The most abundant phenolic biopolymer in the biosphere is the lignin.This phenolic biopolymer commonly exists in combination with polysaccharides and other cell wall components.In this study,the solvent system dioxane...The most abundant phenolic biopolymer in the biosphere is the lignin.This phenolic biopolymer commonly exists in combination with polysaccharides and other cell wall components.In this study,the solvent system dioxane-water is used to extract lignin,which is considered as unaltered native lignin.The dioxane lignin extracted from fig stems was characterized regarding to its structural feature,quantification of its functional groups,molecular weight,and evaluation of its thermal properties.Purity and molecular weight distribution of the studied lignin indicated that isolated lignin contained a low amount of sugar(c.a.19%)and had a high weight-average molecular weight(10068 g.mol-1).Lignin sample had approximately the same amounts of guaiacyl(G)and p-hydroxyphenyl(H)units with relatively fewer syringyl(S)units.The isolated lignin revealed good antioxidant properties.Therefore,it proved to have a high potential of application in new antioxidants formulations.展开更多

A system based on DNA sequences—which is k...Molecular markers provide a useful method for genotype characterization and allow a high precision determination of the genetic relationship between cultivars and varieties. A system based on DNA sequences—which is known as DNA barcoding—will choose one or several standard loci which can be sequenced and compared to differentiate between species. In this research, the ITS, matK, and trnH-psbA sequences were evaluated for the molecular identification of seven F. carica genotypes, generating complete sequences for the first two loci, but unable to produce bidirectional sequences by using the trnH-psbA sequence. The ITS sequence presented the highest variation rates, while the phylogeny constructed with the matK sequence obtained the highest percentage of solved monophyletic groups. Through Pearson’s correlation analysis, it was possible to determine the existence of a significant correlation between the ITS region and psbA-trnH, and the matK and psbA-trnH sequences, but not between ITS and matK. The phylogenies constructed with the ITS + matK barcodes and ITS + matK + psbA-trnH presented the highest percentage for resolution. Although we t...Both morphological characters and PCR-based RAPD approaches were used to determine the genetic diversity and relatedness among nine fig genotypes grown at the northern region of the West-Bank, Palestine. Although we tested 28 primers for the RAPD technique, only 9 produced reasonable amplification products. A total of 57 DNA loci were detected in which 70.2% were polymorphic. DNA fragments presented a minimum of 3 and a maximum of 9 polymorphic bands using primers OPT-10 and OPA-18, respectively. Primers exhibited collective resolving power values (Rp) of 18.826. The Mwazi genotype showed the highest genetic distances among all of the other genotypes. Morphologically, considerable variations were found using 41 quantitative and qualitative traits. Adloni could be a very promising genotype for fresh consumption due to its very late maturation period, extended harvesting period, variable fruit size, and easy skin peeling. In addition, 7 genotypes presented firm fruits, which are a very important criterion for exporting purposes. Dendrogram constructed by UPGMA based on RAPD banding patterns appear somewhat contradictory to the morphological descriptors particularly with Swadi and Biadi genotypes (closed genetically and distanced morphologically), which might be attributed to the phenotypic modifications caused by environmental differences across regions. These preliminary results will make a fundamental contribution to further genetic improvement of fig crops for the region.展开更多
Background:Exploring natural sources to identify new active ingredients for cosmeceutical and pharmaceutical applications is of interest to many researchers.Methods:Flavonoid-rich extracts from dark Ficus carica peels...Background:Exploring natural sources to identify new active ingredients for cosmeceutical and pharmaceutical applications is of interest to many researchers.Methods:Flavonoid-rich extracts from dark Ficus carica peels were investigated in the present study.Extracts were prepared from the peels of four dark cultivars(Bakor Noir,Bouankik,Azenjer and Tazegaght)using acidified methanol.The antimicrobial activity of the extracts was evaluated by the agar well diffusion assay followed by the minimal inhibitory concentration determination using the microdilution method against Gram-positive and Gram-negative pathogenic bacteria and two fungi.Cosmeceutical properties were evaluated by measuring antioxidant activity,ultraviolet absorption characteristics and sun protection factor values.The antioxidant activity of the extracts was measured using aβ-carotene-linoleic acid system.The toxicity of the extracts was assessed using a brine shrimp lethality assay.Results:The extracts contained high amounts of phenolic compounds(3.85–8.63 g/100 g),mainly flavonoids(up to 5 g/100 g).Multiple antibacterial activities were recorded against Gram-positive and Gram-negative bacteria,with the best actions against Bacillus subtilis(minimal