BACKGROUND Diabetes is known damage the liver and kidney,leading to hepatic dysfunction and kidney failure.Honey is believed to help in lowering the blood glucose levels of diabetic patients and reducing diabetic comp...BACKGROUND Diabetes is known damage the liver and kidney,leading to hepatic dysfunction and kidney failure.Honey is believed to help in lowering the blood glucose levels of diabetic patients and reducing diabetic complications.However,the effect of stingless bee honey(SBH)administration in relieving liver and kidney damage in diabetes has not been well-studied.AIM To investigate the effect of SBH administration on the kidney and liver of streptozotocin-induced(STZ;55 mg/kg)diabetic Sprague Dawley rats.METHODS The rats were grouped as follows(n=6 per group):non-diabetic(ND),untreated diabetic(UNT),metformin-treated(MET),and SBH+metformin-treated(SBME)groups.After successful diabetic induction,ND and UNT rats were given normal saline,whereas the treatment groups received SBH(2.0 g/kg and/or metformin(250 mg/kg)for 12 d.Serum biochemical parameters and histological changes using hematoxylin and eosin(H&E)and periodic acid–Schiff(PAS)staining were evaluated.RESULTS On H&E and PAS staining,the ND group showed normal architecture and cellularity of Bowman’s capsule and tubules,whereas the UNT and MET groups had an increased glomerular cellularity and thickened basement membrane.The SBH-treated group showed a decrease in hydropic changes and mild cellularity of the glomerulus vs the ND group based on H&E staining,but the two were similar on PAS staining.Likewise,the SBME-treated group had an increase in cellularity of the glomerulus on H&E staining,but it was comparable to the SBH and ND groups on PAS staining.UNT diabetic rats had tubular hydropic tubules,which were smaller than other groups.Reduced fatty vacuole formation and dilated blood sinusoids in liver tissue were seen in the SBH group.Conversely,the UNT group had high glucose levels,which subsequently increased MDA levels,ultimately leading to liver damage.SBH treatment reduced this damage,as evidenced by having the lowest fasting glucose,serum alanine transaminase,aspartate transaminase,and alkaline phosphatase levels compared to other groups,although the levels of liver enzymes were not statistically significant.CONCLUSION The cellularity of the Bowman’s capsule,as well as histological alteration of kidney tubules,glomerular membranes,and liver tissues in diabetic rats after oral SBH resembled those of ND rats.Therefore,SBH exhibited a protective hepatorenal effect in a diabetic rat model.展开更多
BACKGROUND In recent years,studies have found that the occurrence and development of diabetic cardiomyopathy(DCM)is closely related to an increase in polyadenosine diphosphate-ribose polymerase-1(PARP-1)activity.PARP-...BACKGROUND In recent years,studies have found that the occurrence and development of diabetic cardiomyopathy(DCM)is closely related to an increase in polyadenosine diphosphate-ribose polymerase-1(PARP-1)activity.PARP-1 activation could be involved in the pathophysiological process of DCM by promoting oxidative stress,the inflammatory response,apoptosis and myocardial fibrosis.AIM To investigate the mechanism of liraglutide in improving myocardial injury in type 2 diabetic rats,further clarified the protective effect of liraglutide on the heart,and provided a new option for the treatment of DCM.METHODS Forty healthy male SD rats aged 6 wk were randomly divided into two groups,a normal control group(n=10)and a model group(n=30),which were fed an ordinary diet and a high-sugar and high-fat diet,respectively.After successful modeling,the rats in the model group were fed a high-glucose and high-fat diet for 4 wk and randomly divided into a model group and an intervention group(further divided into a high-dose group and a low-dose group).The rats were fed a high-glucose and high-fat diet for 8 wk and then started drug intervention.Blood samples were collected from the abdominal aorta to detect fasting blood glucose and lipid profiles.Intact heart tissue was dissected,and its weight was used to calculate the heart weight index.Haematoxylin and eosin staining was used to observe the pathological changes in the myocardium and the expression of PARP-1 in the heart by immunohistochemistry.RESULTS The body weight and heart weight index of rats in the model group were significantly increased compared with those in the normal control group,and those in the intervention group were decreased compared with those in the model group,with a more obvious decrease observed in the high-dose group(P<0.05).In the model group,myocardial fibers were disordered,and inflammatory cells and interstitial fibrosis were observed.The cardiomyopathy of rats in the intervention group was improved to different degrees,the myocardial fibers were arranged neatly,and the myocardial cells were clearly striated;the improvement was more obvious in the high-dose group.Compared with the normal control group,the expression of PARP-1 in myocardial tissue of the model group was increased,and the difference was statistically significant(P<0.05).After liraglutide intervention,compared with the model group,the expression of PARP-1 in myocardial tissue was decreased,and the reduction was more obvious in the high-dose group(P<0.05)but still higher than that in the normal control group.CONCLUSION Liraglutide may improve myocardial injury in type 2 diabetic rats by inhibiting the expression of myocardial PARP-1 in a dose-dependent manner.展开更多

