In recent years,wearable devices-based Human Activity Recognition(HAR)models have received significant attention.Previously developed HAR models use hand-crafted features to recognize human activities,leading to the e...In recent years,wearable devices-based Human Activity Recognition(HAR)models have received significant attention.Previously developed HAR models use hand-crafted features to recognize human activities,leading to the extraction of basic features.The images captured by wearable sensors contain advanced features,allowing them to be analyzed by deep learning algorithms to enhance the detection and recognition of human actions.Poor lighting and limited sensor capabilities can impact data quality,making the recognition of human actions a challenging task.The unimodal-based HAR approaches are not suitable in a real-time environment.Therefore,an updated HAR model is developed using multiple types of data and an advanced deep-learning approach.Firstly,the required signals and sensor data are accumulated from the standard databases.From these signals,the wave features are retrieved.Then the extracted wave features and sensor data are given as the input to recognize the human activity.An Adaptive Hybrid Deep Attentive Network(AHDAN)is developed by incorporating a“1D Convolutional Neural Network(1DCNN)”with a“Gated Recurrent Unit(GRU)”for the human activity recognition process.Additionally,the Enhanced Archerfish Hunting Optimizer(EAHO)is suggested to fine-tune the network parameters for enhancing the recognition process.An experimental evaluation is performed on various deep learning networks and heuristic algorithms to confirm the effectiveness of the proposed HAR model.The EAHO-based HAR model outperforms traditional deep learning networks with an accuracy of 95.36,95.25 for recall,95.48 for specificity,and 95.47 for precision,respectively.The result proved that the developed model is effective in recognizing human action by taking less time.Additionally,it reduces the computation complexity and overfitting issue through using an optimization approach.展开更多
3D human pose estimation is a major focus area in the field of computer vision,which plays an important role in practical applications.This article summarizes the framework and research progress related to the estimat...3D human pose estimation is a major focus area in the field of computer vision,which plays an important role in practical applications.This article summarizes the framework and research progress related to the estimation of monocular RGB images and videos.An overall perspective ofmethods integrated with deep learning is introduced.Novel image-based and video-based inputs are proposed as the analysis framework.From this viewpoint,common problems are discussed.The diversity of human postures usually leads to problems such as occlusion and ambiguity,and the lack of training datasets often results in poor generalization ability of the model.Regression methods are crucial for solving such problems.Considering image-based input,the multi-view method is commonly used to solve occlusion problems.Here,the multi-view method is analyzed comprehensively.By referring to video-based input,the human prior knowledge of restricted motion is used to predict human postures.In addition,structural constraints are widely used as prior knowledge.Furthermore,weakly supervised learningmethods are studied and discussed for these two types of inputs to improve the model generalization ability.The problem of insufficient training datasets must also be considered,especially because 3D datasets are usually biased and limited.Finally,emerging and popular datasets and evaluation indicators are discussed.The characteristics of the datasets and the relationships of the indicators are explained and highlighted.Thus,this article can be useful and instructive for researchers who are lacking in experience and find this field confusing.In addition,by providing an overview of 3D human pose estimation,this article sorts and refines recent studies on 3D human pose estimation.It describes kernel problems and common useful methods,and discusses the scope for further research.展开更多

