Sox9a is a new duplication of C.semilaevis Sox9 gene. The complete cDNA of Sox9a gene was 1 842 bp long coding for 487 amino acids and Foxl2 gene was 1 817 bp coding for 308 amino acids. Sox9a was expressed higher in male brain, pituitary and gonad and Foxl2 were higher in female brain, pituitary and gonad. The expression of Sox9a gene in gonads of neo-males was higher than that of normal females. Sox9a and Foxl2 were expressed higher in gastrula stage than in other stages. In the period of sex differentiation, the expression of Sox9a was first going up and then going down and Foxl2 was higher expressed at 37 dph. The highest expressions of Sox9a and Foxl2 genes occurred in nine-month and 12-month old gonad tissues, respectively. Sox9a gene was considered to have inevitable links with sex reversal, sex differentiation and cell differentiation of embryos and formation of spermatogenic ceils. Foxl2 was considered to play a role in sex differentiation, cell differentiation of embryos but not to be necessary for sex determination and sex reversal.展开更多

CSEC cells grow actively and stably more than 50 passages for over 200 d in DMEM medium supplemented with 15% FBS (fetal bovine serum), 2.5 ng/cm^3 bFGF (basic fibroblast growth factor), 1 ng/cm^3 LIF (leukemia inhibitory factor) and 50 mmol/dm^3 2-ME (2-mecaptoethanol). The cells grew well in the temperature range of 24-30 ℃ and the optimal growth temperature was 24 ℃. FBS and bFGF concentrations are the two key components for CSEC cells proliferation. Chromosome analysis reveals that CSEC cells have a normal diploid karyotype with 2n=42t. The significant fluorescent signals were observed in CSEC after transfection with the GFP reporter gene, suggesting that the CSEC cell line can be used as a useful tool for transgenic and genetic manipulation studies. CSEC cells showed the cytopathic effect (CPE) after infection with lymphosystis disease virus (LCDV) in 2 d. Moreover, the LCDV particles can be observed in the cytoplasm of virus-infected cells by electron microscopy. It suggests that CSEC could be potentially used for the study of aquatic virus.展开更多

Seventy healthy male minks were randomly divided into five groups of 14 minks each. The minks were fed in five kinds of experiment diets (HP, LP, LP+M1, LP+M2 and LP+M3). The dietary protein levels, expressed as percentage of dry matter (DM), were 32% (high protein, HP) and 24% (low protein, LP). LP was supple- mented with Met 0.4% (M1), 0.8% (M2) and 1.2% (M3) DM. From mid of September to December 10, apparent digestibility of CP (crude pro- tein), N intake and urinary N excretion were decreased with declining dietary protein levels (p 〈 0.05) and N retained was the highest in treat- ment LP+M2. No significant difference was found in total serum protein (TP) and serum urea nitrogen (SUN) among all treatment groups (p 〉 0.05). Skin length of treatment HP and LP^M2 was higher than that of other groups (p 〈 0.05). Body length, skin weight, length of guard hair and under hair were not affected by different dietary protein levels (p〉0.05). The best performance could be observed in treatment LP+M2. In diet, 24% (DM) protein level with 1.54% Met supplementing was enough for minks during growing-farring period. Dietary protein lowered from 32% to 24% with supplementing Met in diets would result in a37.9% decrease in urinary N excretion. Furthermore, addition of Met in diets for minks would be beneficial in terms of reducing feed expenses and lessening nitrogen emissions to the environment.展开更多

It is found that the eastern equatorial Pacific cold tongue is sensitive to the adjustment. Compared with the simulation of the original level scheme, the adjusting yields a more realistic structure of cold tongue extending from the coast of Peru to the equator, as well as a temperature minimum at Costa Rica coast, north of the cold tongue. In the original scheme experiment, the sharp heating by net surface heat flux at the beginning of spin-up leads to a great warm- ing in the eastern equatorial Pacific Ocean. The weak vertical advection due to a too thick mixed layer in the coarse vertical structure also accounts for the warm bias. The fact that most significant improvements of the upper 50 m temperature appear at the region of the thinnest mixed layer indicates the necessity of fine vertical resolution for the eastern equatorial Pacific Ocean. However, the westward extension of equatorial cold tongue, a defect in the original scheme, gets even more serious in the adjusting scheme due to the intensi- fied vertical velocity and hence vertical advection in the central-eastern equatorial Pacific Ocean.展开更多

