Meyer) cells via cellulase degradation. CDW activated the NADPH oxidase activity of isolated plasma membranes and stimulated in vivo H2O2 generation in ginseng cell suspensions. CDW also increased the activity of phenylalanine ammonia lyase (PAL), expression of a P. ginseng squalene epoxidase (sqe) gene and saponin synthesis. NADPH oxidase inhibitors inhibited both in vitro NADPH oxidase activity and in vivo H2O2 generation. Induction of PAL activity, saponin synthesis and sqe gene expression were all inhibited by such inhibitor treatments and reduced by incubation with catalase and HA scavengers. These data indicate that activation of NADPH oxidase and generation of H2O2 are essential signalling events mediating defence responses induced by the endogenous elicitor(s) present in CDW.展开更多

Methods: A total of 60 patients presenting mild or mild to moderate ED were enrolled in a double-blind, placebo-controlled study in which the efficacies of KRG and a placebo were compared. The patients received either 1 000 mg (3 times daily) of KRG or a placebo. Results: The five-item version of the International Index of Erectile Function (IIEF-5) score after the treatment was significantly higher in the KRG group compared with that before the treatment (from 16.4±2.9 to 21.0±6.3, P 〈 0.0001). In contrast, there was no difference before and after the treatment in the placebo group (from 17.0±3.1 to 17.7±5.6, P 〉 0.05). In the KRG group, 20 patients (66.6%), reported improved erection, significant in the global efficacy question (P 〈 0.01); in the placebo group there was no significance. Scores on questions 2 (rigidity), 3 (penetration), 4 and 5 (maintenance), were significantly higher for KRG than those for the placebo when those questions were answered after 12 weeks of each treatment (P 〈 0.01). When the score in the KRG group was compared to the placebo group after the treatment, there was a significant improvement in total score (IIEF-5 score) in questions 3 and 5 for the KRG-treated group (P 〈 0.001 and P 〈 0.0001, respectively). The levels of serum testosterone, prolactine and cholesterol after the treatment were not statistically significant different between the KRG and the placebo group (P 〉 0.05). Conclusion: Our data show that KRG can be an effective alternative to the invasive approaches for treating male ED.展开更多

A total of 25 ginsenosides were extracted include of which 10 low-polar ginsenosides, such as ginsenosides F4, Rk3, Rh4, 20S-Rg3, 20R-Rg3 and so on, were identified according to their HPLC retention time and MS/MS data. The results indicated that the low polar ginsenosides were seldom found in RPG. For the exploration of the transformation pattern of the ginsenosides in steam processing, the standards of ginsenosides Re, Rg1, Rb1, Rc, Rb2, Rb3 and Rd were selected and hydrolyzed at a temperature of 120 oC. The results show that these polar ginsenosides can be converted to low-polar ginsenosides such as Rg2, Rg6, F4, Rk3 and Rg5 by hydrolyzing the sugar chains.展开更多

We investigated the effects of tissue-cultured mountain ginseng extract (TMGE) on male patients with erectile dysfunction (ED). A double-blind, placebo-controlled study was conducted with 143 patients experiencing ED. Over the course of 8 weeks, one group took 1 000 mg of TMGE twice a day, and the other group took 1 000 mg of placebo twice a day. The effects of the TMGE and the placebo were analyzed using the Korean version of the International Index of Erectile Function (IIEF) questionnaire. A total of 86 patients completed 8 weeks of treatment. The scores on the five domains of the IIEF after medication were significantly higher than the baseline scores in the group treated with TMGE (P 〈 0.05), whereas no significant improvement was observed in the placebo group (P 〉 0.05). Erectile function and overall satisfaction scores after medication were significantly higher in the TMGE group than in the placebo group (P 〈 0.05). Erectile function of patients in the TMGE-treated group significantly improved, suggesting that TMGE could be utilized for improving erectile function in male patients.展开更多

Recent studies showed that ginseng berries produce more ginsenosides than KRG; thus, we investigated the ability of the Korean ginseng berry extract GB0710 to relax the penile corpus cavernosum smooth muscle (CCSM) in this study. As a comparative control, the results were compared to those obtained using KRG. In addition, possible mechanisms of action for GB0710 were investigated. While KRG and GB0710 both displayed dose-dependent relaxation effects on precontracted rabbit CCSM in vitro, GB0710 was shown to be more potent than KRG. The GBO710-induced relaxation could be partially reduced by removing the endothelium. In addition, pre-treatment with several nitric oxide (NO) inhibitors significantly inhibited the relaxation of muscle strips. Furthermore, administration of GB0710 increased intracavernosal pressure (ICP) in a rat in vivo model in both a dose- and duration-dependent manner. Intracellular NO production in human microvascular endothelial cells could be induced by GB0710 and inhibited by NG-monomethyI-L-arginine. In conclusion, GB0710 had a greater relaxation effect on rabbit CCSM than did KRG extract, and increased ICP in a rat model in both a dose- and a duration-dependent manner. This relaxine effect might be mediated bv NO production.展开更多