Currently, some methodsfor 3D human pose estimation in monocular video employ temporal convolutional network (TCN) to extractinter-frame feature relationships, but the majority of them suffer from insufficient inter-frame feature relationshipextractions. In this paper, we decompose the 3D joint location regression into the bone direction and length, wepropose the TCG, a temporal convolutional network incorporating Gaussian error linear units (GELU), to solvebone direction. It enablesmore inter-frame features to be captured andmakes the utmost of the feature relationshipsbetween data. Furthermore, we adopt kinematic structural information to solve bone length enhancing the use ofintra-frame joint features. Finally, we design a loss function for joint training of the bone direction estimationnetwork with the bone length estimation network. The proposed method has extensively experimented on thepublic benchmark dataset Human3.6M. Human enterovirus 71(HEV71) is the cause of hand,foot and mouth disease and associated neurological complications in children under five years of age.There has been an increase in HEV71 epidemic activity throughout th...Human enterovirus 71(HEV71) is the cause of hand,foot and mouth disease and associated neurological complications in children under five years of age.There has been an increase in HEV71 epidemic activity throughout the Asia-Pacific region in the past decade,and it is predicted to replace poliovirus as the extant neurotropic enterovirus of highest global public health significance. To date there is no effective antiviral treatment and no vaccine is available to prevent HEV71 infection. The increase in prevalence, virulence and geographic spread of HEV71 infection over the past decade provides increasing incentive for the development of new therapeutic and prevention strategies against this emerging viral infection. The current review focuses on the potential, advantages and disadvantages of these strategies. Since the explosion of outbreaks leading to large epidemics in China, research in natural therapeutic products has identified several groups of compounds with anti-HEV71 activities. Concurrently, the search for effective synthetic antivirals has produced promising results. Other therapeutic strategies including immunotherapy and the use of oligonucleotides have also been explored. A sound prevention strategy is crucial in order to control the spread of HEV71. To this end the ultimate goal is the rapid development, regulatory approval and widespread implementation of a safe and effective vaccine. The various forms of HEV71 vaccine designs are highlighted in this review. Given the rapid progress of research in this area, eradication of the virus is likely to be achieved.展开更多
Objective To develop RT-nPCR assays for amplifying partial and complete VP1 genes of human enteroviruses(HEVs)from clinical samples and to contribute to etiological surveillance of HEV-related diseases.Methods A panel...Objective To develop RT-nPCR assays for amplifying partial and complete VP1 genes of human enteroviruses(HEVs)from clinical samples and to contribute to etiological surveillance of HEV-related diseases.Methods A panel of RT-nPCR assays,consisting of published combined primer pairs for VP1 genes of HEV A–C and in-house designed primers for HEV-D,was established in this study.The sensitivity of each RT-nPCR assay was evaluated with serially diluted virus stocks of five serotypes expressed as CCID50 perμL and copies perμL,and the newly established methods were tested in clinical specimens collected in recent years.Results The sensitivity of RT-nPCR assays for amplifying partial VP1 gene of HEVs was 0.1 CCID50 perμL and 10 virus copies perμL,and for the complete VP1 gene was 1 CCID50 perμL and 100 virus copies perμL,using serially-diluted virus stocks of five serotypes.As a proof-of-concept,25 serotypes were identified and complete VP1 sequences of 23 serotypes were obtained by this system among 858 clinical specimens positive for HEVs during the past eight surveillance seasons.Conclusion This RT-nPCR system is capable of amplifying the partial and complete VP1 gene of HEV A–D,providing rapid,sensitive,and reliable options for molecular typing and molecular epidemiology of HEVs in clinical specimens.展开更多

The one step reaction was perfor...A sensitive reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay for human enterovirus 71 (EV71) and Coxsackievirus A16 (CVA16) infection was further evaluated. The one step reaction was performed in a single tube at 65?C for 45 min for EV71 and 35 min for CVA16. The detection limits of RT-LAMP assays for both EV71 and CVA16 were 0.1 of a 50% tissue culture infective dose (TCID50) per reaction, based on 10—Fold dilutions of a titrated EV71 or CVA16 strain. The specific assay showed there were no cross-reactions with Coxsackievirus A (CVA) viruses (CVA 2, 4, 5, 7, 9, 10, 14, and 25), Coxsackievirus B (CVB) viruses (CVB 1, 2, 3, 4, and 5) or ECHO viruses (ECHO 3, 6, 11, and 19). In parallel with commercial quantitative real-time polymerase chain reaction (qRT-PCR) diagnostic kits for EV71 and CVA16, the RT-LAMP assay was evaluated with 515 clinical specimens, the results showed the RT-LAMP assay and the qRT-PCR assay were in complete agreement for 513/515 (99.6%) of the specimens. Two samples with discrepant results from two methods were further verified by nested reverse transcription polymerase chain reaction (nRT-PCR) assay and sequencing to be true positives for CVA16. In conclusion, RT-LAMP assay is demonstrated to be a sensitive and specific assay and have a great potential for the rapid and visual screening of EV71 and CVA16 in China, especially in those resource-limited hospitals and rural clinics of provincial and municipal regions.展开更多

Our results showed that although most sites were under negative or neutral evolution, four positions of the VP1 genes were under positive selection pressure. This might account for the spread and frequent outbreaks of the viruses and the enhanced neurovirulence. In particular, position 98 might be involved in neutralizing antibodies, modulating the virus-receptor interaction and enhancing the virulence of the viruses. Moreover, both positions 145 and 241 might correlate to determine the receptor specificity. However, these positions did not display much difference in amino acid polymorphism. In addition, no position in the VP1 genes of viruses isolated from China was under positive selection.展开更多

