The application of Penman's method of cell fractionation to plant protoplasts leads to our finding of keratin intermediate filament(IF)-like system in maize protoplasts,which was identified by using immunogold labelling with monoclonal antibody of cytokeratin from animal cells.Many gold particles were found to be bound on filaments,linked by 3 nm filaments.After further digestion and extraction with DNase I and ammonium sulphate.IF-like framework-lamina-nuclear matrix system was shown under electron microscope.That IF system exists in plant protoplasts just like in animal cells,and their main component is keratin-like protein. 展开更多

目的探讨快速心房起搏犬心房肌中电导钙离子激活钾通道蛋白(KCa3.1)对心外膜脂肪巨噬细胞迁移及心房颤动(房颤)易感性的影响。方法本研究为前瞻性对照研究。将18只比格犬(雄,10~12 kg)按随机数法分为假手术组(Sham组,n=6)、起搏组(Pacin... 目的探讨快速心房起搏犬心房肌中电导钙离子激活钾通道蛋白(KCa3.1)对心外膜脂肪巨噬细胞迁移及心房颤动(房颤)易感性的影响。方法本研究为前瞻性对照研究。将18只比格犬(雄,10~12 kg)按随机数法分为假手术组(Sham组,n=6)、起搏组(Pacing组,n=6)、KCa3.1抑制组(TRAM-34组,n=6)。3组犬均植入起搏器同时于左上肺静脉脂肪垫及左下肺静脉脂肪垫注射巨噬细胞示踪剂DIL脂质体(5 mg/ml),Sham组不起搏,Pacing组和TRAM-34组持续左心耳起搏7 d。TRAM-34组静脉注射KCa3.1抑制剂(TRAM-34,10 mg/kg,3次/d),Sham组和Pacing组注射等量生理盐水。7 d后3组犬行腔内电生理检查,检测脂肪垫毗邻心房肌DIL+巨噬细胞数目、炎症因子及迁移相关蛋白表达水平。结果与Sham组比较,Pacing组心房有效不应期(AERP)显著缩短[(100.8±3.9)ms对(120.8±5.6)ms,P<0.001],房颤诱发次数及持续时间明显增多[(5.0±0.9)次对0次;(12.5±1.9)s对0 s,P<0.001],从脂肪垫迁移至毗邻心房肌的DIL+巨噬细胞明显增多(92.3±8.5对8.0±3.0,P<0.001),脂肪垫毗邻心房肌炎症因子、KCa3.1、趋化因子配体2(CCL2)水平显著增加(P<0.050或P<0.001)。与Pacing组比较,TRAM-34组AERP显著延长[(116.3±8.1)ms对(100.8±3.9)ms,P<0.010],房颤诱发次数及持续时间明显减少[(1.2±0.8)次对(5.0±0.9)次;(4.1±3.2)s对(12.5±1.9)s P<0.010],从脂肪垫迁移至毗邻心房肌的DIL+巨噬细胞明显减少(40.0±5.0对92.3±8.5,P<0.001),脂肪垫毗邻心房肌炎症因子、KCa3.1、CCL2水平显著减少(P<0.050或P<0.001)。结论快速心房起搏引起心房电重构和房颤诱发与心房肌KCa3.1上调增加CCL2分泌,促进心外膜脂肪巨噬细胞向毗邻心房肌迁移有关。 展开更多
