A high sensitive flow-injection chemiluminescence (FI-CL) method for the determination of tryptophan has been developed. The method is based on the chemiluminescence reaction of galangin-potassium permanganate-tryptophan in polyphosphoric acid (PPA) media. Under the optimized conditions, tryptophan was determined in the range 0.05-10 μg/mL with the detection limit (3tr) of 5.0 × 10^-3 μg/mL. The relative standard deviation (RSD) was 1.0% for 11 replicate determinations of 1.0 μg/mL tryptophan. Three synthetic samples were determined selectively with recoveries in the range from 99.6% to 102.0% in the presence of other amino acids. 展开更多

A new flow-injection procedure has been developed for the determination of penicillamine based on the enhancement of the chemiluminescence(CL) reaction of the penicillamine-cerium(Ⅳ)-hydrocortisone system. The method was simple, selective and sensitive with a detection limit of 6.2×10-5 mol·L-1(S/N=3). It was applicable to the determination of penicillamine in the concentration range of 2×10-6~5×10-4 mol·L-1. The method was successfully applied to the determination of penicillamine in tablets. The relative standard deviation (RSD) was 1.42% (n=7) and the recovery was 97%~103% (n=4). 展开更多

Preconcentration and determination are based on (i) the quantitative and fast adsorption of chromium (VI) on the high surface area nanometer-size TiO2 (anatase) powders, which prepared by a sol-gel method from hydrolysis of TiCI4 and (ii) the quantitative and reproducible elution of Cr (VI) by 2. 0 mol. L-1 HCI. A mini-column system for preconcentration is developed, Cr(VI)on the mini-column is eluted and merged with a stream water and DPCB (1, 5-diphenylcarbazide ) as the chromogenic reagent. The Proposed system permits throughputs of 6 sample h--l (0. 001 μg mL-1 Cr(VI) ) or 20 sample h-1 (0. 1 μg mL-1Cr (VI) . The preconcentration factor is 55. Objective To establish a rapid and precise continuous flow-injection chemiluminescence method for the determination of tetracycline and oxytetracycline. Methods In NaOH solution, tetracycline and oxytetracycline can sensitize obviously the chemiluminesence (CL) intensity of the reaction of luminol with KIO_4, the sensitized CL intensity is proportional to the concentration of tetracycline and oxytetracycline. So, a new flow-injection CL method has been developed. The optimum chemical conditions for the CL reaction were investigated. Results Under the optimized conditions (KIO_ 4 concentration: 1.0×10 -5 mol/L; NaOH concentration: 0.1 mol/L; luminol concentration: 1.0×10 -4mol/L), tetracycline and oxytetracycline were determined. The linear range of the working curves was 1.0×10 -7 - 1.0×10 -4g/mL, the detection limits was 1.0×10 -8g/mL and 1.1×10 -8g/mL, and the relative standard deviation was 2.6% (C_S=1.0×10 -6g/mL; n=11) and 2.0% (C_S=1.0×10 -6g/mL; n=11) respectively. Conclusion The method is simple, rapid, and sensitive, and it has been successfully applied to the the determination of tetracycline and oxytetracycline tablets, the mean recoveries being 99.7% and 98.8% respectively. 展开更多

Methods The chemiluminescence of cerium (Ⅳ)-sulfite system was obviously sensitized by adding anyone of three β-blockers in acid media. A new chemiluminescence method was set up by combining with flow-injection technique and used to determine the three β-blockers. Results Good linear ranges were obtained at the concentrations of 2.0×10-7g/mL-4.0×10-5g/mL,1.0×10-7g/mL-3.0×10-5g/mL and 7.0×10-7g/mL-1.0×10-5g/mL,respectively,with the detection limits of 5.0×10-8g/mL,7.0×10-8g/mL and 5.0×10-8g/mL (S/N=3),respectively,and the relative standard deviations for 11 times consecutive injections of 1.0×10-6g/mL bisoprolol,atenolol and propranolol were 3.57%,2.21% and 2.26%,respectively. Conclusion The developed method is sensitive,accurate,rapid and of low cost. And it can be applied to determine bisoprolol,atenolol and propranolol in pharmaceutical preparations. 展开更多

Antimony(... On-line ion-exchange separation and preconcentration were combined with flow-injection hydride generation atomic absorption spectrometry (HGAAS) to determine ultra-trace amounts of antimony in water samples. Antimony(Ⅲ) was preconcentrated on a micro-column packed with CPG-8Q chelating ion-exchanger using time-based sample loading and eluted by 4 mol l^(-1) HCl directly into the hydride generation AAS system. A detection limit (3σ) of 0.0015μg l^(-1) Sb(Ⅲ) was obtained on the basis of a 20 fold enrichment and with a sampling frequency of 60h^(-1). The precision was 1.0% r.s.d.(n=11) at the 0.5μg l^(-1) Sb(Ⅲ) level. Recoveries for the analysis of antimony in tap water, snow water and sea water samples were in the range 97-102%. 展开更多