Epidemiological investigations are carried o...Rabies is an infectious disease that almost always leads to death as soon as the first signs appear, and is still endemic in several regions, including Côte d’Ivoire. Epidemiological investigations are carried out and response measures implemented in the event of any death from human rabies. However, the quality of these investigations does not always allow the adoption of response measures. The aim of this work was to evaluate the reports of epidemiological investigations and response actions carried out during the occurrence of human rabies deaths from 2016 to 2018. This was a cross-sectional study that took place from April to September 2021 at the National Institute of Public Hygiene and focused on human rabies deaths notified from 206 to 2018. Of sixty-one (61) human rabies deaths recorded, sixty (60) were investigated, among which fifty-nine (59) deaths had been subject to response. Most deaths occurred in rural areas (75%), and the main vector was the dog (98%). Several shortcomings were noted: response actions were not carried out promptly;joint interventions bringing together human and animal health professionals were rare, there was little communication about rabies to communities (36%), and very little vaccination of the vector, the source of the disease (12%);an almost total absence of capacity-building for local players (7%);the health districts of Abobo-Ouest, Bondoukou, Divo and Fresco, which had regularly recorded human rabies deaths, had not benefited from dog vaccination, which is supposed to break the circulation of the virus. The persistence of rabies deaths in the health districts could probably be linked to these shortcomings. Faced with these realities, it is essential to strengthen the epidemiological investigation capacities of the players involved, in order to improve the quality of investigations and ensure an effective response.展开更多

Reports of the effects of vitamin D on osteoclasts, however, are conflicting, showing both inhibition and stimulation. Finding that resorbing osteoclasts in human bone express vitamin D receptor (VDR), we examined their response to different concentrations of 25-hydroxy vitamin D3 [25(OH)D3] (100 or 500 nmol·L^-1) and 1,25-dihydroxy vitamin D3 [1,25(OH)2D3] (0.1 or 0.5 nmol·L^-1) metabolites in cell cultures. Specifically, CD14+ monocytes were cultured in charcoal-stripped serum in the presence of receptor activator of nuclear factor kappa-B ligand (RANKL) and macrophage colony-stimulating factor (M-CSF). Tartrate-resistant acid phosphatase (TRAP) histochemical staining assays and dentine resorption analysis were used to identify the size and number of osteoclast cells, number of nuclei per cell and resorption activity. The expression of VDR was detected in human bone tissue (ex vivo) by immunohistochemistry and in vitro cell cultures by western blotting. Quantitative reverse transcription-PCR (qRT-PCR) was used to determine the level of expression of vitamin D-related genes in response to vitamin D metabolites. VDR-related genes during osteoclastogenesis, shown by qRT-PCR, was stimulated in response to 500 nmol·L^-1 of 25(OH)D3 and 0.1-0.5 nmol·L^-1 of 1,25(OH)2D3, upregulating cytochrome P450 family 27 subfamily B member I (CYP27B1) and cytochrome P450 family 24 subfamily A member I (CYP24A1). Osteoclast fusion transcripts transmembrane 7 subfamily member 4 (tm7sf4) and nuclear factor of activated T-cell cytoplasmic 1 (nfatcl) where downregulated in response to vitamin D metabolites. Osteoclast number and resorption activity were also increased. Both 25(OH)D3 and 1,25(OH)2D3 reduced osteoclast size and number when co-treated with RANKL and M-CSF. The evidence for VDR expression in resorbing osteoclasts in vivo and low-dose effects of 1,25(OH)2D3 on osteoclasts in vitro may therefore provide insight into the effects of clinical vitamin D treatments, further providing a counterpoint to the high-dose effects reported from in vitro experiments.展开更多

From January 2011 to July 2015, EV-D68 was identified in 12 individuals with respiratory infections in Beijing, China. The phylogenetic relationships based on the genomic sequence alignment showed that there were two lineages circulating in Beijing from 2011 to 2015. Eight EV-D68 strains belonged to group 1 and four belonged to group 3. All EV-D68 strains from Beijing in 2014 were separately clustered into subgroup II of group 1. Based on these results, we concluded that the Beijing EV-D68 strains had little association with the EV-D68 strains circulating in the 2014 USA outbreak.展开更多

The known anti-proliferative and pro-apoptotic actions of the active metabolite of vitamin D, 1,25-dihydroxy-vitamin D [1,25(OH)2D] are mediated through binding to the vitamin D receptor (VDR). Here, we report on the unexpected finding that stable knockdown of VDR expression in the human breast and prostate cancer cell lines, MDA-MB-231 and PC3, strongly induces cell apoptosis and inhibits cell proliferation in vitro. Implantation of these VDR knockdown cells into the mammary fat pad (MDA-MB-231), subcutaneously (PC3) or intra-tibially (both cell lines) in immune-incompetent nude mice resulted in reduced tumor growth associated with increased apoptosis and reduced cell proliferation compared with controls. These growth-retarding effects of VDR knockdown occur in the presence and absence of vitamin D and are independent of whether cells were grown in bone or soft tissues. Transcriptome analysis of VDR knockdown and non-target control cell lines demonstrated that loss of the VDR was associated with significant attenuation in the Wnt/0-catenin signaling pathway. In particular, cytoplasmic and nuclear β-catenin protein levels were reduced with a corresponding downregulation of downstream genes such as Axin2, Cyclin D1, interleukin-6 (IL-6), and IL-8. Stabilization of 0-catenin using the GSK-3β inhibitor BIO partly reversed the growth-retarding effects of VDR knockdown. Our results indicate that the unliganded VDR possesses hitherto unknown functions to promote breast and prostate cancer growth, which appear to be operational not only within but also outside the bone environment. These novel functions contrast with the known anti-proliferative nuclear actions of the liganded VDR and may represent targets for new diagnostic and therapeutic approaches in breast and prostate cancer.展开更多

