白茅是一种恶性杂草,对极端环境条件具有很强的适应能力.作为自然生态恢复过程中的优势物种,白茅在铜陵铜尾矿废弃地上形成大面积的白茅群落.本研究以铜陵杨山冲尾矿废弃地自然生长的白茅为对象,采用野外调查和室内分析相结合的方法,研... 白茅是一种恶性杂草,对极端环境条件具有很强的适应能力.作为自然生态恢复过程中的优势物种,白茅在铜陵铜尾矿废弃地上形成大面积的白茅群落.本研究以铜陵杨山冲尾矿废弃地自然生长的白茅为对象,采用野外调查和室内分析相结合的方法,研究了在异质微环境下白茅形态和生理可塑性的表达及其适应性.结果表明:(1)不同样方苗的数量(SN)、苗高(SH)、根茎间隔子长(RD)、根茎总长(TLR)具较大差异,变异系数分别为74%、59%、40%、78%;随基质养分水平的增加,SN、TLR显著增加,而SH、DR则明显减少,表现出很强的形态可塑性;(2)不同样方中,白茅根茎中还原糖(RS)、总磷(TPR)和总氮(TNR)含量差异较大,变异系数分别为111%、74%和40%;根茎中的TPR和SS含量随基质养分水平的增加而增加.与形态可塑性相比,白茅根茎生理可塑性较小,生长尾矿上的白茅主要通过形态可塑性适应尾矿环境. 展开更多

Background: Herbal medicinal preparations are used as dietary supplements for disease prevention and as alternative/complementary medicine. The growing interest in herbal medicine raises the question of its safety and efficacy. Numerous recorded cases of intoxication and toxicological studies reveal that medicinal plants can be toxic, which hinders their safe use. Plant intoxications related to a variety of factors include plant organs and many extraction solvents. Little toxicological data on medicinal plants is available. The need to investigate is important for safe use. Imperata cylindrica, a poaceae, is one of the medicinal plants for which few toxicological data are available. Materials & Methods: To expand toxicological data, water, 70% ethanol, and 30% acetone extracts of the leaves and roots, obtained by maceration and reflux methods, were used for phytochemicals molecules qualitative detection and toxicity test by the brine shrimp lethality assay. Results: The qualitative analysis of the different extracts revealed the presence of alkaloids, polyphenolic compounds, saponins, and polyterpenoids. The toxicity endpoint was lethal concentration 50 (LC<sub>50</sub>). The leaves’ extracts LC<sub>50</sub> was between 489.78 μg/mL and 1066.6 μg/mL. As for the root extracts, the LC<sub>50</sub> was between 341.98 μg/mL and 1530 μg/mL. Discussion: The different compounds’ presence justifies the use of Imperata cylindrica as a medicinal plant. According to Clarkson classification the root extracts are moderately toxic (LC<sub>50</sub>: 168.47 μg/mL), and leaf extracts are weakly toxic (LC<sub>50</sub>: 527.25 μg/mL). The extrapolation made in relation to the Gosselin, Smith, and Hodge scale, allows us to characterize the Imperata cylindrica root and leaf extracts as non-toxic to humans by oral route. Conclusion: This result can be a base for more precise toxicological studies. 展开更多

Objective:To assess the effect of short-term gavage of ethanolic extract of Imperata cylindrica L root on the ovarian activity and estrus behavior of female mice.Methods:Eighteen virgin female ddY mice,8 to 10 weeks of age,weighing 22-25 g with regular estrus cycle,were divided into three groups.Group 1 received 0.5%carboxymethylcellulose,whereas Groups 2 and 3 received the ethanolic extract of Imperata cylindrica L root at 90 and 115 mg/kg body weight(b.w.)per day by gavage for 20 days,respectively.All of the groups were checked before 9 a.m.daily for vaginal cytology to determine the estrus phase.On day 21,the mice were sacrificed to collect serum samples to quantify the concentrations of reproductive hormones using enzyme-linked immunosorbent assay and to determine changes in the reproductive organs based on their reproductive organ weight,histomorphology,and histomorphometry of ovarium and uterus.Results:The reproductive organ weight in the treatment groups was similar compared with that in the control group.The 90 mg/kg b.w.treatment group showed an increase in corpus luteum number when compared with the control group,with few degenerated follicles and diminished oocytes.Moreover,the 115 mg/kg b.w.treatment group showed fewer primordial and primary follicles and an increase in corpus luteum number and a prolonged diestrus phase compared to the control and 90 mg/kg b.w.treatment groups.The histomorphology examination of the uterus showed that the thickness of myometrium and epithelium in the treated animals was similar to the control group.In addition,there was a significant decrease in follicle-stimulating hormone level in the 115 mg/kg b.w.treatment group(P<0.05).Conclusions:Short-term gavage of ethanolic extract of Imperata cylindrica L root reduces the follicle-stimulating hormone serum level and folliculogenesis. 展开更多

