BACKGROUND Dermal papillae(DP)and outer root sheath(ORS)cells play important roles in hair growth and regeneration by regulating the activity of hair follicle(HF)cells.AIM To investigate the effects of human mesenchymal stem cell-derived extracellular vesicles(hMSC-EVs)on DP and ORS cells as well as HFs.EVs are known to regulate various cellular functions.However,the effects of hMSC-EVs on hair growth,particularly on human-derived HF cells(DP and ORS cells),and the possible mechanisms underlying these effects are unknown.METHODS hMSC-EVs were isolated and characterized using transmission electron microscopy,nanoparticle tracking analysis,western blotting,and flow cytometry.The activation of DP and ORS cells was analyzed using cellular proliferation,migration,western blotting,and real-time polymerase chain reaction.HF growth was evaluated ex vivo using human HFs.RESULTS Wnt3a is present in a class of hMSC-EVs and associated with the EV membrane.hMSC-EVs promote the proliferation of DP and ORS cells.Moreover,they translocateβ-catenin into the nucleus of DP cells by increasing the expression ofβ-catenin target transcription factors(Axin2,EP2 and LEF1)in DP cells.Treatment with hMSC-EVs also promoted the migration of ORS cells and enhanced the expression of keratin(K)differentiation markers(K6,K16,K17,and K75)in ORS cells.Furthermore,treatment with hMSC-EVs increases hair shaft elongation in cultured human HFs.CONCLUSION These findings suggest that hMSC-EVs are potential candidates for further preclinical and clinical studies on hair loss treatment. 展开更多

Thus, the present study aimed to investigate the effect of Trichoxidil<sup>TM</sup>, a phytocomplex obtained from a blend of essential oils, in the treatment of hair loss caused by AGA. <strong>Methods:</strong> The CCD1072Sk cells were cultured for the 24-hour cell viability assessment and cytotoxicity of Trichoxidil<sup>TM</sup>. The expression of mRNA levels from KGF, IGF-1, and VEGF in fibroblasts was evaluated by RT-qPCR. Thirty-three volunteers, diagnosed with AGA, men and women, aged between 25 and 50 years, were divided into Control group, without treatment (n = 5);Trichosol<sup>TM</sup> vehicle group, without active (n = 5);Hydroalcoholic vehicle group, without active (n = 4);Trichosol<sup>TM</sup> vehicle group, with 2.5% minoxidil (n = 5);Hydroalcoholic vehicle group, with 2.5% minoxidil (n = 5);Trichosol<sup>TM</sup> group with 2.5% Trichoxidil<sup>TM</sup> (n = 5) and Hydroalcoholic vehicle group with 2.5% Trichoxidil<sup>TM</sup> (n = 4) to dermoscopic and histologic. <strong>Results:</strong> Fibroblasts exhibited higher proliferation when treated with higher concentrations of Trichoxidil<sup>TM</sup>. Trichoxidil<sup>TM</sup> significantly increased the expression of KGF, IGF-1, and VEGF mRNA in fibroblasts cells. Analysis of the capillary density showed that Trichoxidil<sup>TM</sup> associated with Trichosol<sup>TM</sup> vehicle, was the most effective association. In addition, it was observed an increased more effectively the percentage of anagen phase and reduction of the telogen when compared to other formulations. <strong>Conclusion:</strong> Trichoxidil<sup>TM</sup> promoted proliferative effects and positively modulated the expression of growth factors IGF-1, VEGF, and KGF, being a promising candidate for the treatment of hair loss caused by AGA. 展开更多

Hair loss can cause psychological distress.Here,red organic light-emitting diode(OLED)light source is first introduced as the photobiomodulation therapy(PBMT)for hair growth and demonstrated as a promising and non-invasive therapeutic modality for alopecia.OLED exhibits unique advantages of homogeneous irradiation,flexible in form factor,less heat generation.These features enable OLED to be an ideal candidate for wearable PBMT light sources.A systematic study of using red OLEDs to facilitate hair growth was conducted.The results show that OLEDs excellently promote hair regrowth.OLED irradiation can increase the length of the hair by a factor of 1.5 as compared to the control,the hair regrowth area is enlarged by over 3 times after 20 days of treatments.Moreover,the mechanism of OLED that stimulates hair follicle regeneration is investigated invivo by conducting a systematic controlled experiments on mice with or without OLED PBMT.Based on the comprehensive histological and immunofluorescence staining studies,two key factors are identified for red OLEDs to facilitate hair follicle regeneration:(i)increased autophagy during the anagen phase of the hair growth cycle;(ii)increased blood oxygen content promoted by the accelerated microvascular blood flow. 展开更多