[Method] With ovaries of Drosophila melanogaster as materials,after fixation with Bouin's fluid,the ovaries were sectioned and stained with HE,haematoxylin or eosin,respectively. The specimens were observed and photographed under fluorescent microscope in bright field,and fluorescent fields after excitation with green,blue and UV light,respectively. [Result] After staining by three methods,germ cells and somatic cells emitted different colors of fluorescence after excitation by different lights; lipids,nucleic acids and proteins in cells could also emit their special fluorescence. [Conclusion] Conventional dyes could give different fluorescence characteristics to germ cells and somatic cells,which can also give special fluorescence characteristics to different cellular components. Thus,the fluorescence histology will provide broad prospect for more convenient study on different cell types and cellular components.展开更多
Cytomegalovirus(CMV) infection is common in humans.The virus then enters a "latency phase" and can reactivate to different stimuli such as immunosuppression.The clinical significance of CMV infection in infl...Cytomegalovirus(CMV) infection is common in humans.The virus then enters a "latency phase" and can reactivate to different stimuli such as immunosuppression.The clinical significance of CMV infection in inflammatory bowel disease is different in Crohn's disease(CD) and ulcerative colitis(UC).CMV does not interfere in the clinical course of CD.However,CMV reactivation is frequent in severe or steroid-resistant UC.It is not known whether the virus exacerbates the disease or simply appears as a bystander of a severe disease.Different methods are used to diagnose CMV colitis.Diagnosis is classically based on histopathological identification of viral-infected cells or CMV antigens in biopsied tissues using haematoxylin-eosin or immunohistochemistry,other tests on blood or tissue samples are currently being investigated.Polymerase chain reaction performed in colonic mucosa has a high sensitivity and a positive result could be associated with a worse prognosis disease;further studies are needed to determine the most appropriate strategy with positive CMV-DNA in colonic mucosa.Specific endoscopic features have not been described in active UC and CMV infection.CMV colitis is usually treated with ganciclovir for several weeks,there are different opinions about whether or not to stop immunosuppressive therapy.Other antiviral drugs may be used.Multicenter controlled studies would needed to determine which subgroup of UC patients would benefit from early antiviral treatment.展开更多

Two adjacent tissue sections were separated subjected to histopathological examination by hematoxylin and eosin staining or THz transmission examination, and THz images were compared with pathologically mapped images. We determined the typical tumor and normal liver tissue regions by pathological examination; the corresponding areas of adjacent sections were examined by THz transmission.RESULTS The transmission rate of HCC tissue was 0.15-0.25, and the transmission rate of typical HCC tissue was about 0.2. THz transmittance in normal liver tissue is slightly higher than 0.4, but there were many influencing factors, including the degree of liver cirrhosis, fat components, ice crystals in frozen sections, and apoptosis.CONCLUSION In conclusion, this study shows that THz imaging can detect HCC tissue. Further research will yield more detailed data of the THz transmission rates of HCC tissue with different degrees of differentiation.展开更多

Experimental data on optical density of blank solutions of different concentrations ranging from 10 to 100 mg/L and optical density of solutions after adsorption on activated carbon samples were taken and analyzed. Calibration curves were plotted and the amount of dye adsorbed was calculated. The data was fitted to Langmuir and Freundlich isotherms for two different carbon samples and different concentration values. Constants were calculated from the slope and intercept values of the isotherms. Coefficient of correlation R2 and Standard Deviation SD were also noted. The data fitted well to the iso- therms. It was observed that adsorption decreased with increase in ppm concentrations. Carbon sample C2 showed higher potential to adsorb the dye Eosin as compared to carbon sample C1. Further Carbon sample C2 showed better adsorption in acidic pH as compared to in alkaline pH. From the analysis of the data it is shown that C2 sample has a good capacity to remove the textile dye from the residue water.展开更多

Cascading events after injury can lead to cell death. Heat shock protein 70 and other endogenous injury-signaling molecules are released by damaged cells, which can lead to systemic stress reactions. Protecting the brain through repair begins with the stress-injury-repair signaling chain. This study aimed to verify whether acupuncture acts through this chain to facilitate effective treatment of ischemic stroke. Rat models of cerebral ischemia/reperfusion injury were established by Zea Longa's method, and injury sites were identified by assessing neurological function, 2,3,5-triphenyltetrazolium chloride staining, and hematoxylin-eosin staining. Electroacupuncture at acupoints Baihui(DU20) and Zusanli(ST36) was performed in the model rats with dilatational waves, delivered for 20 minutes a day at 2–100 Hz and an amplitude of 2 m A. We analyzed the blood serum from the rats and found that inflammatory cytokines affected the levels of adrenotrophin and heat shock protein 70, each of which followed a similar bimodal curve. Specifically, electroacupuncture lowered the peak levels of adrenocorticotrophic hormone and heat shock protein 70. Thus, electroacupuncture was able to inhibit excessive stress, reduce inflammation, and promote the repair of neurons, which facilitated healing of ischemic stroke.展开更多

The effects of various parameters on the removal efficiency were studied. It was found that the eosin degradation could be raised considerably by increasing the applied voltage and the initial concentration, or by decreasing pH of the aqueous solution. Fe^2+ ion had an evident accelerating effect on the eosin degradation. The degradation process of eosin obeyed a pseudo-first-order reaction. The relationship between the degradation rate constant k and the reaction temperature T could be expressed by Arrhenius equation with which the apparent activation energy Ea of 14.110 kJ· mol^-1 and the pre-exponential factor k0 of 2.065× 10^-1 min^-1 were obtained, too. The determination of hydroxyl radical was carried out by using N, N-dimethyl -p-nitrosoaniline (RNO) as a scavenger. The results showed that the hydroxyl radical plays an important role in the degradation process.展开更多

