大量研究证实“微生物-肠-脑轴”在抑郁症的发生发展中发挥重要作用,而肠道微生物产生的短链脂肪酸(short chain fatty acids,SCFAs),更是参与神经调控作用的关键分子。该研究首次采用SCFA-酰化淀粉在大肠定点释放5类短链脂肪酸,探究其... 大量研究证实“微生物-肠-脑轴”在抑郁症的发生发展中发挥重要作用,而肠道微生物产生的短链脂肪酸(short chain fatty acids,SCFAs),更是参与神经调控作用的关键分子。该研究首次采用SCFA-酰化淀粉在大肠定点释放5类短链脂肪酸,探究其对慢性应激诱导的抑郁症状的缓解及机制。以雄性C57BL/6 J小鼠为研究对象,通过为期5周的慢性不可预知温和应激构建抑郁小鼠模型,通过旷场实验、高架十字迷宫实验和强迫游泳实验对小鼠的抑郁样行为进行评价。同时对小鼠应激激素、神经递质水平及相关酶的表达水平进行检测。结果显示,乙酸、异丁酸和丁酸能在不同程度上减轻小鼠抑郁样行为,其可能机制与SCFAs修复下丘脑-垂体-肾上腺轴反馈调节障碍、恢复5-羟色胺的正常代谢相关。该研究为抑郁症的营养干预提供了新的理论依据。 展开更多

Schizophrenia(SCZ)is the most common serious mental illness with a high disability rate and heavy social and family burdens.At present,there is no clear etiology and pathogenesis of schizophrenia.Studies have shown that the occurrence of schizophrenia may be related to the abnormality of the hypothalamic-pituitary-adrenal(HPA)axis.The LIM-homeobox gene 3(LHXS)and early growth response 1(EGR1)can affect pituitary function.Because the synapsin 2(SYN2)gene polymorphism regulates the activity of LHX3 and EGR1,it may cause the occurrence of schizophrenia.This article will review the possible involvement of SYN2 gene polymorphism in the pathogenesis of schizophrenia via regulating the activity of LHX3 and EGR1,then to afiect the HPA axis. 展开更多

目的探讨丙戊酸钠对慢性不可预见性温和刺激(chronic unpredicted mild stress,CUMS)所致抑郁大鼠行为学及下丘脑-垂体-肾上腺皮质轴(hypothalamic-pituitary-adrenal axis,HPA)功能的影响及其机制。方法将SD大鼠随机分为正常对照组(CG... 目的探讨丙戊酸钠对慢性不可预见性温和刺激(chronic unpredicted mild stress,CUMS)所致抑郁大鼠行为学及下丘脑-垂体-肾上腺皮质轴(hypothalamic-pituitary-adrenal axis,HPA)功能的影响及其机制。方法将SD大鼠随机分为正常对照组(CG)、模型组(MG)、丙戊酸钠给药对照组(VCG)和丙戊酸钠给药模型组(VMG),每组15只。模型组采用孤养结合CUMS的方式建立大鼠抑郁模型,建模周期为28 d。VCG和VMG组灌胃给予丙戊酸钠300 mg/(kg·d),CG和MG组灌胃等体积的0.5%CMC-Na溶液。采用旷场试验和强迫游泳试验评价大鼠抑郁行为,试剂盒测定大鼠大脑皮层丙二醛(malonaldehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)和血清皮质酮(concertmaster,CORT)含量,RT-PCR和Western blot法检测大鼠下丘脑促肾上腺皮质激素释放激素(corticotrophin releasing factor,CRF)基因m RNA和蛋白的表达水平。结果与CG组相比,MG组大鼠水平活动和垂直活动水平得分明显下降(P<0.01),不动时间明显延长(P<0.01),大脑皮层MDA含量明显升高(P<0.05),SOD活力显著下降(P<0.05),血清CORT含量明显升高(P<0.05),下丘脑CRF基因m RNA及蛋白表达水平明显升高(P<0.01及<0.05);丙戊酸钠能够明显阻遏上述变化。结论丙戊酸钠能够明显改善CUMS大鼠的抑郁行为,改善机体氧化应激失衡,下调HPA轴功能。 展开更多

The European beaver (Castor fiber L.) is the largest free-living rode nt in Eurasia. The prese nt work aimed to determi ne sex- and seas on-related cha nges in leptin receptor (Ob-R) expression in the hypothalamic-pituitary-gonadal/adrenal axes and uterus of beavers during breeding-(April), postbreeding-(July), and pre-breeding-(November) periods. The expression of Ob-R gene and protein was found in all analyzed tissues. The expression of Ob-R mRNA remained constant in the hypothalamus of both sexes during the analyzed stages. Sex- and season-related changes were found in the pituitary gland;the greatest level was observed in July in both sexes. The same expression pattern was noted in the testis, whereas in the ovary a lack of seasonal changes was found. In uterine tissues, the greatest expression occurred in November. The impact of season was also demonstrated in the adrenal cortex. In females, a higher Ob-R transcript level was noted in April, while in males, an increased mRNA abundanee was noted in November than July. Our study suggests that in the beaver, leptin acting via the Ob-R can be an important endocrine factor engaged in the regulation of reproductive functions and stress response. 展开更多