inhibitory concentration=156.25µg/mL),Staphylococcus aureus and Pseudomonas aeruginosa(minimal inhibitory concentration=312.5µg/mL),but no antifungal activity was recorded.Azenjer extract showed high inhibition of lipid oxidation evaluated by theβ-carotene bleaching assay with a half-maximal inhibitory concentration of 88.84µg/mL.All extracts showed absorption peaks in the ultraviolet range and Azenjer and Tazegaght extracts at 25 mg/mL presented sun protection factor values>15.The brine shrimp toxicity assay results revealed lethal concentration of 50%values greater than 2,000µg/mL,indicating the safety of the extracts.Conclusion:Therefore,Ficus carica peel extracts could be considered original ingredients with potential applications in the formulation of antibacterial drugs and skincare products.展开更多
Objective:To explore the impact of Ficus carica fruit aqueous extract on fertility parameters in streptozotocin(STZ)-induced male rats.Methods:Twenty-four male Sprague-Dawley rats were divided into four different grou...Objective:To explore the impact of Ficus carica fruit aqueous extract on fertility parameters in streptozotocin(STZ)-induced male rats.Methods:Twenty-four male Sprague-Dawley rats were divided into four different groups.All groups except a normal control group were induced with 50 mg/kg of streptozotocin(STZ)intravenously to induce diabetes.A positive control group was treated with an antidiabetic drug,metformin(500 mg/kg)whereas a negative control group remained untreated throughout the experiment.Meanwhile,another diabetic rat group received treatment with 400 mg/kg of aqueous Ficus carica fruit extract.Rats in the treatment group were administered Ficus carica fruit aqueous extract daily through forcefeeding via oral gavage for a 21-day period.Assessments included the sperm quality(count,motility and morphology),histology of the testes,serum testosterone and fasting blood glucose(FBG)level.Results:The FBG level of the Ficus carica-treated rats exhibited a significant decrease compared to the negative control group(P<0.05).Sperm quality analysis also indicated that the aqueous Ficus carica extract had significant positive effects on sperm count and motility(P<0.05).The histology of the testes in Ficus caricatreated rats revealed an improved cell arrangement in the germinal cell layer.Furthermore,serum testosterone level showed an increment in the Ficus carica treatment group in comparison to the negative control group.Conclusions:Our findings provide compelling evidence for the profertility and anti-hyperglycemic properties of aqueous Ficus carica fruit extract in diabetic-induced male rats.展开更多
Vibrational spectroscopy analysis of full-ripened fig fruits(Ficus carica L.)was acquired using Fourier-transformed infrared(FTIR)spectroscopy by attenuated total reflectance(ATR).The objective of this study was to in...Vibrational spectroscopy analysis of full-ripened fig fruits(Ficus carica L.)was acquired using Fourier-transformed infrared(FTIR)spectroscopy by attenuated total reflectance(ATR).The objective of this study was to investigate accurate discrimination of 25 fig cultivars using chemometric analysis of both ATR-FTIR fingerprinting and in-vitro antioxidant activity along with chromatic coordinates color.In-vitro antioxidant activity was assessed through three methods and displayed statistically significant differences across cultivars and between the fruit parts,with promising antioxidant potency.Infrared spectra were measured separately on both peels and pulp to better know which fruit part provides high discrimination throughput between cultivars.The results showed vibration intensities of five fingerprint regions displaying statically significant differences.The highest vibration intensity occurred in the region of 1175-940 cm^(-1)including the phenols fingerprint.Principal components analysis based on integrated intensities corresponding to fingerprint regions,revealed two main groups in pulp samples,while five groups in peels samples.The pulp extracts of’Nabout’showed a typical vibration in 3000-2800 cm^(-1)and 1775-1725 cm^(-1)regions and,therefore was classified as single subset in cultivars scatterplot.This cultivar had a lightcolored fig and exhibited the highest antioxidant potency for all essays particularly in peel extracts.The biochemical assessment and FTIR fingerprinting data were involved in PCA analysis,and that displayed some dissimilarities in the classification patterns,given the fact that antioxidant activities and chromaticity together could not totally explain the classification based on FTIR fingerprinting between peel and pulp.Application of FTIR-ATR spectroscopy to discriminate fig cultivars seems to be a rapid,accurate and cost-effective alternative to laborious measurement techniques,in fig quality screening and preselection,such as chromatography analysis.This study suggests the use of fig peels for a high-level discrimination while using ATR-FTIR spectroscopy,since it provides high-throughput screening framework.展开更多

