Preliminary observations on the result of infection by parvovirus of primary cultured cells of human hepatoma and parahepatoma tissue were reported. Human hepatoma and parahepatoma tissue were obtained by the culture method of tissue fragment plating. It was observed that their respective sensitivity to parvovirus H-1 was different. H-1 had inhibitory effect on epithelial cells of hepatoma tissue but without any significant effect on fibroblasts simultaneously, H-l had no effect on epithelial cells and fibroblasts of parahepatoma tissue. Further investigation of the relationship between parvovirus and hepatoma would be helpful not only in elucidating the mechanism of carcinogenesis and its suppression but also in the search of new approach for the treatment of hepatoma. 展开更多

The present study was to use liver cancer cell line to explore ... BACKGROUND: The oncogenesis of hepatocellular carcinoma(HCC) is not clear. The current methods of the pertinent studies are not precise and sensitive. The present study was to use liver cancer cell line to explore the bio-compatibility and cytotoxicity of ternary quantum dots(QDs) probe and to evaluate the possible application of QDs in HCC.METHODS: CuInS_2-ZnS-AFP fluorescence probe was designed and synthesized to label the liver cancer cell HepG 2. The cytotoxicity of CuInS_2-ZnS-AFP probe was evaluated by MTT experiments and flow cytometry. RESULTS: The labeling experiments indicated that CuInS_2-ZnS QDs conjugated with AFP antibody could enter HepG 2 cells effectively and emit intensive yellow fluorescence by ultraviolet excitation without changing cellular morphology. Toxicity tests suggested that the cytotoxicity of CuInS_2-ZnS-AFP probe was significantly lower than that of CdT e-ZnS-AFP probe(t test, F=0.8, T=-69.326, P〈0.001). For CuInS_2-ZnS-AFP probe, timeeffect relationship was presented in intermediate concentration(〉20%) groups(P〈0.05) and dose-effect relationship was presented in almost all of the groups(P〈0.05). CONCLUSION: CuInS_2-ZnS-AFP QDs probe had better biocompatibility and lower cytotoxicity compared with CdT e-ZnS-AFP probe, and could be used for imaging the living cells in vitro. 展开更多

Anticancer Effects of Crude Extract from Melia toosendan Sieb.et Zucc on Hepatocellular Carcinoma In Vitro and In Vivo 被引量:13

Objective: To investigate the anti-cancer effects of crude extract from Melia toosendan Sieb. et Zucc and its possible molecular mechanisms in vitro and in vivo. Methods: Transonic alcohol-chloroform extraction method was used to extract toosendanin from the bark of Melia toosendan Sieb. et Zucc, and the content of toosendanin in the crude extract was measured by high performance liquid chromatography (HPLC). Anti-cancer effects of crude extract from Melia toosendan Sieb. et Zucc were investigated in in vivo and in vitro studies. In the in vitro experiment, human hepatocellular carcinoma cell lines SMMC-7721 and Hep3B were co-incubated with toosendanin crude extract of different concentrations, respectively. In the in vivo experiment, BALB/c mice were subcutaneously inoculated with mouse hepatocellular carcinoma H22 cells and treated with crude extract. Results: HPLC revealed the content of toosendanin was about 15%. Crude extract from Melia toosendan Sieb. et Zucc inhibited cancer cells growth in a dose- and time-dependent manner. The 50% inhibitory concentration (IC50, 72 h) was 0.6 mg/L for SMMC-7721 cells and 0.8 mg/L for Hep3B cells. Both high-dose [0.69 mg/(kg·d)] and low-dose [0.138 mg/(kg·d)] crude extract could markedly suppress cancer growth, and the inhibition rate was greater than 50%. Hematoxylin and eosin staining showed necrotic area in cancers and transmission electron microscopy displayed necrotic and apoptotic cancer cells with apoptotic bodies. Immunohistochemistry showed that the expression of Bax and Fas increased and the expression of Bcl-2 reduced. Conclusions: Toosendanin extract has potent anti-cancer effects via suppressing proliferation and inducing apoptosis of cancer cells in vivo and in vitro. The mechanism of apoptosis involves in mitochondrial pathway and death receptor pathway. 展开更多