However, the expression and potential role of iASPP in oral tongue squamous cell carcinoma (OTSCC) have not been addressed. In our study, we detected iASPP expression in OTSCC by immunohistochemistry, iASPP expression is up-regulated in OTSCC tissues. Moreover, in clinical pathology specimens, we found that increased iASPP expression correlates with poor differentiation and lymph node metastasis. Using multicellular tumor spheroids (MTS) and flow cytometry, we demonstrated that iASPP down-regulation arrests OTSCC cells at the G0/G1 phase, induces OTSCC cell apoptosis and inhibits OTSCC cell proliferation. These results indicate that iASPP plays a significant role in the progression of OTSCC and may serve as a biomarker or therapeutic target for OTSCC patients.展开更多

Methodology Human tongue carcinoma Tca8113 cells were cultured with DAPT. Cell growth was determined using Indigotic Reduction method. The cell cycle and apoptosis were analyzed by flow cytometry. Real-time PCR and Immuno-Fluorescence (IF) were employed to determine the intracellular expression levels. Results DAPT inhibited the growth of human tongue carcinoma Tca8113 cells by inducing G0-G1 cell cycle arrest and apoptosis, The mRNA levels of Hairy/Enhancer of Split-1 (Hes-1), a target of Notch activation, were reduced by DAPT in a dose-dependent manner. Coincident with this observation, DAPT induced a dose-dependent promotion of constitutive Caspase-3 in Tca8113 cells. Conclusion DAPT may have a therapeutic value for human tongue carcinoma. Moreover, the effects of DAPT in tumor inhibition may arise partly via the modulation of Notch- 1 and Caspase-3.展开更多

The negative transfer of Chinese into English is the major cause of the so-called Chinglish. The paper states some forms of the negative transfer.
文摘Due to the impact of the negative transfer of mother tongue, students often think in their mother tongue when they write international business letters.Consequently, they may have difficulties communicating with the intended customers. In a bid to improve students'skills in international business letter writing, this article analyzes the impact of the negative transfer from three aspects:the word order in English, the position of modifiers and the use of conjunctions.
文摘By investigating the types of 371 non-English major freshmen's use of mother-tongue-reliance strategies in English languagelearning, the study showed that students' use of mother-tongue-reliance strategies would vary significantly with their different L2 profi-ciency.And it was negatively correlated with L2 proficiency. And the study discussed the implications to guide English language teach-ing and learning.

文摘Two transcription factors, Sox9a and Foxl2 were cloned from half-smooth tongue sole (Cynoglossus semilaevis). Foxl2 was considered to play a role in sex differentiation, cell differentiation of embryos but not to be necessary for sex determination and sex reversal.

文摘A new cell line, CSEC, has been successfully established from embryos at gastrula stage of a cultured marine fish, half smooth tongue sole (Cynoglossus sernilaevis). It suggests that CSEC could be potentially used for the study of aquatic virus.

文摘A study was conducted to evaluate production performance of minks in growing-furring period with supplementing DL-Methinnine (Met) in low protein diet. Furthermore, addition of Met in diets for minks would be beneficial in terms of reducing feed expenses and lessening nitrogen emissions to the environment.
文摘The vertical resolution of LICOM1.0 (LASG/IAP Climate System Ocean Model, version 1.0) is adjusted by increasing the level amount within the upper 150 m while keeping the total of levels. However, the westward extension of equatorial cold tongue, a defect in the original scheme, gets even more serious in the adjusting scheme due to the intensi- fied vertical velocity and hence vertical advection in the central-eastern equatorial Pacific Ocean.

文摘iASPP is an inhibitory member of the apoptosis-stimulating proteins of P53 (ASPP) family. iASPP is over expressed in several malignant tumors and potentially affects cancer progression. These results indicate that iASPP plays a significant role in the progression of OTSCC and may serve as a biomarker or therapeutic target for OTSCC patients.

文摘Aim To investigate the effect of DAPT (γ-secretase inhibitor) on the growth of human tongue carcinoma cells and to determine the molecular mechanism to enable the potential application of DAPT to the treatment of tongue carcinoma. Moreover, the effects of DAPT in tumor inhibition may arise partly via the modulation of Notch- 1 and Caspase-3.
