The catalytic effects of the organic compounds of iron,tin and manganese on the degradation of low density polyethylene (LDPE) at compost temperature are discussed.A series of samples were aged in a simulating compost environment.The mechanical properties,viscosity average molecular weight (M η) of PE and hydroperoxide (POOH) concentration in the samples were measured.FT IR and DSC were also applied to characterize some samples.It was shown that the above mentioned metallic organic compounds can catalyze the degradation of LDPE efficiently.After 2 months aging,all samples with catalysts became fragile and the M η of the material decreased dramatically.Furthermore,the concentration of carbonyl and the degree of crystallinity of the material increased with the aging time. 展开更多

In this work,a series of acidic montmorillonite/cordierite monolithic catalysts were prepared by a coating method using silica sol as the binder.The morphology and structure of the acidic montmorillonite/cordierite samples were characterized by means of X-ray diffraction(XRD),N_2 adsorption/desorption isotherms,and scanning electron microscope(SEM).The cleavage of cumene hydroperoxide(CHP) in a conventional fixed-bed reactor was chosen as a model reaction to evaluate the catalytic activity of the monolithic catalysts.The influences of acidic montmorillonite loading,reaction temperature.CHP concentration,and weight hourly space velocity(WHSV) on the catalytic activity and selectivity of phenol were studied.The results indicated that the obtained acidic montmorillonite/cordierite monolithic catalysts were firm and compact,and the loading of acidic montmorillonite was found to reach 40%(by mass) after three coating operations.The surface area of acidic montmorillonite/cordierite catalysts increases greatly as acidic montmorillonite loading increases due to higher surface area of acidic montmorillonite.Under the optimal reaction conditions(acidic montmorillonite loading of 32.5%(by mass),temperature of 80 ℃,a mass ratio of CHP to acetone of 1:3,and WHSV of CHP of 90 h^(-1)),the conversion of CHP can reach 100%,and the selectivity of phenol is up to 99.8%. 展开更多

Homogeneous oxidation using an oil-soluble oxidant, tert-amyl hydroperoxide (TAHP), for ultra-deep desulfurization was performed under mild conditions in the presence of molybdenum oxide catalysts. Dibenzothio- phene (DBT), benzothiophene (BT) and 4, 6-dimethyl-dibenzothiophene (DMDBT), which are the refractory sulfur compounds for hydrodesulfurization (HDS), were employed as model substrates for a simulated diesel fuel. Activity of molybdenum oxide supported on a macroporous weak acidic resin was investigated. The mass conversion of DBT reached near 100% at 90℃ and a TAHP/DBT molar ratio of 3 with 1% of molybdenum oxide supported on Amberlite IRC-748 resin for 1 h. It was further found that the activities of the model substrates decreased in the or- der of DMDBT 〉DBT 〉BT. In the flow oxidation using TAHP as the oxidant, mass conversion of DBT increased remarkably from 61.3% to 98.5% when dropping the weight hourly space velocity (WHSV) from 40 h^-1 to 10 h^-1. A series of experiments dealt with selectivity of this oxidation using TAHP revealed that the model unsaturated compounds, i.e. 4, 6, 8-trimethyl-2-nonylene, and 2-methylnaphthalene did not affect the oxidation of DBT. Carbazole had nearly no effect on the conversion of DBT using TAHP, but had some influence on the one using tert-butyl hydroperoxide (TBHP). The mass conversion of DBT decreased remarkably from 75.2% to 3.6% when the content of carbazole increased from 0 to 500μg·g^-1. In the flow oxidation using TAHP as the oxidant, the concentration of DBT in model fuels was reduced from 500 μg·g^-1 to 7.2 μg·g^-1 at WHSV of 10 h^-1, and then reduced to 3.8 μg·g^-1 by adsorntion of Al2O3. 展开更多

AIM: To investigate catalase (KatA) and alkyl hydroperoxide reductase (AhpC) antibodies of Helicobacter pylori as biomarkers for gastric cancer (GC). METHODS: This study included 232 cases and 264 controls. Recombinant KatA and AhpC proteins were constructed and the levels of antibodies were tested by indirect enzyme-linked immunosorbent assay (ELISA). Logistic regression was applied to analyze the relationships between KatA, AhpC and GC. The chi(2) trend test was used to evaluate the dose-response relationships between serum KatA and AhpC antibody levels and GC. Receiver operating characteristic (ROC) curve was used to evaluate the screening accuracy of KatA and AhpC as biomarkers. Combined analysis was used to observe screening accuracy of predictors for GC. RESULTS: In all subjects, the association between KatA and AhpC and GC risk was significant (P < 0.001) with odds ratio (OR) = 12.84 (95%CI: 7.79-21.15) and OR = 2.4 (95%CI: 1.55-3.73), respectively. KatA and AhpC antibody levels were strongly related to GC risk with a dose-dependent effect (P for trend < 0.001). The area under the ROC (AUC) for KatA was 0.806, providing a sensitivity of 66.81% and specificity of 86.36%; and the AUC for AhpC was 0.615, with a sensitivity of 75.65% and specificity of 45.49%. The AUC was 0.906 for KatA and flagella protein A (FlaA) combined analysis. CONCLUSION: Serum KatA and AhpC antibodies are associated with GC risk and KatA may serve as a biomarker for GC. KatA/FlaA combined analysis improved screening accuracy. 展开更多

