血清CHI3L1水平与肝脏疾病之间的关系

目的 通过壳多糖酶3样蛋白1(chitinase 3-like protein 1,CHI3L1)在慢性乙型肝炎(chronic hepatitis B,CHB)、自身免疫性肝病(autoimmune liver disease, AILD)、肝硬化(liver cirrhosis, LC)、肝癌(hepatocellular carcinoma, HC)中的... 目的 通过壳多糖酶3样蛋白1(chitinase 3-like protein 1,CHI3L1)在慢性乙型肝炎(chronic hepatitis B,CHB)、自身免疫性肝病(autoimmune liver disease, AILD)、肝硬化(liver cirrhosis, LC)、肝癌(hepatocellular carcinoma, HC)中的水平,为临床监测肝脏疾病进展提供依据。方法 选取菏泽市立医院2022年6月至2023年12月健康体检者60例、慢性乙型肝炎患者60例、自身免疫性肝病患者60例、肝硬化患者60例、肝癌患者60例作为研究对象,应用酶联免疫吸附(ELISA)法检测所有研究对象血清中CHI3L1表达水平;应用流式细胞检测细胞因子表达水平;生化分析仪检测生化指标水平。结果 CHI3L1水平,健康体检组(47.00±20.50ng/mL)<CHB组(91.75±35.58ng/mL)<AILD组(128.30±41.92ng/mL)<LC组(131.29±46.52ng/mL)<HC组(154.07±59.92ng/mL)。以健康体检组为对照,CHI3L1诊断LC的ROC曲线显示,AUC为0.965;CHI3L1诊断HC的ROC曲线显示AUC为0.974。以慢性乙型肝炎组为对照,CHI3L1诊断肝硬化的值AUC为0.749(P<0.001),CHI3L1+AST诊断肝硬化AUC值为0.845(P<0.001),优于CHI3L1检测;以慢性乙型肝炎组为对照,AFP诊断HC的AUC为0.783,CHI3L1诊断HC的AUC为0.800(P<0.001);CHI3L1+AFP诊断HC的AUC为0.891,CHI3L1+AST+IL-17+IL-6诊断肝癌的AUC为0.937(P<0.001),优于CHI3L1和AFP检测。结论 血清CHI3L1表达水平在肝癌组中最高,慢性乙型肝炎组最低,血清CHI3L1表达水平随疾病进展升高;血清CHI3L1可作为辅助诊断肝硬化和肝癌的无创指标;血清CHI3L1、AST、IL-6、IL-17、IL-8、IFN-γ联合检测能够提高对肝硬化和肝癌的诊断。 展开更多

目的探讨慢性肾小球肾炎(CGN)患者血清壳多糖酶3样蛋白1(CHI3L1)、热休克蛋白60(HSP60)表达水平及其临床意义。方法选择97例CGN患者为CGN组,另选取体检健康者120例为健康组。对CGN组随访12个月,根据预后分为预后良好组(n=79)和预后不良... 目的探讨慢性肾小球肾炎(CGN)患者血清壳多糖酶3样蛋白1(CHI3L1)、热休克蛋白60(HSP60)表达水平及其临床意义。方法选择97例CGN患者为CGN组,另选取体检健康者120例为健康组。对CGN组随访12个月,根据预后分为预后良好组(n=79)和预后不良组(n=18)。采用酶联免疫吸附法检测血清CHI3L1、HSP60水平,采用全自动生化分析仪检测肾功能指标,分析CHI3L1、HSP60与肾功能指标的相关性。采用ROC分析CHI3L1、HSP60对CGN患者预后的预测价值。结果CGN组血清CHI3L1、HSP60高于健康组(P<0.05)。CGN组、预后不良组血尿素氮、血肌酐、血尿酸分别高于健康组、预后良好组(P<0.05)。Pearson相关分析结果显示,血清CHI3L1、HSP60与血尿素氮、血肌酐、血尿酸均呈正相关(P<0.001)。血清CHI3L1、HSP60联合预测CGN患者预后的效能最高(P<0.001)。结论CGN患者血清CHI3L1、HSP60水平升高,其可作为预测患者预后的潜在生物标志物。 展开更多

