Combination therapy using evening primrose oil and electrical stimulation to improve nerve function following a crush injury of sciatic nerve in male rats 被引量:7

Peripheral nerve injuries with a poor prognosis are common.Evening primrose oil(EPO) has beneficial biological effects and immunomodulatory properties.Since electrical activity plays a major role in neural regeneration,the present study investigated the effects of electrical stimulation(ES),combined with evening primrose oil(EPO),on sciatic nerve function after a crush injury in rats.In anesthetized rats,the sciatic nerve was crushed using small haemostatic forceps followed by ES and/or EPO treatment for 4 weeks.Functional recovery of the sciatic nerve was assessed using the sciatic functional index.Histopathological changes of gastrocnemius muscle atrophy were investigated by light microscopy.Electrophysiological changes were assessed by the nerve conduction velocity of sciatic nerves.Immunohistochemistry was used to determine the remyelination of the sciatic nerve following the interventions.EPO + ES,EPO,and ES obviously improved sciatic nerve function assessed by the sciatic functional index and nerve conduction velocity of the sciatic nerve at 28 days after operation.Expression of the peripheral nerve remyelination marker,protein zero(P0),was increased in the treatment groups at 28 days after operation.Muscle atrophy severity was decreased significantly while the nerve conduction velocity was increased significantly in rats with sciatic nerve injury in the injury + EPO + ES group than in the EPO or ES group.Totally speaking,the combined use of EPO and ES may produce an improving effect on the function of sciatic nerves injured by a crush.The increased expression of P0 may have contributed to improving the functional effects of combination therapy with EPO and ES as well as the electrophysiological and histopathological features of the injured peripheral nerve. 展开更多

Nrf2 modulates the benefits of evening exercise in type 2 diabetes

Exercise has well-characterized therapeutic benefits in the management of type 2 diabetes mellitus(T2DM).Most of the beneficial effects of exercise arise from the impact of nuclear factor erythroid 2 related factor-2(Nrf2)activation of glucose metabolism.Nrf2 is an essential controller of cellular anti-oxidative capacity and circadian rhythms.The circadian rhythm of Nrf2 is influenced by circadian genes on its expression,where the timing of exercise effects the activation of Nrf2 and the rhythmicity of Nrf2 and signaling,such that the timing of exercise has differential physiological effects.Exercise in the evening has beneficial effects on diabetes management,such as lowering of blood glucose and weight.The mechanisms responsible for these effects have not yet been associated with the influence of exercise on the circadian rhythm of Nrf2 activity.A better understanding of exercise-induced Nrf2 activation on Nrf2 rhythm and signaling can improve our appreciation of the distinct effects of morning and evening exercise.This review hypothesizes that activation of Nrf2 by exercise in the morning,when Nrf2 level is already at high levels,leads to hyperactivation and decrease in Nrf2 signaling,while activation of Nrf2 in the evening,when Nrf2 levels are at nadir levels,improves Nrf2 signaling and lowers blood glucose levels and increases fatty acid oxidation.Exploring the effects of Nrf2 activators on rhythmic signaling could also provide valuable insights into the optimal timing of their application,while also holding promise for timed treatment of type 2 diabetes. 展开更多

Plants initiate leaf senescence to reallocate energy and nutrients from aging to developing tissues for optimizing growth fitness and reproduction at the end of the growing season or under stress. Jasmonate (JA), a lipid-derived phytohormone, is known as an important endogenous signal in inducing leaf senescence. However, whether and how the circadian clock gates JA signaling to induce leaf senescence in plants remains elusive. In this study, we show that Evening Complex (EC), a core component of the circadian oscillator, negatively regulates leaf senescence in Arabidopsis thaliana. Transcriptomic profiling analysis reveals that EC is closely involved in JA signaling and response, consistent with accelerated leaf senescence unanimously displayed by EC mutants upon JA induction. We found that EC directly binds the promoter of MYC2, which encodes a key activator of JA-induced leaf senescence, and represses its expression. Ge- netic analysis further demonstrated that the accelerated JA-induced leaf senescence in EC mutants is abrogated by myc2 myc3 myc4 triple mutation. Collectively, these results reveal a critical molecular mechanism illustrating how the core component of the circadian clock gates JA signaling to regulate leaf senescence. 展开更多