In this manuscript, a series of catalyst SG n-[VVO2-PAMAM-MSA] (SG silica gel, PAMAM polyamidoamine, MSA 5-methyl salicylaldehyde, n=0, 1, 2, 3) was prepared and their structures were fully characterized by Fourier transform-infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), X-ray photoelectron spectroscopy (XPS) and inductive coupled plasma emission spectrometer (ICP) etc. XPS revealed that the metal V and SG n-PAMAM-MSA combined more closely after the formation of Schiff base derivatives. Their catalytic activities for oxidation of dibenzothiophene were evaluated using tert-butyl hydroperoxide as oxidant. The results showed that the catalyst SG 2.0-[VVO2-PAMAM-MSA] presented good catalytic activity and recycling time. Meanwhile, the optimal condition for the catalytic oxidation of SG 2.0-[VVO2-PAMAM-MSA] was also investigated, which showed that when the oxidation temperature was 90 °C, time was 60 min, the O/S was 3:1, and the mass content of catalyst was 1%, the rate of desulfurization could reach 85.2%. Moreover, the catalyst can be recycled several times without significant decline in catalytic activity. 展开更多

The combination of relatively low-cost ionic liquids, simple copper salt, and terminal oxidant tert-butyl hydroperoxide provided an efficient and environmentally friendly approach to the preparation of ionone-like dienones. Six pyridinium ionic liquids were evaluated in allylic oxidation of α-ionone and β-ionone. The 60%-70% yields of 3-oxo-α-ionone were obtained with 0.02 0.20 mmol of CuCl2·2H2O as catalyst, 3-5 mmol of tert-butyl hydroperoxide as oxidant and 1 g of [Bpy]PF6 as solvent for 4-20 h at 60℃. The facile recovery and recycle of catalyst were also achieved. More significantly, peculiar phase behaviors of [Bpy]PF6 and [Epy]PF6 offered the catalytic system advantages of homogeneous reaction and heterogeneous separation. Scanning electron microscope (SEM) images of [Bpy]PF6 provided evidences for the behaviors. Transmission electron microscope (TEM) micrographs showed copper salt nanoparticles catalyst formed and stabilized in pyridinium ionic liquids. 展开更多

The potential of carrying out oxidative desulfurization(ODS) using oxygen as an oxidant was explored in this work. n-Octane firstly reacted with oxygen to produce hydroperoxides in-situ, which were then used as oxidants to oxidize the dibenzothiophene(DBT) in the absence of catalysts. The hydroperoxides generated in-situ were effective in oxidizing DBT to its corresponding dibenzothiophene sulfone(DBTO_2) which was characterized by FT-IR and ~1H-NMR. The removal rate of DBT could reached 98.4% under conditions covering a temperaure of 140℃, a rection duration of 4 h, and an oxygen partial pressure of 0.4 MPa. The influences of different hydrocarbon components in diesel on DBT removal were investigated. The results showed that cyclohexane and n-dodecane had no effect on the removal of DBT, but xylene had a slight negative effect on DBT removal. A possible oxidation mechanism was proposed and the concentration of hydroperoxides in both O_2-oxidized octane and model diesel were detected. 展开更多

