BACKGROUND Dry eye is a common eye disease.Artificial tears supplements are widely used for the treatment of dry eyes.However,multiple adverse effects have been observed in patients receiving long-term treatment with artificial tears,which may affect the therapeutic effect.AIM To analyze the characteristics of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-alpha(TNF-α)levels in patients with dry eye and the therapeutic effect of artificial tears combined with cyclosporine A.METHODS A total of 124 dry eye patients treated at The First People’s Hospital of Xining from April 2020 to April 2022 were selected as the observation group,while 20 healthy individuals served as the control group during the same period.Levels of inflammatory markers,including IL-1β,IL-6,and TNF-α,were analyzed.The observation group was further divided into a study group and a control group,each consisting of 62 patients.The control group received artificial tears,whereas the study group received a combination of artificial tears and cyclosporine A.Inflammatory markers,Schirmer’s test(SIT),tear break-up time(TBUT),corneal fluorescein staining(CFS),National Eye Institute Visual Function Questionnaire-25(NEI-VFQ-25)scores,and adverse events(AEs)were compared between the two groups.RESULTS The observation group exhibited significantly elevated serum levels of IL-1β,IL-6,and TNF-αin comparison to the healthy group.Following treatment,the study group demonstrated substantial reductions in IL-1β,IL-6,and TNF-αlevels relative to the control group.Moreover,after treatment,the study group experienced a marked decrease in CFS scores and significant increases in both SIT and BUT levels when compared to the control group.Additionally,significant improvements were observed in the primary symptom of dry eye and secondary symptoms such as photophobia,foreign body sensation,fatigue,red eye,and burning sensation within the study group.Furthermore,post-treatment NEI-VFQ-25 scores across all dimensions exhibited significant enhancements in the study group compared to the control group(P<0.05).It is noteworthy that significant AEs were reported in both groups throughout the treatment period.CONCLUSION Cyclosporine A combined with artificial tears is effective in treating dry eye,yielding enhanced outcomes by improving SIT and TBUT levels,reducing CFS scores,and ameliorating vision-related quality of life. 展开更多

BACKGROUND Acute pancreatitis(AP)is an inflammatory disease,which presents with epigastric pain and is clinically diagnosed by amylase and lipase three times the upper limit of normal.The 2012 Atlanta classification stratifies the severity of AP as one of three risk categories namely,mild AP(MAP),moderately severe AP(MSAP),and severe AP(SAP).Challenges in stratifying AP upon diagnosis suggest that a better understanding of the underlying complex pathophysiology may be beneficial.AIM To identify the role of the chemokine receptor 8(CCR8),expressed by T-helper type-2 Lymphocytes and peritoneal macrophages,and its possible association to Interleukin(IL)-6 and AP stratification.METHODS This study was a prospective case-control study.A total of 40 patients were recruited from the Chris Hani Baragwanath Academic Hospital and the Charlotte Maxeke Johannesburg Academic Hospital.Bioassays were performed on 29 patients(14 MAP,11 MSAP,and 4 SAP)and 6 healthy controls as part of a preliminary study.A total of 12 mL of blood samples were collected at Day(D)1,3,5,and 7 post epigastric pain.Using multiplex immunoassay panels,real-time polymerase chain reaction(qRT-PCR)arrays,and multicolour flow cytometry analysis,immune response-related proteins,genes,and cells were profiled respectively.GraphPad Prism^(TM) software and fold change(FC)analysis was used to determine differences between the groups.P<0.05 was considered significant.RESULTS The concentration of IL-6 was significantly different at D3 post epigastric pain in both the MAP group and MSAP group with P=0.001 and P=0.013 respectively,in a multiplex assay.When a FC of 2 was applied to identify differentially expressed genes using RT2 Profiler,CCR8 was shown to increase steadily with disease severity from MAP(1.33),MSAP(38.28)to SAP(1172.45)median FC.Further verification studies using RT-PCR showed fold change increases of CCR8 in MSAP and SAP ranging from 1000 to 1000000 times when represented as Log10,compared to healthy control respectively at D3.The findings also showed differing lymphocyte and monocyte cell frequency between the groups.With monocyte population frequency as high as 70%in MSAP at D3.CONCLUSION The higher levels of CCR8 and IL-6 in the severe patients and immune cell differences compared to MAP and controls provide an avenue for exploring AP stratification to improve management. 展开更多

