目的 探讨circRNA CCND1对人肝癌HepG2细胞增殖、凋亡、侵袭和迁移的影响。方法 采用荧光原位杂交(FISH)实验及实时荧光定量聚合酶链反应(qRT-PCR)检测circRNA CCND1在HepG2细胞及L02细胞中的表达情况;进一步干扰及过表达circRNA CCND1... 目的 探讨circRNA CCND1对人肝癌HepG2细胞增殖、凋亡、侵袭和迁移的影响。方法 采用荧光原位杂交(FISH)实验及实时荧光定量聚合酶链反应(qRT-PCR)检测circRNA CCND1在HepG2细胞及L02细胞中的表达情况;进一步干扰及过表达circRNA CCND1后,通过qRT-PCR检测circRNA CCND1在HepG2细胞中的表达情况,CCK-8法和EDU法检测HepG2细胞增殖情况,流式细胞术检测细胞凋亡、细胞周期,Transwell实验检测细胞侵袭情况,划痕实验检测细胞迁移情况。结果 HepG2细胞circRNA CCND1相对表达量较LO2细胞高(P <0.05)。Si-circRNA CCND1组circRNA CCND1相对表达量较Control组和Si-NC组下降(P <0.05),Oe-circRNA CCND1组较Control组和Oe-NC组升高(P <0.05)。各组细胞培养0、24、48、72和96 h后细胞增殖活性比较,经重复测量设计的方差分析,结果:(1)不同时间点细胞增殖活性比较,差异有统计学意义(P <0.05);(2)各组细胞增殖活性比较,差异有统计学意义(P <0.05);(3)各组细胞增殖活性变化趋势比较,差异有统计学意义(P <0.05)。与Control组及Si-NC组比较,Si-circRNA CCND1组细胞凋亡率升高(P <0.05),G_(0)/G_(1)期比值升高(P <0.05),S及G_(2)/M期比值下降(P <0.05);与Control组及Oe-NC组比较,OecircRNA CCND1组细胞凋亡率下降(P <0.05),G_(0)/G_(1)期比值下降(P <0.05),S及G_(2)/M期比值升高(P <0.05)。Si-circRNA CCND1组细胞侵袭数、细胞迁移数较Control组、Si-NC组少(P <0.05),Oe-circRNA CCND1组较Control组及Oe-NC组多(P <0.05)。结论 circRNA CCND1能够促进人肝癌细胞HepG2细胞增殖、侵袭和迁移,抑制细胞凋亡。 展开更多

Background:Traditional Chinese medicine is promising for managing challenging and complex disorders,including cancer,and in particular,saffron is applied in treating various cancer types.However,its potential therapeutic efficacy and active components in managing squamous cell carcinoma of the head and neck(HNSCC)remain unclear yet.Methods:Using network pharmacology approaches,active ingredients of saffron,their target genes,and HNSCC-related genes were identified.Enrichment analyses were conducted for determining molecular functions and pathways enriched by genes that overlapped between the saffron target gene set and the HNSCC gene set.Among the four known active ingredients of saffron,crocetin was found to have the strongest inhibitory impact on HNSCC,based on the findings of cell viability and migration assays.Therefore,the potential target genes of crocetin in HNSCC cells were examined using molecular docking experiments and were confirmed by qPCR.Result s:Four active ingredients of saffron and 184 of their target genes were identified.Further,a total of 34 overlapping saffron-/HNSCC-associated targets related to the four active ingredients were screened,and crocetin was chosen for further investigation because it had the strongest inhibitory effect on HNSCC cells.Molecular docking experiments indicated that ESR1 and CCND1 were the target genes of crocetin.These results were confirmed through qPCR analysis,in which crocetin was found to lower the expression of the ESR1 and CCND1 genes in AMC-HN-8 and FaDu cells.Conclusion:According to our results,crocetin is a primary active anti-cancer component of saffron that may have potential in the development of novel HNSCC-treating medications.However,more thorough molecular research is necessary for confirming these results and elucidating the anti-cancer mechanism underlying saffron. 展开更多