In initial studies, however, a soft catheter type endocytoscope was used, while only methylene blue dye was used for the staining of Barrett's mucosa. Integrated type endocytoscopes (GIF-Q260 EC, Olympus Corp, Tokyo, Japan) have been recently developed, with the incorporation of a high-power magnifying endocytoscope into a standard endoscope together with narrow-band imaging (NBI). Moreover, double staining with a mixture of 0.05% crystal violet and 0.1% of methylene blue (CM) during ECS enables higher quality images comparable to conventional hematoxylin eosin histopathological images.RESULTS: In vivo endocytoscopic visualization of papillary squamous cell islands within glandular Barrett's epithelium in a patient with long-segment Barrett's esophagus is reported. Conventional white light endoscopy showed typical long-segment Barrett's esophagus, with small squamous cell islands within normal Barrett's mucosa, which were better visualized by NBI endoscopy. ECS after double CM staining showed regular Barrett's esophagus, while higher magnification (×480) revealed the orifices of glandular structures better. Furthermore, typical squamous cell papillary protrusion, classified as endocytoscopic atypia classification (ECA) 2 according to ECA, was identified within regular glandular Barrett's mucosa. Histological examination of biopsies taken from the same area showed squamous epithelium within glandular Barrett's mucosa, corresponding well to endocytoscopic findings. CONCLUSION: To our knowledge, this is the first report of in vivo visualization of esophageal papillary squamous cell islands surrounded by glandular Barrett's epithelium.展开更多

Diffuse reflecta...The role of oxygen and the generation of active radicals in the photocatalitic degradation of phenol were investigated using the eosin sensitized TiO2 as photocatalyst under visible light irradiation. Diffuse reflectance spectra show that the absorbancy range of eosin/TiO2 is expanded from 378 nm (TiO2 ) to about 600 nm. The photocatalitic degradation of phenol is almost stopped when the eosin/TiO2 system is saturated with N2 , which indicates the significance of O2 . The addition of NaN 3 (a quencher of single oxygen) causes about a 62% decrease in the phenol degradation. The phenol degradation ratio is dropped from 92% to 75% when the isopropanol (a quencher of hydroxyl radical) is present in the system. The experimental results show that there are singlet oxygen and hydroxyl radical generated in the eosin/TiO2 system under visible light irradiation. The changes of absorbancy indicate that the hydrogen peroxide might be produced. Through the analysis and comparison, it is found that the singlet oxygen is the predominant active radical for the degradation of phenol.展开更多

We report a case of CG from China.CASE SUMMARY A 24-year-old woman presented with recurrent abdominal distension and discomfort for 3 mo. Upper gastrointestinal endoscopy found diffuse nodular elevation-depression changes in the mucosa of the entire gastric corpus.Endoscopic ultrasound showed predominant involvement of the lamina propria and submucosa, and computed tomography imaging showed mild enhancement of the gastric wall. Pathological histology revealed that the thickness of the subepithelial collagen band was about 40 μm, and the Masson trichrome staining result was positive and the Congo red staining result was negative. This case is consistent with the child-adolescent type of CG.CONCLUSION Serum pepsinogen Ⅰ, pepsinogen Ⅱ, pepsinogen Ⅰ/Ⅱ ratio, and gastrin-17 may be potential non-invasive monitoring markers. Currently, treatments for CG vary,and the likely prognosis is unknown. Individual cases of gastric cancer in patients with CG have been reported.展开更多
Objective The successful establishment of a tumor cell bank is based on the premise that the target cells can be cultured by a legitimate approach.In this experiment,we used primary culture to select and detect breast...Objective The successful establishment of a tumor cell bank is based on the premise that the target cells can be cultured by a legitimate approach.In this experiment,we used primary culture to select and detect breast cancer cells in vitro,which can provide experimental ideas and methods for the establishment of a living tumor tissue cell bank.Methods Fifty-two specimens were collected over a two-year period from people with breast cancer who needed surgical treatment in our hospital.Cells were isolated and used to establish successful cell culture.Cell activity and cell purity were measured before liquid nitrogen cryopreservation.Results(1)At the initial culture stage,cells grew with adherence.Cell multiplication could be seen after the cell medium was exchanged three times.Cell viability was above 86%,while the viability of the target cells was above 75%,as detected by hematoxylin and eosin(HE)staining.(2)The number of breast cancer cells decreased,while the number of fibroblasts increased after five rounds of passage.(3)The success rate was 73.08%,which did not include polluted cells and those that were not successfully cryopreserved.Conclusion(1)breast cancer cells could be selected from primary culture in vitro through an appropriate method.(2)Exchange of the cell medium and further cell passage improved cell multiplication.(3)The experimental results could be monitored using trypan blue and HE staining.(4)The success of breast cancer cell culture in vitro could be used as a reference for other cell culture,so as to establish a tumor tissue cell bank.展开更多