Multi-spectroscopic techniques were used to investigate the binding mode of PAH to HSA and results revealed the presence of static type of quenching mechanism. The number of binding sites, binding constants and thermodynamic parameters were calculated. The results showed a spontaneous binding of PAH to HSA and hydrophobic interactions played a major role. In addition, the distance between PAH and the Trp–214 was estimated employing the F?rster's theory. Site marker competitive experiments indicated that the binding of PAH to HSA primarily took place in subdomain IIA(Sudlow's site I). The influence of interference of some common metal ions on the binding of PAH to HSA was studied. Synchronous fluorescence spectra(SFS), 3D fluorescence spectra and circular dichroism(CD) results indicated the conformational changes in the structure of HSA.展开更多

BACKGROUND Acute flaccid paralysis(AFP)and neurogenic respiratory failure rarely occur in children.At the end of 2018,some children with such symptoms were admitted to our hospital.In this study,we aimed to assess two...BACKGROUND Acute flaccid paralysis(AFP)and neurogenic respiratory failure rarely occur in children.At the end of 2018,some children with such symptoms were admitted to our hospital.In this study,we aimed to assess two children with AFP and neurogenic respiratory failure associated with enterovirus D68(EV-D68).CASE SUMMARY Two children admitted to our hospital presented with symptoms and imaging results different from those of acute disseminated encephalomyelitis and hand,foot,and mouth disease.Their main symptoms were AFP and neurogenic respiratory failure.Magnetic resonance imaging showed severe inflammatory injury mainly to the anterior horn cells of the spinal cord.Blood and cerebrospinal fluid samples were collected to assess for pathogens,including bacteria,tuberculosis,cryptococcus,herpes virus,and coxsackie virus,and the results were negative.At the beginning,the two cases were not assessed for EV-D68 in the nasopharyngeal,blood,and cerebrospinal fluid specimens.About 2 mo later,EVD68 was detected in the stool sample of one of the cases.The symptom of AFP was caused by injury to the anterior horn cells at levels C5-L5 of the spinal cord,while neurogenic respiratory failure was at levels C3-C5.CONCLUSION We should pay attention to the detection and diagnosis of EV-D68 and make efforts to develop antivirus drugs and vaccines.展开更多

文摘The human pose paradigm is estimated using a transformer-based multi-branch multidimensional directed the three-dimensional(3D)method that takes into account self-occlusion,badly posedness,and a lack of depth data in the per-frame 3D posture estimation from two-dimensional(2D)mapping to 3D mapping.Firstly,by examining the relationship between the movements of different bones in the human body,four virtual skeletons are proposed to enhance the cyclic constraints of limb joints.Then,multiple parameters describing the skeleton are fused and projected into a high-dimensional space.Utilizing a multi-branch network,motion features between bones and overall motion features are extracted to mitigate the drift error in the estimation results.Furthermore,the estimated relative depth is projected into 3D space,and the error is calculated against real 3D data,forming a loss function along with the relative depth error.This article adopts the average joint pixel error as the primary performance metric.Compared to the benchmark approach,the estimation findings indicate an increase in average precision of 1.8 mm within the Human3.6M sample.

文摘Human pose estimation is a basic and critical task in the field of computer vision that involves determining the position(or spatial coordinates)of the joints of the human body in a given image or video.It is widely used in motion analysis,medical evaluation,and behavior monitoring.In this paper,the authors propose a method for multi-view human pose estimation.Two image sensors were placed orthogonally with respect to each other to capture the pose of the subject as they moved,and this yielded accurate and comprehensive results of three-dimensional(3D)motion reconstruction that helped capture their multi-directional poses.Following this,we propose a method based on 3D pose estimation to assess the similarity of the features of motion of patients with motor dysfunction by comparing differences between their range of motion and that of normal subjects.We converted these differences into Fugl–Meyer assessment(FMA)scores in order to quantify them.Finally,we implemented the proposed method in the Unity framework,and built a Virtual Reality platform that provides users with human–computer interaction to make the task more enjoyable for them and ensure their active participation in the assessment process.The goal is to provide a suitable means of assessing movement disorders without requiring the immediate supervision of a physician.

