Over higher plant would easily result in plant lodging and output reducing. On the other hand, the dwarf varieties with proper plant height had higher lodging resistance and a greater harvest index, allowing for the increased use of nitrogen fertilizer. Dwarf breeding had made a great breakthrough in the rice breeding. The breeding and extension of excellent dwarf varieties remarkably improved the yield potential of rice. Polymorphic analysis of 221 SSR loci demonstrated that 162d derived from a semidwarf variety Shuhui 162 through mutation, and 162d and Shuhui 162 were just a pair of near isogenic lines. Genetic analysis of F_1 and F_2 populations suggested that dwarfism in 162d was controlled by a single recessive gene. Phenotypic characteristics of the mutant gene were that plant height was about a quarter of normal height, grain size about a quarter of normal size, leaf was short and broad, and seed setting rate was very low, compared with the near isogenic line Shuhui 162. The mutant gene was sensitive to gibberellin (GA_3) treatment and did not located on the region near the centromere of rice chromosome 5, where dl gene located. Therefore, it was concluded that the mutant gene of 162d was a new small grain dwarf gene in rice. 展开更多

The lengths of the 4-5th internode, the 3rd, 2nd, 1st internodes from the top and the panicle length in the six dwarf near isogenic lines were 97.2%, 53.3%, 65.1%, 61.9% and 94.7% of those in the six tall ones, respectively, indicating that the dominant semi-dwarfing gene significantly inhibited the internode elongation. Moreover, Y98149 (mutant type) was more sensitive to GA3 than Y98148 (wild type), and had a lower GA3 concentration in plant, about 78% of Y98148. 展开更多

Understanding the effects of wheat dwarfing genes on the coleoptile length and plant height is crucial for the proper utilization of dwarfing genes in the improvement of wheat yield. Molecular marker analysis combined with pedigree information were used to classify wheat cultivars widely planted in major wheat growing regions in China into different categories based on the dwarfing genes they carried. The effects of the dwarfing genes with different sensitivity to gibberellins (GA3) on the coleoptile length and plant height were analyzed. Screening of 129 cultivars by molecular marker analysis revealed that 58 genotypes of wheat contained the dwarfing gene Rht-B1b, 24 genotypes of wheat contained Rht-D1b gene and 73 genotypes of wheat possessed Rht8 gene. In addition, among these 129 cultivars, 35 genotypes of wheat cultivars contained both Rht-B1b and Rht8 genes and 16 genotypes of wheat cultivars contained both Rht-D1b and Rht8 genes. Wheat cultivars with the dwarfing genes Rht-B1b or Rht-D1b were insensitive to GA3, while the cultivars with the dwarfing gene Rht8 were sensitive to GA3. Most of the wheat genotypes containing combination of Rht8 gene with either Rht-B1b or Rht-D1b gene were insensitive to GA3. The plant height was reduced by 24.6, 30.4, 28.2, and 32.2%, respectively, for the wheat cultivars containing Rht-B1b, Rht-D1b, Rht-B1b + Rht8, and Rht-D1b + Rht8 genes. The plant height was reduced by 14.3% for the wheat cultivar containing GA3-sensitive gene Rht8. The coleoptile length was shortened by 25.4, 31.3, 28.4 and 31.3%, respectively, in the wheat cultivars containing Rht-B1b, Rht-D1b, Rht-B1b +Rht8 and Rht-D1b + Rht8 genes, while the coleoptile length was shortened only by 6.2% for the wheat cultivar containing Rht8 gene. We conclude that GA3-insensitive dwarfing genes (Rht-B1b and Rht-D1b) are not suitable for the wheat improvement in dryland because these two genes have effect on reducing both plant height and coleoptile length. In contrast, GA3- sensitive dwarfing gene (Rht8) is a relatively ideal candidate for the wheat improvement since it significantly reduces the plant height of wheat, but has less effect on the coleoptile length. 展开更多

The combination of the Rht2 and Rht3 geneshowed significantly negative effect on yield components. All of these 5 dwarfing orsemidwarfing genotypes mentioned above had a significantly negative effect on plantheight and no significant effect on the area of flag leaf, spikelets per spike and spikelength. 展开更多

