先天性巨结肠患儿术后肠道组织中CAD、SOX2的表达水平及其临床意义

目的探讨先天性巨结肠(HD)患儿术后肠道组织蛋白酶D(CAD)、性别决定区Y框蛋白2(SOX2)的表达水平及其临床意义。方法选取2015年5月至2019年3月郑州大学第一附属医院收治的56例HD患儿结肠组织(HD组)和23例因肠道梗阻或穿孔实施造瘘手术获... 目的探讨先天性巨结肠(HD)患儿术后肠道组织蛋白酶D(CAD)、性别决定区Y框蛋白2(SOX2)的表达水平及其临床意义。方法选取2015年5月至2019年3月郑州大学第一附属医院收治的56例HD患儿结肠组织(HD组)和23例因肠道梗阻或穿孔实施造瘘手术获取的结肠组织标本(对照组)。采用免疫组化、Western-blot技术检测结肠组织中CAD、SOX2的表达水平,并采用Pearson线性相关分析法分析CAD、SOX2表达与病变肠段肌间神经丛直径、神经节细胞数的关系。结果HD患儿的狭窄段组织中CAD蛋白、SOX2蛋白阳性表达率分别为7.14%、12.50%,移行段肠组织中CAD蛋白、SOX2蛋白阳性表达率分别为32.14%、35.71%,明显低于对照组的78.26%、82.61%,而狭窄段组织中CAD蛋白、SOX2蛋白阳性表达率明显低于移行段,差异均有统计学意义(P<0.05);HD患儿的狭窄段肠组织中肌间神经丛直径、神经节细胞数目分别为(30.66±5.14)μm、(0.83±0.24)个/视野,移行段肠组织中肌间神经丛直径、神经节细胞数目分别为(31.20±5.83)μm、(1.94±0.56)个/视野,明显短(少)于对照组的(54.29±8.50)μm、(5.40±1.84)个/视野,狭窄段肠组织中肌间神经丛直径、神经节细胞数目明显短(少)于移行段,差异均有统计学意义(P<0.05);Pearson相关分析结果显示,HD患儿狭窄段、移行段肠组织中肌间神经丛直径、神经节细胞数目与CAD蛋白、SOX2蛋白表达强度均呈显著正相关(P<0.05)。结论HD患儿病变结肠组织中CAD蛋白、SOX2蛋白的表达强度下调,可能与肌间神经丛直径、神经节细胞数目的减少有关。 展开更多

[目的]研究庆阳驴养殖群体的遗传多样性与母系起源,了解其遗传信息,为保护庆阳驴种质资源、选育和遗传改良工作提供理论依据。[方法]随机选取133头庆阳驴,对其线粒体DNA(mitochondrial DNA,mtDNA)D-loop区序列进行PCR扩增、测序及比对,并探讨庆阳驴的遗传多样性与母系起源。[结果]在获得的520 bp D-loop碱基序列中,AT含量(57.3%)高于GC含量(42.8%),表现出碱基的偏倚性;检测到38个变异位点,包含8个碱基对的转换;其核苷酸多样性(Pi)、单倍型多样性(Hd)、平均核苷酸差异(K)分别为0.01591、0.895和8.274,与欧洲家驴和中国家驴研究的平均值相比较低,说明该驴品种核苷酸变异较为贫乏。庆阳驴mtDNA D-loop区存在35个单倍型,单倍型之间的遗传距离为0.002~0.042。系统进化结果显示,庆阳驴存在2个线粒体支系,表明其具有2个母系起源,且遗传距离表明,庆阳驴与克罗地亚家驴之间的遗传距离较近。[结论]本研究从分子水平初步揭示庆阳驴核苷酸变异比较贫乏,杂交程度高,mtDNA遗传多态性正逐步丧失,应加强庆阳驴品种的遗传资源保护工作。 展开更多