Further understanding of the formation and progression of TAAs may provide novel, less invasive therapeutic strategies in patients with this devastating disease. Aneurysm formation is a complicated, dynamic process involving both cellular and extracellular processes. The mechanisms of TAA formation are poorly understood, mainly due to the lack of a useful and reproducible model. In 2003, Ikonomidis in South Carolina developed a murine model of TAA.The mouse model was less satisfactory because the small size of the murine aorta. The rat model has the advantage of larger aorta and the ability to do more research. Proper animal models are useful to understand the aetiology and evolution and they can be used to evaluate the efficacy of new treatments.Objective The goal of this study was to test the hypothesis that abluminal calcium chloride application could creat TAAs in the SD rats.MethOds Eight-week-old male Sprague-Dawley(SD) rats(n=60) were divided into two groups equally. All rats were anesthetized. An angiocath was then advanced through the mouth into the trachea under direct vision and connected to a rodent ventilator set at a respiratory rate of 180 breaths per minute. And their thoracic aortas were exposed via left thoracotomy. In the experiment group,a sponge soaked in 0.5 mol/L CaCl2 was placed on the distal half of the exposed descending thoracic aorta under direct vision. As control group, 0.5 mol/L CaCl2 was instead of 0.9% NaCl. After 15 min, the sponge was removed. Then the chest was closed. At 4,8,12 weeks, all animals undergoing fixation and aortic harvest were reanesthetized and the thorax was then opened beneath the xiphoid process. Potassium chloride was injected into the left ventricle under direct vision to arrest the heart. Then it was perfusion program and the entire animal was immersed in formalin. Seven days after fixation, the animal was removed from formalin, and the aorta was carefully harvested from its root to the renal arteries. The aorta was then immersed in an individually labeled jar containing a mixture of 80% methanol and 20% dimethyl sulfoxide (Dent’s solution).Diameter measurements were made using Axioplan 2 imaging system. Hematoxylin and eosin staining and Van Gieson and Verhoeff staining showed the elastic architecture and collagen structure. Immunohistochemical staining was performed to evaluate the presence of lymphocytes and macrophages. Results 12 weeks of CaCl2 treatment resulted in visually obvious dilation of the exposed distal descending thoracic aortic segment.The distal descending thoracic aortic diameter at 4,8,12 weeks was (0.82±0.13), (1.12±0.43), (1.65±0.58)μm respectively. The control group at 4,8,12 weeks was (0.81±0.04) , (0.94±0.12), (1.09±0.03)μm respectively. The diameter of the ascending aorta (used as an internal control) were not significantly different between groups. Hematoxylin and eosin stained slides showed disruption of the normal elastic lamellar architecture of the CaCl2-exposed aortic wall compared to the contralateral wall or untreated control. Van Gieson-stained sections showed decreased collagen content in the exposed region of aortic wall. Immunohistochemical study indicated significantly more CD3+cells and CD68+ cells in the aortas of CaCl2 treated groups than in control aortas. CD3+ cells were located in the media and surrounding the vasa vasorum in the adventitia. Conclusion We conclude that abluminal application of CaCl2 to the thoracic aorta reliably produces dilation, wall-thinning, and disruption of mural architecture, the hallmark signs of aneurysm formation. These results suggest that the rat is a useful and technically feasible model for the study of TAAs. To our knowledge, these findings describe for the first time the generation of a reproducible model of isolated TAA formation in SD rat.展开更多
AIM: To clarify the diagnostic values of hematoxylin and eosin (HE), D2-40, CD31, CD34, and HHV-8 immunohistochemical (IHC) staining in gastrointestinal Kaposi's sarcoma (GI-KS) in relation to endoscopic tumor sta...AIM: To clarify the diagnostic values of hematoxylin and eosin (HE), D2-40, CD31, CD34, and HHV-8 immunohistochemical (IHC) staining in gastrointestinal Kaposi's sarcoma (GI-KS) in relation to endoscopic tumor staging. METHODS: Biopsy samples (n = 133) from 41 human immunodeficiency virus-infected patients were reviewed. GI-KS was defined as histologically negative for other GI diseases and as a positive clinical response to KS therapy. The receiver operating characteristic area under the curve (ROC-AUC) was compared in relation to lesion size, GI location, and macroscopic appearances on endoscopy. RESULTS: GI-KS was confirmed in 84 lesions (81.6%). Other endoscopic findings were polyps (n = 9), inflammation (n = 4), malignant lymphoma (n = 4), and condyloma (n = 2), which mimicked GI-KS on endoscopy. ROC-AUC of HE, D2-40, blood vessel markers, and HHV-8 showed results of 0.83, 0.89, 0.80, and 0.82, respectively. For IHC staining, the ROC-AUC of D2-40 was significantly higher (P < 0.05) than that of HE staining only. In the analysis of endoscopic appearance, the ROC-AUC of HE and IHC showed a tendency toward an increase in tumor staging (e.g. , small to large, patches, and polypoid to SMT appearance). D2-40 was significantly (P < 0.05) advantageous in the upper GI tract and for polypoid appearance compared with HE staining. CONCLUSION: The diagnostic value of endothelial markers and HHV-8 staining was found to be high, and its accuracy tended to increase with endoscopic tumor staging. D2-40 will be useful for complementing HE staining in the diagnosis of GI-KS, especially in the upper GI tract and for polypoid appearance.展开更多