白茅根多糖理化性质及对HepG2细胞的降糖作用研究

该文以高温高压超声波辅助法提取白茅根多糖(Rhizoma imperatae polysaccharides,RPS),通过响应面法优化RPS提取工艺。经Sevag法脱蛋白,DEAE-DE纤维素52层析柱和Sephadex G-100凝胶层析柱分离纯化,得到2种RPS(RPS-DS 0和RPS-DS 0.1)。... 该文以高温高压超声波辅助法提取白茅根多糖(Rhizoma imperatae polysaccharides,RPS),通过响应面法优化RPS提取工艺。经Sevag法脱蛋白,DEAE-DE纤维素52层析柱和Sephadex G-100凝胶层析柱分离纯化,得到2种RPS(RPS-DS 0和RPS-DS 0.1)。采用α-淀粉酶抑制实验,测定RPS的体外降糖作用。构建胰岛素抵抗HepG2细胞(insulin-resistant HepG2,IR-HepG2)模型,测定RPS-DS 0.1对IR-HepG2细胞中葡萄糖消耗量、糖原含量、己糖激酶和丙酮酸激酶活性的影响。结果表明,在液料比(mL∶g)为20∶1、提取温度112℃、提取时间15 min、超声波功率410 W、超声波处理时间25 min时,RPS质量分数得率为14.79%。RPS-DS 0和RPS-DS 0.1是分子质量分别为179.14 kDa和29.79 kDa的均一中性多糖。RPS-DS 0由阿拉伯糖、半乳糖、葡萄糖、木糖和甘露糖组成,其物质的量比为6.79∶9.68∶75.52∶4.38∶1.88。RPS-DS 0.1由鼠李糖、阿拉伯糖、半乳糖、葡萄糖、木糖、甘露糖和葡萄糖醛酸组成,其物质的量比为2.48∶11.76∶20.08∶50.92∶10.25∶2.08∶2.12。RPS-DS 0和RPS-DS 0.1对α-淀粉酶的半数抑制浓度分别为0.40和0.21 mg/mL。细胞实验结果表明,RPS-DS 0.1能显著提高IR-HepG2细胞中葡萄糖消耗量、糖原含量、己糖激酶和丙酮酸激酶活性,调节糖代谢,从而达到降糖作用。 展开更多

用15—20%(V/V)丝瓜伤流液加入培养基可以显著提高水稻花药培养力和花粉植株自然加倍的频率。诱导培养基中附加15—20%(V/V)丝瓜伤流液,形成愈伤组织百分率显著高于对照;粳稻的愈伤组织分化率高达95%,籼稻愈伤组织分化率可达70%,比对照... 用15—20%(V/V)丝瓜伤流液加入培养基可以显著提高水稻花药培养力和花粉植株自然加倍的频率。诱导培养基中附加15—20%(V/V)丝瓜伤流液,形成愈伤组织百分率显著高于对照;粳稻的愈伤组织分化率高达95%,籼稻愈伤组织分化率可达70%,比对照提高1倍左右。花粉植株自然加倍的频率显著(p>0.01)高于对照。附加10%、15%、20%、25%(V/V)丝瓜伤流液诱导成的愈伤组织经继代培养后的重量除了25%(V/V)浓度外,其余都比对照增重5.7—18.09毫克/10块。绿苗的鲜重和干重分别增重20—89.0毫克/10株,13.0—30.0毫克/10株,在10—20(V/V)浓度的丝瓜伤流液范围内,对植物细胞分裂和生长有促进作用。其中以15—20%(V/V)浓度的丝瓜伤流液效果最好,经紫外光谱初步测定表明,丝瓜伤流液含有类似细胞激动素以及培养基中有机生长物质。 展开更多

A Higher Plant Myosin in Luffa cylindrica: Electron Microscopic Visualization

The myosin exhibited actin_activated MgATP... The molecular structure of a higher plant myosin with two 174 kD heavy chains purified from the tendrils of Luffa cylindrica (L.) Roem. was viewed by electron microscopy. The myosin exhibited actin_activated MgATPase activity and could be recognized immunologically by a monoclonal antibody against the skeletal muscle myosin. Electron micrographs of rotary shadowed images of this protein revealed that it had two heads with size and shape similar to those of the skeletal muscle myosin and a relatively short tail in comparison with the conventional myosin. Luffa tendril actin filaments were also visualized and occasionally other Luffa myosin_like proteins with globular structure at the tail ends were also observed. The structural similarity and immunological cross reactivity with antibodies against muscle myosin demonstrate that the 174 kD Luffa tendril myosin is a double_headed myosin. The possible involvement of myosin_actin interactions in Luffa tendril contact coiling will be the subject of further research. 展开更多

