic extracts of L.pacari with respect to ameliorating the inflammatory and nociceptive symptoms of acute experimental colitis in mice.METHODS Keto-alcoholic extracts of L.pacari leaves and bark were administered to male andfemale Swiss mice weighing 25 g to 30 g(n=8 male mice and n=8 female mice).The effect of these extracts was observed in an acetic acid-induced acute experimental model of colitis with regard to antinociception/analgesia and inflammatory tissue damage.Recorded macroscopic indices included the Wallace score and the colon weight obtained using a precision scale.Mechanical hyperalgesia was determined using an electronic analgesimeter.Behavior related to overt pain was determined by quantifying the number of writhing instances within 20 min of administration of acetic acid.Molecular docking was performed using human and murine cyclooxygenase-2(COX-2)with 3 flavonoids(ellagic acid,kaempferol,and quercetin)on the AutoDock Vina software.Analysis of variance followed by Tukey’s posttest was used with P<0.05 indicating significance.RESULTS In this murine model of colitis,administration of extracts from L.pacari ameliorated acetic acidinduced writhing and colitis-associated inflammatory pain.These improvements may be attributable to the reduction in edema,inflammation(e.g.,ulcers,hyperemia,and bowel wall damage),and the intensity of abdominal hyperalgesia.The keto-alcoholic extracts of L.pacari leaves and bark administered at a dose of either 100 mg/kg or 300 mg/kg significantly reduced the number of writhing events when compared to the negative control(P<0.05).Additionally,extracts of L.pacari bark also performed better than Dipyrone.Leaf extracts administered at 10 mg/kg,30 mg/kg,and 100 mg/kg and bark extracts administered at 30 mg/kg significantly reduced or prevented the development of edema in the colon of treated mice,while mesalazine did not.Moreover,using molecular docking,we observed that the flavonoids present in L.pacari extracts bind to COX-2,an event not unique to ellagic acid.CONCLUSION The results of this study demonstrate a potential novel application of L.pacari extracts for the reduction of inflammation and promotion of antinociception/analgesia as demonstrated by our findings in a murine model of colitis.These findings were also corroborated by in silico analyses,and suggest that L.pacari extracts may be a promising therapeutic agent in the treatment of IBD. 展开更多

Clinical implications of forkhead box M1, cyclooxygenase-2, and glucose-regulated protein 78 in breast invasive ductal carcinoma

BACKGROUND Breast infiltrating ductal carcinoma(BIDC)represents the largest heterotypic tumor group,and an in-depth understanding of the pathogenesis of BIDC is key to improving its prognosis.AIM To analyze the expression profiles and clinical implications of forkhead box M1(FOXM1),cyclooxygenase-2(COX-2),and glucose-regulated protein 78(GRP78)in BIDC.METHODS A total of 65 BIDC patients and 70 healthy controls who presented to our hospital between August 2019 and May 2021 were selected for analysis.The peripheral blood FOXM1,COX-2,and GRP78 levels in both groups were measured and the association between their expression profiles in BIDC was examined.Additionally,we investigated the diagnostic value of FOXM1,COX-2,and GRP78 in patients with BIDC and their correlations with clinicopathological features.Furthermore,BIDC patients were followed for 1 year to identify factors influencing patient prognosis.RESULTS The levels of FOXM1,COX-2,and GRP78 were significantly higher in BIDC patients compared to healthy controls(P<0.05),and a positive correlation was observed among them(P<0.05).Receiver operating characteristic analysis demonstrated that FOXM1,COX-2,and GRP78 had excellent diagnostic value in predicting the occurrence of BIDC(P<0.05).Subsequently,we found significant differences in FOXM1,COX-2,and GRP78 levels among patients with different histological grades and metastasis statuses(with vs without)(P<0.05).Cox analysis revealed that FOXM1,COX-2,GRP78,increased histological grade,and the presence of tumor metastasis were independent risk factors for prognostic death in BIDC(P<0.001).CONCLUSION FOXM1,COX-2,and GRP78 exhibit abnormally high expression in BIDC,promoting malignant tumor development and closely correlating with prognosis.These findings hold significant research implications for the future diagnosis and treatment of BIDC. 展开更多

