Domestication of Marama Bean in Arid Namibia: Challenges and Opportunities in a Climate Changing Agroecology

Tylosema esculentum (Burch.) A. Schreib. (Marama bean), referred to as marama in sections of this article, is an obligate outcrossing native plant with a yield potential of 2 ton/hectare which grows naturally in the deep sandy soils of the Kalahari Desert. It has adapted to the low precipitation levels in that agro-ecosystem. Marama serves as a staple food for the San and Bantus in that area. In Namibia, in the past you could find wild stands of marama in the Khomas region, Omaheke region, and the Otjozondjupa region without must struggle. It is renowned for its brown seeds, which are rich in high-quality oils and proteins. The tuberous root contains a significant amount of starch. The objective of domesticating orphaned marama is to provide farmers in this climate change-prone region with a viable alternative for food and nutrition security. This program, initiated in 2008 with an open-minded mindset, required swift implementation using harsh and occasionally unconventional methods. To introduce indigenous tools for resource-poor farmers, the domestication program prioritized the utilization of farmer-participatory methodologies. It was crucial to integrate old and new approaches to ensure learning from past and present experiences, leading to innovative solutions. There is little research and development of native crops in Africa because most of the currently cultivates crops were brought for use from abroad. Only a few numbers of indeginous African crops can be named. The arid Kalahari region, susceptible to climate change, necessitates the revival of indigenous crops like marama, which are resilient and well-adapted to the region’s conditions and have thrived for centuries. In many discussions regarding the health and nutrition of Africa, the recommendation to consume traditional foods to avoid exposure to modern foods, which may not be genetically compatible, is frequently emphasized. Regardless of their validity, these opinions must be acknowledged, and steps need to be taken to ensure a positive legacy for future generations. However, this chapter will address the limitations and challenges that exist in this regard. This article will summarize the progress made in the domestication program of the marama bean in Namibia thus far. Furthermore, this article will highlight the challenges that have been faced during the domestication journey for marama bean and other orphaned crops. The domestication program commenced with a broad germplasm collection, characterization, and preselection for breeding. Crop selection in this program was influenced by climate change-related concerns of shorter and uncertain rain seasons, and recurrent droughts. Selection included but was not limited to identifying marama genotypes with superior characteristics, early germination and many seeds per pod were among some of the identified and selected characteristics. The Namibia University of Science and Technology (NUST) has compiled a list of potential marama bean varieties and is currently testing marama seeds in anticipation of their introduction as a new crop alternative with good adaptation to the effects of climate change, since conventional crops like maize underperform due to persistent droughts. Marama bean, if properly developed, holds significant potential to address issues of hunger and malnutrition in arid regions of Southern Africa and other similar territories. The findings presented here are the result of ongoing field research and experiments conducted at multiple sites using superior marama bean varieties. 展开更多

Soybean(Glycine max L.)is a protein and oil crop grown worldwide.Its fitness may be reduced by deleterious mutations,whose identification and purging is desirable for crop breeding.In the published whole-genome re-sequenced data of 2214 soybean accessions,including 221 wild soybean,1132 landrace cultivars and 861 improved soybean lines,we identified 115,275 deleterious single-nucleotide polymorphisms(SNPs).Numbers of deleterious alleles increased from wild soybeans to landraces and decreased from landraces to modern improved lines.Genes in selective-sweep regions showed fewer deleterious mutations than the remaining genes.Deleterious mutations explained 4.3%-48%more phenotypic variation than randomly selected SNPs for resistance to soybean cyst nematode race 2(SCN2),soybean cyst nematode race 3(SCN3)and soybean mosaic virus race 3(SMV3).These findings illustrate how mutation load has shifted during soybean domestication,expansion and improvement and provide candidate sites for breeding out deleterious mutations in soybean by genome editing and/or conventional breeding focused on the selection of progeny with fewer deleterious alleles. 展开更多

The demand for medicinal plants at the origin of phyto-drugs,like all ethnobotanical plants,threatens their existence today.The aim of this work was to determine suitable methods for optimizing the possibilities of regenerating Stereospermum kunthianum.To achieve this objective,3 levels of mechanical scarification(light scarification,medium scarification and deep scarification)were applied to the seeds of this species using a rough instrument,and 5 days after germination,the seedlings were transplanted into 5 types of substrate to study their effects on seedling growth.The length of the seedlings was measured using a graduated ruler.The results obtained showed that the best latency time was 2 days for average seed scarifications.The highest germination half-time was 4.00±0.06 days observed for average seed scarification,the highest germination speed was 22.50±1.00 and the highest germination rate was 100.00%±0.00%for average scarification.The homogeneous substrate mixes black earth/woodsilk and Fine sand/woodsilk were more effective for seedling growth(1.9 cm/day in the juvenile state)and development.Medium seed scarification is the most effective method for regenerating Stereospermum kunthianum,and the black earth/wood shavings substrate mix is the best for its cultivation. 展开更多