[Objectives]To study the anti-inflammatory effects of extracts from different parts of Ficus pumila L.[Methods]Inflammatory models of xylene-induced ear swelling,glacial acetic acid-induced celiac capillary permeabili...[Objectives]To study the anti-inflammatory effects of extracts from different parts of Ficus pumila L.[Methods]Inflammatory models of xylene-induced ear swelling,glacial acetic acid-induced celiac capillary permeability and carrageenan-induced toe swelling were established to evaluate the effects of extracts from different parts of F.pumila L.on inflammation.The inflammatory model of ear swelling in mice with bilateral adrenalectomy induced by xylene was established to determine whether its anti-inflammatory effect depends on hypothalamus-pituitary-adrenal axis(HPAA)system.[Results]Compared with the blank control group,except that there was no statistical significance in the low dose group(P>0.05),the high and middle dose groups of extracts from different parts of F.pumila L.had significant inhibitory effects on xylene-induced ear swelling in mice.The high,middle and low dose groups of F.pumila L.extract could significantly increase the celiac capillary permeability of mice induced by glacial acetic acid,and significantly inhibit the toe swelling induced by carrageenan.Compared with the blank control group,the high and middle dose groups of extracts from different parts of F.pumila L.could significantly inhibit xylene-induced ear swelling and carrageenan-induced toe swelling in bilateral adrenalectomy mice.[Conclusions]The extracts from different parts of F.pumila L.had anti-inflammatory activity,and its anti-inflammatory effect was independent of hypothalamus-pituitary-adrenal axis(HPAA)system.展开更多
Papaya (Carica papaya L.) is one of the most economically, medicinally and nutritionally important tropical fruit crops. Expressed sequence tags (ESTs) derived simple sequence repeat (SSR) markers are more valuable as...Papaya (Carica papaya L.) is one of the most economically, medicinally and nutritionally important tropical fruit crops. Expressed sequence tags (ESTs) derived simple sequence repeat (SSR) markers are more valuable as they are derived from conserved genic portion. Development of EST-SSRs markers through in silico approach is cheaper, less time consuming and labour-intensive. In this study, we aimed to mine SSRs and developed EST-SSR primers from papaya floral ESTs. A total of 75,846 papaya floral ESTs were downloaded from public database National Centre for Biotechnology Information (NCBI). A total of 26,039 floral unigenes (7961 contigs and 18,078 singletons) were generated after assembly of these ESTs. From these floral unigenes, 433,782 perfect SSRs, 204,968 compound SSRs and 6061 imperfect SSRs were mined, respectively. In perfect SSRs, mononucleotide repeats were most abundant (94.7%) followed by tri- (3.1%) and di-nucleotide repeats (1.7%). The frequencies of tetra-, hexa- and penta-nucleotide repeats accounted for only (0.17%), (0.04%) and (0.03%), respectively. In mononucleotide repeats, the most abundant motif was A/T (69.3%) and in di- and tri-nucleotide repeats were AG/CT (61%) and AAG/CTT (31%), respectively. In imperfect SSRs, mononucleotide repeats (56.5%) were most abundant. 176 different types of motifs were identified. A total of 3807 primer pairs for floral papaya ESTs were successfully designed. These developed EST-SSR primers are being used for the genetic improvement of papaya such as study of cross-transferability across genera/species, evaluation of genetic diversity, and identification of sex-specific markers. These EST derived SSRs can also be used in filling gaps in existing linkage maps in papaya.展开更多

The mechanical extraction increased the mucilage yield to 30% dry weight. Physical characterization concerns pH, viscosity, free acidity and density, useful for emulsifying capacity. Spectrophotometric analysis was applied to assess total carbohydrates, proteins, uronic acids, total polyphenols content and antioxidant capacity. DART-MS and SEM-EDX were performed to evaluate functional mucilage components and relative amounts of minerals, respectively. From the main results <em>Opuntia ficus-indica</em> by-product, in addition to the preponderant total carbohydrates content, shows the high concentration in calcium and potassium and a fair amount of health-promoting phytochemicals, which make it a good candidate for the different type of industrial applications.展开更多

文摘Objective:To determine the content of benzyl glueosinolate(BG) in the pulp and the seed and investigate the anti-cuncer activity of its hydrolysis product in Curica papaya L.Methods: Determination of BG was performed on an Hypersil BDS C<sub>18</sub> column at the wavelength of 214 nm with 0.1%trifluoroacelic acid(TFA) aqueous solution(A) and 0.1%TFA acelonilrile(B) as the mobile phase.In vitro activity test was adopted with cidtured human lung cancer H69 cell in vitro to investigate the inhibition rate of cell proliferation of benzyl isothiocyanale(BITC) againsl H69 cell.Results:The pulp has more BG before the maturation of papaya and it nearly disappeared after papaya matured,while the seed contains BG at every stage.Activity test demonstrated that the a higher concentration of BITC would have betler inhibition rate of cell proliferation on 1169 cell,and the IC<sub>50</sub> was 6.5μmol/L.Conclusions:BG also can be produced in the pulp of papaya and it will be stored in the seed after the fruit has been matured.The hydrolysis product of BG has certain cancer-prevention anti-cancer activities for human.