BACKGROUND Inflammatory cytokines play a vital role in the occurrence of osteoarticular injury and inflammation. Whether inflammation-associated factors interleukin-1β(IL- 1β), IL-6, tumor necrosis factor-α(TNF-α) and vascular endothelial growth factor (VEGF) are involved in the pathogenesis of keen articular cartilage injury remains poorly understood. AIM To measure the levels of inflammatory factors [IL-1β, IL-6, TNF-α and VEGF] in patients with knee articular cartilage injury. METHODS Fifty-five patients with knee articular cartilage injury were selected as patient groups, who were divided into three grades [mild (n = 20), moderate (n = 19) and severe (n = 16)] according to disease severity and X-ray examinations. Meanwhile, 30 healthy individuals who underwent physical examination were selected as the control group. The levels of IL-1β, IL-6, TNF-α and VEGF were measured by ELISA and immunohistochemical staining. RESULTS Compared with the control group, patient groups displayed significantly higher levels of IL-1β, IL-6, TNF-α and VEGF, and the extent of increase was directly proportional to the severity of injury (P < 0.05). In addition, the number of cells with positive staining of IL-1β, IL-6, TNF-α and VEGF in the synovial membrane were significantly increased, along with increased disease severity (P < 0.05). After treatment, the scores of visual analogue scale and the Western Ontario and McMaster University of Orthopaedic Index in patient groups were 2.26 ± 1.13 and 15.56 ± 7.12 points, respectively, which were significantly lower than those before treatment (6.98 ± 1.32 and 49.48 ± 8.96). Correlation analysis suggested that IL-1β and TNF-α were positively correlated with VEGF. CONCLUSION IL-1β, IL-6, TNF-α and VEGF levels are increased in patients with knee articular cartilage injury, and are associated with the disease severity, indicating they might play an important role in the occurrence and development of knee articular cartilage injury. Furthermore, therapeutically targeting them might be a novel approach for the treatment of keen articular cartilage injury. 展开更多

Objective To investigate the effect of interleukin-6 ( IL-6 ) on the apoptosis of annulus fibrosus (AF) cell induced by intedeukin-1β (IL-1β). Methods Cultured AF cells were divided into 6 groups and treated with no drug, 10 ng/mL IL-6, 10 ng/mL IL-1β, 10 ng/mL IL-1β and Z-VAD-FMK ( a caspase-9 inhibitor), 10 ng/mL IL-1β and 10 ng/mL IL-6, 10 ng/mL IL-1β and 100 ng/mL IL-6, respectively. After three days of culture, the apoptosis rate, the positive rates of caspase-3, -8, and -9 of AF cells were detected with flow cytometry. Results The apoptosis rates of cells in group 1 to 6 were 2.67% ± 1.08%, 2.71% ± 0.53%, 20. 37% ± 1.57 %, 11.34% ± 0.67 %, 18.17 % ± 0.74%, and 9.42 % ± 1.08 %, respectively. There was no significant difference between group 1 and 2, while the apoptosis rates of group 4, 5, and 6 were significantly lower than group 3 ( P = 0. 001, P =0. 172, and P =0. 001, respectively). Positive rates of caspase-3 in group 5 ( 12. 35% ±0.64% ) and 6 (9.26% ±0. 36% ) were significantly lower than group 3 ( 17.14% ±0. 72% ; P =0. 001 and P 〈0.001, respectively). And positive rates of caspase-9 in group 5 ( 15.13% ± 1.45% ) and 6 ( 10.17% ± 2.50% ) were significantly lower than group 3 ( 19.4% ±0.98% ; P =0. 014 and P =0. 004, respectively). But there was not obvious change of caspase-8 activity after IL-6 was added. Conclusion IL-6 is capable of protecting AF cells from IL-1β induced apoptosis in vitro. Mechanism of the protection is related with the inhibition of caspase-3 and -9 activities. 展开更多

在研究并分析了断路器状态评估方法的基础上,提出了一种基于断路器维修曲线的多信息融合评估方法。根据断路器状态评估方法和IEC 61850建模规范,绘制了断路器分/合操作次数相对于分断跳闸动作电流的曲线图。结合断路器的预估寿命和最大... 在研究并分析了断路器状态评估方法的基础上,提出了一种基于断路器维修曲线的多信息融合评估方法。根据断路器状态评估方法和IEC 61850建模规范,绘制了断路器分/合操作次数相对于分断跳闸动作电流的曲线图。结合断路器的预估寿命和最大通断电流等因素评估断路器触头寿命,形成了断路器的维修预警机制,从而实现了断路器的在线监视、保护、测控等设备功能的集成。该方案将原断路器智能组件功能集成到保护测控装置,通过制造信息声明(MMS)服务,将装置现有逻辑功能状态信息上传至后台服务器,以实现实时监视功能。将该方法集成于保护测控装置智能电子设备(IED)中,实现了断路器触头机械寿命预警监视功能。与现有其他断路器在线监测设备相比,该方法可降低站端设备的数量和成本。同时,IEC 61850建模的规范化有利于装置的功能扩展。 展开更多