Application and functional study of dwarf and semi-dwarf genes are of great importance to both crop breeding and molecular biology. A new semi-dwarf gene, sd-t(t), non-allelic to sd-1,had been identified in an indica rice variety, Aitaiyin 2. In this study the gene was genetically mapped by using an F2 population, which consisted of 474 individuals developed from a cross between Aitaiyin 2 and B30. The sd-t(t) gene was located between the RFLP markers R514 and R1408B with a distance of 1.1 cM to R514, and 4.5 cM to R1408B on chromosome 4. A physical contig covering the sd-t(t) mapping region was further constructed by screening a BAC library with R514 and R1408B as probes, and the physical distance between R514 and R1408B was estimated at approximately 147 kb. This result will facilitate map-based cloning of the sd-t(t) gene. 展开更多

Using direct cross and para-back-cross methods, allelic tests were carried out not only between the dwarfing genes in Zhepi 1 and Aizao 3, but also with br, uzu, denso, or sdwl and the six novel dwarfing genes that have been recently identified in barley in China. The results showed that the plant height was attributed to Mendelian inheritance of a recessive dwarfing gene both in Zhepi 1 and Aizao 3. The dwarfing genes carried by the two cultivars were the same, but different from the three known and the six novel dwarfing genes. On the basis of the present study, only two dwarfing genes have been used in barley breeding in China since 1950. 展开更多

A segregating population with 410 F2 individuals from the cross MERCIA (Rht-Bla)×Dwarf 123 was made to identify a new major dwarfing gene carrying by novel wheat germplasm Dwarf 123. Combination of bulk segerant analysis method was used. A total of 145 SSR markers were tested for polymorphisms among parental lines and DNA bulks of F2 population. Out of 145 primer pairs only three markers revealed corresponding polymorphism among parental lines and F2 DNA bulks. The marker Barc20 was close to the dwarfing gene with a genetic distance of 1.8 cM, and markers Gwm513 and Gwm495 were linked to the gene with genetic distance of 6.7 and 13 cM, respectively. Linkage analysis mapped the dwarfing gene to the long arm of chromosome 4B with the order of Barc20-dwarfing gene-Gwm513-Gwm495. The Comparision between the new gene and the known Rht-B1 alleles showed that dwarfing gene Rht-Ai123 was different from the others. The identification of the new dwarfing gene and its linked markers will greatly facilitate its utilization in wheat high yield breeding for reducing plant height. 展开更多

A novel rice d1 mutant was identified using map-based cloning and comparative analysis of known dl mutants.The mutant[d1-a) shows a mild dwarf trait,which differs only slightly from the wildtype in plant height at the tillering stage.The dl-a mutant is different from other dl mutants.We found that it was interrupted by an Osr4 long terminal repeat(LTR)-retrotransposon,which resulted in the loss of exon 7 in the mutant D1 mRNA.A paralog of the D1 gene.D1-like,was revealed.D1-like is a truncated gene that might have resulted from recombination between retrotransposons.We identified 65 Osr4LTR-retrotransposons in Nipponbare,and found more LTR variants in contrast to coding DNA sequence(CDS) in the retrotransposons.We also identified five possible regulatory motifs in LTRs which may control the expression of the retrotransposons.In addition,we predicted six putative functional Osr4 retrotransposons that contain complete CDSs and all important elements.Osr4 retrotransposons were classified into 4 groups,and this type of retrotransposon only appears to be present in monocots.Members of group 1-1,which included all putative functional retrotransposons,showed a high similarity with each other.The retrotransposons were expressed in all tissues,at especially higher levels in some leaves and seeds.These findings imply that transpositions of group 1-1 members might have occurred frequently and recently. 展开更多