文摘[Objectives]To study the anti-inflammatory effects of extracts from different parts of Ficus pumila L.[Methods]Inflammatory models of xylene-induced ear swelling,glacial acetic acid-induced celiac capillary permeability and carrageenan-induced toe swelling were established to evaluate the effects of extracts from different parts of F.pumila L.on inflammation.The inflammatory model of ear swelling in mice with bilateral adrenalectomy induced by xylene was established to determine whether its anti-inflammatory effect depends on hypothalamus-pituitary-adrenal axis(HPAA)system.[Results]Compared with the blank control group,except that there was no statistical significance in the low dose group(P>0.05),the high and middle dose groups of extracts from different parts of F.pumila L.had significant inhibitory effects on xylene-induced ear swelling in mice.The high,middle and low dose groups of F.pumila L.extract could significantly increase the celiac capillary permeability of mice induced by glacial acetic acid,and significantly inhibit the toe swelling induced by carrageenan.Compared with the blank control group,the high and middle dose groups of extracts from different parts of F.pumila L.could significantly inhibit xylene-induced ear swelling and carrageenan-induced toe swelling in bilateral adrenalectomy mice.[Conclusions]The extracts from different parts of F.pumila L.had anti-inflammatory activity,and its anti-inflammatory effect was independent of hypothalamus-pituitary-adrenal axis(HPAA)system.
文摘Papaya (Carica papaya L.) is one of the most economically, medicinally and nutritionally important tropical fruit crops. Expressed sequence tags (ESTs) derived simple sequence repeat (SSR) markers are more valuable as they are derived from conserved genic portion. Development of EST-SSRs markers through in silico approach is cheaper, less time consuming and labour-intensive. In this study, we aimed to mine SSRs and developed EST-SSR primers from papaya floral ESTs. A total of 75,846 papaya floral ESTs were downloaded from public database National Centre for Biotechnology Information (NCBI). A total of 26,039 floral unigenes (7961 contigs and 18,078 singletons) were generated after assembly of these ESTs. From these floral unigenes, 433,782 perfect SSRs, 204,968 compound SSRs and 6061 imperfect SSRs were mined, respectively. In perfect SSRs, mononucleotide repeats were most abundant (94.7%) followed by tri- (3.1%) and di-nucleotide repeats (1.7%). The frequencies of tetra-, hexa- and penta-nucleotide repeats accounted for only (0.17%), (0.04%) and (0.03%), respectively. In mononucleotide repeats, the most abundant motif was A/T (69.3%) and in di- and tri-nucleotide repeats were AG/CT (61%) and AAG/CTT (31%), respectively. In imperfect SSRs, mononucleotide repeats (56.5%) were most abundant. 176 different types of motifs were identified. A total of 3807 primer pairs for floral papaya ESTs were successfully designed. These developed EST-SSR primers are being used for the genetic improvement of papaya such as study of cross-transferability across genera/species, evaluation of genetic diversity, and identification of sex-specific markers. These EST derived SSRs can also be used in filling gaps in existing linkage maps in papaya.
文摘The present work focuses on the proximate physical and chemical profile of <em>Opuntia ficus-indica</em> mucilage, mechanically extracted from cladodes, a waste of pruning in traditional organic cactus pear orchards in Italy (San Cono, Sicily). The mechanical extraction increased the mucilage yield to 30% dry weight. Physical characterization concerns pH, viscosity, free acidity and density, useful for emulsifying capacity. Spectrophotometric analysis was applied to assess total carbohydrates, proteins, uronic acids, total polyphenols content and antioxidant capacity. DART-MS and SEM-EDX were performed to evaluate functional mucilage components and relative amounts of minerals, respectively. From the main results <em>Opuntia ficus-indica</em> by-product, in addition to the preponderant total carbohydrates content, shows the high concentration in calcium and potassium and a fair amount of health-promoting phytochemicals, which make it a good candidate for the different type of industrial applications.
