目的运用网络药理学方法对达原饮进行分析,探索达原饮治疗发热的相关靶点与通路,明确其作用机制与潜在治疗疾病,为后续研究提供指导。方法通过中药系统药理学数据库与分析平台(TCMSP)检索达原饮各组成药物的主要化学成分及作用靶点,参...目的运用网络药理学方法对达原饮进行分析,探索达原饮治疗发热的相关靶点与通路,明确其作用机制与潜在治疗疾病,为后续研究提供指导。方法通过中药系统药理学数据库与分析平台(TCMSP)检索达原饮各组成药物的主要化学成分及作用靶点,参考类药五原则并结合口服生物利用度对成分靶点数据进行筛选,通过String数据库检索筛选后的靶点,得到达原饮作用靶点基因,在CTD数据库中检索发热(fever)相关作用靶点基因,对二者进行比较分析,取重复值,得到达原饮治疗发热相关靶点;在String数据库中检索达原饮治疗发热相关靶点,得到达原饮治疗发热相关通路;利用Cytoscape3.7.0软件对通路进行模块化分析,选择分值最高者,在WEB-based GEne SeT AnaLysis Toolkit中进行可视化分析。结果得到达原饮治疗发热相关靶点432个,相关通路75条,其中特异性疾病相关通路21条,非特异性疾病相关通路54条。结论达原饮治疗发热的作用机制涉及以代谢为中心的多种途径,可能通过多靶点复杂通路的共同干预起效。本研究就达原饮对于癌性发热的治疗补充了数据支持,并推测达原饮治疗神经退行性疾病具有可能性。展开更多