In this work, the influence of phenol-enriched olive mill wastewater (OMWW) extract on hair growth was investigated <em>in vitro</em> on human follicle dermal papilla cells. OMWW has already shown great potential for use in skincare products, and its high polyphenol content is predestined to have a positive effect on hair growth. The studies included caffeine, a positive modulator of hair growth, and dihydrotestosterone, which causes hair loss <em>in vivo</em>, as controls. The impact of the investigated compounds on hair growth was evaluated by studies on cell viability and proliferation, the release of growth factors (insulin-like growth factor-1 and vascular endothelial growth factor), and the reduction of reactive oxygen species formation. OMWW showed a positive influence on the proliferation of the human follicle dermal papilla cells. Moreover, the extract leads to a significantly increased secretion of insulin-like growth factor-1, and a considerable reduction in reactive oxygen species formation was observed. Overall, our results show that the investigated phenol-enriched OMWW extract is a promising ingredient for hair care to improve hair growth, prevent hair loss due to oxidative stress and maintain a healthy scalp. 展开更多

Background and Objectives: Micrograft transplantation is accompanied by a transient induction of telogen in transplanted hair follicles (HF), which might be avoided by supporting the metabolic pathways of the micrograft during the ex vivo period. Vitamin B12 (cobalamin) has been suggested to influence HF growth and cycling in humans, but the mechanisms are unclear. Method: HFs were obtained from patients undergoing routine micrograft transplantation and were cultured for 5 days in Dulbecco’s modified Eagles Medium, supplemented with different amounts of vitamin B12. Hair shaft elongation (HSE) of the isolated HFs as well as quantitative changes of mRNA for beta-catenin, glykogensynthase kinase-3 (GSK-3) and TCF/Lef-1 in HF cells were determined. Results: In vitro HSE demonstrated a dose dependent induction of HSE after stimulation with 2.5 ug/ml and 25 ug/ml vitamin B12 (6.2 +/- 2.1% and 15.4 +/- 3.8% respectively). A dose dependent induction of beta-catenin-mRNA could be demonstrated in cultured HFs after stimulation with 2.5 ug/ml and 25 ug/ml vitamin B12 (fold change compared to DMEM: 9.5 +/- 2.7, p < 0.05 and 23.1 +/- 7.4, p < 0.01;respectively). Concomitantly the amounts of GSK-3 were significantly reduced after stimulation with 25 ug/ml vitamin B12 (fold change compared to DMEM: 0.76 +/- 0.12, p < 0.05). Conclusions: Our data demonstrate a hair growth promoting effect of vitamin B12 in vitro. This effect is accompanied by the modulation of intracellular signal transduction molecules of the wnt-pathway and might promote hair growth after micrograft transplantation. 展开更多

背景:在生物医学界,自噬是一个快速发展的研究热点,许多研究人员正在积极探究多种疾病中自噬的分子调控机制,但是自噬在毛发生长中所起的作用仍存在许多未知。目的:综述目前自噬在毛发生长与再生中的研究进展及应用价值,了解自噬在毛发... 背景:在生物医学界,自噬是一个快速发展的研究热点,许多研究人员正在积极探究多种疾病中自噬的分子调控机制,但是自噬在毛发生长中所起的作用仍存在许多未知。目的:综述目前自噬在毛发生长与再生中的研究进展及应用价值,了解自噬在毛发生长中的作用,以探究自噬在病理性脱发中的发病机制,为研究治疗脱发的药物提供新思路。方法:应用计算机在中国知网(CNKI)、PubMed数据库中,以“毛囊生长,毛发生长,毛发再生,自噬相关蛋白,自噬活性,自噬相关基因,自噬”或“hair growth,hair follicle,hair regeneration,autophagy”为检索词进行检索。查阅国内外近年来关于自噬、毛发生长和自噬在毛发生长中作用的研究进展进行总结,排除与文章主题内容不相符的文献,最终纳入78篇文献进行结果分析。结果与结论:(1)自噬是真核生物的正常代谢过程,具有复杂的分子机制和功能特性,既有利于细胞生存,又可促进细胞死亡。(2)脱发相关疾病与自噬活性的改变有关,且自噬活性可调控毛发生长周期;敲除或过表达自噬相关基因可改变毛发生长状态,已发现有多条自噬相关信号通路与毛囊生长有关;自噬的激活剂或抑制剂可用于治疗或预防脱发疾病。 展开更多