Activity of bc1 complex kinase(ABC1K)is an atypical protein kinase(aPK)that plays a crucial role in plant mitochondrial and plastid stress responses,but little is known about the responses of ABC1Ks to stress in cotton(Gossypium spp.).Here,we identified 40 ABC1Ks in upland cotton(Gossypium hirsutum L.)and found that the Gh ABC1Ks were unevenly distributed across 17 chromosomes.The GhABC1K family members included 35 paralogous gene pairs and were expanded by segmental duplication.The GhABC1K promoter sequences contained diverse cis-acting regulatory elements relevant to hormone or stress responses.The qRT-PCR results revealed that most Gh ABC1Ks were upregulated by exposure to different stresses.Gh ABC1K2-A05 and Gh ABC1K12-A07 expression levels were upregulated by at least three stress treatments.These genes were further functionally characterized by virus-induced gene silencing(VIGS).Compared with the controls,the Gh ABC1K2-A05-and Gh ABC1K12-A07-silenced cotton lines exhibited higher malondialdehyde(MDA)contents,lower catalase(CAT),peroxidase(POD)and superoxide dismutase(SOD)activities and reduced chlorophyll and soluble sugar contents under NaCl and PEG stress.In addition,the expression levels of six stress marker genes(Gh DREB2A,Gh SOS1,Gh CIPK6,Gh SOS2,Gh WRKY33,and Gh RD29A)were significantly downregulated after stress in the Gh ABC1K2-A05-and Gh ABC1K12-A07-silenced lines.The results indicate that knockdown of Gh ABC1K2-A05 and Gh ABC1K12-A07 make cotton more sensitive to salt and PEG stress.These findings can provide valuable information for intensive studies of Gh ABC1Ks in the responses and resistance of cotton to abiotic stresses. 展开更多

Tazarotene-induced gene 1(TIG1)is induced by a derivative of vitamin A and is known to regulate many important biological processes and control the development of cancer.TIG1 is widely expressed in various tissues;yet in many cancer tissues,it is not expressed because of the methylation of its promoter.Additionally,the expression of TIG1 in cancer cells inhibits their growth and invasion,suggesting that TIG1 acts as a tumor suppressor gene.However,in some cancers,poor prognosis is associated with TIG1 expression,indicating its protumor growth characteristics,especially in promoting the invasion of inflammatory breast cancer cells.This review comprehensively summarizes the roles of the TIG1 gene in cancer development and details the mechanisms through which TIG1 regulates cancer development,with the aim of understanding its various roles in cancer development. 展开更多

Ethylene plays essential roles in plant growth,development and stress responses.The ethylene signaling pathway and molecular mechanism have been studied extensively in Arabidopsis and rice but limited in peanuts.Here,we established a sand-culture method to screen pingyangmycin mutagenized peanut lines based on their specific response to ethylene(“triple response”).An ethylene-insensitive mutant,inhibition of peanut hypocotyl elongation 1(iph1),was identified that showed reduced sensitivity to ethylene in both hypocotyl elongation and root growth.Through bulked segregant analysis sequencing,a major gene related to iph1,named AhIPH1,was preliminarily mapped at the chromosome Arahy.01,and further narrowed to a 450-kb genomic region through substitution mapping strategy.A total of 7014 genes were differentially expressed among the ACC treatment through RNA-seq analysis,of which only the Arahy.5BLU0Q gene in the candidate mapping interval was differentially expressed between WT and mutant iph1.Integrating sequence variations,functional annotation and transcriptome analysis revealed that a predicated gene,Arahy.5BLU0Q,encoding SNF1 protein kinase,may be the candidate gene for AhIPH1.This gene contained two single-nucleotide polymorphisms at promoter region and was more highly expressed in iph1 than WT.Our findings reveal a novel ethylene-responsive gene,which provides a theoretical foundation and new genetic resources for the mechanism of ethylene signaling in peanuts. 展开更多