Compared with wild-type plants, the d63 mutant showed multiple abnormal phenotypes, such as dwarfism, more tillers, smaller flag leaf and reduced seed-setting rate and 1000-grain weight. In this study, two F2 populations were developed by crossing between d63 and Nipponbare, d63 and 93-11. Genetic analysis indicated that d63 was controlled by a single recessive gene, which was located on the short arm of chromosome 8, within the genetic distance of 0.40 cM from RM22195. Hence, D63 might be a new gene as there are no dwarf genes reported on the short arm of chromosome 8. 展开更多

Expression profiles of ten pathogenesis-related (PR) genes during plant defense against Fusarium, Yellow dwarf virus (YDV) aphid-transmitted and Hessian fly (Hf) were compared temporally in both resistant and susceptible genotypes following pathogen infection or insect infestation. Quantitative real-time PCR (qRT-PCR) revealed that PR1, PR2, PR3, PR5, PR6, PR8, PR9, and PR15 appeared to be induced or suppressed independently in response to Fusarium, YDV aphid-transmitted or Hf during the interactions. The PR gene(s) essential to defense against one organism may play little or no role in defense against another pathogen or pest, suggesting the alternative mechanisms may be involved in different interactions of wheat- Fusarium, wheat-YDV aphid-transmitted and wheat-Hf. However, strong up- or down-regulation of PRl2 and PR14 encoding low molecular membrane acting protein, defensin and lipid transfer protein (LTP), respectively, had been detected after either pathogen infection or insect infestation, therefore showed broad responses to pathogens and insects. It was postulated that low molecular proteins such as defensins and LTPs might play a role in the early stages of pathogenesis in the signaling process that informs plants about the attack from biotic stresses. In addition, a synergistic action between different PR genes might exist in plants to defense certain pathogens and insects on the basis of comprehensive expression profiling of various pathogenesis-related genes revealed by qRT-PCR in this study. 展开更多

Wheat dwarf virus (WDV), an important cereal pathogen, is closely related to Maize streak virus (MSV), a model virus of the Mastrevirus genus. Based on its similarity to known MSV resistance strategies, a truncated part of the WDV replication- associated (RepA) gene (WDVRepA215) and the WDV RepA gene with a mutated retinoblastoma-related protein (RBR) interaction domain (WDVRepA215RBRre^t) were cloned into the plPKb002 expression vector and transformed into immature embryos of spring barley cv. Golden Promise plants through Agrobacterium-mediated transformation. A detailed study of T1-generation plants infected by leafhoppers (Psammotettix alienus) fed on infection sources of variable strength was performed over a 5-week period encompassing the initial stages of virus infection. A DNA WDV TaqMan qPCR assay normalized using the DNA puroindoline-b SYBR Green qPCR assay for samples on a per week basis revealed an approximately 2-week delay in WDVRepA215RBR^mut plants to WDVRepA215 plants before significant increases in the WDV viral levels occurred. Both WDVRepA215 and WDVRepA215RBR^mut plants showed similar levels of transgenic transcripts over the screened period; however, the transgenic plants also showed increased numbers of infected plants compared to the control plants. 展开更多

地方种是小麦育种的重要种质资源,为了解矮秆基因在地方种中的分布,本研究检测了甘肃省地方种矮秆基因等位变异类型及其在不同麦区的分布频率。结果表明:(1)地方种Rht-B1b和Rht-D1b的频率极低;41.4%的地方种携带Rht8,且春麦区高于冬麦区... 地方种是小麦育种的重要种质资源,为了解矮秆基因在地方种中的分布,本研究检测了甘肃省地方种矮秆基因等位变异类型及其在不同麦区的分布频率。结果表明:(1)地方种Rht-B1b和Rht-D1b的频率极低;41.4%的地方种携带Rht8,且春麦区高于冬麦区;46.7%的地方种含Rht24b,春麦区低于冬麦区。Ppd-D1a的频率仅17.8%,且春麦区低于冬麦区。另外,仅检测到Rht-D1b/Rht8、Rht-D1b/Rht24b和Rht8/Rht24b 3种组合,频率分别为0.2%、0.5%和12.8%。(2)地方种携带的矮秆基因及其组合分布频率低于育成种,且差异较大。不同来源育成品种携带的优势矮秆等位变异和频率不同,清水试验站的品种以Rht-D1b、Rht8和Rht24b为主,黄羊试验站的品种以Rht-B1b、Rht-D1b、Rht8和Rht24b为主,甘谷试验站的品种以Rht8和Rht24b为主。清水和黄羊试验站的品种秆矮、丰产性好,可在河西、沿黄灌区、陇南、陇东的小麦育种中应用;甘谷试验站的品种茎秆高,抗病性突出,可应用于定西、天水、陇南和陇东等旱地小麦的抗病改良。(3)基于分子标记检测结果,筛选出15份地方种和31份育成种,以上材料均携带2个及以上降秆基因(包括矮秆基因或Ppd-D1a),可为甘肃不同麦区小麦矮秆育种提供亲本材料。 展开更多