可变剪接(Alternative splicing,AS)是在转录后水平调控基因表达的一种重要方式。为挖掘调控绒山羊毛囊周期性生长相关的关键可变剪接,通过比较绒山羊绒毛不同生长时期的皮肤转录组数据,以P<0.05,|Inc‐LevelDifference|>0.1为标... 可变剪接(Alternative splicing,AS)是在转录后水平调控基因表达的一种重要方式。为挖掘调控绒山羊毛囊周期性生长相关的关键可变剪接,通过比较绒山羊绒毛不同生长时期的皮肤转录组数据,以P<0.05,|Inc‐LevelDifference|>0.1为标准筛选差异的可变剪接,并进行功能分析。结果显示,转换期Ⅰ、Ⅱ、Ⅲ、Ⅳ中分别鉴定到1615、1667、1387和1522个差异可变剪接,分别对应了1202、1268、1073、1170个基因;这些基因主要富集于Notch、Hedgehog、mTOR、AMPK、PI3K-Akt、Wnt、TGF-β等与毛囊发育相关的信号通路;蛋白互作网络结合富集分析表明,Ctnnb1、Gsk3b、Tgfbr1、Vegfa、Gli1等基因产生的可变剪接可能参与调控毛囊周期性生长。综上可知,可变剪接在绒山羊毛囊周期性生长过程中普遍存在,且在不同发育阶段间存在差异;鉴定到调控绒山羊毛囊周期性生长发育相关的关键差异可变剪接基因及其重要信号通路。 展开更多

目的 探讨氧化铈(CeO_(2))纳米酶对环磷酰胺所致斑秃小鼠模型毛发生长的影响。方法 将雌性C57BL/6J小鼠30只随机分为空白组、模型组和CeO_(2)纳米酶组(300μg/mL),每组10只。模型组和CeO_(2)纳米酶组小鼠采用单次腹腔内注射环磷酰胺注射... 目的 探讨氧化铈(CeO_(2))纳米酶对环磷酰胺所致斑秃小鼠模型毛发生长的影响。方法 将雌性C57BL/6J小鼠30只随机分为空白组、模型组和CeO_(2)纳米酶组(300μg/mL),每组10只。模型组和CeO_(2)纳米酶组小鼠采用单次腹腔内注射环磷酰胺注射液,构建斑秃小鼠模型。CeO_(2)纳米酶组小鼠给予300μg/mL CeO_(2)纳米酶外涂于小鼠背部剃毛区,空白组和模型组小鼠均予以等体积的PBS溶液外涂,1次/d,连续27 d。实验结束后(第28天)肉眼观察小鼠背部秃发区毛发生长情况并进行评分;HE染色小鼠背部经处理的皮肤组织计算单位视野内毛囊数量;酶联免疫吸附试验(ELISA)法检测小鼠血清细胞间黏附分子-1(ICAM-1)及内皮细胞白细胞黏附分子-1(ELAM-1)水平;水溶性四唑盐-1法(WST-1)检测皮肤组织丙二醛(MDA)及超氧化物歧化酶(SOD)含量。结果 与空白组相比,模型组小鼠毛发生长及毛发评分显著下降(P<0.05),毛囊数量明显减少(P<0.05),血清ICAM-1及ELAM-1水平升高(P<0.05),皮肤组织MDA含量升高而SOD含量降低(P<0.05);与模型组相比,CeO_(2)纳米酶组毛发生长及毛发评分显著增加(P<0.05),毛囊数量明显增多(P<0.05),血清ICAM-1及ELAM-1水平降低(P<0.05),皮肤组织MDA含量降低而SOD含量升高(P<0.05)。结论 外用CeO_(2)纳米酶对环磷酰胺所致斑秃小鼠模型具有促进毛发再生的作用,为CeO_(2)纳米酶外用治疗斑秃提供了实验依据。 展开更多