Chronic enteropathy associated with the SLCO2A1 gene(CEAS)is a complex gastroenterological condition characterized by multiple ulcers in the small intestine with chronic bleeding and protein loss.This review explores the potential mechanisms underlying the pathogenesis of CEAS,focusing on the role of SLCO2A1-encoded prostaglandin transporter OATP2A1 and its impact on prostaglandin E2(PGE2)levels.Studies have suggested that elevated PGE2 levels contribute to mucosal damage,inflammation,and disruption of the intestinal barrier.The effects of PGE2 on macrophage activation and Maxi-Cl channel functionality,as well as its interaction with nonsteroidal anti-inflammatory drugs play crucial roles in the progression of CEAS.Understanding the balance between its protective and pro-inflammatory effects and the complex interactions within the gastrointestinal tract can shed light on potential therapeutic targets for CEAS and guide the development of novel,targeted therapies. 展开更多

目的探讨CCND1基因长短两种转录本及其蛋白产物cyclinD1在宫颈癌组织中的表达情况及对宫颈癌生物学行为的影响。方法采用实时定量RT-PCR和免疫组化S-P法检测CCND1基因两种转录本及CyclinD1蛋白在宫颈癌组织和正常组织中的表达。结果 CC... 目的探讨CCND1基因长短两种转录本及其蛋白产物cyclinD1在宫颈癌组织中的表达情况及对宫颈癌生物学行为的影响。方法采用实时定量RT-PCR和免疫组化S-P法检测CCND1基因两种转录本及CyclinD1蛋白在宫颈癌组织和正常组织中的表达。结果 CCND1基因的短转录本在正常组织和宫颈癌组织中相对表达量有统计学意义(P<0.05)而长转录本则无统计学意义(P>0.05);并且在宫颈癌组织中发现短转录本转录量在低分化组织中降低明显,有统计学意义(P<0.05);同时发现cyclinD1蛋白在宫颈癌组织中部分高表达(57.14%),在正常宫颈组织中仅在具有分化潜能的基底细胞中有所表达(13.33%)。结论 CCND1基因的短转录本对于宫颈癌可能存在保护作用,同时,CCND1基因的两种转录本的相对表达量的差异不会影响cyclinD1蛋白总的表达量。 展开更多

目的探讨细胞周期相关蛋白Cyclin D1、CDK4、p16蛋白及Ig H/CCND1融合基因在套细胞淋巴瘤(mantle cell lymphoma,MCL)中的表达及相互关系。方法采用荧光原位杂交(fluorescence in situ hybridization,FISH)法及免疫组化En Vision两步法... 目的探讨细胞周期相关蛋白Cyclin D1、CDK4、p16蛋白及Ig H/CCND1融合基因在套细胞淋巴瘤(mantle cell lymphoma,MCL)中的表达及相互关系。方法采用荧光原位杂交(fluorescence in situ hybridization,FISH)法及免疫组化En Vision两步法联合检测40例已利用基因重排技术及免疫组化确诊的MCL石蜡包埋组织(实验组)及20例淋巴结反应性增生的石蜡包埋组织(对照组),并利用对照组建立MCL的Ig H/CCND1融合基因阈值。结果实验组中Cyclin D1蛋白阳性率为100%,CDK4蛋白阳性率为87.50%,p16蛋白阳性率为17.50%;实验组中Ig H/CCND1融合基因的阳性率为100%;对照组中Ig H/CCND1融合基因均阴性。结论 MCL细胞周期调控中Cyclin D1-CDK4-p16通路关系符合肿瘤细胞周期调控原理的阐述;采用FISH技术建立检测Ig H/CCND1融合基因阈值,为国内该融合基因的FISH法判断提供依据。 展开更多

目的检测CCND1基因rs9344位点在东北地区汉族女性中的基因型分布,探讨CCND1基因多态性与东北地区汉族人群人乳头瘤病毒(HPV)感染及HPV感染后宫颈癌前病变发病风险的相关性。方法应用限制性片段长度多态法(PCR-RFLP)及测序方法,对东北地... 目的检测CCND1基因rs9344位点在东北地区汉族女性中的基因型分布,探讨CCND1基因多态性与东北地区汉族人群人乳头瘤病毒(HPV)感染及HPV感染后宫颈癌前病变发病风险的相关性。方法应用限制性片段长度多态法(PCR-RFLP)及测序方法,对东北地区汉族健康女性(n=533)和宫颈癌前病变CIN3患者(n=241)的CCND1基因rs9344位点的基因型进行检测。结果 CCND1基因rs9344位点不同基因型与东北地区汉族女性HPV感染风险无关,rs9344位点不同基因型并不影响非HPV感染者的CIN3患病风险,但HPV阳性者中,AA基因型对宫颈CIN3存在保护价值(OR=0.494;95%CI=0.255-0.955;P=0.036)。从整体来看,AA基因型降低宫颈CIN3的发病风险(OR=0.618;95%CI=0.390-0.980;P=0.041)。结论 CCND1基因rs9344位点多态性影响汉族女性对宫颈癌前病变CIN3的易感性,AA基因型具有保护价值。对于HPV感染后的人群来说,AA基因的保护价值更为显著。 展开更多