白茅根提取物抑菌抗炎活性在口腔护理品中研究与分析

采用体外评价方法对白茅根提取物在口腔护理品中的抑菌抗炎活性进行研究。采用水、50%乙醇、乙酸乙酯、丙酮、石油醚作为溶剂提取白茅根提取物,将其加入到二氧化硅体系牙膏中,通过“划割牙龈+钢丝结扎+滴入牙周炎患者细菌培养物+高糖黏... 采用体外评价方法对白茅根提取物在口腔护理品中的抑菌抗炎活性进行研究。采用水、50%乙醇、乙酸乙酯、丙酮、石油醚作为溶剂提取白茅根提取物,将其加入到二氧化硅体系牙膏中,通过“划割牙龈+钢丝结扎+滴入牙周炎患者细菌培养物+高糖黏性食物饲养”构建20只5周龄SD大鼠牙周炎模型,测定白茅根提取物对大鼠牙周炎模型中的牙周致病菌[伴放线杆菌(Aa)、牙龈卟林单胞菌(Pg)、具核梭杆菌(Fn)、中间普菌(Pi)、硒赛类标菌(Tf)、变黑普菌(Pn)]、外周血、龈沟液内炎性因子[C反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白介素-6(IL-6)、前列腺素E_(2)(PGE_(2))]及牙周健康指标[菌斑指数(PLI)、探诊深度(PD)、出血指数(BI)]的影响,采用小白鼠急性经口毒性实验验证白茅根提取物应用于口腔护理品中的安全性。结果显示,白茅根提取物能明显减少Aa、Pg、Fn、Pi、Tf、Pn,降低CRP、TNF-α、IL-1β、IL-6、PGE_(2)表达,具有抑制牙周致病菌活性,抵抗牙周炎症反应作用,能改善牙周健康,且安全性可靠。 展开更多

为丰富果冻种类,以白茅根和金银花为主要原料,以可得然胶、黄原胶和魔芋胶为复配胶,以感官评分、色泽、质构特性及析水率为指标,通过单因素试验探究金银花提取液添加量、复配胶添加量、赤藓糖醇添加量对白茅根金银花复合果冻品质的影响... 为丰富果冻种类,以白茅根和金银花为主要原料,以可得然胶、黄原胶和魔芋胶为复配胶,以感官评分、色泽、质构特性及析水率为指标,通过单因素试验探究金银花提取液添加量、复配胶添加量、赤藓糖醇添加量对白茅根金银花复合果冻品质的影响;在此基础上,以感官评分为响应值,通过响应面法优化白茅根金银花复合果冻的配方。结果表明,白茅根金银花复合果冻的最佳配方为金银花提取液添加量47.6%、复配胶添加量2.1%(可得然胶、黄原胶和魔芋胶的质量比为30∶7∶3)、赤藓糖醇添加量17.0%;在此条件下制得的果冻呈乳黄色,富有光泽,清甜可口,白茅根和金银花的特征风味突出且协调、富有层次,口感细腻,弹性、咀嚼性及硬度适中,总体可接受性极好,感官评分为8.70分。本研究可为白茅根金银花复合保健果冻的开发提供参考。 展开更多

Sponge gourd(Luffa cylindrica)is an important cultivated vegetable and medicinal plant in the family Cucurbitaceae.In this study,a draft genome sequence of the sponge gourd inbred line P93075 was analyzed.Using Illumina,PacBio,and 10×Genomics sequencing techniques as well as new assembly techniques such as FALCON and chromatin interaction mapping(Hi-C),a chromosome-scale genome of approximately 656.19 Mb,with an N50 scaffold length of 48.76 Mb,was generated.From this assembly,25,508 protein-coding gene loci were identified,and 63.81%of the whole-genome consisted of transposable elements,which are major contributors to the expansion of the sponge gourd genome.According to a phylogenetic analysis of conserved genes,the sponge gourd lineage diverged from the bitter gourd lineage approximately 41.6 million years ago.Additionally,many genes that respond to biotic and abiotic stresses were found to be lineage specific or expanded in the sponge gourd genome,as demonstrated by the presence of 462 NBS-LRR genes,a much greater number than are found in the genomes of other cucurbit species;these results are consistent with the high stress resistance of sponge gourd.Collectively,our study provides insights into genome evolution and serves as a valuable reference for the genetic improvement of sponge gourd. 展开更多

Extent of the inhibition was enchanced with increased KCl concentrations. The biggest decrease of nitrogen-fixing activity was between 0.02–0.05 mol/L KCl. The same decline on growth was between 0.05–0.10 mol/L KCl. The longer treating-time was, the lower nitrogen-fixing activity was. A short time of KCl-treatment (for 4 h) resulted in 66.4%decrease of the activity. Under effecting of KCl for three days, the frequency of heterocyst differentiation decreased from 7.2%to 2.1%. 展开更多

