Objective To investigate the effects of erigeron breviscapus (Vant.) Hand-Mazz (erigeron breviscapus) pretreatment on pathology and oxyradical level in the spinal cord after ischemia-reperfusion (I/R) injury in rabbits. Methods A total of 40 New Zealand white rabbits were randomly divided into three groups: sham-operation group with 10 rabbits treated with only abdominal aorta exposure without occlusion,control group with 15 rabbits that underwent ischemia for 50 minutes and treated with matched saline,and experimental group with 15 rabbits that underwent ischemia for 50 minutes and treated with erigeron breviscapus (9mg/kg) injection before ischemia. Malondialdehyde (MDA) level and superoxide dismutase (SOD) activity in the spinal cord were examined at 6 and 24 hours after I/R,respectively. The morphological changes and the number of the spinal cord anterior horn motor neurons were observed and counted under the light microscope and electron microscope,respectively. Results The level of MDA was markedly decreased and SOD activity was increased in the experimental group compared with those in the control group (P<0.01). Compared with that in the control group,the number of motor neurons in the experimental group significantly increased at 24h after I/R (P<0.01) and the morphous of the motor neurons improved. Conclusion Erigeron breviscapus can reduce oxyradical production and the apoptosis of nerve cells,and protect nerve tissue structure and function after spinal cord I/R. 展开更多

目的:探讨中药灯盏细辛对大鼠肾冷缺血再灌注损伤(IRI)的保护作用及机制。方法:采用随机数字表将36只SD大鼠分为假手术组、模型对照组和实验组3组(n=12);实验组术前15min尾静脉注射灯盏细辛注射液1.2mL/100g,其他2组给予生理盐水。假手... 目的:探讨中药灯盏细辛对大鼠肾冷缺血再灌注损伤(IRI)的保护作用及机制。方法:采用随机数字表将36只SD大鼠分为假手术组、模型对照组和实验组3组(n=12);实验组术前15min尾静脉注射灯盏细辛注射液1.2mL/100g,其他2组给予生理盐水。假手术组切除右肾并游离左肾蒂;模型对照组和实验组制作冷IRI模型。3组大鼠均在术后24h再次手术切除左肾,采用透射电镜观察超微结构的变化,并制作肾脏组织匀浆,检测肾组织匀浆中超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量。手术前后取血标本检测血浆中尿素氮(BUN)、肌酐(Cr)以评估肾脏功能。结果:①假手术组肾小球及肾小管及细胞器结构正常;模型对照组肾小管细胞呈现典型IRI,线粒体肿胀明显,部分线粒体膜撕裂,微绒毛脱落,胞质内空泡变性,部分细胞核可见凋亡迹象;实验组病变较模型对照组明显减轻。②3组肾组织匀浆SOD活及MDA含量差异有统计学意义(F分别为44.534和145.535,P<0.001)。③IRI后模型对照组、实验组血浆BUN、Cr均高于术前,且模型对照组高于实验组(BUN:F组间=88.647,F时间=188.620,F交互=88.331,P<0.001;Cr:F组间=200.470,F时间=555.184,F交互=375.640,P<0.001)。结论:灯盏细辛注射液对大鼠肾脏冷IRI具有保护作用。 展开更多

目的观察中药灯盏细辛对大鼠肾脏冷缺血再灌注损伤(IRI)中肾脏细胞凋亡及相关基因表达的影响。方法封闭群SD大鼠36只,随机分为3组,每组12只。假手术组(A组),对照组(B组),实验组(C组)。药物应用:c组术前15min,灯盏细辛注... 目的观察中药灯盏细辛对大鼠肾脏冷缺血再灌注损伤(IRI)中肾脏细胞凋亡及相关基因表达的影响。方法封闭群SD大鼠36只,随机分为3组,每组12只。假手术组(A组),对照组(B组),实验组(C组)。药物应用:c组术前15min,灯盏细辛注射液按1.2ml/100g通过尾静脉注射,A、B组按相应剂量注射生理盐水。动物手术:A组,切除右肾。B、C组采用的是冷IRI模型,3组大鼠均在术后24h再次手术切除左肾进行检测。透射电镜检查肾组织形态学,免疫组织化学检测凋亡相关的基因bcl-2与bax的表达,原位末端标记法(TUNEL)检测细胞凋亡。结果(1)超微结构检查(透射电镜):A组结构正常;B组细胞呈损伤形态:线粒体肿胀,微绒毛减少,胞质内空泡形成,部分细胞核可见凋亡迹象。C组病变较B组显著减轻。(2)免疫组织化学蛋白阳性染色指数(PI):缺血再灌注后B、C组的bcl-2表达分别为(21.21±1.18)%和(35.52±1.94)%,较A组(4.95±0.77)%均增多(P〈0.05),B组低于C组(P〈0.05)。B、C组的bax表达分别为(58.55±2.90)%和(45.90±3.14)%,较A组(4.67±0.67)%增多(P〈0.05),而且B组高于C组(P〈0.05)。A组的蛋白阳性染色指数的比值bcl-2/bax为(1.06±0.07)高于B组(0.35±0.03)和C组(0.78±0.07,P〈0.05),而且C组高于B组(P〈0.05)。(3)细胞凋亡检测(TUNEL):细胞凋亡指数B组(28.57±3.58)%和C组(19.99±3.37)%均显著大于A组(2.33±0.42)%(P〈0.01),C组小于B组(P〈0.01)。结论灯盏细辛减少大鼠肾脏冷IRI诱导的肾脏细胞的凋亡,与调节凋亡相关基因bcl-2与bax表达有关。 展开更多