目的研究细胞周期素D1(Cyclin D1,CCND1)基因G870A多态性与肺癌(Lung cancer)危险性的关系。方法检索中国医学文献数据库和PubMed,获取所有关于CCND1基因G870A与肺癌危险性的病例-对照研究,使用Meta分析方法合并G870A与肺癌危险性... 目的研究细胞周期素D1(Cyclin D1,CCND1)基因G870A多态性与肺癌(Lung cancer)危险性的关系。方法检索中国医学文献数据库和PubMed,获取所有关于CCND1基因G870A与肺癌危险性的病例-对照研究,使用Meta分析方法合并G870A与肺癌危险性关系的OR值。同时进行异质性分析、亚组分析和发表偏倚检验。采用固定效应模型和随机效应模型进行分析。结果纳入符合标准的文献有8篇,共包括4 747例病例和4 512例对照,在显性模型下,G870A多态性能够增加罹患肺癌的风险(OR=1.12,95%CI=1.02~1.23)。种族分层分析发现,此危险在高加索人群中更为显著,而在亚洲人群中并未显著。结论 CCND1基因G870A多态性是肺癌的易感性位点,而且在高加索人群中更为显著。 展开更多

In this editorial we comment on the article by Wei et al,published in the recent issue of the World Journal of Clinical Oncology.The authors investigated the role of Transmembrane 9 superfamily member 1(TM9SF1)protein in bladder cancer(BC)carcinogenesis.Lentiviral vectors were used to achieve silencing or overexpression of TM9SF1 gene in three BC cell lines.These cell lines were then subject to cell counting kit 8,wound-healing assay,transwell assay,and flow cytometry.Proliferation,migration,and invasion of BC cells were increased in cell lines subjected to TM9SF1 overexpression.TM9SF1 silencing inhibited proliferation,migration and invasion of BC cells.The authors conclude that TM9SF1 may be an oncogene in bladder cancer pathogenesis. 展开更多

AIM To study the correlationship between the changes of p53, Waf1p21 and the cell proliferation determined by PCNA at different stages of human esophageal carcinogenesis. METHODS Biopsied and resected esophageal tissues from a high risk population for esophageal cancer in northern China were used in this study. All the specimens were fixed with 85% alcohol and further processed with routine histology. The avidin biotin peroxidase complex (ABC) method was used for the detection of p53, Waf1p21 and PCNA. RESULTS The strong nuclear staining for p53, Waf1p21 and PCNA was observed in the normal esophageal epithelium and the epithelia with different severities of lesions. As the lesions progressed to dysplasia (DYS) and to esophageal squamons cell carcinoma (SCC), the percentage of Waf1p21 immunoreactivity decreased. The number of Waf1p21 immunostaining positive cells increased slightly from normal to basal cell hyperplasia (BCH), but there was no further increase in DYS and in SCC. The total number of positive cells for Waf1p21 stain appeared to be lower than that of p53 in normal and BCH esophageal epithelia and much lower in DYS and SCC. The Waf1p21 positive immunostaining cells were located at the third and forth cell layers in half of the samples examined, which was 2~4 cell layers higher than that of PCNA and p53 in the same histological categories of normal, BCH and DYS. CLNCLUSION The low levels of Waf1p21 at the stage of DYS may be related to a functional loss of p53. Other mechanisms may also be responsible to the lack of Waf1p21 expression in DYS and SCC. 展开更多