目的探讨低剂量CT结合Ras相关区域家族蛋白1A(RASSF1A)、矮小同源盒基因2(SHOX2)甲基化在肺癌早期预测中的应用价值。方法选取2021年1月~2023年1月我院90例拟行肺结节手术患者,根据手术病理学分为肺良性结节组和肺癌组。2组均于术前行... 目的探讨低剂量CT结合Ras相关区域家族蛋白1A(RASSF1A)、矮小同源盒基因2(SHOX2)甲基化在肺癌早期预测中的应用价值。方法选取2021年1月~2023年1月我院90例拟行肺结节手术患者,根据手术病理学分为肺良性结节组和肺癌组。2组均于术前行低剂量CT检查、SHOX2、RASSF1A甲基化检测,采用Kappa指数分析上述检查结果与手术病理学一致性,分析低剂量CT、SHOX2、RASSF1A甲基化与血清肿瘤标志物[癌胚抗原(CEA)、神经元特异性烯醇化酶(NSE)、鳞状细胞癌抗原(SCC-Ag)、细胞角蛋白19片段(CYFRA21)]对肺癌诊断效能,采用Spearman低剂量CT检查、SHOX2、RASSF1A甲基化与临床病理特征相关性。结果低剂量CT、SHOX2、RASSF1甲基化及三者联合分别确定40例、43例、46例、58例肺癌,三者联合与手术病理学诊断肺癌效能一致性Kappa值为0.951;三者联合诊断肺癌敏感度96.67%、准确度97.78%均高于三者单一诊断效能(P<0.05);肺癌患者血清CEA、SCC、NSE、CYFRA21水平均高于肺良性结节患者(P<0.05);低剂量CT联合SHOX2、RASSF1甲基化诊断肺癌效能的AUC为0.983,近似于四种血清肿瘤标志物诊断肺癌效能的AUC 0.933;不同肿瘤直径、临床分期、组织学分化肺癌患者低剂量CT检出率及SHOX2、RASSF1A甲基化阳性率比较差异有统计学意义(P<0.05);肺癌患者低剂量CT检出率、SHOX2及RASSF1A甲基化阳性率与肿瘤直径、临床分期呈正相关,与组织学分化呈负相关(P<0.05)。结论低剂量CT联合SHOX2及RASSF1A甲基化可用于肺癌早期预警中,临床可通过其进行早期诊断、评估病情进展程度,以针对性展开后续治疗,改善预后。 展开更多