Polyploidy plays a major role in genome evolution,which corresponds to environmental changes over millions of years.The mechanisms of genome evolution,particularly during the process of domestication,are of broad interest in the fields of plant science and crop breeding.Upland cotton is derived from the hybridization and polyploidization of its ancient A and D diploid ancestors.As a result,cotton is a model for polyploid genome evolution and crop domestication.To explore the genomic mysteries of allopolyploid cotton,we investigated asymmetric evolution and domestication in the A and D subgenomes.Interestingly,more structural rearrangements have been characterized in the A subgenome than in the D subgenome.Correspondingly,more transposable elements,a greater number of lost and disrupted genes,and faster evolution have been identified in the A subgenome.In contrast,the centromeric retroelement(RT-domain related) sequence of tetraploid cotton derived from the D subgenome progenitor was found to have invaded the A subgenome centromeres after allotetrapolyploid formation.Although there is no genome-wide expression bias between the subgenomes,as with expression-level alterations,gene expression bias of homoeologous gene pairs is widespread and varies from tissue to tissue.Further,there are more positively selected genes for fiber yield and quality in the A subgenome and more for stress tolerance in the D subgenome,indicating asymmetric domestication.This review highlights the asymmetric subgenomic evolution and domestication of allotetraploid cotton,providing valuable genomic resources for cotton research and enhancing our understanding of the basis of many other allopolyploids. 展开更多

Kokwaro].These include the contribution of this species towards health,nutrition,food security and conservation by sharing local skills and knowledge related to it.Marula fruits can be eaten fresh,squeezed to make juice,brewed in traditional beer or used to make jam and jelly.The kernels are also edible and can be pressed to extract oil for cooking and cosmetics,i.e.,for skin and hair application.The bark,roots,seeds and leaves are exploited for traditional medicinal purposes.Marula has acquired significant commercial value since its fruits and other products have entered local,regional and international trade in southern Africa.To diver-sify fruit production and satisfy the increasing demands for this resource,several domestication initiatives have been carried out at regional and international levels.The accumulated knowledge and skills relevant to the establishment and commercialization of marula therefore provide an effective guide in areas where marula remains undomesticated or underutilized.We discuss the great importance of marula in southern Africa to reveal its great potential to regions where it remains unexploited. 展开更多

RESULTS: Trained Tibetan monkeys calmly accepted IOP measurement by the TonoVete (R) rebound tonometer without sedation or anesthesia and their IOP values were similar to other primates. CONCLUSION: Human-cultivated Thibetan monkeys are tamable, and can be used for biomedical research such as ophthalmic research without anesthesia. 展开更多

Tomato(Solanum lycopersicum) stress resistance and fruit total soluble solid(TSS) content have changed dramatically during selective breeding, and transcriptome variation has played a critical role in this rewiring. However, the single tomato reference genome impedes characterization of whole-transcriptome variation during domestication and breeding at the population level. Here, we constructed a pan-transcriptome of orange-stage tomato fruit, and investigated global expression presence/absence variation(e PAV) and differentially expressed genes(DEGs) based on RNA sequencing(RNA-seq) data from 399 tomato accessions. A total of 7 181 genes absent from the reference genome were identified, 6 122 of which were e PAV genes during tomato domestication and breeding including resistance genes such as late blight resistance gene PIM_DN29746_c0_g3_i1 and peroxidase P7-like gene PIM_DN30274_c0_g2_i1. In addition, 3 629 genes were significantly differentially expressed during tomato selection, among which 19 genes were associated with the reduced fruit TSS content of modern tomato cultivars, including LIN5, TIV1, and seven novel sugar transporter genes. Our results indicate that natural and artificial selection greatly shaped the tomato transcriptome, thereby altering the fruit TSS content and resistance to abiotic and biotic stresses. 展开更多