文摘Background Intelligent garments,a burgeoning class of wearable devices,have extensive applications in domains such as sports training and medical rehabilitation.Nonetheless,existing research in the smart wearables domain predominantly emphasizes sensor functionality and quantity,often skipping crucial aspects related to user experience and interaction.Methods To address this gap,this study introduces a novel real-time 3D interactive system based on intelligent garments.The system utilizes lightweight sensor modules to collect human motion data and introduces a dual-stream fusion network based on pulsed neural units to classify and recognize human movements,thereby achieving real-time interaction between users and sensors.Additionally,the system incorporates 3D human visualization functionality,which visualizes sensor data and recognizes human actions as 3D models in real time,providing accurate and comprehensive visual feedback to help users better understand and analyze the details and features of human motion.This system has significant potential for applications in motion detection,medical monitoring,virtual reality,and other fields.The accurate classification of human actions contributes to the development of personalized training plans and injury prevention strategies.Conclusions This study has substantial implications in the domains of intelligent garments,human motion monitoring,and digital twin visualization.The advancement of this system is expected to propel the progress of wearable technology and foster a deeper comprehension of human motion.
文摘In recent years,wearable devices-based Human Activity Recognition(HAR)models have received significant attention.Previously developed HAR models use hand-crafted features to recognize human activities,leading to the extraction of basic features.The images captured by wearable sensors contain advanced features,allowing them to be analyzed by deep learning algorithms to enhance the detection and recognition of human actions.Poor lighting and limited sensor capabilities can impact data quality,making the recognition of human actions a challenging task.The unimodal-based HAR approaches are not suitable in a real-time environment.Therefore,an updated HAR model is developed using multiple types of data and an advanced deep-learning approach.Firstly,the required signals and sensor data are accumulated from the standard databases.From these signals,the wave features are retrieved.Then the extracted wave features and sensor data are given as the input to recognize the human activity.An Adaptive Hybrid Deep Attentive Network(AHDAN)is developed by incorporating a“1D Convolutional Neural Network(1DCNN)”with a“Gated Recurrent Unit(GRU)”for the human activity recognition process.Additionally,the Enhanced Archerfish Hunting Optimizer(EAHO)is suggested to fine-tune the network parameters for enhancing the recognition process.An experimental evaluation is performed on various deep learning networks and heuristic algorithms to confirm the effectiveness of the proposed HAR model.The EAHO-based HAR model outperforms traditional deep learning networks with an accuracy of 95.36,95.25 for recall,95.48 for specificity,and 95.47 for precision,respectively.The result proved that the developed model is effective in recognizing human action by taking less time.Additionally,it reduces the computation complexity and overfitting issue through using an optimization approach.

文摘3D human pose estimation is a major focus area in the field of computer vision,which plays an important role in practical applications.This article summarizes the framework and research progress related to the estimation of monocular RGB images and videos.An overall perspective ofmethods integrated with deep learning is introduced.Novel image-based and video-based inputs are proposed as the analysis framework.From this viewpoint,common problems are discussed.The diversity of human postures usually leads to problems such as occlusion and ambiguity,and the lack of training datasets often results in poor generalization ability of the model.Regression methods are crucial for solving such problems.Considering image-based input,the multi-view method is commonly used to solve occlusion problems.Here,the multi-view method is analyzed comprehensively.By referring to video-based input,the human prior knowledge of restricted motion is used to predict human postures.In addition,structural constraints are widely used as prior knowledge.Furthermore,weakly supervised learningmethods are studied and discussed for these two types of inputs to improve the model generalization ability.The problem of insufficient training datasets must also be considered,especially because 3D datasets are usually biased and limited.Finally,emerging and popular datasets and evaluation indicators are discussed.The characteristics of the datasets and the relationships of the indicators are explained and highlighted.Thus,this article can be useful and instructive for researchers who are lacking in experience and find this field confusing.In addition,by providing an overview of 3D human pose estimation,this article sorts and refines recent studies on 3D human pose estimation.It describes kernel problems and common useful methods,and discusses the scope for further research.