浓缩生长因子治疗雄激素性秃发的回顾性研究

目的:回顾性分析浓缩生长因子(concentrated growth factor,CGF)治疗雄激素性秃发(androgenetic alopecia,AGA)的疗效与安全性。方法:收集使用CGF治疗的60例AGA患者资料。12例为CGF治疗组,48例为联合治疗组。CGF组和联合治疗组均给予CG... 目的:回顾性分析浓缩生长因子(concentrated growth factor,CGF)治疗雄激素性秃发(androgenetic alopecia,AGA)的疗效与安全性。方法:收集使用CGF治疗的60例AGA患者资料。12例为CGF治疗组,48例为联合治疗组。CGF组和联合治疗组均给予CGF治疗,连续注射3次,联合组另给予米诺地尔酊外用,口服非那雄胺(男性)或螺内酯(女性)。通过毛发密度和毛囊单位密度改善程度、毛发镜图像特征变化、治疗前后照片以及医生和患者满意度评价综合评估有效性。同时,记录发生局部及全身不良反应的患者例数,以评估安全性。结果:CGF联合治疗组和CGF组治疗后的平均毛囊单位密度较治疗前分别增加了(18.04±11.15)和(8.81±5.77)个/cm 2(均P<0.05),平均毛发密度较治疗前分别增加了(36.43±21.18)和(16.19±12.62)根/cm 2(均P<0.05)。两组之间在改善毛发密度上差异具有统计学意义(P<0.05),在改善毛囊单位密度上差异无统计学意义(P>0.05)。两组毛囊周围褐色征、黄点征、空毛囊的治疗后改善率均在70%以上,但两组之间差异均无统计学意义(均P>0.05)。两组医生与患者治疗后满意率均在90%以上。60例患者中仅有2例局部注射部位疼痛明显,3例患者头皮瘙痒。结论:CGF治疗AGA安全有效,联合使用米诺地尔酊和非那雄胺/螺内酯,效果更佳。 展开更多

提出了超高效液相色谱-串联质谱法测定洗发水中西咪替丁、米诺地尔、美拉酮宁、香草酸甲酯、酮康唑、比马前列素、非那雄胺、塞替匹仑、尼鲁米特、氟罗地尔、螺内酯、氟他胺、拉坦前列素、度他雄胺等14种防脱育发类药物含量的方法。样... 提出了超高效液相色谱-串联质谱法测定洗发水中西咪替丁、米诺地尔、美拉酮宁、香草酸甲酯、酮康唑、比马前列素、非那雄胺、塞替匹仑、尼鲁米特、氟罗地尔、螺内酯、氟他胺、拉坦前列素、度他雄胺等14种防脱育发类药物含量的方法。样品经水溶解、乙腈提取后,用氨基固相萃取柱净化。以Waters Accquity UPLC BEH C18色谱柱为固定相,以乙腈-含0.1%(体积分数)甲酸的2 mmol·L^(-1)乙酸铵溶液为流动相体系进行梯度洗脱;分离后的14种目标物经电喷雾离子源正、负离子模式扫描,多反应监测模式检测。结果表明:14种目标物的质量浓度在1.0~50.0μg·L^(-1)内与对应的峰面积呈线性关系,检出限(3S/N)为0.05~3.0μg·kg^(-1);对空白样品进行3个浓度水平的加标回收试验,回收率为83.6%~99.0%,测定值的相对标准偏差(n=6)为5.6%~15%;方法用于10个洗发水样品分析,一个样品中检出米诺地尔、西咪替丁、塞替匹仑、酮康唑、尼鲁米特、氟罗地尔,另一个样品中检出美拉酮宁,检出量为0.4~47.8μg·kg^(-1)。 展开更多
