Background Poly(ADP-ribose) polymerase (PARP) plays an important role in the death of retinal capillary cells in diabetic retinopathy (DR) partly via its regulation of nuclear factor kappa B (NF-κB). The current study investigated the effect of the regimen of Gaoshan Hongjingtian (RG) on the mechanism of PARP regulation of NF-κB, and demonstrated the possible impact of the RG and Gaoshan Hongjingtian (Rhodiola sachalinensis, RS) on diabetic retinopathy. Methods Wistar rats were made diabetic by administering streptozotocin. They were then assigned to three groups at random. After 2 months, the three groups of these diabetic rats were treated with RS or RG, or untreated. Analyses of expression levels of PARP, NF-κB, and intercellular adhesion molecule-1 (ICAM-1) in the retinas of rats in different groups were performed by Western blotting and immunohistochemical assays, and mRNA levels of NF-κB and ICAM-1 were determined by real-time polymerase chain reaction (PCR). In addition, the basement membranes of capillaries in the rats' retinas were observed using electron microscopy, and diabetes-induced capillary degeneration (ghost pericytes and acellular capillaries) were quantitated. Results From the third month after the injection of streptozotocin, the diabetic rats were given daily RG, RS or tap water separately. The diabetic rats failed to gain weight compared with normal age-matched rats, whereas their glycated hemoglobin levels were significantly increased. After 5 months, the mRNA levels of NF-KB and ICAM-1 and the protein expression of PARP, NF-κB, and ICAM-1 were significantly increased in the retinas of diabetic rats in the untreated group compared with the nondiabetic controls. After 8 months, the number of degenerated retinal capillaries (ghost pericytes and acellular capillaries) was significantly increased in the diabetic rats in the untreated group compared with normal age-matched rats. RG and RS inhibited diabetes-induced over-expression of PARP, NF-KB, and ICAM-1 in the retinas of diabetic rats at the end of 5-month diabetic duration. Treatment using RG and RS significantly inhibited increases in the number of acellular capillaries and pericyte ghosts and suppressed the basement membrane thickening in the retinas of rats with diabetes for 8 months compared with the control diabetic rats. Conclusions These results indicate that PARP plays an important role in the pathogenesis of diabetic retinopathy. RS and RG may have acted on the mechanism of PARP regulation of NF-κB, which suppressed the expression of NF-KB and ICAM-1, and led to the inhibition of retinal capillary degeneration. 展开更多

目的研究高山红景天口服液对瘀血阻滞证冠心病心绞痛患者症状的改善效果。方法方便选取2017年1月-2018年12月该院收治的瘀血阻滞证冠心病心绞痛患者208例,随机分为对照组和实验组各104例。对照组行常规治疗,实验组在对照组的基础上使用... 目的研究高山红景天口服液对瘀血阻滞证冠心病心绞痛患者症状的改善效果。方法方便选取2017年1月-2018年12月该院收治的瘀血阻滞证冠心病心绞痛患者208例,随机分为对照组和实验组各104例。对照组行常规治疗,实验组在对照组的基础上使用高山红景天口服液治疗,统计患者心绞痛发作情况及治疗效果,检测hsCRP、IL-18表达水平、冠状动脉主要供血动脉血流速度。结果治疗后实验组患者心绞痛发作频次(3.12±0.24)次/周、hs-CRP(3.99±0.12)mg/L、IL-18(22.45±2.46)μg/L、中医证候评分(4.25±0.57)分优于对照组患者心绞痛发作频次(5.15±0.68)次/周、hs-CRP(7.89±0.67)mg/L、IL-18(39.68±4.45)μg/L、中医证候评分(6.25±1.03)分,差异有统计学意义(t=28.712、58.431、34.559、17.331,P=0.001、0.001、0.001、0.001<0.05);心绞痛持续时间(2.31±0.06)min、心绞痛间隔时间(3.01±0.05)d短于对照组患者心绞痛持续时间(4.16±0.28)min、心绞痛间隔时间(5.12±0.24)d,差异有统计学意义(t=65.879、87.773,P<0.05);冠状动脉主要供血动脉血流速度,收缩期峰值速度(16.65±2.69)cm/s,舒张期峰值速度(31.28±5.69)cm/s、治疗总有效率(97.12%)高于对照组冠状动脉主要供血动脉血流速度,收缩期峰值速度:(14.36±1.45)cm/s,舒张期峰值速度:(28.77±4.56)cm/s、治疗总有效率(86.54%),差异有统计学意义(t=7.642、3.513,P=0.001、0.001<0.05;χ^2=7.751,P=0.005<0.05)。结论高山红景天口服液可有效缓解瘀血阻滞证冠心病心绞痛患者心绞痛症状、增加冠状动脉主要供血动脉血流速度,治疗效果显著。 展开更多

保元咀嚼片的制备及其抗疲劳作用评价

目的:筛选保元咀嚼片的制备工艺并初步探究其抗疲劳作用及机制。方法:以挥发油包合率、浸膏得率、人参皂苷和甘草酸等有效成分提取率等为指标,采用单因素和正交试验设计优选保元咀嚼片的挥发油包合工艺、水煎煮工艺和制剂成型工艺。ICR... 目的:筛选保元咀嚼片的制备工艺并初步探究其抗疲劳作用及机制。方法:以挥发油包合率、浸膏得率、人参皂苷和甘草酸等有效成分提取率等为指标,采用单因素和正交试验设计优选保元咀嚼片的挥发油包合工艺、水煎煮工艺和制剂成型工艺。ICR小鼠随机分为空白组、模型组、高山红景天口服液组(6.01 mL·kg^(-1))、保元咀嚼片低、中、高剂量(2.1、4.2、8.4 g·kg^(-1)),每组8只,每日按相应剂量灌胃给药1次,空白组和模型组给予等体积生理盐水,持续15 d,末次给药30 min后在小鼠尾部负重体质量5%的铅负荷,游泳至力竭建立疲劳模型,并记录各组小鼠负重游泳时间,同时对小鼠肌肉组织切片,苏木素-伊红(HE)染色并进行病理观察,测定血清中血尿素氮(BUN)、乳酸(LA)、肝糖原(LG)的水平,乳酸脱氢酶(LDH)和肌酸激酶(CK)的活性。结果:保元咀嚼片中肉桂挥发油的最佳包合工艺为主客比(β-环糊精-挥发油)10∶1,采用饱和水溶液法50℃包合2 h;最佳水提工艺为煎煮2次,首次加10倍水煎煮50 min,再次加9倍水煎煮40 min;保元汤提取物与微晶纤维素、麦芽糊精、蔗糖、甘露醇、柠檬酸、硬脂酸镁比例为63∶13∶8∶17∶17∶1∶1,采用湿法制粒压片,每片1.2 g,硬度为60~80 N。与模型组比较,保元咀嚼片低、中、高剂量组均能明显延长小鼠负重游泳至力竭的时间(P<0.05),提高机体的运动耐力,HE染色结果表明保元咀嚼片各剂量组均能明显改善由运动导致的肌肉组织损伤,显著降低血清BUN、LA的水平(P<0.01),和血清LDH、CK的活性(P<0.01),并明显提高LG含量(P<0.05,P<0.01)。结论:筛选所得保元咀嚼片制备工艺稳定可行,该制剂可通过增加肝组织中LG水平以提高运动耐力、加速清除体内的LA以缓解肌肉酸痛、降低CK和LDH活力以发挥抗疲劳作用。 展开更多