文摘With the advancement of image sensing technology, estimating 3Dhuman pose frommonocular video has becomea hot research topic in computer vision. 3D human pose estimation is an essential prerequisite for subsequentaction analysis and understanding. It empowers a wide spectrum of potential applications in various areas, suchas intelligent transportation, human-computer interaction, and medical rehabilitation. Currently, some methodsfor 3D human pose estimation in monocular video employ temporal convolutional network (TCN) to extractinter-frame feature relationships, but the majority of them suffer from insufficient inter-frame feature relationshipextractions. In this paper, we decompose the 3D joint location regression into the bone direction and length, wepropose the TCG, a temporal convolutional network incorporating Gaussian error linear units (GELU), to solvebone direction. It enablesmore inter-frame features to be captured andmakes the utmost of the feature relationshipsbetween data. Furthermore, we adopt kinematic structural information to solve bone length enhancing the use ofintra-frame joint features. Finally, we design a loss function for joint training of the bone direction estimationnetwork with the bone length estimation network. The proposed method has extensively experimented on thepublic benchmark dataset Human3.6M. Both quantitative and qualitative experimental results showed that theproposed method can achieve more accurate 3D human pose estimations.
文摘Human enterovirus 71(HEV71) is the cause of hand,foot and mouth disease and associated neurological complications in children under five years of age.There has been an increase in HEV71 epidemic activity throughout the Asia-Pacific region in the past decade,and it is predicted to replace poliovirus as the extant neurotropic enterovirus of highest global public health significance. To date there is no effective antiviral treatment and no vaccine is available to prevent HEV71 infection. The increase in prevalence, virulence and geographic spread of HEV71 infection over the past decade provides increasing incentive for the development of new therapeutic and prevention strategies against this emerging viral infection. The current review focuses on the potential, advantages and disadvantages of these strategies. Since the explosion of outbreaks leading to large epidemics in China, research in natural therapeutic products has identified several groups of compounds with anti-HEV71 activities. Concurrently, the search for effective synthetic antivirals has produced promising results. Other therapeutic strategies including immunotherapy and the use of oligonucleotides have also been explored. A sound prevention strategy is crucial in order to control the spread of HEV71. To this end the ultimate goal is the rapid development, regulatory approval and widespread implementation of a safe and effective vaccine. The various forms of HEV71 vaccine designs are highlighted in this review. Given the rapid progress of research in this area, eradication of the virus is likely to be achieved.

文摘Objective To develop RT-nPCR assays for amplifying partial and complete VP1 genes of human enteroviruses(HEVs)from clinical samples and to contribute to etiological surveillance of HEV-related diseases.Methods A panel of RT-nPCR assays,consisting of published combined primer pairs for VP1 genes of HEV A–C and in-house designed primers for HEV-D,was established in this study.The sensitivity of each RT-nPCR assay was evaluated with serially diluted virus stocks of five serotypes expressed as CCID50 perμL and copies perμL,and the newly established methods were tested in clinical specimens collected in recent years.Results The sensitivity of RT-nPCR assays for amplifying partial VP1 gene of HEVs was 0.1 CCID50 perμL and 10 virus copies perμL,and for the complete VP1 gene was 1 CCID50 perμL and 100 virus copies perμL,using serially-diluted virus stocks of five serotypes.As a proof-of-concept,25 serotypes were identified and complete VP1 sequences of 23 serotypes were obtained by this system among 858 clinical specimens positive for HEVs during the past eight surveillance seasons.Conclusion This RT-nPCR system is capable of amplifying the partial and complete VP1 gene of HEV A–D,providing rapid,sensitive,and reliable options for molecular typing and molecular epidemiology of HEVs in clinical specimens.
文摘A sensitive reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay for human enterovirus 71 (EV71) and Coxsackievirus A16 (CVA16) infection was further evaluated. The one step reaction was performed in a single tube at 65?C for 45 min for EV71 and 35 min for CVA16. The detection limits of RT-LAMP assays for both EV71 and CVA16 were 0.1 of a 50% tissue culture infective dose (TCID50) per reaction, based on 10—Fold dilutions of a titrated EV71 or CVA16 strain. The specific assay showed there were no cross-reactions with Coxsackievirus A (CVA) viruses (CVA 2, 4, 5, 7, 9, 10, 14, and 25), Coxsackievirus B (CVB) viruses (CVB 1, 2, 3, 4, and 5) or ECHO viruses (ECHO 3, 6, 11, and 19). In parallel with commercial quantitative real-time polymerase chain reaction (qRT-PCR) diagnostic kits for EV71 and CVA16, the RT-LAMP assay was evaluated with 515 clinical specimens, the results showed the RT-LAMP assay and the qRT-PCR assay were in complete agreement for 513/515 (99.6%) of the specimens. Two samples with discrepant results from two methods were further verified by nested reverse transcription polymerase chain reaction (nRT-PCR) assay and sequencing to be true positives for CVA16. In conclusion, RT-LAMP assay is demonstrated to be a sensitive and specific assay and have a great potential for the rapid and visual screening of EV71 and CVA16 in China, especially in those resource-limited hospitals and rural clinics of provincial and municipal regions.
文摘Human enterovirus 71 viruses have been long circulating throughout the world. In this study, we performed a positive selection analysis of the VP1 genes of capsid proteins from Enterovirus 71 viruses. Our results showed that although most sites were under negative or neutral evolution, four positions of the VP1 genes were under positive selection pressure. This might account for the spread and frequent outbreaks of the viruses and the enhanced neurovirulence. In particular, position 98 might be involved in neutralizing antibodies, modulating the virus-receptor interaction and enhancing the virulence of the viruses. Moreover, both positions 145 and 241 might correlate to determine the receptor specificity. However, these positions did not display much difference in amino acid polymorphism. In addition, no position in the VP1 genes of viruses isolated from China was under positive selection.