目的:探讨精子线粒体DNAD-Loop区基因突变和弱精症的相关性。方法:收集2019年7月至2021年3月温州市中医院诊治的134例弱精症患者(弱精症组)和129例同期健康男性(对照组)的精液,提取精子细胞DNA,采用聚合酶链反应(PCR)对线粒体DNA D-Loo... 目的:探讨精子线粒体DNAD-Loop区基因突变和弱精症的相关性。方法:收集2019年7月至2021年3月温州市中医院诊治的134例弱精症患者(弱精症组)和129例同期健康男性(对照组)的精液,提取精子细胞DNA,采用聚合酶链反应(PCR)对线粒体DNA D-Loop区进行扩增并测序,测序结果与剑桥标准序列(r CRS)进行比对,分析弱精症患者精子细胞线粒体DNA D-Loop区的基因突变情况。结果:两组中发生率>5%的突变有62种,大部分位于HV-1区和HV-2区(87.78%),突变类型以替换突变为主(87.03%)。分析显示,有11种突变在弱精症组和对照组之间的差异有统计学意义(P<0.05),这些突变可能增加或降低弱精症风险。结论:精子线粒体DNA D-Loop区具有较高突变率,可能在弱精症发生发展中发挥重要作用。 展开更多

基于线粒体Cytb基因和D-loop区的鸭绿江流域细鳞鱼群体遗传多样性

旨在研究鸭绿江流域(丹东段)细鳞鱼野生群体与养殖群体的遗传多样性,并探究其种质资源情况。基于细鳞鱼线粒体DNA Cytb基因和D-loop区序列,对宽甸(KD)和丹东江域(DD)2个野生细鳞鱼群体和1个养殖群体的遗传结构与遗传多样性等进行比较研... 旨在研究鸭绿江流域(丹东段)细鳞鱼野生群体与养殖群体的遗传多样性,并探究其种质资源情况。基于细鳞鱼线粒体DNA Cytb基因和D-loop区序列,对宽甸(KD)和丹东江域(DD)2个野生细鳞鱼群体和1个养殖群体的遗传结构与遗传多样性等进行比较研究。结果表明:Cytb基因1141 bp,A+T含量53.55%,G+C含量46.45%,D-loop区序列988 bp,A+T含量62.75%,G+C含量37.25%;基于Cytb和D-loop序列在90个样本中分别检测到42和56个多态性位点,定义了21和26种单倍型,其中KD、DD和YZ群体Cytb基因的单倍型数分别为14、10和7,KD、DD和YZ群体D-loop区单倍型数分别为16、15和10;遗传多样性分析Cytb基因表现出高单倍型多样性(0.986)和低核苷酸多样性(0.00466),而D-loop区表现出高单倍型多样性(0.981)和高水平的核苷酸多样性(0.01982),基于Cytb基因和D-loop区序列KD和DD群体分别具有4个和2个相同单倍型,说明2个细鳞鱼野生群体间的分化程度较小,而养殖群体与野生群体间遗传分化为中度;分子方差分析(AMOVA)发现群体内部的遗传变异(Cytb:73.38%;D-loop:85.91%)占比明显高于群体间的遗传变异(Cytb:26.62%;D-loop:14.09%),表明鸭绿江流域(丹东段)细鳞鱼群体遗传变异主要来源于群体内,不同地理群体间的遗传变异不大;群体单倍型系统发育树表明,KD、DD和YZ群体间互有交叉,均未表现出明显的地理聚集,3个群体间尚未出现明显的地理谱系结构,结果与遗传多样性研究一致。Tajima’s D和Fu and Li’s中性检验显著偏离中性,推测细鳞鱼KD和DD群体可能经历过群体扩张事件,随后处于相对平衡稳定状态。 展开更多

皖南牛mtDNA D-loop区遗传多样性和系统进化分析

采用DNA提取及PCR扩增技术,对30头皖南母牛的mtDNA D-loop区的全序列进行扩增与分析。利用Genbank数据库,下载国内外部分黄牛品种mtDNA D-loop区的全序列,再使用Clustalx、Dnasp5.0等生物软件,对其全序列进行比对和对单倍型多样度、核... 采用DNA提取及PCR扩增技术,对30头皖南母牛的mtDNA D-loop区的全序列进行扩增与分析。利用Genbank数据库,下载国内外部分黄牛品种mtDNA D-loop区的全序列,再使用Clustalx、Dnasp5.0等生物软件,对其全序列进行比对和对单倍型多样度、核苷酸多样度等展开分析。皖南牛mtDNA D-loop区序列长度均在962~1052 bp的区间内,共发现68个变异位点,其中单一信息位点共有18个,简约信息位点共有50个。构成13种单倍型,其单倍型多样性(Hd)的均值为0.6483,核苷酸多样性(Pi)的均值为0.0131,其序列之间核苷酸的差异平均数(k)为11.589,A、C、G、T碱基含量平均占比分别为33.26%,25.23%,13.39%,28.12%。通过遗传距离和N-J系统进化树得知,皖南牛和威宁黄牛、湘西牛、大别山牛和吉安黄牛亲缘关系较近。皖南牛的遗传多样性和群体变异程度较低。研究结果可为皖南牛遗传资源的保护开发提供科学依据和理论参考。 展开更多

