The objective of this work is to present an appropriate set of RAPD (random amplified polymorphic DNA) markers using single and multiplex PCR analysis suitable for the characterization of the endemic Cuban species Car...The objective of this work is to present an appropriate set of RAPD (random amplified polymorphic DNA) markers using single and multiplex PCR analysis suitable for the characterization of the endemic Cuban species Carica cubensis and the establishment of genetic relationships with the cultivated species Carica papaya. RAPD markers presented a high level of polymorphism. In addition, the incorporation of more than one RAPD primer in the PCR analysis increased the number of obtained bands and the polymorphism of these bands. A total of 73 RAPD bands were detected (45 of them polymorphic) with the nine RAPD markers assayed using single and multiplex PCR analysis. Results demonstrated a reduced genetic variability within the tested Carica cubensis accessions. The observed clustering in this species could be better explained according to geographic proximity and can indicate the similar precedence of the isolated studied populations. C. cubensis seem to be subspecies of C. papaya adapted to the environmental conditions of the mountains of Cuba or a endemic species close to C. papaya. The implications of these results in the creation of effective germplasm core collection in Carica species have been also discussed.展开更多

In 1988, this disease caused a loss of seasonal production amounted 70% on wheat variety Mexipak in Syria, and recurrent infection in 2010, caused by a virulent race called Yr27, caused a considerable loss in the production of bread wheat cultivars (Cham 8, Cham 6 particularly) amounted 90%. Recently, 15 races of yellow rust had been addressed in Syria for seasons 2010-2014; 159E256, 166E254, 166E256, 255 E112, 0 E0, 64 E 6, 230 El50, 0 E 18, 198 El30, 166 El50, 102 El60, 128 E0, 126 El50, 214E150, and 6E16. The race 6E16 was the most frequent during the two seasons, while the race 255El12 was the most virulent, followed by the race 230E222 and the race 0E0 was the weakest one. This study revealed the presence of fourteen newly observed races in Syria. Molecular Variance Analysis of Molecular Variance (AMOVA) of 55 yellow rust Puccinia striiformis f.sp tritici isolates examined by Amplify Fragment Length Polymorphism (AFLP) revealed high genetic variation within population, and the dimensional scale analysis (MSD) and tree diagram showed that the Syrian yellow rust isolates were clustered in three groups: the first group contained isolates derived from durum wheat, the second one contained bread wheat isolates, but the third was made of isolates derived from both durum and bread wheat species.展开更多

文摘耐旱苔藓植物常常单个个体矮小、生物量低,如何从细小的单个个体中有效提取总DNA是进一步开展居群遗传多样性研究的关键。本研究以古尔班通古特沙漠广泛分布的刺叶墙藓(Tortula desertorum)为对象,使用快速提取法、2×CTAB法及DNeasy plant mini kit试剂盒提取法等3种方法对刺叶墙藓单个个体的总DNA进行提取。结果表明,2×CTAB法提取的DNA纯度高,凝胶电泳显示无明显降解现象,适宜作为PCR扩增的模板。利用所提取的单个个体DNA为模板,建立了优化的RAPDI、SSR反应体系。