AIM: To identify alkyl hydroperoxide reductase subunit C(AhpC) homologs in Bacillus subtilis(B. subtilis) and to characterize their structural and biochemical properties. AhpC is responsible for the detoxification of reactive oxygen species in bacteria.METHODS: Two AhpC homologs(AhpC_H1 and AhpC_H2) were identified by searching the B. subtilis database; these were then cloned and expressed in Escherichia coli. AhpC mutants carrying substitutions of catalytically important Cys residues(C37S, C47 S, C166 S, C37/47 S, C37/166 S, C47/166 S, and C37/47/166 S for AhpC_H1; C52 S, C169 S, and C52/169 S for AhpC_H2) were obtained by site-directed mutagenesis and purified, and their structure-function relationship was analyzed. The B. subtilis ahp C genes were disrupted by the short flanking homology method, and the phenotypes of the resulting AhpC-deficient bacteria were examined.RESULTS: Comparative characterization of AhpC homologs indicates that AhpC_H1 contains an extra C37, which forms a disulfide bond with the peroxidatic C47, and behaves like an atypical 2-Cys AhpC, while AhpC_H2 functions like a typical 2-Cys AhpC. Tryptic digestion analysis demonstrated the presence of intramolecular Cys37-Cys47 linkage, which could be reduced by thioredoxin, resulting in the association of the dimer into higher-molecular-mass complexes. Peroxidase activity analysis of Cys→Ser mutants indicated that three Cys residues were involved in the catalysis. AhpC_H1 was resistant to inactivation by peroxide substrates, but had lower activity at physiological H2O2 concentrations compared to AhpC_H2, suggesting that in B. subtilis, the enzymes may be physiologically functional at different substrate concentrations. The exposure to organic peroxides induced AhpC_H1 expression, while AhpC_H1-deficient mutants exhibited growth retardation in the stationary phase, suggesting the role of AhpC_H1 as an antioxidant scavenger of lipid hydroperoxides and a stress-response factor in B. subtilis. CONCLUSION: AhpC_H1, a novel atypical 2-Cys AhpC, is functionally distinct from AhpC_H2, a typical 2-Cys AhpC. 展开更多

Lipids oxidation, histamine production and quality loss were studied according to storage time and temperature (ambient temperature (Tamb) 26 ℃, 4 ℃ and -18℃) in Atlantic mackerel (Scomber scombrus). Muscle pH, hydrolysis of phospholipids, content of primary (hydroperoxides), secondary lipid oxidation products (TBARS) and histamine were determined and compared with a sensory assessment. Atlantic mackerel is sensory acceptable, less than 24 hours at Tamb, for up to 3 days at 4 ℃ and 3 months at-18 ℃. Evolution of biochemical parameters with storage time and temperature showed significant differences (P 〈 0.05). Muscle pH increased from 5.99 to 6.13 at Tamb, to 6.23 at 4 ℃ and to 6.04 at -18 ℃. The highest content of TBARS is associated with a decrease in phospholipids and hydroperoxides contents and highest levels of sensory alteration. Histamine content exceeded the limit recommended by the Trade Algerian Ministry (10 mg/100g), after 24 hours at Tamb, 5 days of storage at 4 ~C only. Therefore, freezing storage has a preserving effect on lipid damage and histamine production and seems the best means of storage; if these species are not consumed during the two days following capture. Moreover, monitoring histamine production is more useful as sanitary index rather than spoilage parameter and the strategy used for measuring kinetic of lipid oxidation appear pertinent for determining the degree of oxidation. 展开更多

Flavonoids are a major component in the traditional Chinese medicine Radix Ilicis Pubescentis.Previous studies have shown that the administration of Radix Ilicis Pubescentis total flavonoids is protective in cerebral ischemia.However,to our knowledge,no studies have examined whether the total flavonoids extracted from Radix Ilicis Pubescentis prevent or ameliorate neuronal damage following transient ischemic attacks.Therefore,Radix Ilicis Pubescentis total flavonoids question and the potential underlying mechanisms.Thus,beginning 3 days before the induction of a mouse model of transient ischemic attack using tert-butyl hydroperoxide injections,mice were intragastrically administered 0.3,0.15,or 0.075 g/kg of Radix Ilicis Pubescentis total flavonoids daily for 10 days.The results of spectrophotometric analyses demonstrated that Radix Ilicis Pubescentis total flavonoids enhanced oxygen free radical scavenging and reduced pathological alterations in the brain.Hematoxylin-eosin staining results showed that Radix Ilicis Pubescentis total flavonoids reduced hippocampal neuronal damage and cerebral vascular injury in this mouse model of transient ischemic attack.These results suggest that the antioxidant effects of Radix Ilicis Pubescentis total flavonoids alleviate the damage to brain tissue caused by transient ischemic attack. 展开更多

Five fucoidan fractions from Laminaria japonica with different sulfate content and molecular weight were prepared by anion-exchange chromatography and mild acid hydrolysis. Their antioxidant effects on azo radicals 2-2'-Azobis (2-amidinopropane) dihydrochloride (AAPH) induced oxidation of human low-density lipoprotein (LDL) were evaluated by monitoring cholesteryl ester hydroperoxides (CHL-OOH) formation kinetics through reverse-phase high performance liquid chromatography (RP-HPLC) analysis. Fucoidan F - C with a low molecular mass of 2 000 ~8 000 and a sulfate content of 24.3% had much stronger protective antioxidant effect than other four fucoidan fractions on the hydrophilic radical AAPH induced LDL oxidation. However, the highly sulfated fucoidan fraction L - B with a molecular mass of 20 000 was completely ineffective in protecting LDL from the AAPH induced oxidation. The results suggested that both molecular mass and sulfate content of fucoidan played very important roles in their effects on the AAPH induced LDL oxidation. 展开更多