Around 18 million people are estimated to be infected with HDV. An important trend in HDV infection is global decline. HDV prevalence has decreased significantly in Europe since the 1970s and 1980s when it was f irst reported. The Asia-Pacif ic region now seems to be where HDV is a major health concern. There is a lack of available data from most of the countries from this region; hence, the true status of HDV cannot be determined. In South Asia, most of the countries have conditions that are favorable for the spread of hepatitis B and other related infections. Countries like Pakistan and Iran have shown an increase in HDV prevalence over a period of time. Other countries and region like China, Turkey, Australia, Japan, India and Taiwan, some of which had very high HDV prevalence in the past, have shown a decline in the incidence, but high prevalence persists in some. Intravenous drug abusers, homosexual men and women, prostitutes, and people on hemodialysis are the groups with very high HDV prevalence. 展开更多

Two species Lefua echigonia and Lefua sp. 2 of the eight-barbel loach inhabit the Tokai region of Honshu, Japan. We determined sequences of the mitochondrial D-loop region to elucidate intraspecific phylogenetic relationships and variation in these two species. Lefua sp. 2 represented high intraspecific genetic similarity and complicated haplotype network, but three assemblages were recognized, including specimens mainly from Yahagi, Toyo, and Tenryu River systems, respectively, and named Groups 1 to 3. Divergence of Group 1 from the others was marginally supported, but Group 2 was paraphyletic to Group 3, suggesting the existence of two populations, i.e. Yahagi River population and Toyo-Tenryu River population. Lefua echigonia also represented high intraspecific genetic similarity, and two assemblages with slight genetic differentiation were discernible, including specimens from Shizuoka and southeastern Aichi prefectures and those from northwestern Aichi, Gifu, and Mie prefectures, respectively, and named Groups A and B. Star-like relationships of haplotypes suggested the dispersal origin located in eastern Aichi prefecture. The two species are threatened to extinction and thus we proposed evolutionary significant units for conservation. 展开更多

The magma system of Changbaishan-Tianchi Volcanic region is studied with three-dimensional deep seismic sounding (DSS) technique. The results show that the magma system of Changbaishan-Tianchi volcanic region, mainly characterized by low velocity of P wave, can be divided into three parts in terms of depth. At the depth range of 9-15 km, the distribution of the magma system is characterized by extensiveness, large scale and near-SN orientation. This layer is the major place for magma storage. From the depth of 15 km down to the lower crust, it is characterized by small lateral scale, which indicates the "trace" of magma intrusion from the upper mantle into the crust and also implies that the magma system most probably extends to the upper mantle, or even deeper.(less than 8-9 km deep), the range of magma distribution is even smaller, centering on an SN-oriented area just north of the Tianchi crater. If low velocity of P wave is related to the magma system, it then reflects that the magma here is still in a state of relatively high temperature. In this sense, the magma system of Changbaishan-Tianchi volcanic region is at least not "remains", in other words, it is in an "active" state. 展开更多

物体区域信息引导下的RGB-D场景3D目标检测

针对室内场景RGB-D数据的3D目标检测是图形学与三维视觉中的重要问题。针对RGB-D场景中3D目标检测对复杂背景的适应性较差、目标检测中难以有效利用物体区域信息及场景点云特征信息等缺陷,基于物体区域信息引导,提出一种融合全局和局部... 针对室内场景RGB-D数据的3D目标检测是图形学与三维视觉中的重要问题。针对RGB-D场景中3D目标检测对复杂背景的适应性较差、目标检测中难以有效利用物体区域信息及场景点云特征信息等缺陷,基于物体区域信息引导,提出一种融合全局和局部点云特征并排除背景干扰的3D目标检测框架。该框架以场景RGB-D数据作为输入,首先提取彩色图像中待检测目标对象2D区域并为对象进行粗分类,再将对象区域二维边界框提升到三维斜锥体区域并转化形成点云数据;然后在斜锥体点云上利用物体区域分类信息进行特征提取,并利用特征变换与最大池聚合操作将点云全局特征和局部特征有效融合;接着利用融合特征以预测各采样点与前景背景相关程度的概率分数,依据此概率分数分割场景前景点与背景点,并通过场景背景点剔除以形成屏蔽性点云;最终在屏蔽性点云中投票产生物体中心点并借助物体区域信息提出建议及3D目标预测,此外,还加入了一个角点损失,对边界框精度进行优化。针对SUN RGB-D数据集进行网络训练,实验结果表明,与传统方法相比,所提框架的目标检测结果准确率得到有效提升,同一评估指标下的点云目标检测准确率达到59.1%,并且在强遮挡或稀疏采样点区域下亦能够精确估计三维物体的边界框。 展开更多