文摘BACKGROUND Diabetes is known damage the liver and kidney,leading to hepatic dysfunction and kidney failure.Honey is believed to help in lowering the blood glucose levels of diabetic patients and reducing diabetic complications.However,the effect of stingless bee honey(SBH)administration in relieving liver and kidney damage in diabetes has not been well-studied.AIM To investigate the effect of SBH administration on the kidney and liver of streptozotocin-induced(STZ;55 mg/kg)diabetic Sprague Dawley rats.METHODS The rats were grouped as follows(n=6 per group):non-diabetic(ND),untreated diabetic(UNT),metformin-treated(MET),and SBH+metformin-treated(SBME)groups.After successful diabetic induction,ND and UNT rats were given normal saline,whereas the treatment groups received SBH(2.0 g/kg and/or metformin(250 mg/kg)for 12 d.Serum biochemical parameters and histological changes using hematoxylin and eosin(H&E)and periodic acid–Schiff(PAS)staining were evaluated.RESULTS On H&E and PAS staining,the ND group showed normal architecture and cellularity of Bowman’s capsule and tubules,whereas the UNT and MET groups had an increased glomerular cellularity and thickened basement membrane.The SBH-treated group showed a decrease in hydropic changes and mild cellularity of the glomerulus vs the ND group based on H&E staining,but the two were similar on PAS staining.Likewise,the SBME-treated group had an increase in cellularity of the glomerulus on H&E staining,but it was comparable to the SBH and ND groups on PAS staining.UNT diabetic rats had tubular hydropic tubules,which were smaller than other groups.Reduced fatty vacuole formation and dilated blood sinusoids in liver tissue were seen in the SBH group.Conversely,the UNT group had high glucose levels,which subsequently increased MDA levels,ultimately leading to liver damage.SBH treatment reduced this damage,as evidenced by having the lowest fasting glucose,serum alanine transaminase,aspartate transaminase,and alkaline phosphatase levels compared to other groups,although the levels of liver enzymes were not statistically significant.CONCLUSION The cellularity of the Bowman’s capsule,as well as histological alteration of kidney tubules,glomerular membranes,and liver tissues in diabetic rats after oral SBH resembled those of ND rats.Therefore,SBH exhibited a protective hepatorenal effect in a diabetic rat model.

文摘BACKGROUND In recent years,studies have found that the occurrence and development of diabetic cardiomyopathy(DCM)is closely related to an increase in polyadenosine diphosphate-ribose polymerase-1(PARP-1)activity.PARP-1 activation could be involved in the pathophysiological process of DCM by promoting oxidative stress,the inflammatory response,apoptosis and myocardial fibrosis.AIM To investigate the mechanism of liraglutide in improving myocardial injury in type 2 diabetic rats,further clarified the protective effect of liraglutide on the heart,and provided a new option for the treatment of DCM.METHODS Forty healthy male SD rats aged 6 wk were randomly divided into two groups,a normal control group(n=10)and a model group(n=30),which were fed an ordinary diet and a high-sugar and high-fat diet,respectively.After successful modeling,the rats in the model group were fed a high-glucose and high-fat diet for 4 wk and randomly divided into a model group and an intervention group(further divided into a high-dose group and a low-dose group).The rats were fed a high-glucose and high-fat diet for 8 wk and then started drug intervention.Blood samples were collected from the abdominal aorta to detect fasting blood glucose and lipid profiles.Intact heart tissue was dissected,and its weight was used to calculate the heart weight index.Haematoxylin and eosin staining was used to observe the pathological changes in the myocardium and the expression of PARP-1 in the heart by immunohistochemistry.RESULTS The body weight and heart weight index of rats in the model group were significantly increased compared with those in the normal control group,and those in the intervention group were decreased compared with those in the model group,with a more obvious decrease observed in the high-dose group(P<0.05).In the model group,myocardial fibers were disordered,and inflammatory cells and interstitial fibrosis were observed.The cardiomyopathy of rats in the intervention group was improved to different degrees,the myocardial fibers were arranged neatly,and the myocardial cells were clearly striated;the improvement was more obvious in the high-dose group.Compared with the normal control group,the expression of PARP-1 in myocardial tissue of the model group was increased,and the difference was statistically significant(P<0.05).After liraglutide intervention,compared with the model group,the expression of PARP-1 in myocardial tissue was decreased,and the reduction was more obvious in the high-dose group(P<0.05)but still higher than that in the normal control group.CONCLUSION Liraglutide may improve myocardial injury in type 2 diabetic rats by inhibiting the expression of myocardial PARP-1 in a dose-dependent manner.

文摘[Objective] In order to provide firsthand information for the development of fluorescent histology,the paper studied the fluorescent features of ovarioles and germline cysts of fruit fly after common fixation and staining. [Method] With ovaries of Drosophila melanogaster as materials,after fixation with Bouin's fluid,the ovaries were sectioned and stained with HE,haematoxylin or eosin,respectively. The specimens were observed and photographed under fluorescent microscope in bright field,and fluorescent fields after excitation with green,blue and UV light,respectively. [Result] After staining by three methods,germ cells and somatic cells emitted different colors of fluorescence after excitation by different lights; lipids,nucleic acids and proteins in cells could also emit their special fluorescence. [Conclusion] Conventional dyes could give different fluorescence characteristics to germ cells and somatic cells,which can also give special fluorescence characteristics to different cellular components. Thus,the fluorescence histology will provide broad prospect for more convenient study on different cell types and cellular components.
文摘Cytomegalovirus(CMV) infection is common in humans.The virus then enters a "latency phase" and can reactivate to different stimuli such as immunosuppression.The clinical significance of CMV infection in inflammatory bowel disease is different in Crohn's disease(CD) and ulcerative colitis(UC).CMV does not interfere in the clinical course of CD.However,CMV reactivation is frequent in severe or steroid-resistant UC.It is not known whether the virus exacerbates the disease or simply appears as a bystander of a severe disease.Different methods are used to diagnose CMV colitis.Diagnosis is classically based on histopathological identification of viral-infected cells or CMV antigens in biopsied tissues using haematoxylin-eosin or immunohistochemistry,other tests on blood or tissue samples are currently being investigated.Polymerase chain reaction performed in colonic mucosa has a high sensitivity and a positive result could be associated with a worse prognosis disease;further studies are needed to determine the most appropriate strategy with positive CMV-DNA in colonic mucosa.Specific endoscopic features have not been described in active UC and CMV infection.CMV colitis is usually treated with ganciclovir for several weeks,there are different opinions about whether or not to stop immunosuppressive therapy.Other antiviral drugs may be used.Multicenter controlled studies would needed to determine which subgroup of UC patients would benefit from early antiviral treatment.