High-energy-density fuels are important for volume-limited aerospace vehicles,but the increase in fuel energy density always leads to poor cryogenic performance.Herein,we investigated the transposed Paternò-Büchi reaction of biomass cyclic ketone and cyclic alkene to synthesize a new kind of alkyl-substituted polycyclic hydrocarbon fuel with high energy density and good cryogenic performance.The triplet-energy-quenching results and phosphorescent emission spectra reveal the sensitization mechanism of the reaction,including photosensitizer excitation,triplettriplet energy transfer,cyclization,and relaxation,and the possible reaction path was revealed by the density functional theory(DFT)calculations.The reaction conditions of photosensitizer type and addition,molar ratio of substrates,reaction temperature,and incident light intensity were optimized,with the target product yield achieving 65.5%.Moreover,the reaction dynamics of the reaction rate versus the light intensity are established.After the hydrogenation-deoxygenation reaction,three fuels with a high density of 0.864-0.938 g·ml^(-1) and a low freezing point of<-55℃ are obtained.This work provides a benign and effective approach to synthesize high-performance fuels. 展开更多

Through the use of a survey and statistical methods, this study explores the effects and interventions of handheld Tai Chi water resistance fitness balls on the elderly with Parkinson’s disease. Firstly, a questionnaire on exercise compliance for patients with Parkinson’s disease was developed, and its reliability and validity were tested. Then, a survey was conducted to investigate the current status of exercise compliance among Parkinson’s disease patients, including general information, scoring status, and single and multiple factor analyses of influencing factors [1]. The results of the study show that through qualitative research, the dimensions and item pools of the questionnaire were initially constructed, and the reliability analysis of the questionnaire was conducted through Delphi expert consultation, with favorable results in terms of its reliability and validity [2]. Regarding the current status of exercise compliance among Parkinson’s disease patients, the study found that the level of exercise compliance needs improvement, and there are significant differences in exercise compliance levels among patients under different circumstances. Finally, the research results were discussed and conclusions were drawn. The innovation of this study lies in the development of a questionnaire on exercise compliance for patients with Parkinson’s disease and the preliminary qualitative research and Delphi expert consultation conducted on it, providing new ideas and methods for the study of exercise compliance. However, the study also has limitations as it did not examine the effects of other interventions on Parkinson’s disease, so further research should be conducted [3]. 展开更多

[Objectives]This study was conducted to explore active components and molecular biological mechanisms of Camellia nitidissima Chi in the treatment of premature ovarian failure(POF).[Methods]The active components and corresponding targets of C.nitidissima Chi were retrieved through literature and the TCMSP database.POF-related disease targets were identified using the OMIM and Genecards databases.A PPI network was constructed using the STRING database and Cytoscape 3.10.2.A herb-active component-target-pathway network diagram was also constructed with Cytoscape 3.10.2.The CytoNCA plugin was used to screen out the top five core targets and core active components.GO and KEGG enrichment analyses of the intersecting targets were performed using DAVID.Finally,molecular docking was conducted using Auto Dock to verify the interaction between core targets and active components,and visualization was done using PyMol.[Results]A total of 26 active components and 461 targets of C.nitidissima Chi were identified,with 154 intersecting targets related to POF.The core components of the herb included 1,1'-bi-2-naphthol,3',4-O-dimethylcedrusin,eriodictyol,quercetin,and vanillin.The PPI network revealed that the main targets were epidermal growth factor receptor(EGFR),protein kinase B1(AKT1),proto-oncogene Src(SRC),hypoxia-inducible factor 1α(HIF1A),and estrogen receptor 1(ESR1).KEGG enrichment analysis revealed 10 pathways closely related to POF,mainly involving the PI3K-Akt signaling pathway,chemical carcinogenesis-reactive oxygen species,endocrine resistance,and the HIF-1 signaling pathway.Molecular docking results showed that the core active components had strong binding activity with the targets.[Conclusions]C.nitidissima Chi has multi-component,multi-target,and multi-pathway characteristics in the comprehensive treatment of POF,providing informational support for its clinical application. 展开更多