背景 帕金森病(Parkinson’s disease,PD)是一种多发于中老年的神经退行性疾病,线粒体DNA(mtDNA)甲基化可能参与PD的发生和发展,目前相关研究极少。目的 观察PD患者外周血mtDNA D环区的甲基化水平及其拷贝数的变化,分析D环区甲基化与mt... 背景 帕金森病(Parkinson’s disease,PD)是一种多发于中老年的神经退行性疾病,线粒体DNA(mtDNA)甲基化可能参与PD的发生和发展,目前相关研究极少。目的 观察PD患者外周血mtDNA D环区的甲基化水平及其拷贝数的变化,分析D环区甲基化与mtDNA拷贝数的相关性,探讨PD的发病机制。方法 选取2020-2022年内蒙古科技大学包头医学院第一附属医院PD患者(PD组)和健康体检者(对照组)各30例[(66.63±7.95)岁vs (63.50±4.96)岁],每组男性、女性各15例,采用质谱法(MassARRAY)检测外周血mtDNA D环区的甲基化水平,采用实时定量PCR法(qRT-PCR)检测mt DNA拷贝数。结果 PD组与对照组的外周血mtDNA中D环区总体甲基化率及各CpG位点甲基化率差异无统计学意义(P>0.05),早期PD患者与中晚期PD患者的外周血mtDNA中D环区总体甲基化率及各CpG位点甲基化率差异无统计学意义(P>0.05)。PD患者外周血mtDNA拷贝数低于对照组(P=0.02),中晚期PD患者外周血mtDNA拷贝数低于早期PD患者,差异有统计学意义(P=0.012)。mtDNA D环区第1、15个CpG位点(即人mtDNA序列第112、413位的核苷酸)甲基化率与mtDNA拷贝数呈负相关(r=-0.257,P=0.047;r=-0.517,P<0.001)。Logistic回归分析显示,mtDNA拷贝数越高,PD发生概率越低(OR=0.119,P=0.023)。结论 PD患者外周血mtDNA D环区甲基化水平未发生显著改变,但其外周血mt DNA拷贝数降低,且随着PD病程的进展呈动态变化,mtDNA拷贝数有望成为预测PD发生及病程进展的指标。D环区的甲基化可能会负向调控mtDNA的复制与转录。 展开更多

Shear wave anisotropy in D” region beneath the western Pacific

Using seismic shear phases from 47 Tonga-Fiji and its adjacent region events recorded by the CENC and IRIS, and from 26 northeast Asia and north Pacific events recorded by IRIS, we studied the shear wave anisotropy in D" region beneath the western Pacific utilizing the ScS-S differential travel time method and obtained the splitting time values between the radial and transverse components of each ScS wave corresponding to each core-mantle boundary (CMB) reflection point. We found that most shear waves involved horizontally polarized shear wave components traveling faster than vertically polarized shear wave components through the D" region. The splitting time values of ScS wave range from ?0.91 s to 3.21 s with an average value of 1.1 s. The strength of anisotropy varies from ?0.45% to 1.56% with an average value of 0.52%. The observations and analyses show that in the D" region beneath the western Pacific the lateral flow is expected to be dominant and the vertical transverse isotropy may be the main anisotropic structure. This structure feature may be explained by the shape preferred orientation of the CMB chemical reaction products or partial melt and the lattice preferred orientation of the lower mantle materials caused by the lateral flow at lowermost mantle. 展开更多