文摘BACKGROUND Hepatocellular carcinoma(HCC) is one of the most common malignant tumors worldwide, and novel methods for early/rapid diagnosis of HCC are needed.Terahertz(THz) spectroscopy is considered to have the potential to distinguish between normal liver tissue and HCC tissue; however, there are few reports on it.We conduct this observational study to explore the feasibility of THz imaging for the diagnosis of HCC.AIM To evaluate the feasibility of THz for discriminating between HCC and normal liver tissues using fresh tissue specimens obtained from HCC patients who had undergone surgery.METHODS Normal liver tissue and HCC tissue were cryosectioned into 50 μm-thick slicesand placed on cover glass. Two adjacent tissue sections were separated subjected to histopathological examination by hematoxylin and eosin staining or THz transmission examination, and THz images were compared with pathologically mapped images. We determined the typical tumor and normal liver tissue regions by pathological examination; the corresponding areas of adjacent sections were examined by THz transmission.RESULTS The transmission rate of HCC tissue was 0.15-0.25, and the transmission rate of typical HCC tissue was about 0.2. THz transmittance in normal liver tissue is slightly higher than 0.4, but there were many influencing factors, including the degree of liver cirrhosis, fat components, ice crystals in frozen sections, and apoptosis.CONCLUSION In conclusion, this study shows that THz imaging can detect HCC tissue. Further research will yield more detailed data of the THz transmission rates of HCC tissue with different degrees of differentiation.

文摘Removal of dye Eosin on two different samples of activated carbon by static batch method was studied. Experimental data on optical density of blank solutions of different concentrations ranging from 10 to 100 mg/L and optical density of solutions after adsorption on activated carbon samples were taken and analyzed. Calibration curves were plotted and the amount of dye adsorbed was calculated. The data was fitted to Langmuir and Freundlich isotherms for two different carbon samples and different concentration values. Constants were calculated from the slope and intercept values of the isotherms. Coefficient of correlation R2 and Standard Deviation SD were also noted. The data fitted well to the iso- therms. It was observed that adsorption decreased with increase in ppm concentrations. Carbon sample C2 showed higher potential to adsorb the dye Eosin as compared to carbon sample C1. Further Carbon sample C2 showed better adsorption in acidic pH as compared to in alkaline pH. From the analysis of the data it is shown that C2 sample has a good capacity to remove the textile dye from the residue water.

文摘Inflammation after stroke is the main cause of cerebral ischemia/reperfusion injury. Cascading events after injury can lead to cell death. Heat shock protein 70 and other endogenous injury-signaling molecules are released by damaged cells, which can lead to systemic stress reactions. Protecting the brain through repair begins with the stress-injury-repair signaling chain. This study aimed to verify whether acupuncture acts through this chain to facilitate effective treatment of ischemic stroke. Rat models of cerebral ischemia/reperfusion injury were established by Zea Longa's method, and injury sites were identified by assessing neurological function, 2,3,5-triphenyltetrazolium chloride staining, and hematoxylin-eosin staining. Electroacupuncture at acupoints Baihui(DU20) and Zusanli(ST36) was performed in the model rats with dilatational waves, delivered for 20 minutes a day at 2–100 Hz and an amplitude of 2 m A. We analyzed the blood serum from the rats and found that inflammatory cytokines affected the levels of adrenotrophin and heat shock protein 70, each of which followed a similar bimodal curve. Specifically, electroacupuncture lowered the peak levels of adrenocorticotrophic hormone and heat shock protein 70. Thus, electroacupuncture was able to inhibit excessive stress, reduce inflammation, and promote the repair of neurons, which facilitated healing of ischemic stroke.

文摘This paper describes a novel method for the degradation of eosin by using glow discharge electrolysis (GDE). The effects of various parameters on the removal efficiency were studied. It was found that the eosin degradation could be raised considerably by increasing the applied voltage and the initial concentration, or by decreasing pH of the aqueous solution. Fe^2+ ion had an evident accelerating effect on the eosin degradation. The degradation process of eosin obeyed a pseudo-first-order reaction. The relationship between the degradation rate constant k and the reaction temperature T could be expressed by Arrhenius equation with which the apparent activation energy Ea of 14.110 kJ· mol^-1 and the pre-exponential factor k0 of 2.065× 10^-1 min^-1 were obtained, too. The determination of hydroxyl radical was carried out by using N, N-dimethyl -p-nitrosoaniline (RNO) as a scavenger. The results showed that the hydroxyl radical plays an important role in the degradation process.
文摘AIM: To study the endocytoscopic visualization of squamous cell islands within Barrett's epithelium. METHODS: Endocytoscopy (ECS) has been studied in the surveillance of Barrett's esophagus, with controversial results. In initial studies, however, a soft catheter type endocytoscope was used, while only methylene blue dye was used for the staining of Barrett's mucosa. Integrated type endocytoscopes (GIF-Q260 EC, Olympus Corp, Tokyo, Japan) have been recently developed, with the incorporation of a high-power magnifying endocytoscope into a standard endoscope together with narrow-band imaging (NBI). Moreover, double staining with a mixture of 0.05% crystal violet and 0.1% of methylene blue (CM) during ECS enables higher quality images comparable to conventional hematoxylin eosin histopathological images.RESULTS: In vivo endocytoscopic visualization of papillary squamous cell islands within glandular Barrett's epithelium in a patient with long-segment Barrett's esophagus is reported. Conventional white light endoscopy showed typical long-segment Barrett's esophagus, with small squamous cell islands within normal Barrett's mucosa, which were better visualized by NBI endoscopy. ECS after double CM staining showed regular Barrett's esophagus, while higher magnification (×480) revealed the orifices of glandular structures better. Furthermore, typical squamous cell papillary protrusion, classified as endocytoscopic atypia classification (ECA) 2 according to ECA, was identified within regular glandular Barrett's mucosa. Histological examination of biopsies taken from the same area showed squamous epithelium within glandular Barrett's mucosa, corresponding well to endocytoscopic findings. CONCLUSION: To our knowledge, this is the first report of in vivo visualization of esophageal papillary squamous cell islands surrounded by glandular Barrett's epithelium.