Postoperative cognitive dysfunction is a seve re complication of the central nervous system that occurs after anesthesia and surgery,and has received attention for its high incidence and effect on the quality of life of patients.To date,there are no viable treatment options for postoperative cognitive dysfunction.The identification of postoperative cognitive dysfunction hub genes could provide new research directions and therapeutic targets for future research.To identify the signaling mechanisms contributing to postoperative cognitive dysfunction,we first conducted Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses of the Gene Expression Omnibus GSE95426 dataset,which consists of mRNAs and long non-coding RNAs differentially expressed in mouse hippocampus3 days after tibial fracture.The dataset was enriched in genes associated with the biological process"regulation of immune cells,"of which Chill was identified as a hub gene.Therefore,we investigated the contribution of chitinase-3-like protein 1 protein expression changes to postoperative cognitive dysfunction in the mouse model of tibial fractu re surgery.Mice were intraperitoneally injected with vehicle or recombinant chitinase-3-like protein 124 hours post-surgery,and the injection groups were compared with untreated control mice for learning and memory capacities using the Y-maze and fear conditioning tests.In addition,protein expression levels of proinflammatory factors(interleukin-1βand inducible nitric oxide synthase),M2-type macrophage markers(CD206 and arginase-1),and cognition-related proteins(brain-derived neurotropic factor and phosphorylated NMDA receptor subunit NR2B)were measured in hippocampus by western blotting.Treatment with recombinant chitinase-3-like protein 1 prevented surgery-induced cognitive impairment,downregulated interleukin-1βand nducible nitric oxide synthase expression,and upregulated CD206,arginase-1,pNR2B,and brain-derived neurotropic factor expression compared with vehicle treatment.Intraperitoneal administration of the specific ERK inhibitor PD98059 diminished the effects of recombinant chitinase-3-like protein 1.Collectively,our findings suggest that recombinant chitinase-3-like protein 1 ameliorates surgery-induced cognitive decline by attenuating neuroinflammation via M2 microglial polarization in the hippocampus.Therefore,recombinant chitinase-3-like protein1 may have therapeutic potential fo r postoperative cognitive dysfunction. 展开更多

Tai Chi,a practice that combines elements of both exercise and mindfulness,offers a wide range of health benefits.The body of evidence concerning the impact of Tai Chi on diabetes has recently been growing.This editorial aims to provide a concise summary of the current state of evidence for Tai Chi's effects on individuals with type 2 diabetes(T2D).The review includes 3 randomized controlled trials(RCTs)and 5 systematic reviews and meta-analyses,all of which investigate the effectiveness of Tai Chi on various health outcomes in individuals with T2D.Tai Chi demonstrates a significant effect to enhance glycemic control,lower blood pressure,improve serum lipid profiles,reduce insulin resistance,positively influence obesity-related indices,and improve overall quality of life in individuals with T2D.However,it is noteworthy that recent RCTs have reported inconsistent findings regarding the effects of Tai Chi on glycemic control and insulin resistance.The author also delves into potential mechanisms by which Tai Chi may exert its influence on the human body.Finally,the editorial highlights the critical issues that warrant further exploration in the future. 展开更多

目的:探讨2型糖尿病(DM)患者血清分泌型卷曲蛋白-4(SFRP-4)、壳多糖3样蛋白1(CHI3L1)水平与糖尿病视网膜病变(DR)的相关性。方法:前瞻性研究。选取2018-10/2023-10本院收治的DR患者103例为DR组,其中DR早期39例,DR中期42例,DR晚期22例;... 目的:探讨2型糖尿病(DM)患者血清分泌型卷曲蛋白-4(SFRP-4)、壳多糖3样蛋白1(CHI3L1)水平与糖尿病视网膜病变(DR)的相关性。方法:前瞻性研究。选取2018-10/2023-10本院收治的DR患者103例为DR组,其中DR早期39例,DR中期42例,DR晚期22例;选取单纯2型糖尿病患者98例为DM组,同期选取健康体检者101例为对照组。收集各组受试者基线资料及临床指标。采用酶联免疫吸附(ELISA)法检测血清SFRP-4、CHI3L1水平。结果:DR组患者血清SFRP-4、CHI3L1水平及TG、LDL-C、HbA1C、FPG、HOMA-IR均高于DM组和对照组(均P<0.05);DM组患者血清SFRP-4、CHI3L1水平及TG、LDL-C、HbA1C、FPG、HOMA-IR均高于对照组(均P<0.05)。DR晚期患者病程、TG、LDL-C、HbA1C、FPG、HOMA-IR、SFRP-4、CHI3L1均高于DR中期、DR早期(均P<0.05)。DR患者血清SFRP-4、CHI3L1水平与病程、TG、LDL-C、HbA1C、FPG、HOMA-IR、DR分期均呈正相关(均P<0.05)。血清SFRP-4、CHI3L1及联合诊断DR的曲线下面积(AUC)分别为0.809、0.801、0.898。血清SFRP-4、CHI3L1水平是影响DR发生的危险因素(P<0.05)。结论:血清SFRP-4、CHI3L1水平与2型糖尿病患者DR关系密切,二者水平升高提示患者发生DR的风险较高。 展开更多