AGAP2-AS1 affects TNM staging and prognosis of lung cancer patients by acting on SLC7A11 mRNA stability and ferroptosis

Objective The initiation and progression of lung carcinomas are critically regulated by long non-coding RNAs(lncRNAs).However,the role of lncRNAs in the pathways causing lung cancer remains unknown.Methods Cell morphology was regularly observed using an inverted phase-contrast microscope.Cell viability was assessed using CCK-8 according to the manufacturer’s instructions.Total RNA was retrotranscribed from each specimen using the RNAiso Plus Kit.The RT-PCR data were calculated using the Ct approach for comparison.Flow cytometric analyses were prepared by Click-iT™Plus TUNEL Assay for In Situ apoptosis detection,with Alexa Fluor^(TM)594 dye,as instructed.RNA immunoprecipitation assays were used to determine RNA concentration.Results Activated natural killer cells repeat and PH domain-containing protein 2 antisense RNA 1(AGAP2-AS1)levels in cancerous tissues were significantly correlated with cancerous tumor node metastasis(TNM)stage,with cancerous AGAP2-AS1 levels being higher in cancerous tissues than healthy tissues.Patients withelevated AGAP2-AS1 levels had considerably worse outcomes than those with reduced AGAP2-AS1 levels,regardless of the progression-free or overall survival.Functionally,AGAP2-AS1 downregulation represseslung cancer cell growth.AGAP2-AS1 elimination induces erastin-mediated ferroptosis in lung cancer cells.However,the ferritin inhibitor FERSINT-1 negated this result,whereas ERASTIN induced lung cancer cellmortality.After AGAP2-AS1 silencing,erastin-treated lung cancer cells showed a remarkable decrease inGSH levels.These results indicated that AGAP2-AS1 enhanced the stabilization of SLC7A11 mRNA via Recombinant Insulin Like Growth Factor Binding Protein 2(IGF BP2).Patients with elevated AGAP2-AS1 had considerably worse outcomes.Down-regulating AGAP2-AS1 was able to repress lung cancer cell growth and induce greater Erastin-mediated ferroptosis.Lungcancer cells treated with Erastin exhibited a remarkable decrease inglutathione(GSH)levels.The mechanical findingsindicated that AGAP2-AS1 enhanced the stabilization of SLC7A11 mRNA via the IGF2BP2.Conclusion We identified a novel effect of AGAP2-AS1 on TNM staging and the prognosis of patientswith lungcancer by modulating SLC7A11 mRNA stability and ferroptosis. 展开更多

BACKGROUND A 70-year-old man with hepatitis C virus-related recurrent hepatocellular carcinoma was admitted for further diagnosis of a 1 cm iso-hyperechoic nodule in segment(S)5.CASE SUMMARY Gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid-enhanced magnetic resonance imaging(EOB-MRI)revealed the nodule in S5 with a defect at the hepatobiliary phase,hyperintensity on diffusion weighted imaging(DWI)and hypointensity on apparent diffusion coefficient(ADC)map.Contrast-enhanced computed tomography revealed hypervascularity at the early phase,and delayed contrast-enhancement was observed at the late phase.Contrast-enhanced ultrasound(US)revealed incomplete defect at the late vascular phase.Inflammatory liver tumor,lymphoproliferative disease,intrahepatic cholangiocarcinoma(small duct type)and bile duct adenoma were suspected through the imaging studies.US guided biopsy,however,showed a noncaseating hepatic sarcoid-like epithelioid granuloma(HSEG),and histopathological analysis disclosed spindle shaped epithelioid cells harboring Langhans-type multinucleated giant cells.One month after admission,EOB-MRI signaled the disappearance of the defect at the hepatobiliary phase,of hyperintensity on DWI,of hypointensity on ADC map,and no stain at the early phase.CONCLUSION That the patient had received BNT162b2 messenger RNA(mRNA)coronavirus disease 2019 vaccination 3 mo before the occurrence of HSEG,and that its disappearance was confirmed 4 mo after mRNA vaccination suggested that the drug-induced sarcoidosis-like reaction(DISR)might be induced by the mRNA vaccination.Fortunately,rechallenge of drug-induced DISR with the third mRNA vaccination was not confirmed. 展开更多