Phospholipid hydroperoxide glutathione peroxidase is an antioxidant enzyme that has the highest capability of reducing membrane-bound hydroperoxy lipids as compared to free organic and inorganic hydroperoxides amongst the glutathione peroxidases.In this study,urea-induced effects on the inactivation and unfolding of a recombinant phospholipid hydroperoxide glutathione peroxidase(PHGPx)from Oryza sativa were investigated by means of circular dichroism and fluorescence spectroscopy.With the increase of urea concentration,the residual activity of OsPHGPx decreases correspondingly.When the urea concentration is above 5.0 mol/L,there was no residual activity.In addition,the observed changes in intrinsic tryptophan fluorescence,the binding of the hydrophobic fluorescence probe ANS,and the far UV CD describe a common dependence on the concentration of urea suggesting that the conformational features of the native OsPHGPx are lost in a highly cooperative single transition.The unfolding process comprises of three zones:the native base-line zone between 0 and 2.5 mol/L urea,the transition zone between 2.5 and 5.5 mol/L urea,and the denatured base-line zone above 5.5 mol/L urea.The transition zone has a midpoint at about 4.0 mol/L urea. 展开更多

The results showed that PSK could enhance the gene expression of SeGSHPx in macrophages,this corresponded with our previous work that PSK could increase the activity of SeGSHPx in macrophages. Small dosage of tbOOH had no marked effect on the gene expression of SeGSHPx in macrophages from mice treated by PSK,but could significantly induce the gene expression of SeGSHPx in macrophages from non-PSK treated mice. 展开更多

The results showed that MMP decreased when macrophageswere treated by Ox-LDL. If LOOH in Ox-LDL was pre cleared by ebselen plus GSH. the decreased MMP could be recovered by about 20 %. Lipid moiety alone had no effect on IMP, but protein moiety could cause decrease ofMMP, the extent of the decrease was equivalent to that caused by Ox-LDL in which LOOH was pre-cleared by ebselen plus GSH. 展开更多

The family of glutathione peroxidases encompasses, as far, three tetrameric glutathione'peroxidases (GPx) and the monomeric PHGPx. Although the overall homology between tetrameric enzymes and PHGPx is less than 30%, a pronounced similarity has been detected on clusters involved in the active site and a common catalytic triad (selenocysteine glutamine and tryptophan) has been defined by structural and kinetic data.A major peculiar feature of the reaction catalyzed by PHGPx is the possibility to accommodate large lipophilic substrates. This accounts for the observed dramatic antiperoxidant effect and the synergism with vitamin E.Moreover, the reduction of lipid hydroperoxides accounts also for the observed modulation of cycloxygenase and inhibition of 15-lipoxygenase.On the other hand, structural and kinetic data indicate that also the specificity of PHGPx for the donor substrate is not restricted to GSH and the recent observation the PHGPx binds to specific mitochondrial proteins, from which it is released by ionic strength and thiols, suggests a possible fole of this seleooenzyme'in catalyzing the specific oxidation of protein thiols,thus modulating the activity of cellular regulatory elements. on this light, the selenium mojety of PHGPx, reacting much faster that thiols with a peroxide, and then oxidizing specific protein thiols, would channel the oxidation toward protein targets, thus providing, by protein-protein interaction, the specificity of the redox transition 展开更多

Does Ethanol Play a Pro-Oxidant Role during Oxidative Stress in the Liver?

Oxidative stress has been implicated in the pathophysiology of liver injury during xenobiotic and alcohol metabolism, ischemia/reperfusion injury. In this study we examined if ethanol acted as a pro-oxidant making cells become more sensitive to tert-butylhydroperoxide (tBH) killing. Cell viability was determined in a rat hepatoma cell line (FTO2B) and rat primary hepatocytes in culture in the presence or absence of ethanol pretreatment. To elucidate the contribution of labile iron, deferoxamine (DF, an iron chelator) or lipid free radicals, N,N-diphenyl-p-phenylenediamine (DPPD, a lipid scavenger) were added to the ethanol tBH co-treatment. The levels of glutathione (GSH) and glutathione disulfide (GSSG) in the hepatocytes were also measured. Ethanol treatment (both pretreatment and co-treatment during the 3-hr tBH exposure) increased cell killing dramatically in both FTO2B cells and primary rat hepatocytes. Both DF and DPPD decreased ethanol-enhanced tBH cell killing in hepatocytes. These results demonstrated that co-treatment of FTO2B cells and primary rat hepatocytes with ethanol and tBH increased cell killing. The GSH level was dramatically reduced while GSSG level rose. Both DFP and DPPD reversed or protected the cells from this insult, indicating that ethanol was a pro-oxidant. 展开更多