目的制备黏膜免疫佐剂Chi-CpG NP,并检测其对重组B群脑膜炎奈瑟菌0315蛋白(rNMB0315)的免疫效果。方法用壳聚糖纳米颗粒(Chi NP)包裹免疫刺激分子(CpG-ODN)合成Chi-CpG NP,随后检测其理化性质和安全性。将不同黏膜免疫佐剂与rNMB0315充... 目的制备黏膜免疫佐剂Chi-CpG NP,并检测其对重组B群脑膜炎奈瑟菌0315蛋白(rNMB0315)的免疫效果。方法用壳聚糖纳米颗粒(Chi NP)包裹免疫刺激分子(CpG-ODN)合成Chi-CpG NP,随后检测其理化性质和安全性。将不同黏膜免疫佐剂与rNMB0315充分混合、吸附,滴鼻免疫雌性BALB/c小鼠,检测PBS组、rNMB0315组、rNMB0315+CpG组、rNMB0315+Chi NP组、rNMB0315+Chi-CpG NP组小鼠免疫42天后的体液免疫(IgG、IgG1、IgG2a、sIgA)、细胞免疫水平[白细胞介素(IL)-4、γ干扰素(IFN-γ)、IL-17A]、免疫保护效果以及血清体外杀菌活性。结果成功制备的Chi-CpG NP呈球形,大小均一,有较好的吸附性能和良好的安全性。小鼠免疫42天后,体液免疫和细胞免疫水平rNMB0315+Chi-NP组、rNMB0315+Chi-CpG NP组高于rNMB0315组(P<0.05);rNMB0315+Chi-CpG NP组高于rNMB0315+CpG组和rNMB0315+Chi-NP组(P<0.05)。致死剂量B群脑膜炎奈瑟菌攻击各组小鼠后,rNMB0315+Chi-CpG NP组小鼠存活率最高;血清体外杀菌抗体滴度为1∶4。结论成功制备了Chi-CpG NP。Chi-CpG NP能增强rNMB0315蛋白的免疫效果,提高小鼠的存活率。 展开更多

为研究白三叶类黄酮合成途径的关键基因TrCHI在白三叶(Trifolium repens)花色形成中的作用,以白三叶野生型和红花突变体为材料,通过同源克隆得到TrCHI基因并进行转录分析和CHI酶活性测定。结果表明,白三叶野生型TrCHI基因开放阅读框长66... 为研究白三叶类黄酮合成途径的关键基因TrCHI在白三叶(Trifolium repens)花色形成中的作用,以白三叶野生型和红花突变体为材料,通过同源克隆得到TrCHI基因并进行转录分析和CHI酶活性测定。结果表明,白三叶野生型TrCHI基因开放阅读框长660 bp,编码219个氨基酸,其红花突变体TrCHI开放阅读框长度为669 bp,编码222个氨基酸。7处氨基酸突变包括:第13号位的谷氨酸突变成天冬氨酸,第75号位的谷氨酸突变成谷氨酰胺,第154号位的丝氨酸突变成苏氨酸,第218号位的赖氨酸突变成精氨酸,并在末尾增加了甘氨酸、蛋氨酸和赖氨酸。进化分析发现,CHI基因进化具有物种保守性,Tr CHI属于Ⅱ型CHI。花瓣、根和叶柄中,红花突变体TrCHI转录水平显著高于野生型;但在茎、叶和花梗中,红花突变体TrCHI基因转录水平低于野生型。突变体花瓣中CHI酶活性极显著高于野生型(P<0.001)。推测白三叶突变体的红花表型与TrCHI基因突变并对白三叶中的类黄酮化合物的合成调控有关。本研究为进一步研究TrCHI在白三叶上的潜在功能提供了一定的价值。 展开更多