黄淮南部地区小麦品种矮秆基因的组成及其对茎秆特性的影响

为研究矮秆基因及其组合对小麦茎秆特性的影响,以黄淮南部地区490个小麦品种(系)为材料,采用分子标记检测了小麦矮秆基因Rht-B1b、Rht-D1b、Rht8、Rht9、Rht12和Rht24,并连续2年测定了小麦株高、重心高度、节间长度、直径、折断力等茎... 为研究矮秆基因及其组合对小麦茎秆特性的影响,以黄淮南部地区490个小麦品种(系)为材料,采用分子标记检测了小麦矮秆基因Rht-B1b、Rht-D1b、Rht8、Rht9、Rht12和Rht24,并连续2年测定了小麦株高、重心高度、节间长度、直径、折断力等茎秆特性,分析了矮秆基因的组成及其对茎秆特性的影响。结果表明,在490份材料中,携带Rht-B1b、Rht-D1b、Rht8、Rht9、Rht12和Rht24的材料分布频率分别为7.25%、92.24%、39.18%、42.86%、72.65%和95.31%,共得到了40种基因组合,其中分布频率较高的组合有Rht-D1b+Rht9+Rht12+Rht24(20.2%)、Rht-D1b+Rht12+Rht24(19.8%)、Rht-D1b+Rht8+Rht12+Rht24(16.3%)、Rht-D1b+Rht8+Rht9+Rht12+Rht24(8.4%)、Rht-D1b+Rht24(7.3%)、Rht-D1b+Rht9+Rht24(6.5%)、Rht-D1b+Rht8+Rht24(4.7%)和Rht-D1b+Rht8+Rht9+Rht24(3.9%)。矮秆基因对降低小麦株高的效应大小依次是Rht-D1b、Rht9、Rht8、Rht12和Rht-B1b,且基因间存在累加效应,但Rht8与Rht9或Rht12结合不能起到明显的降秆作用。基因组合Rht-D1b+Rht9+Rht12+Rht24对降低茎秆重心高度和基部前3节间长度以及增强基部前3节间折断力的效应最大,基因组合Rht-D1b+Rht8+Rht9+Rht24对降低穗下节长度的效应最大,基因组合Rht-D1b+Rht8+Rht12+Rht24对增大茎秆基部前3节间直径的效应最大。矮秆基因可以有效降低小麦的株高和重心高度,缩短基部第2、3节间长度,提高基部前3节间直径和折断力,可作为小麦的抗倒伏育种的重要依据。 展开更多

甘肃冬小麦品种(系)主要矮秆基因型及其育种利用进展

适当降低株高,增强品种抗旱性,提高品种丰产性是甘肃旱地冬小麦育种新策略。为了明确近20年甘肃冬小麦品种(系)中矮秆基因的分布以及株高变化情况,本研究利用6个主要矮秆基因的特异性分子标记对92份冬小麦品种(系)进行基因型检测,并对... 适当降低株高,增强品种抗旱性,提高品种丰产性是甘肃旱地冬小麦育种新策略。为了明确近20年甘肃冬小麦品种(系)中矮秆基因的分布以及株高变化情况,本研究利用6个主要矮秆基因的特异性分子标记对92份冬小麦品种(系)进行基因型检测,并对矮秆基因的利用情况进行分析。结果表明,供试品种(系)中均含有Rht-D1b基因,其次是Rht8、Rht13和Rht5(分布频率分别为59.8%、29.3%和10.9%),但并未检测到Rht-B1b和Rht12。Rht5和Rht13基因主要分布在陇南(天水市和陇南市)的山旱地品种(系)中,频率分别为16.7%和36.7%,Rht8主要分布在陇南川区品种(系)中,频率为77.4%。同时含有2个矮秆基因的材料有39份,分布频率为42.4%,其中Rht-D1b+Rht8的分布频率最高(33.7%);同时含有3个矮秆基因的材料有25份,分布频率为27.2%,其中Rht-D1b+Rht8+Rht13的分布频率最高(19.6%);同时含有4个矮秆基因Rht-D1b+Rht5+Rht8+Rht13的材料只有1份,分布频率为1.1%。含多矮秆基因组合的品种(系)在陇南川区的分布频率最高。结合表型鉴定,陇南山旱地、川区和陇东(平凉市和庆阳市)区域推广应用品种(系)的平均株高分别是96.8、80.9和98.9 cm,变异范围分别在80.5~111.0、60.0~105.0和80.0~115.0 cm。对不同年份育成的冬小麦品种(系)株高分析发现,自2000年至2022年陇南山旱地、陇南川区和陇东区域推广应用品种(系)的株高分别下降了16.9%、15.2%和6.7%。不同矮秆基因组合在品种(系)中的降秆效应由强到弱依次为Rht-D1b+Rht5+Rht8(20.5%)>Rht-D1b+Rht5+Rht8+Rht13(20.3%)>Rht-D1b+Rht8+Rht13(15.0%)>Rht-D1b+Rht5+Rht13(14.8%)>Rht-D1b+Rht8(10.1%)>Rht-D1b+Rht5(6.4%)>Rht-D1b+Rht13(1.1%)。多样化的矮秆基因在甘肃冬小麦育种中得到应用,说明近年来旱地冬小麦育种在适当降低株高、防止倒伏、提高品种丰产性方面有了新的进展,为进一步适当降低甘肃冬小麦品种株高的同时保持生产品种的抗旱性奠定了基础。 展开更多

