AIM:To investigate the presence of mixed infection and discrepancy between hepatitis C virus(HCV) genotypes in plasma,peripheral blood mononuclear cells(PBMCs),and liver biopsy specimens.METHODS:From September 2008 up...AIM:To investigate the presence of mixed infection and discrepancy between hepatitis C virus(HCV) genotypes in plasma,peripheral blood mononuclear cells(PBMCs),and liver biopsy specimens.METHODS:From September 2008 up to April 2009,133 patients with chronic hepatitis C referred to Firouzgar Hospital for initiation of an antiviral therapy were recruited in the study.Five milliliters of peripheral blood was collected from each patient and liver biopsy was performed in those who gave consent or had indications.HCV genotyping was done using INNO-LiPATM HCV in serum,PBMCs,and liver biopsy specimens and then conf irmed by sequencing of 5'-UTR fragments.RESULTS:The mean age of patients was 30.3 ± 17.1 years.Multiple transfusion was seen in 124(93.2%) of patients.Multiple HCV genotypes were found in 3(2.3%) of 133 plasma samples,9(6.8%) of 133 PBMC samples,and 8(18.2%) of 44 liver biopsy specimens.It is notable that the different genotypes found in PBMCs were not the same as those found in plasma and liver biopsy specimens.CONCLUSION:Our study shows that a signif icant proportion of patients with chronic hepatitis C are affected by multiple HCV genotypes which may not be detectable only in serum of patients.展开更多

Objective:To observe the effect of warm-unblocking acupuncture plus fluticasone propionate nasal spray on the pulmonary ventilation,level of interferon-y(IFN-y)and sleep quality in parents with allergic rhinitis(AR).M...Objective:To observe the effect of warm-unblocking acupuncture plus fluticasone propionate nasal spray on the pulmonary ventilation,level of interferon-y(IFN-y)and sleep quality in parents with allergic rhinitis(AR).Methods:A total of 112 AR patients were enrolled between January 2013 and August 2018 and were divided into an observation group and a control group by the random number table method,with 56 cases in each group.Patients in the observation group received warm-unblocking acupuncture plus fluticasone propionate nasal spray,and patients in the control group only received fluticasone propionate nasal spray.The nasal symptom score,pulmonary function indexes,the levels of IFN-y and interleukin(IL)-4 in serum,and sleep quality in the two groups were compared.Results:After treatment,the total effective rate in the observation group was higher than that in the control group(P<0.05).The nasal symptom score dropped in both groups after treatment(both P<0.05),and the score in the observation group was lower than that in the con trol group(P<0.05).The pulm onary ven tilati on indexes all in creased sign ificantly after treatment in the observation group(all P<0.05);the forced expiratory volume in 1 second(FEV1)to forced vital capacity(FVC)ratio(FEV1/FVC)and the forced expiratory flow at 50%,75%and 25%-75%of the vital capacity(FEF50%,FEF75%,FEF25%-75%)in creased after treatme nt in the control group(all P<0.05);the pulm onary ven tilati on in dexes were higher in the observati on group tha n those in the control group(all P<0.05).The level of IFN-y in creased sign ificantly after t reatment in the two groups(both P<0.05)and the level of IL-4 dropped significantly(both P<0.05);the observation group had a higher IFN-y level(P<0.05)and a lower IL-4 level(P<0.05)compared with the control group.Regarding the Pittsburgh sleep quality index(PSQI),the scores of subjective sleep quality,habitual sleep efficiency and sleep disturbances and the general PSQI score decreased significantly after treatment in both groups(all P<0.05),and the scores in the observation group were significantly lower than those in the control group(all P<0.05).Conclusion:Warm-unblocking acupuncture plus fluticasone propionate nasal spray can effectively control the clinical symptoms and improve pulmonary function in the treatment of AR;this approach can regulate the levels of IFN-y and IL-4 towards the normal range in AR patients;it can also improve patient's sleep quality.This method can produce more significant efficacy than fluticasone propionate nasal spray used alone.展开更多
Aim:Peripheral cytokines contribute to arthritis and bone cancer pain through sensory nerve actions.However,increased spinal cytokine and glial filament expression,coined neuroinflammation,has also been proposed to pl...Aim:Peripheral cytokines contribute to arthritis and bone cancer pain through sensory nerve actions.However,increased spinal cytokine and glial filament expression,coined neuroinflammation,has also been proposed to play a part in chronic pain.Therefore,spinal cord,dorsal root ganglia and circulating cytokines were compared in murine arthritis and bone cancer models in relationship to behavioral signs of pain.Methods:Exploratory behaviors were studied after intra-articular complete Freund’s adjuvant or bone intramedullary sarcoma cell injection.Nervous tissue and blood cytokine expression were determined by real-time polymerase chain reaction(PCR)and multiplex immunoassays,respectively.Results:PCR analysis did not reveal any hallmark of spinal neuroinflammation in spontaneously-behaving mice with cartilage or bone lesions.However,imposed paw stimulation during joint inflammation increased spinal interleukin-1β(IL-1β)expression.Spontaneous paw guarding during rearing was displayed by animals with joint inflammation and bone destruction and was accompanied by increased circulating IL-6 and monocyte chemoattractant protein-1,respectively.In addition,dorsal root ganglia were found to constitutively express receptors for this chemotactic cytokine.Conclusion:Our findings indicate that spinal neuroinflammation is not a necessary condition for chronic pain and suggest that circulating cytokine action in dorsal root ganglia may contribute to experimental joint inflammation and bone cancer pain.展开更多