文摘In this article,a comprehensive survey of deep learning-based(DLbased)human pose estimation(HPE)that can help researchers in the domain of computer vision is presented.HPE is among the fastest-growing research domains of computer vision and is used in solving several problems for human endeavours.After the detailed introduction,three different human body modes followed by the main stages of HPE and two pipelines of twodimensional(2D)HPE are presented.The details of the four components of HPE are also presented.The keypoints output format of two popular 2D HPE datasets and the most cited DL-based HPE articles from the year of breakthrough are both shown in tabular form.This study intends to highlight the limitations of published reviews and surveys respecting presenting a systematic review of the current DL-based solution to the 2D HPE model.Furthermore,a detailed and meaningful survey that will guide new and existing researchers on DL-based 2D HPE models is achieved.Finally,some future research directions in the field of HPE,such as limited data on disabled persons and multi-training DL-based models,are revealed to encourage researchers and promote the growth of HPE research.
文摘Rabies is an infectious disease that almost always leads to death as soon as the first signs appear, and is still endemic in several regions, including Côte d’Ivoire. Epidemiological investigations are carried out and response measures implemented in the event of any death from human rabies. However, the quality of these investigations does not always allow the adoption of response measures. The aim of this work was to evaluate the reports of epidemiological investigations and response actions carried out during the occurrence of human rabies deaths from 2016 to 2018. This was a cross-sectional study that took place from April to September 2021 at the National Institute of Public Hygiene and focused on human rabies deaths notified from 206 to 2018. Of sixty-one (61) human rabies deaths recorded, sixty (60) were investigated, among which fifty-nine (59) deaths had been subject to response. Most deaths occurred in rural areas (75%), and the main vector was the dog (98%). Several shortcomings were noted: response actions were not carried out promptly;joint interventions bringing together human and animal health professionals were rare, there was little communication about rabies to communities (36%), and very little vaccination of the vector, the source of the disease (12%);an almost total absence of capacity-building for local players (7%);the health districts of Abobo-Ouest, Bondoukou, Divo and Fresco, which had regularly recorded human rabies deaths, had not benefited from dog vaccination, which is supposed to break the circulation of the virus. The persistence of rabies deaths in the health districts could probably be linked to these shortcomings. Faced with these realities, it is essential to strengthen the epidemiological investigation capacities of the players involved, in order to improve the quality of investigations and ensure an effective response.

文摘The effects of vitamin D on osteoblast mineralization are well documented. Reports of the effects of vitamin D on osteoclasts, however, are conflicting, showing both inhibition and stimulation. Finding that resorbing osteoclasts in human bone express vitamin D receptor (VDR), we examined their response to different concentrations of 25-hydroxy vitamin D3 [25(OH)D3] (100 or 500 nmol·L^-1) and 1,25-dihydroxy vitamin D3 [1,25(OH)2D3] (0.1 or 0.5 nmol·L^-1) metabolites in cell cultures. Specifically, CD14+ monocytes were cultured in charcoal-stripped serum in the presence of receptor activator of nuclear factor kappa-B ligand (RANKL) and macrophage colony-stimulating factor (M-CSF). Tartrate-resistant acid phosphatase (TRAP) histochemical staining assays and dentine resorption analysis were used to identify the size and number of osteoclast cells, number of nuclei per cell and resorption activity. The expression of VDR was detected in human bone tissue (ex vivo) by immunohistochemistry and in vitro cell cultures by western blotting. Quantitative reverse transcription-PCR (qRT-PCR) was used to determine the level of expression of vitamin D-related genes in response to vitamin D metabolites. VDR-related genes during osteoclastogenesis, shown by qRT-PCR, was stimulated in response to 500 nmol·L^-1 of 25(OH)D3 and 0.1-0.5 nmol·L^-1 of 1,25(OH)2D3, upregulating cytochrome P450 family 27 subfamily B member I (CYP27B1) and cytochrome P450 family 24 subfamily A member I (CYP24A1). Osteoclast fusion transcripts transmembrane 7 subfamily member 4 (tm7sf4) and nuclear factor of activated T-cell cytoplasmic 1 (nfatcl) where downregulated in response to vitamin D metabolites. Osteoclast number and resorption activity were also increased. Both 25(OH)D3 and 1,25(OH)2D3 reduced osteoclast size and number when co-treated with RANKL and M-CSF. The evidence for VDR expression in resorbing osteoclasts in vivo and low-dose effects of 1,25(OH)2D3 on osteoclasts in vitro may therefore provide insight into the effects of clinical vitamin D treatments, further providing a counterpoint to the high-dose effects reported from in vitro experiments.