血清CRP、CHI3L1联合彩超对血栓闭塞性脉管炎的诊断价值

目的分析血清C反应蛋白(CRP)、几丁质酶3样蛋白1(CHI3L1)联合彩超对血栓闭塞性脉管炎的诊断价值。方法选取2020年10月至2022年9月在我院诊治的60例血栓闭塞性脉管炎患者作为观察组,及同期60例健康体检志愿者作为对照组进行研究。受试者... 目的分析血清C反应蛋白(CRP)、几丁质酶3样蛋白1(CHI3L1)联合彩超对血栓闭塞性脉管炎的诊断价值。方法选取2020年10月至2022年9月在我院诊治的60例血栓闭塞性脉管炎患者作为观察组,及同期60例健康体检志愿者作为对照组进行研究。受试者均行彩超检查,酶联免疫吸附(ELISA)法测定血清CRP、CHI3L1水平。相关性采用Pearson法分析;受试者工作特征(ROC)曲线分析血清CRP、CHI3L1水平对血栓闭塞性脉管炎的诊断价值;采用四格表分析血清CRP、CHI3L1水平联合彩超对血栓闭塞性脉管炎的诊断价值。结果与对照组相比,观察组血栓闭塞性脉管炎患者血清CRP、CHI3L1水平均显著升高(P<0.05)。CRP与CHI3L1呈正相关(r=6.396,P<0.001);血清CRP、CHI3L1诊断血栓闭塞性脉管炎的曲线下面积(AUC)分别为0.897、0.833;彩超血栓闭塞性脉管炎的诊断符合率为76.67%;血清CRP、CHI3L1联合彩超诊断血栓闭塞性脉管炎的灵敏度为85.25%、特异性为86.44%、准确度为85.83%,其诊断效能高于彩超、血清CRP、CHI3L1单独检测。结论血栓闭塞性脉管炎患者彩超表现明显,对血栓闭塞性脉管炎具有一定的诊断价值,联合血清CRP、CHI3L1检测可明显提高血栓闭塞性脉管炎的诊断效能。 展开更多

目的:观察川黄方治疗3~4期慢性肾脏病(CKD)患者的临床疗效,及对急性肾损伤(AKI)标志物尿L-FABP、CHI3L1的影响。方法:将3~4期CKD患者92例,按照1∶1的比例随机分到治疗组或对照组,即每组46例。对照组给予内科基础治疗;治疗组在对照组基... 目的:观察川黄方治疗3~4期慢性肾脏病(CKD)患者的临床疗效,及对急性肾损伤(AKI)标志物尿L-FABP、CHI3L1的影响。方法:将3~4期CKD患者92例,按照1∶1的比例随机分到治疗组或对照组,即每组46例。对照组给予内科基础治疗;治疗组在对照组基础上加服川黄方。疗程为12周。进行临床疗效评价,并观察中医证候积分变化。检测治疗前后肾功能(Scr、BUN、UA、eGFR),24 h U-pro,尿L-FABP、尿CHI3L1变化。结果:(1)治疗组总有效率为73.9%,对照组43.5%,治疗组高于对照组(P<0.05)。(2)用药后,两组患者肾Scr、BUN、UA均有一定程度的降低,eGFR均有一定程度的升高,肾功能得到了改善。治疗组更明显(P<0.01)。两组患者UA差异无统计学意义(P>0.05)。(3)治疗后两组患者24 h U-pro均有降低,治疗组更显著(P<0.01)。(4)两组患者尿L-FABP、CHI3L1治疗前后比较,差异均有统计学意义(P<0.01)。治疗组降低更明显。(5)治疗后,两组患者中医证候积分均显著降低(P<0.01),说明两种治疗方法均有利于改善患者临床症状。结论:川黄方联合内科基础治疗的临床疗效优于单纯内科基础治疗,能更好地促进3~4期CKD患者肾功能恢复、延缓CKD进展,并能显著降低患者尿L-FABP和尿CHI3L1。 展开更多