文摘An efficient CuO‐modified zeolitic imidazolate framework‐9(ZIF‐9)photocatalyst is successfully prepared at room temperature under mild conditions.It is observed that the ZIF‐9/CuO photocatalyst is effective for H2generation under visible light with sacrificial agent conditions.When the CuO is introduced,the photocatalytic properties of ZIF‐9are greatly improved and when the content of CuO is40%,the photocatalytic activity reaches a maximum of78.74μmol after5h.This results from the200–300nm cube structure of ZIF‐9being able to adsorb more dye molecules and the CuO,which connects with ZIF‐9,greatly improving the electronic transmission efficiency.Moreover,the interaction between the dye molecule Eosin Y(EY)and the catalyst is also studied by transient fluorescence spectroscopy.A series of characterizations,such as SEM,TEM,XPS,XRD,UV‐vis,FTIR,transient fluorescence and photocurrent,are conducted,and the results are in good agreement with the experimental result.In addition,the possible reaction mechanism over EY‐sensitized ZIF‐9/CuO under visible light irradiation is proposed.

文摘The role of oxygen and the generation of active radicals in the photocatalitic degradation of phenol were investigated using the eosin sensitized TiO2 as photocatalyst under visible light irradiation. Diffuse reflectance spectra show that the absorbancy range of eosin/TiO2 is expanded from 378 nm (TiO2 ) to about 600 nm. The photocatalitic degradation of phenol is almost stopped when the eosin/TiO2 system is saturated with N2 , which indicates the significance of O2 . The addition of NaN 3 (a quencher of single oxygen) causes about a 62% decrease in the phenol degradation. The phenol degradation ratio is dropped from 92% to 75% when the isopropanol (a quencher of hydroxyl radical) is present in the system. The experimental results show that there are singlet oxygen and hydroxyl radical generated in the eosin/TiO2 system under visible light irradiation. The changes of absorbancy indicate that the hydrogen peroxide might be produced. Through the analysis and comparison, it is found that the singlet oxygen is the predominant active radical for the degradation of phenol.

文摘BACKGROUND Collagenous gastritis(CG) is a rare condition whose pathogenesis may be related to immune abnormalities. We report a case of CG from China.CASE SUMMARY A 24-year-old woman presented with recurrent abdominal distension and discomfort for 3 mo. Upper gastrointestinal endoscopy found diffuse nodular elevation-depression changes in the mucosa of the entire gastric corpus.Endoscopic ultrasound showed predominant involvement of the lamina propria and submucosa, and computed tomography imaging showed mild enhancement of the gastric wall. Pathological histology revealed that the thickness of the subepithelial collagen band was about 40 μm, and the Masson trichrome staining result was positive and the Congo red staining result was negative. This case is consistent with the child-adolescent type of CG.CONCLUSION Serum pepsinogen Ⅰ, pepsinogen Ⅱ, pepsinogen Ⅰ/Ⅱ ratio, and gastrin-17 may be potential non-invasive monitoring markers. Currently, treatments for CG vary,and the likely prognosis is unknown. Individual cases of gastric cancer in patients with CG have been reported.

文摘Objective The successful establishment of a tumor cell bank is based on the premise that the target cells can be cultured by a legitimate approach.In this experiment,we used primary culture to select and detect breast cancer cells in vitro,which can provide experimental ideas and methods for the establishment of a living tumor tissue cell bank.Methods Fifty-two specimens were collected over a two-year period from people with breast cancer who needed surgical treatment in our hospital.Cells were isolated and used to establish successful cell culture.Cell activity and cell purity were measured before liquid nitrogen cryopreservation.Results(1)At the initial culture stage,cells grew with adherence.Cell multiplication could be seen after the cell medium was exchanged three times.Cell viability was above 86%,while the viability of the target cells was above 75%,as detected by hematoxylin and eosin(HE)staining.(2)The number of breast cancer cells decreased,while the number of fibroblasts increased after five rounds of passage.(3)The success rate was 73.08%,which did not include polluted cells and those that were not successfully cryopreserved.Conclusion(1)breast cancer cells could be selected from primary culture in vitro through an appropriate method.(2)Exchange of the cell medium and further cell passage improved cell multiplication.(3)The experimental results could be monitored using trypan blue and HE staining.(4)The success of breast cancer cell culture in vitro could be used as a reference for other cell culture,so as to establish a tumor tissue cell bank.