Meyer is an endangered species in Russia. To restore this species, effective protective measures, including the reintroduction into favorable habitats, must be worked out considering the specificity of genetic structure of ginseng populations. One hundred and thirty-nine P. ginseng plants were collected from the forests of nine administrative areas of Primorsky Territory of Russia and transferred to a collection nursery for further investigation. Microsatellite markers were used to study the genetic diversity and the genetic structure of ginseng populations. For populations studied with SSR, the number of observed alleles was ranging from 15 to 25, allelic richness from 1.83 to 3.04, polymorphic loci from 62.5% to 87.7%, observed heterozygosity from 0.410 to 0.512 (an average of 0.453) and expected heterozygosity from 0.304 to 0.479, with an average of 0.393. The values of the inbreeding coefficient within populations (Fis) ranged from -0.447 to 0.056, and their average value was -0.296. Genetic differentiation among populations was significant (Fst = 0.115) but an isolation-by-distance pattern was not detected. UPGMA and MS-tree confirmed the presence of genetic structure within P. ginseng and visualized genetic relationships of populations with similar pattern. STRUCTURE analysis revealed the genetic admixture between different ginseng populations. It was established with SSR markers that P. ginseng still preserves substantial genetic resources although all populations are largely exhausted. Because ginseng populations are significantly differentiated all of them should be restored. Considering the admixture of ginseng populations it would be advisable to apply the individual assignment test to verify the content of indigenous populations and to identify the "true" population plants to serve as stock material for reintroduction.展开更多

文摘Endogenous elicitor, termed cellulase-degraded cell wall (CDW), was prepared from the cell wall of suspension-cultured ginseng (Panax ginseng C.A. These data indicate that activation of NADPH oxidase and generation of H2O2 are essential signalling events mediating defence responses induced by the endogenous elicitor(s) present in CDW.

This relaxine effect might be mediated bv NO production.

文摘[Objectives] To compare the effects of different extraction technologies on the extraction of total flavonoids from ginseng flowers.[Methods]Taking the ginseng flower as raw material, and the total flavonoids of ginseng flower were extracted by water extraction, ethanol extraction,freeze-thaw,ultrasonic extraction, and microwave extraction. The scavenging ability of total flavonoids and Vc to DPPH free radicals and hydroxyl radicals were compared between ultrasonic extraction and water extraction.[Results]The yield of ultrasonic and microwave technology was relatively high.In this study, the ultrasonic extraction was taken as the main method, ethanol aqueous solution was used as the solvent, and single factor experiment was carried out with ultrasonic power, ethanol volume, solid to liquid ratio and time as the variables. The best extraction process of flavonoids was ultrasonic power of 120 Hz,ethanol concentration of 60%,solid to liquid ratio of 1∶ 30,and ultrasonic time of 80 s.Under these conditions, the yield of flavonoid extraction was 6.72 mg/g.[Conclusions] The flavonoids obtained by ultrasonic extraction and microwave extraction have the highest antioxidant capacity,65.4% and 53.5%,respectively,indicating that the ultrasonic extraction and microwave extraction technologies are more suitable for the extraction of flavonoids from the ginseng flower.
文摘Panax ginseng C.A. Considering the admixture of ginseng populations it would be advisable to apply the individual assignment test to verify the content of indigenous populations and to identify the "true" population plants to serve as stock material for reintroduction.

文摘[Objectives] To study the effects of Panax ginseng,Codonopsis pilosula and Pseudostellaria heterophylla on the growth and development of SD female rats. [Methods] The rats were randomly divided into control group,P. ginseng group,C. pilosula group, and P. heterophylla group, intragastrically administered with physiological saline,P. ginseng,C. pilosula, and P. heterophylla for 2 weeks, daily vaginal smear inspection was performed, estradiol( E_2) level before and after intervention was detected, rats were dissected, took out ovary and uterus, and calculated the ovarian index and uterus index. [Results]In 4 groups of rats, there was no significant difference in the time of sexual maturity and ovarian index; compared with the control group,E_2 level of C. pilosula group and P. ginseng group were higher( P < 0. 05);compared with the control group and P. heterophylla group, the ovarian index of P. ginseng group was higher( P < 0. 05); compared with the control group, the body weight of three administration groups increased much more( P < 0. 05). [Conclusions] P. ginseng and C. pilosula may lead to sexual maturity of SD female rats, while P. ginseng,C. pilosula and P. heterophylla can promote increase of body weight of SD female rats.

文摘Gestational diabetes mellitus(GDM)is the most common glucose metabolism complication or cause of potential impaired glucose tolerance that can occur either before or during pregnancy and lactation. The prevalence of GDM and its related complications in young women is increasing, and this condition may cause serious outcomes and health hazards to the foetus. However, traditional oral hypoglycaemic drugs have potential safety hazards;therefore,it is urgent to develop new, safe, effective, and easily administered agents and remedies.Ginseng polysaccharide(GPS),which is isolated from Panax(P.)ginseng C.A.Meyer,exhibits notably promising biological activities and effects;specifically,it has been shown to lower blood glucose with mild, safe, and nontoxic characteristics, and it can also improve human bodily functions.Hence,we hypothesise that GPS might be used as an additional therapy and candidate agent for treating GDM.This review innovatively summarizes the available reports and evidence from basic studies to analyze the potential for and feasibility of using GPS as a new therapeutic agent for treating GDM.Additionally, for the first time, this review provides a rationale for the use of GPS.Our summarized results show that GPS may be developed as a novel antidiabetic drug and a remedy for use in preventing and treating GDM,with great application prospects.