From the case study of Evening Bell Ringing at Nanping Hill,one of the West Lake Cultural Landscapes in Hangzhou,China,we investigated the soundscape of a scenic area with a profound cultural background.First,we conducted the soundscape physical index of the area in both winter and spring seasons to analyze its objective graphical expression.Second,we focused on people's reactions to the soundscape in order to obtain a subjective evaluation of each component in the soundscape and integrated environment.Then,the relationship between the objective data and the subjective evaluation was analyzed.Finally,the impacts of the natural environment,history,and cultural factors on the evaluation of the Jingci Temple soundscape were studied.It was found that natural sounds,cultural sounds,and historic sounds were widely acclaimed in people's subjective feelings,which indicated the close relationships among historical and cultural background,soundscape,and natural environment.Thus,the conclusion was made that soundscape should be consistent with the local natural environment and the historical and cultural background. 展开更多

Soybean,a typical short-day crop,is sensitive to photoperiod,which is a major limiting factor defining its north-to-south cultivation range.The long-juvenile(LJ)trait is controlled primarily by the J locus which has been used for decades by soybean breeders to delay flowering and improve grain yield in tropical regions.The J gene encodes an ortholog of the Arabidopsis Evening Complex(EC)component EARLY FLOWERING 3(ELF3).To identify modifiers of J,we conducted a forward genetic screen and isolated a mutant(eoj57)that in combination with j has longer flowering delay compared with j single mutant plants.Map-based cloning and genome re-sequencing identified eoj57(designated as GmLUX2)as an ortholog of the Arabidopsis EC component LUX ARRHYTHMO(LUX).To validate that GmLUX2 is a modifier of J,we used trans-complementation and identified a natural variant allele with a similar phenotype.We also show that GmLUX2 physically interacts with GmELF3a/b and binds DNA,whereas the mutant and natural variant are attenuated in both activities.Transcriptome analysis shows that the GmLUX2-GmELF3a complex co-regulates the expression of several circadian clock-associated genes and directly represses E1 expression.These results provide mechanistic insight into how GmLUX2-GmELF3 controls flowering time via synergistic regulation of gene expression.These novel insights expand our understanding of the regulation of the EC complex,and facilitate the development of soybean varieties adapted for growth at lower latitudes. 展开更多

The present study attempts to determine diurnal changes by testing the differential sensitivity and temporal resolution in morning-type,evening-type,and intermediate-type individuals with normal hearing.Thirty participants with normal hearing were divided into morningtype,evening-type,and intermediate-type using“MorningnesseEveningness Questionnaire”.The tests of differential sensitivity and temporal resolution were administered to all the participants in the morning and evening,respectively.The differences in the test results between different timing across all the three groups were analyzed.The results of the study showed that there was a significant diurnal effect on psychoacoustic tests as morning-type individuals performed better when tested in morning and evening-type performed better when they were tested in evening.However,there was no diurnal effect found in intermediate group.The lack of inhibitory control and poor cognitive load in off peak time could have lead to poor scores.Thus,the variable of diurnal effects should be considered in further studies on psychoacoustic tests,especially in young adults.Furthermore,studies could be performed to explore diurnal effects on objective audiological tests in a larger group of population in the future. 展开更多

Spatio-temporal evolution analysis of landscape pattern and habitat quality in the Qinghai Province section of the Yellow River Basin from 2000 to 2022 based on InVEST model

Habitat quality is an important indicator for evaluating the quality of ecosystem.The Qinghai Province section of the Yellow River Basin plays an important role in the ecological protection of the upper reaches of the Yellow River Basin.To comprehensively analysis the alterations of habitat quality in the Qinghai Province section of the Yellow River Basin,this study utilized the Integrated Valuation of Ecosystem Services and Tradeoffs(InVEST)model to calculate the habitat quality index and analyze the spatio-temporal variation characteristics of habitat quality in the study area from 2000 to 2022,and calculated seven landscape pattern indices(number of patches,patch density,largest patch index(LPI),landscape shape index(LSI),contagion index(CONTAG),Shannon diversity index,and Shannon evenness index)to research the variation of landscape pattern in the study area.The results showed that the number of patches,patch density,LPI,LSI,Shannon diversity index,and Shannon evenness index increased from 2000 to 2022,while the CONTAG decreased,indicating that the landscape pattern in the Qinghai Province section of the Yellow River Basin changed in the direction of distribution fragmentation,shape complexity,and heterogeneity.The average value of the habitat quality index in the Qinghai Province section of the Yellow River Basin from 2000 to 2022 was 0.90.Based on the value of habitat quality index,we divided the level of habitat quality into five categories:lower(0.00-0.20),low(0.20-0.40),moderate(0.40-0.60),high(0.60-0.80),and higher(0.80-1.00).Most areas were at the higher habitat quality level.The lower habitat quality patches were mainly distributed in Longyang Gorge and Yellow River-Huangshui River Valley.From 2000 to 2022,the habitat quality in most areas was stable;the increase areas were mainly distributed in Guinan County,while the decrease areas were mainly distributed in Xining City,Maqen County,Xinghai County,Qumarleb County,and Darlag County.To show the extent of habitat quality variation,we calculated Sen index.The results showed that the higher habitat quality area had a decrease trending,while other categories had an increasing tendency,and the decreasing was faster than increasing.The research results provide scientific guidance for promoting ecological protection and high-quality development in the Qinghai Province section of the Yellow River Basin. 展开更多