Domestication and genetic improvement of maize improve yield and stress tolerance due to changes in morphological and physiological properties, which likely alter rhizosphere microbial diversity. Understanding how the evolution of maize germplasm impacts its rhizobacterial traits during the growth stage is important for optimizing plant-microbe associations and obtaining yield gain in domesticated germplasms. In this study, a total of nine accessions representing domestication and subsequent genetic improvement were selected. We then sequenced the plant DNA and rhizobacterial DNA of teosinte, landraces and inbred lines at the seedling, flowering and maturity stages in a field trial. Moreover, the soil chemical properties were determined at the respective stages to explore the associations of soil characteristics with bacterial community structures. The results showed that domestication and genetic improvement increased the rhizobacterial diversity and substantially altered the rhizobacterial community composition. The core microbiome in the rhizosphere differed among germplasm groups. The co-occurrence network analysis demonstrated that the modularity in the bacterial network of the inbred lines was greater than those of teosinte and the landraces. In conclusion, the increased diversity of the rhizobacterial community with domestication and genetic improvement may improve maize resilience to biotic stresses and soil nutrient availability to plants. 展开更多

Strong genetic incompatibilities exist between two primary rice subspecies,indica and japonica.However,the wild ancestors of rice,O.nivara Sharma et Shastry and O.rufipogon Griff.,are genetically compatible.How this genetic incompatibility became established has not been clearly elucidated.To provide insights into the process,we analyzed a pair of hybrid sterility genes in rice,DOPPELGANGER 1(DPL1)and DOPPELGANGER 2(DPL2).Either of the two loci can have one defective allele(DPL1-and DPL2-).Hybrid pollen carrying both DPL1-and DPL2-alleles is sterile.To explore the origination of DPL1-and DPL2-,we sequenced the DPL1 and DPL2 genes of 811 individual plants,including Oryza sativa(132),O.nivara(296)and O.rufipogon(383).We then obtained 20 DPL1 and 34 DPL2 sequences of O.sativa from online databases.Using these sequences,we analyzed the genetic and geographic distribution patterns of DPL genes in modern rice and its wild ancestors.Compared with the ancestral populations,DPL1-and DPL2-showed reduced diversity but increased frequency in modern rice.We speculated that the diversity reduction was due to a historic genetic bottleneck,and the frequency had likely increased because the defective alleles were preferred following this artificial selection.Such results indicated that standing variances in ancestral lines can lead to severe incompatibilities among descendants.Haplotype analysis indicated that the DPL1-haplotype of rice emerged from an O.nivara population in India,whereas the DPL2-haplotype emerged from O.rufipogon in South China.Hence,the evolutionary history of DPLs conforms to the presumed multiple domestication events of modern rice. 展开更多

According to the characteristics of oil containing wastewater, four strains of microorganism, named TA-11, TA-17, HA-9, HD-1, were picked out from the oil contaminated soil and activated sludge of biochemical treatment system of an asphalt plant wastewater in Panjin. They can degrade oil and CODCr in oil containing wastewater. The research result showed that each strain of microorganisms can remove oil and CODCr in oil containing wastewater effectively when the pH value was 7.0, the temperature was 30 degree Celsius, the rotation speed was 140r/min and the inoculation amount was 10%. Especially the highest removal ratio of CODCr was 68% after growth of 64 hours. The removal ratio of CODCr in oil containing wastewater of mixed bacilli was much higher than that of unitary bacilli, and mixing a certain amount of domestic sewage with the oil containing wastewater will also improve the removal rate of CODCr. 展开更多

The antioxidant activities of Ocimum basilicum and Ocimum gratissimum consumed as spices in Nigeria were evaluated in a bid to provide better scientific basis for increased domestication efforts on them. Total phenol and flavonoid contents of the spice extracts ranged from 9.09 - 27.41 μg GAE/mg and 5.38 - 22.88 μg RE/mg respectively. The DPPH and hydroxyl radical scavenging activities of the spice extracts ranged from 58.43% - 92.37% and 6.27% - 16.67% respectively. The total antioxidant capacities and reducing powers of the extracts (measured as absorbance values) ranged from 0.137 - 0.160 and 0.130 - 0.158 respectively. Generally, Ocimum basilicum maintained superior antioxidant activities to O. gratissimum in all the test assays, and all the extracts showed dose-dependent antioxidant activities. Ascorbic acid, Gallic acid and Rutin used as reference compounds generally showed higher antioxidant activities to the spice extracts except in the hydroxyl radical scavenging assay. Put together, these results confirm that Ocimum basilicum and O. gratissimum extracts possess appreciable natural antioxidant potentials, thereby providing good justification for their increased domestication and consumption. 展开更多