文摘Backgroud Thoracic aortic aneurysm (TAA) disease is a serious condition with both high mortality and morbidity rates associated with surgical therapy. Further understanding of the formation and progression of TAAs may provide novel, less invasive therapeutic strategies in patients with this devastating disease. Aneurysm formation is a complicated, dynamic process involving both cellular and extracellular processes. The mechanisms of TAA formation are poorly understood, mainly due to the lack of a useful and reproducible model. In 2003, Ikonomidis in South Carolina developed a murine model of TAA.The mouse model was less satisfactory because the small size of the murine aorta. The rat model has the advantage of larger aorta and the ability to do more research. Proper animal models are useful to understand the aetiology and evolution and they can be used to evaluate the efficacy of new treatments.Objective The goal of this study was to test the hypothesis that abluminal calcium chloride application could creat TAAs in the SD rats.MethOds Eight-week-old male Sprague-Dawley(SD) rats(n=60) were divided into two groups equally. All rats were anesthetized. An angiocath was then advanced through the mouth into the trachea under direct vision and connected to a rodent ventilator set at a respiratory rate of 180 breaths per minute. And their thoracic aortas were exposed via left thoracotomy. In the experiment group,a sponge soaked in 0.5 mol/L CaCl2 was placed on the distal half of the exposed descending thoracic aorta under direct vision. As control group, 0.5 mol/L CaCl2 was instead of 0.9% NaCl. After 15 min, the sponge was removed. Then the chest was closed. At 4,8,12 weeks, all animals undergoing fixation and aortic harvest were reanesthetized and the thorax was then opened beneath the xiphoid process. Potassium chloride was injected into the left ventricle under direct vision to arrest the heart. Then it was perfusion program and the entire animal was immersed in formalin. Seven days after fixation, the animal was removed from formalin, and the aorta was carefully harvested from its root to the renal arteries. The aorta was then immersed in an individually labeled jar containing a mixture of 80% methanol and 20% dimethyl sulfoxide (Dent’s solution).Diameter measurements were made using Axioplan 2 imaging system. Hematoxylin and eosin staining and Van Gieson and Verhoeff staining showed the elastic architecture and collagen structure. Immunohistochemical staining was performed to evaluate the presence of lymphocytes and macrophages. Results 12 weeks of CaCl2 treatment resulted in visually obvious dilation of the exposed distal descending thoracic aortic segment.The distal descending thoracic aortic diameter at 4,8,12 weeks was (0.82±0.13), (1.12±0.43), (1.65±0.58)μm respectively. The control group at 4,8,12 weeks was (0.81±0.04) , (0.94±0.12), (1.09±0.03)μm respectively. The diameter of the ascending aorta (used as an internal control) were not significantly different between groups. Hematoxylin and eosin stained slides showed disruption of the normal elastic lamellar architecture of the CaCl2-exposed aortic wall compared to the contralateral wall or untreated control. Van Gieson-stained sections showed decreased collagen content in the exposed region of aortic wall. Immunohistochemical study indicated significantly more CD3+cells and CD68+ cells in the aortas of CaCl2 treated groups than in control aortas. CD3+ cells were located in the media and surrounding the vasa vasorum in the adventitia. Conclusion We conclude that abluminal application of CaCl2 to the thoracic aorta reliably produces dilation, wall-thinning, and disruption of mural architecture, the hallmark signs of aneurysm formation. These results suggest that the rat is a useful and technically feasible model for the study of TAAs. To our knowledge, these findings describe for the first time the generation of a reproducible model of isolated TAA formation in SD rat.

文摘AIM: To clarify the diagnostic values of hematoxylin and eosin (HE), D2-40, CD31, CD34, and HHV-8 immunohistochemical (IHC) staining in gastrointestinal Kaposi's sarcoma (GI-KS) in relation to endoscopic tumor staging. METHODS: Biopsy samples (n = 133) from 41 human immunodeficiency virus-infected patients were reviewed. GI-KS was defined as histologically negative for other GI diseases and as a positive clinical response to KS therapy. The receiver operating characteristic area under the curve (ROC-AUC) was compared in relation to lesion size, GI location, and macroscopic appearances on endoscopy. RESULTS: GI-KS was confirmed in 84 lesions (81.6%). Other endoscopic findings were polyps (n = 9), inflammation (n = 4), malignant lymphoma (n = 4), and condyloma (n = 2), which mimicked GI-KS on endoscopy. ROC-AUC of HE, D2-40, blood vessel markers, and HHV-8 showed results of 0.83, 0.89, 0.80, and 0.82, respectively. For IHC staining, the ROC-AUC of D2-40 was significantly higher (P < 0.05) than that of HE staining only. In the analysis of endoscopic appearance, the ROC-AUC of HE and IHC showed a tendency toward an increase in tumor staging (e.g. , small to large, patches, and polypoid to SMT appearance). D2-40 was significantly (P < 0.05) advantageous in the upper GI tract and for polypoid appearance compared with HE staining. CONCLUSION: The diagnostic value of endothelial markers and HHV-8 staining was found to be high, and its accuracy tended to increase with endoscopic tumor staging. D2-40 will be useful for complementing HE staining in the diagnosis of GI-KS, especially in the upper GI tract and for polypoid appearance.