背景 中老年人的晚睡行为可能影响其血脂代谢,但二者的关联性仍需深入分析。目的 探讨不同性别、中心性肥胖、睡前加餐和吸烟人群睡眠时型与血脂异常的关联性。方法 对2022年3—8月在福泉市第一人民医院体检中心体检的40~65岁人群开展... 背景 中老年人的晚睡行为可能影响其血脂代谢,但二者的关联性仍需深入分析。目的 探讨不同性别、中心性肥胖、睡前加餐和吸烟人群睡眠时型与血脂异常的关联性。方法 对2022年3—8月在福泉市第一人民医院体检中心体检的40~65岁人群开展问卷调查(n=697),收集研究对象的一般资料和睡眠情况,采用“清晨型-夜晚型量表”的单项问题来评价睡眠时型。采用非条件二分类Logistic回归模型评估睡眠时型与血脂异常患病风险的关联,在总人群中以性别、中心性肥胖、睡前加餐进行分层分析,在男性人群中以是否吸烟进行分层分析。采用敏感性分析排除夜班工作的影响。结果 纳入体检者中,睡眠时型为清晨型偏好393人(56.4%)、夜晚型偏好304人(43.6%),检出血脂异常334人(47.9%)。剔除夜班人群后的非条件二分类Logistic回归分析结果显示,睡眠时型是血脂异常的影响因素[OR(95%CI)=1.54(1.10,2.16)](P<0.05),夜晚型人群患高TG血症、低HDL-C血症的风险高于清晨型人群[OR(95%CI)分别为1.48(1.04,2.12)、1.79(1.18,2.72)](P<0.05)。在总人群中进行分层分析结果显示:在男性人群和中心性肥胖人群中,夜晚型人群患低HDL-C血症的风险分别是清晨型的1.80倍[95%CI为(1.12,2.91)](P<0.05)和1.73倍[95%CI为(1.02,2.81)](P<0.05);在睡前加餐人群中,夜晚型人群患高TG血症的风险是清晨型的3.43倍[95%CI为(1.30,8.99)](P<0.05),在女性人群、非中心性肥胖人群中睡眠时型对血脂异常及其各指标均无明显影响(P>0.05)。在男性人群中以吸烟分层,结果显示:在吸烟的男性人群中,夜晚型人群患低HDL-C血症的风险是清晨型的1.83倍[95%CI为(1.03,3.26)](P<0.05),在吸烟和不吸烟的男性人群中,睡眠时型对高TC血症、高TG血症、高LDL-C血症和非HDL-C异常均无明显影响(P>0.05)。结论 在40~65岁人群中,夜晚型睡眠偏好可能是血脂异常的危险因素。不同性别、是否为中心性肥胖、睡前是否加餐、是否吸烟人群的睡眠时型与血脂异常之间的关联性可能存在一定差异。 展开更多

目的为了探讨早晚偏好型新手驾驶员的危险感知特点,为交通安全教育提供启示和参考。方法采用2(早-晚型偏好类型:早间型vs晚间型)×2(实验时间:早间vs晚间)混合实验设计。运用早晚型量表(MEQ),从南通某高校随机选择持有驾照的新手大... 目的为了探讨早晚偏好型新手驾驶员的危险感知特点,为交通安全教育提供启示和参考。方法采用2(早-晚型偏好类型:早间型vs晚间型)×2(实验时间:早间vs晚间)混合实验设计。运用早晚型量表(MEQ),从南通某高校随机选择持有驾照的新手大学生。采用经过汉化修订的英国驾驶理论测试中的危险感知任务检测被试危险感知水平,同时运用Biopac多导生理仪,数据结果采用SPSS20.0进行分析。结果早间型被试危险感知得分显著低于晚间型被试。驾驶员晚间实验时的呼吸频率显著高于早间,早间型驾驶员在晚间实验时的肌电值显著高于其在早间实验时的肌电值。结论驾驶员危险感知过程中的肌电、呼吸受时间的影响。不同偏好新手驾驶员的的危险感知水平受到时间的影响,晚间型驾驶员危险感知能力稍高于早间型驾驶员。 展开更多