【目的】对一个同时导致营养和生殖器官发育异常的水稻突变体进行表型鉴定、基因定位和候选基因分析,为下一步的基因克隆与功能分析奠定基础。【方法】在水稻籼型恢复系602组织培养后代中,发现一个矮化并花发育异常突变体dwarf and defo... 【目的】对一个同时导致营养和生殖器官发育异常的水稻突变体进行表型鉴定、基因定位和候选基因分析,为下一步的基因克隆与功能分析奠定基础。【方法】在水稻籼型恢复系602组织培养后代中,发现一个矮化并花发育异常突变体dwarf and deformed flower 2(ddf2)。抽穗期,以野生型为对照,对ddf2株高、主穗长、节间和功能叶的长宽等性状进行统计分析;同时利用冷冻切片等技术对茎、叶和花器官进行详细的形态和组织学分析。分别以西农1A和中花11为母本,以DDF2/ddf2杂合株系为父本构建2个F2群体进行遗传分析和基因定位,并对候选基因进行实时荧光定量PCR(real-time PCR)分析。【结果】相较于野生型,突变体各节间的长和茎粗均极显著降低,叶片极显著变短、变窄,同时花序也极显著变短。组织细胞学分析发现,突变体大叶脉数目和相邻2个大叶脉之间的小叶脉数都没有明显的变化,但相邻2个大叶脉之间的宽度明显减小,进一步比较2个小叶脉之间的叶肉细胞,发现在突变体中细胞数目和尺寸均显著降低;突变体茎秆维管束的数目与野生型相比没有明显的变化,但统计发现2个大维管束之间基本组织细胞的数量和细胞的大小都显著小于野生型,表明ddf2突变体茎、叶细胞分裂和膨胀都受到了抑制;此外,ddf2突变体的花器官特征发育受到了严重干扰:第一轮外稃顶部弯曲、内稃不同程度退化,第三轮雄蕊器官严重退化,部分甚至转化为雌蕊状器官,另外部分ddf2小穗的护颖过度发育,转变成稃片状,一些小穗还表现分生组织确定性的丢失,发育出2个以上的小花。遗传分析表明该突变性状受1对隐性基因控制。利用中花11/ddf2的1 024株F2分离群体,最终将DDF2精细定位在第11染色体短臂近着丝粒位置处,位于insertion/deletion(in/del)标记S-11和S-14之间,遗传距离分别为0.049和0.098c M,物理距离为90.295 kb,并与标记S-24共分离。分析定位区间的基因,发现共有MSU注释基因12个,其中一个编码Sec3_C蛋白的LOC_Os11g17600内部包含共分离标记S-24,进一步对该基因进行表达分析,发现该基因在突变体的叶、茎和穗中都表现出明显的下调,初步将LOC_Os11g17600作为DDF2候选基因。【结论】DDF2是一个同时控制水稻茎/叶和花器官发育的新基因。 展开更多

