Objective:To evaluate the effect of Grifola frondosa polysaccharides(GFP)in a rat model of Alzheimer’s disease(AD).Methods:Seventy-five rats were divided into five groups:the normal control group and the AD group tre...Objective:To evaluate the effect of Grifola frondosa polysaccharides(GFP)in a rat model of Alzheimer’s disease(AD).Methods:Seventy-five rats were divided into five groups:the normal control group and the AD group treated with or without GFP(100,200,and 400 mg/kg).Behavioral responses in the open field test and elevated plus maze test were assessed.Additionally,the levels of malondialdehyde and ferric-reducing ability of plasma,and the mRNA expressions of TNF-α,IL-6,and IL-1βin the hippocampus were measured.Results:Treatment with GFP significantly improved AD-induced behavioral changes in the open field test and elevated plus maze test(P<0.05).In addition,the level of malondialdehyde and the mRNA expressions of TNF-α,IL-6,and IL-1βwere decreased by GFP treatment in a dose-dependent manner in AD rats(P<0.05),while the level of ferric-reducing ability of plasma was significantly increased(P<0.05).Conclusions:Oral administration of GFP can reduce inflammation and oxidative stress,as well as improve behavioral responses associated with AD,suggesting its potential use in AD treatment.However,additional studies are needed to elucidate its underlying mechanisms and efficacy.展开更多

The Box-Behnken design was used to determine the optimal time,temperature and solvent∶solid ratio for extracting intracellular polysaccharide from G.frondosa mycelium produced under submerged culture conditions.Appli...The Box-Behnken design was used to determine the optimal time,temperature and solvent∶solid ratio for extracting intracellular polysaccharide from G.frondosa mycelium produced under submerged culture conditions.Application of response surface methodology predicted a maximum polysaccharide extraction yield of 7.28% using an extraction time of 2.24 h,an extraction temperature of 92.53 ℃,and a water∶mycelium ratio of 35.76∶1.Under these conditions,the actual yield was 7.15%.Both low(200 mg/kg/d)and high(500 mg/kg/d)doses of G.frondosa polysaccharide,and positive controls administered 20 mg/kg/d cyclophosphamide,significantly inhibited the growth of S180 tumors with inhibition values of 61.5%,44.42% and 73.8%,respectively compared with negative controls administered physiological saline.展开更多
In this paper,the liquid seed medium of Grifola frondosa was optimized by single factor and orthogonal experiments using the shake flask culture method.The single factor experiments results showed that soybean powder ...In this paper,the liquid seed medium of Grifola frondosa was optimized by single factor and orthogonal experiments using the shake flask culture method.The single factor experiments results showed that soybean powder and maltose were the optimal nitrogen and carbon sources,and MgSO4 was the optimal inorganic salt.The orthogonal experiment results showed that the optimal combination of liquid seed culture medium for Grifola frondosa was:20%potato,0.75%soybean powder,5%maltose,0.1%KH2PO4,0.05%MgSO4,0.001%VB1.Using the optimized medium,shaking culture for 10 d under the inoculum amount of 5%,the temperature of 28℃,and the rotation speed of 150 r/min,the dry weight of mycelium exceeded 1.3228 g/100mL.展开更多