The Alpine region of Uvira District in South-Kivu Province faces low agricultural productivity challenges largely due to human population pressure on the natural resource base. During the dry season, conflicts between crop farmers and livestock breeders are common. This region is located 2500 - 3020 m a.s.l. and is almost inaccessible due to poor road infrastructure. To understand the interactions between natural resources (forests, pastures, soils) and sedentary agriculture, and to propose solutions for sustainable natural resource management, Participatory Rural Appraisal (PRA) sessions covering three farmer group interviews were conducted in July 2010 in Marungu, Kitembe and Kahololo location. The livelihoods of the Banyamulenge have been transformed from pastoralism to a sedentary system with the introduction of crop cultivation after 1980. Maize, bush beans and Irish potatoes are cultivated with limited success because of late crop maturity under the prevailing climatic conditions. Livestock, mainly cattle, sheep and goats is managed under extensive system, relying on natural pasture far from the human settlements. Livestock plays an important role to define people’s wealth and is mainly managed by men. There is a real need of forages during the dry season;however, cultivation of forages is unknown. On the other hand, pastures appear to be degenerating due to overgrazing. Access by farmers to efficient and effective agricultural extension services is fundamental in order to promote locally adapted and profitable crop and livestock production while at the same time, sustaining the productive resource base and security. 展开更多

This study analyzed the mitochondrial DNA D-loop hypervariable region 601 bp sequence in 12 Eutamias sibiricus from Heilongjiang area.The result showed that the average contents of A,T,G and C were 33.2%,30.5%,11.8%and 24.5%respectively,the A+T content(63.7%)was obviously higher than the G+C content(36.3%).Thirty-six mutation(approximately 6.0%)sites were found and 9 haplotypes were defined.The mutations types,including transition,transversion and deletion were all found in the detected mtDNA D-loop regions,most of which was transition.The average nucleotide mutational ratio was 1.22%.The nucleotide mutation sites affected the restriction site appearance or disappearance of the restriction site.The research on mtDNA D-loop is focused on the domestic animals and there is no report on Eutamias sibiricus.This study analyzed the mitochondrial DNA D-loop hypervariable in Eutamias sibiricus so as to provide some useful informations for related research in the future. 展开更多

Based on the first arrival P and S data of 4 625 regional earthquakes recorded at 174 stations dispersed in the Yunnan and Sichuan Provinces, the 3-D velocity structure of crust and upper mantle in the region is determined, incorporating with previous deep geophysical data. In the upper crust, a positive anomaly velocity zone exists in the Sichuan basin, whereas a negative anomaly velocity zone exists in the western Sichuan plateau. The boundary between the positive and negative anomaly zones is the Longmenshan fault zone. The images of lower crust and upper mantle in the Longmenshan fault, Xianshuihe fault, Honghe fault and others show the characteristic of tectonic boundary, indicating that the faults likely penetrate the Moho discontinuity. The negative velocity anomalies at the depth of 50 km in the Tengchong volcanic area and the Panxi tectonic zone appear to be associated with the temperature and composition variations in the upper mantle. The overall features of the crustal and the upper mantle structures in the SichuanYunnan region are the lower average velocity in both crust and uppermost mantle, the large crustal thickness variations, and the existence of high conductivity layer in the crust or/and upper mantle, and higher geothermal value. All these features are closely related to the collision between the India and the Asia plates. The crustal velocity in the SichuanYunnan rhombic block generally shows normal value or positive anomaly, while the negative anomaly exists in the area along the large strike-slip faults as the block boundary. It is conducive to the crustal block side-pressing out along the faults. In the major seismic zones, the seismicity is relative to the negative anomaly velocity. Most strong earthquakes occurred in the upper-mid crust with positive anomaly or normal velocity, where the negative anomaly zone generally exists below. 展开更多

POM97, an oceanic model, has been used for the first time to the numerical study on the tidal waves of the as Regions around Taiwan. In this paper, we have got the result that the semidiurnal tidal waves of these area mainly are the co-operating tides which come from the south of 23’N of the western Pacific. Those semidiurnal tidal waves affecting the Taiwan Straits come respectively from the south and the north entrance of the channel, and the north tidal wave is stronger than the south one. The strongest tidal field is the area from the Meizhou Bay to the Xinhua Bay along the coast of Fujian Province, where the biggest amplitude of the M2 partial tide can reach 240 cm. The strongest tidal cur- rent fields lie in the Penghu watercourse, where the maximum velocity of the M2 partial tide can arrive at 196 m/s. In the horizontal structure of the tidal currets, we have found that there is a stream dot in the north of the channel, besides, there still exist four new ones. As for the vertical structure, it mainly is biassed to the right at the surface, and to the left near the bottom layer. 展开更多