目的:探究高分辨CT三维重建联合肺泡灌洗液(BALF)中矮小同源盒基因2(SHOX2)、Ras相关区域家族A1(RASSF1A)基因甲基化检测诊断早期肺结节的价值。方法:选取2021年5月—2022年5月某院就诊并经纤维支气管镜(FB)检查的肺结节患者100例,FB下... 目的:探究高分辨CT三维重建联合肺泡灌洗液(BALF)中矮小同源盒基因2(SHOX2)、Ras相关区域家族A1(RASSF1A)基因甲基化检测诊断早期肺结节的价值。方法:选取2021年5月—2022年5月某院就诊并经纤维支气管镜(FB)检查的肺结节患者100例,FB下收集患者的BALF,通过实时荧光定量PCR法测定BALF中SHOX2和RASSF1A基因甲基化状态,同时收集高分辨CT三维重建检查、BALF检测结果。依据病检结果将患者分为恶性结节组(n=40)和良性结节组(n=60),分析高分辨CT三维重建检查、BALF中SHOX2、RASSF1A基因甲基化检测对早期肺结节的诊断价值,并绘制ROC曲线评价各检测方法在早期肺结节诊断中的效能。结果:SHOX2甲基化诊断恶性结节的敏感度为50.00%,AUC为0.708,RASSF1A甲基化诊断恶性结节的敏感度为52.50%,AUC为0.713,2基因甲基化联合诊断恶性结节的敏感度为75.00%,AUC为0.767。高分辨CT三维重建诊断恶性结节的敏感度为72.50%,特异度为73.33%,AUC为0.729,BALF对恶性结节的诊断敏感度为25.00%,特异度为100.00%,AUC为0.625。2基因甲基化联合+高分辨CT三维重建诊断恶性肺结节的AUC为0.890,敏感度与特异度分别为90.00%和73.33%,其诊断效能高于2基因甲基化联合+BALF细胞学分析和高分辨CT三维重建+BALF细胞学分析(Z=2.453、2.736,P均<0.05)。结论:高分辨CT三维重建联合BALF中SHOX2、RASSF1A基因甲基化检测在肺结节良恶性诊断中的鉴别效能较高,值得在临床中应用。 展开更多

小麦骨干亲本周8425B矮秆基因在其衍生品种(系)中的遗传解析

利用4个主要矮秆基因[Rht1(Reduced height 1)、Rht2、Rht8和Rht24]的分子标记对219个周8425B衍生品种(系)进行检测,研究矮秆基因分布、组合类型及其对小麦株高、产量相关性状的影响,为骨干亲本周8425B的持续遗传研究、小麦矮秆基因分... 利用4个主要矮秆基因[Rht1(Reduced height 1)、Rht2、Rht8和Rht24]的分子标记对219个周8425B衍生品种(系)进行检测,研究矮秆基因分布、组合类型及其对小麦株高、产量相关性状的影响,为骨干亲本周8425B的持续遗传研究、小麦矮秆基因分子标记辅助选择育种的利用奠定基础。结果表明,矮秆基因Rht1、Rht2、Rht8和Rht24在周8425B衍生品种(系)中的分布频率分别为94.52%、100.00%、42.01%和97.26%。在219份材料中,没有发现单独存在某一矮秆基因的类型,各矮秆基因均以组合形式存在,4个矮秆基因的组合类型共有7种,分别为Rht1+Rht2、Rht2+Rht8、Rht2+Rht24、Rht1+Rht2+Rht8、Rht1+Rht2+Rht24、Rht2+Rht8+Rht24和Rht1+Rht2+Rht8+Rht24,以Rht1+Rht2+Rht24组合分布频率最高,为53.42%;Rht2+Rht8组合分布频率最低,为0.46%。对于单个矮秆基因,Rht8基因的降秆作用最强,Rht1基因最弱;对于矮秆基因组合,Rht1+Rht2组合降秆作用最强,Rht1+Rht2+Rht8组合最弱。对于单个矮秆基因,Rht2基因型小麦产量最高,株高为77.38 cm;Rht8基因型小麦产量最低,株高为76.71 cm。对于矮秆基因组合,Rht2+Rht8组合产量最高,株高为78.21 cm;Rht1+Rht2组合产量最低,株高为75.85 cm。产量相关性状与4个矮秆基因的相关性分析结果表明,在一定范围内,适当提升株高有利于提高植株的生物量,增加收获指数,实现产量增加。筛选出3份产量较高的小麦材料,分别是郑麦103、存麦13和中育1220,其株高分别为82.79 cm、78.71 cm和82.50 cm,矮秆基因组合均为Rht1+Rht2+Rht24。 展开更多