A flow injection chemiluminescence (CL) method for the determination of catecholamines is described which is based on their inhibition of the intensity of CL from the luminol-hypochlorite system The hypochlorite was electrogenerated on-line by a constant current electrolysis, resulting in the elimination of the instability of the hypochlorite solution prepared from a commercial reagent of sodium hypochlorite. The detection limits are 0.6 ng/ml for dopamine, 0.8 ng/ml for adrenaline and isoprenaline. 第三次全国土壤普查意义重大,为确保普查数据真实可靠,需对行业标准检测方法进行反复验证以优选出适合在全国范围内推广的方法。由于NY/T 1121.14—2006比浊法测定土壤有效硫标准曲线线性较差,亟须优化其提取、测定条件以提高方法的准... 第三次全国土壤普查意义重大,为确保普查数据真实可靠,需对行业标准检测方法进行反复验证以优选出适合在全国范围内推广的方法。由于NY/T 1121.14—2006比浊法测定土壤有效硫标准曲线线性较差,亟须优化其提取、测定条件以提高方法的准确度和精密度。鉴于此,主要探究了沉淀剂的状态、品牌及线性拟合方式对比浊法测定土壤有效硫标准曲线线性的影响,结果表明,有效硫标准曲线改用现用现配的40 g/L氯化钡溶液定容可显著提高标准曲线的线性,使其相关系数r>0.999,满足分析测试的要求。同时本实验室对电感耦合等离子体光谱法(ICP-OES)测定土壤有效硫进行方法验证、对比,比浊法、ICP-OES法的检出限分别为1.20、0.80 mg/kg,均满足分析测试的要求。方法准确度和精密度验证结果表明,二者均可用于土壤有效硫的测定并且测定结果无显著性差异。其中,比浊法适用于测定低含量(<48 mg/kg)样品,ICP-OES法适用于含量范围在0~150 mg/kg的样品。结合本实验室在第三次全国土壤普查内业检测试点工作中的经验,建议将ICP-OES法作为第三次全国土壤普查有效硫测定的首选方法,以期为第三次全国土壤普查内业检测提供参考和借鉴。 展开更多

目的对AG800全自动血小板聚集仪光学比浊法检测血小板聚集功能进行评价,并测试仪器的检测性能。方法参照美国临床实验室标准化委员会(Clinical and Laboratory Standards Institute,CLSI)及《光学比浊法检测血小板聚集标准化专家共识》... 目的对AG800全自动血小板聚集仪光学比浊法检测血小板聚集功能进行评价,并测试仪器的检测性能。方法参照美国临床实验室标准化委员会(Clinical and Laboratory Standards Institute,CLSI)及《光学比浊法检测血小板聚集标准化专家共识》等相关文件,使用AG800全自动血小板聚集仪和美国Chrono-log700型血小板聚集仪,从样本精密度检验、二磷酸腺苷(adenosine diphosphate,ADP)和花生四烯酸(arachidonic acid,AA)相关性及一致性检验评估两台仪器对血小板聚集功能的检测。对AG800全自动血小板聚集仪进行了性能评估,包括加样、温控及光学检测精密度检验。结果通过7 d的连续跟踪结果显示,Chrono-log700血小板聚集仪低水平样本总体检测精密度CV值为10.99%,AG800自动血小板聚集仪的低水平总体精密度为4.95%,Chrono-log700血小板聚集仪的高水平样本总体检测精密度CV值为9.00%,而AG800全自动血小板聚集仪的高水平总体精密度为4.91%。ADP检测结果统计分析,Pearson相关系数r=0.9915,相关程度高。P<0.0001,表示存在显著性,Bland-Altman一致性检验统计结果显示0个点在95%一致性界限以外,符合一致性分析原则,说明两机检测ADP结果一致性好;AA检测结果统计分析,Pearson相关系数r=0.9832,相关程度高。P<0.0001,表示存在显著性;Bland-Altman一致性检验统计结果显示1个点(2.86%)在95%一致性界限以外,符合一致性分析原则,说明两机检测AA结果一致性好。性能评估结果分析,AG800全自动血小板聚集仪的加样均数(Mean)=9.99μL,均数标准误(Standard error of mean)=0.006,偏度系数(Skewness)=0.119,加样精密度CV为0.48%,离散度(平均绝对偏差,AVEDEV)为0.038μL;AG800全自动血小板聚集仪的温度均数=36.98℃,均数标准误=0.041,偏度系数=-0.293,加样精密度CV为0.93%,离散度为0.284℃;AG800全自动血小板聚集仪对低水平标本的检测均数=20.02%,均数标准误=0.099,偏度系数=0.733,加样精密度CV为4.94%,离散度为0.74%。对高水平标本检测的均数=44.13%,均数标准误=0.184,偏度系数=0.416,加样精密度CV为4.14%,离散度为1.49%。结论AG800全自动血小板聚集仪和Chrono-log700血小板聚集仪检测的ADP和AA值之间存在强线性相关性,说明两机检测结果一致性好;AG800全自动血小板聚集仪在对临床样本低水平和高水平标本的检测CV不超过5%,加样、温控及对样本的浊度变化检测精密度高,可满足临床检测的精密度要求,临床适应性好,具有自动化程度高、操作简便、人为影响因素小等优点。 展开更多