Objective:To investigate the curative effect of Rhizoma Imperata in the treatment of patients with community-acquired pneumonia and its influence on inflammatory factors related to TLR4/NF-κB signaling pathway,so as to seek new drugs for the treatment of community-acquired pneumonia.Methods:120 patients with community-acquired pneumonia who were treated in Department of respiratory medicine,the First Affiliated Hospital of Hainan Medical College from November 2019 to September 2020 were collected and randomly divided into control group and experimental group,with 60 patients in each group.The control group was given cefuroxime sodium injection,and the experimental group was given cefuroxime sodium injection+Baimao root decoction.The levels of high-sensitivity C-reactive protein(CRP)and the level of serum CRP in the two groups before and after treatment were compared Objective To observe the expression of procalcitonin(PCT),IL-6,IL-8,TNF-αand TLR4/NF-κB mRNA levels,and to observe the effect of Radix Imperata Rubra on community-acquired pneumonia.Results:The fever,cough,expectoration subsided time and chest CT inflammation absorption time of the experimental group were shorter than those of the control group,the difference was statistically significant(P<0.05);the total effective rate of the experimental group(96.67%)was higher than that of the control group(85%),the difference was statistically significant(P<0.05);after treatment,the serum CRP,PCT,IL-6,IL-8,TNF-αin the two groups were lower than before,and the experimental group was significantly lower than the control group,the difference was statistically significant(P<0.05),There was statistical significance(P<0.05);after treatment,TLR4/NF-κB mRNA of the two groups were decreased,and the experimental group was significantly lower than the control group,the difference was statistically significant(P<0.05).Conclusion:In the treatment of cap,Rhizoma Imperata can reduce the levels of CRP,PCT,IL-6,IL-8 and TNF-α.The mechanism is related to inhibiting the activation of TLR4/NF-κB signaling pathway,so as to reduce the release of inflammatory factors and improve clinical symptoms. 展开更多

采用大孔树脂纯化白茅根黄酮提取物,并比较不同产物的抗运动疲劳活性。通过静态吸附-解吸试验筛选适宜的色谱柱型号后,采用单因素试验探讨大孔树脂纯化白茅根黄酮提取物的最佳工艺条件,并利用动物试验考察不同产物的抗运动疲劳活性。结... 采用大孔树脂纯化白茅根黄酮提取物,并比较不同产物的抗运动疲劳活性。通过静态吸附-解吸试验筛选适宜的色谱柱型号后,采用单因素试验探讨大孔树脂纯化白茅根黄酮提取物的最佳工艺条件,并利用动物试验考察不同产物的抗运动疲劳活性。结果表明,AB-8型大孔树脂纯化白茅根黄酮的最佳条件为上样液浓度2.0 mg/mL、上样液pH 5.0、上样液体积60 mL、上样流速2.0 mL/min、洗脱液乙醇的体积分数70%、洗脱流速1.0 mL/min、洗脱液体积100 mL,所得产物中黄酮化合物的含量为(64.2±1.5)%,约为纯化前2.24倍。动物试验结果显示,中、高剂量的白茅根黄酮纯化产物可明显延长小鼠的力竭游泳时间,降低体内乳酸与尿素氮的生成量并增加肝、肌糖原储备量,同时显著改善体内超氧化物歧化酶与谷胱甘肽过氧化物酶的活力,从而可知纯化后的白茅根黄酮产物具有较好缓解运动疲劳的作用。 展开更多

以清凉峰国家级自然保护区黄山玉兰(Yulania cylindrica)为研究对象,对其种群分布、物种多样性、生活型与生境进行调查与分析。结果表明:保护区内黄山玉兰主要集中分布在干坑林区,数量相对不多;群落内共有50种植物(含变种),以蔷薇科(Ros... 以清凉峰国家级自然保护区黄山玉兰(Yulania cylindrica)为研究对象,对其种群分布、物种多样性、生活型与生境进行调查与分析。结果表明:保护区内黄山玉兰主要集中分布在干坑林区,数量相对不多;群落内共有50种植物(含变种),以蔷薇科(Rosaceae)、菊科(Asteraceae)、百合科(Liliaceae)、樟科(Lauraceae)为主,乔木层优势种是杉木(重要值43.86)与黄山玉兰(重要值25.81),灌木层优势种是阔叶箬竹(Indocalamus latifolius)(重要值17.06);群落垂直结构明显,物种生活型谱中高位芽占一半以上;群落内草本层的Simpson指数(0.897)、Shannon-Wiener指数(3.632),Pielou均匀度指数(1.234)均最高,乔木层的3个指数最低。综上所述,保护区内黄山玉兰资源较为珍稀,虽然目前群落结构较为稳定,但仍存在被群落内其他物种演替取代的可能,建议采取措施对其进行保护。 展开更多