文摘A retrospective surveillance study on enterovirus D68 was performed in Beijing, China, following the largest and most widespread EV-D68 infection, which occurred in the USA. From January 2011 to July 2015, EV-D68 was identified in 12 individuals with respiratory infections in Beijing, China. The phylogenetic relationships based on the genomic sequence alignment showed that there were two lineages circulating in Beijing from 2011 to 2015. Eight EV-D68 strains belonged to group 1 and four belonged to group 3. All EV-D68 strains from Beijing in 2014 were separately clustered into subgroup II of group 1. Based on these results, we concluded that the Beijing EV-D68 strains had little association with the EV-D68 strains circulating in the 2014 USA outbreak.

文摘Vitamin D co-regulates cell proliferation, differentiation and apoptosis in numerous tissues, including cancers. The known anti-proliferative and pro-apoptotic actions of the active metabolite of vitamin D, 1,25-dihydroxy-vitamin D [1,25(OH)2D] are mediated through binding to the vitamin D receptor (VDR). Here, we report on the unexpected finding that stable knockdown of VDR expression in the human breast and prostate cancer cell lines, MDA-MB-231 and PC3, strongly induces cell apoptosis and inhibits cell proliferation in vitro. Implantation of these VDR knockdown cells into the mammary fat pad (MDA-MB-231), subcutaneously (PC3) or intra-tibially (both cell lines) in immune-incompetent nude mice resulted in reduced tumor growth associated with increased apoptosis and reduced cell proliferation compared with controls. These growth-retarding effects of VDR knockdown occur in the presence and absence of vitamin D and are independent of whether cells were grown in bone or soft tissues. Transcriptome analysis of VDR knockdown and non-target control cell lines demonstrated that loss of the VDR was associated with significant attenuation in the Wnt/0-catenin signaling pathway. In particular, cytoplasmic and nuclear β-catenin protein levels were reduced with a corresponding downregulation of downstream genes such as Axin2, Cyclin D1, interleukin-6 (IL-6), and IL-8. Stabilization of 0-catenin using the GSK-3β inhibitor BIO partly reversed the growth-retarding effects of VDR knockdown. Our results indicate that the unliganded VDR possesses hitherto unknown functions to promote breast and prostate cancer growth, which appear to be operational not only within but also outside the bone environment. These novel functions contrast with the known anti-proliferative nuclear actions of the liganded VDR and may represent targets for new diagnostic and therapeutic approaches in breast and prostate cancer.
文摘Interaction of procainamide hydrochloride(PAH) with human serum albumin(HSA) is of great significance in understanding the pharmacokinetic and pharmacodynamic mechanisms of the drug. Multi-spectroscopic techniques were used to investigate the binding mode of PAH to HSA and results revealed the presence of static type of quenching mechanism. The number of binding sites, binding constants and thermodynamic parameters were calculated. The results showed a spontaneous binding of PAH to HSA and hydrophobic interactions played a major role. In addition, the distance between PAH and the Trp–214 was estimated employing the F?rster's theory. Site marker competitive experiments indicated that the binding of PAH to HSA primarily took place in subdomain IIA(Sudlow's site I). The influence of interference of some common metal ions on the binding of PAH to HSA was studied. Synchronous fluorescence spectra(SFS), 3D fluorescence spectra and circular dichroism(CD) results indicated the conformational changes in the structure of HSA.
文摘In this paper, a 3-D video encoding scheme suitable for digital TV/HDTV (high definition television) is studied through computer simulation. The encoding scheme is designed to provide a good match to human vision. Basically, this involves transmission of low frequency luminance information at full frame rate for good motion rendition and transmission of high frequency luminance signal at reduced frame rate for good detail in static images.

文摘BACKGROUND Acute flaccid paralysis(AFP)and neurogenic respiratory failure rarely occur in children.At the end of 2018,some children with such symptoms were admitted to our hospital.In this study,we aimed to assess two children with AFP and neurogenic respiratory failure associated with enterovirus D68(EV-D68).CASE SUMMARY Two children admitted to our hospital presented with symptoms and imaging results different from those of acute disseminated encephalomyelitis and hand,foot,and mouth disease.Their main symptoms were AFP and neurogenic respiratory failure.Magnetic resonance imaging showed severe inflammatory injury mainly to the anterior horn cells of the spinal cord.Blood and cerebrospinal fluid samples were collected to assess for pathogens,including bacteria,tuberculosis,cryptococcus,herpes virus,and coxsackie virus,and the results were negative.At the beginning,the two cases were not assessed for EV-D68 in the nasopharyngeal,blood,and cerebrospinal fluid specimens.About 2 mo later,EVD68 was detected in the stool sample of one of the cases.The symptom of AFP was caused by injury to the anterior horn cells at levels C5-L5 of the spinal cord,while neurogenic respiratory failure was at levels C3-C5.CONCLUSION We should pay attention to the detection and diagnosis of EV-D68 and make efforts to develop antivirus drugs and vaccines.