A rapid method is described for the determination of petroleum hydrocarbons in soil samples. The method is based on the extraction of hydrocarbons by a solvent and the treatment of the solution with an aqueous solution of a surfactant to release the hydrocarbons to the water phase in the form of a stable emulsion. The emulsion is then utilized to measure the hydrocarbon content by turbidimetry. The effects of various operating parameters including the surfactant solution composition and time of extraction and time of mixing with the releasing solution are investigated. The stability of the emulsion was improved in acid environment containing and electrolyte. The turbidity values (T) were related with hydrocarbon concentration in the extract (C) by the following equation. Turbidity = 2.75 C + 205.7. With R2 = 0.9929. The soil hydrocarbon content (SHC) measured in μg/g can then be calculated using the formula: SHC = [Extract Vol. (mL) x C]/Sample Wt (g). The results correlated well with the results of total hydrocarbons in soils determined by standard methods. The method was applied for the estimation of hydrocarbons in Passaic river sediments taken from various locations and depths. For field work the method was used to supply data on the hydrocarbon contamination of soil samples taken within an oil refinery and a monitoring well drilled within heavy hydrocarbon waste dumping location. 展开更多

Coagulation-flocculation is a proven technique for the removal of suspended solids in wastewater, through the application of single angle turbidimetric measurement. Through this measurement, the coag-flocculation kinetics and functional parameters behavior of Detarium microcarpumin brewery effluent with respect to pH, dosage and time were followed at room temperature. Process parameters such as order of reaction α, rate constant (K), coagulation period τ1/2 etc were determined. Results indicated that reaction order, rate constant, period, pH and dosage recorded optimum values at 2, 1.8 × 10-2 L/mg.min, 0.152 min, 4 and 100 mg/L, respectively. Maximum efficiency recorded was 96.07% at 30 min. Detarium microcarpum has shown potential as an effective bio-coagulant for the removal of turbidity from brewery effluent. 展开更多

利用血清指数比较溴甲酚绿法和免疫比浊法检测清蛋白的抗干扰能力

目的利用血清指数(SI)作为量化指标,探讨内源性干扰物血红蛋白(Hb)、结合胆红素(Bil-C)、游离胆红素(Bil-F)和乳糜微粒(CM)对溴甲酚绿(BCG)法和免疫比浊法检测血清清蛋白(ALB)的影响。方法统计临床生化标本SI波动范围。配制含不同水平... 目的利用血清指数(SI)作为量化指标,探讨内源性干扰物血红蛋白(Hb)、结合胆红素(Bil-C)、游离胆红素(Bil-F)和乳糜微粒(CM)对溴甲酚绿(BCG)法和免疫比浊法检测血清清蛋白(ALB)的影响。方法统计临床生化标本SI波动范围。配制含不同水平干扰物的干扰标本和对照标本。分别采用BCG法和免疫比浊法检测血清ALB,计算干扰标本与对照标本血清ALB检测结果的相对偏倚。以1/2国家卫生健康委临床检验中心室间质评允许总误差(3%)作为临床可接受标准。结果SI波动范围:溶血指数、黄疸指数、脂浊指数的P2.5和P97.5分别为0和24、0和2、1和27。不同水平Hb、Bil-C、CM对两种方法检测血清ALB均无干扰,所有偏倚均在允许范围内。Bil-F对BCG法检测血清ALB正常值、低值标本均产生正干扰。低值标本的检测中,当黄疸指数≥6(Bil-F≥72.04μmol/L)时,偏倚超出允许范围,正常值标本的检测中,则黄疸指数≥17(Bil-F≥324.18μmol/L)时,偏倚超出允许偏倚范围,而且随着Bil-F水平升高,干扰逐步增加。Bil-F对免疫比浊法检测血清ALB正常值、低值标本产生正干扰,两个水平标本,均在黄疸指数≥19(Bil-F≥360.2μmol/L)时,检测偏倚超出允许范围。临床常见水平的Bil-F对结果的影响均在可接受范围内。结论SI可以对干扰程度进行量化,便于临床实验室决定是否需要采取针对性措施降低干扰物的影响或据此建立标本退检界值。BCG法和免疫比浊法检测血清ALB具有相似的抗干扰能力,临床常见的溶血、黄疸、脂浊不会影响检测结果。 展开更多