Vascular endothelial growth factor (VEGF, namely VEGF-A) is an angiogenic polypeptide and VEGF-C is a lymphangiogenic polypeptide that has been implicated in cancer growth, invasion and metastasis. Several cytokines and growth factors play an important part in cancer progression. These cytokines and growth factors are the principal mediators of cancer cells-stromal cell interaction , which is critical for invasion of cancer cells to the surrounding tissues and metastatic dissemination to distant organs. In this study, we studied VEGF-A, C expression in cultured human pancreatic cancer cell lines and whether the presence of VEGF-A, C in the cell lines is regulated by cytokines interleukin-lct (EL-1α), and interleukin-6 (IL-6). METHODS: We used Northern blot and Western blot methods to analyze expression of the gene and protein of VEGF-A, C in all 6 tested cell lines (ASPC-1, CAPAN-1, MIA-PaCa-2, PANC-1, COLO-357 and T3M4) respectively. To analyze what is the regulator for this VEGF-A, C expression in pancreatic cancer,we used the reverse transcription -polymerase chain reaction (RT-PCR) method to analyze VEGF-A, C expression in cultured human pancreatic cancer cell lines (CAPAN-1 and COLO-357) under the stimulation with IL-1α (10μg/L) or IL-6 (100 μg/L). RESULTS:Northern blot analysis revealed the presence of the 4.1-kb VEGF-A mRNA transcript and 2.4-kb VEGF-C mRNA transcript in all 6 tested cell lines. Immunoblotting with highly specific anti-VEGF-A, anti-VEGF-C antibody revealed the presence of a molecular weight of 43-kDa VEGF-A protein and 55-kDa VEGF-C protein in all the cell lines. RT-PCR analysis revealed the levels of the VEGF-A and VEGF-C gene were 1-2 fold and a 1-fold increase in the COLO-357 cell line by stimulation with IL-la, however, no effect was found in the CAPAN-1 cell line. The levels of the VEGF-A and VEGF-C gene were 2-5 fold and a 1-fold increase in the CAPAN-1 cell line by stimulation with IL-6, but, no effect was found in the COLO-357 cell line. CONCLUSION:These findings suggested that the expression of VEGF-A, C and their regulation by IL-1α, IL-6 in pancreatic cancer contributes to the lymphatic and distant metastasis and the disease progression. 展开更多

AIM: To identify whether Aspergillus fumigatus(A.fumigatus) hyphae antigens induced the release of interleukin-8(IL-8) in anti-fungal innate immunity of cultured human corneal epithelial cells(HCECs) and determine the involvement of intracellular signalling pathways. METHODS: HCECs were treated with A. fumigatus hyphae antigens with different concentrations and time.The cytoplasmic calcium of HCECs were assessed by fluorescence imaging. Western blot was used to detect the expression of Ca2 +-dependent protein kinase C(PKC). The IL-8 levels were determined by specific human IL-8 enzyme-linked immunosorbent assay(ELISA) and reverse transcriptase polymerase chain reaction(RT-PCR). Using a series of pharmacological inhibitors, we examined the upstream signalling pathway responsible for IL-8 expression in response to A.fumigatus hyphae antigens. RESULTS: Cells exposed to A. fumigatus hyphae antigens showed higher level of IL-8 m RNA expression and protein production. We demonstrated here that stimulation of HCECs with A. fumigatus hyphae triggers an intracellular Ca2 +flux and results in the activation of Ca2 +-dependent PKC(α, βⅠ and βⅡ) which can be attenuated by pre-treatment of cells with laminarin,suggesting that Dectin-1 signals pathway induced cytoplasmic calcium and influence the activation of PKC in HCECs. Inhibitors of Ca2 +-dependent PKC(Ro-31-8220 and Go6976) significantly abolished hyphae-induced expression of IL-8.CONCLUSION: Our findings suggest that A. fumigatushyphae-induced IL-8 expression was regulated by the activation of Dectin-1-mediated Ca2 +-dependent PKC in HCECs. 展开更多

Objectives: The aim of this study was to assess the levels of Y-box binding protein 1 (YBX-1) and interleukin 6 (IL-6) in the sera of metastatic and non-metastatic breast cancer patients (BC), investigate their clinicopathological significance and to analyze their potential use as biomarkers of breast cancer metastasis. Methods: The study included ninety subjects sub-grouped equally into metastatic BC, non-metastatic BC and healthy volunteers. Serum YBX-1 and IL-6 were quantified using ELISA technique while CA 15-3 was quantified using IRMA kit. Clinical data were collected from patients’ records. Results: YBX-1 (p < 0.001), IL-6 (p < 0.001) and CA15-3 (p = 0.017, 0.001) were significantly elevated in metastatic and non-metastatic BC patients compared to healthy controls, however, only YBX-1 (p 0.001) showed a significant difference with cancer metastasis. Generally, YBX-1 and IL-6 were correlated with worse histological grade and late clinical stage in breast cancer patients and they were also associated with axillary lymph nodes involvement and positive vascular invasion in metastatic BC patients. Serum YBX-1 and IL-6 levels were positively correlated to each other (rs = 0.615, p < 0.001) and they showed high sensitivity and specificity compared to CA 15-3 (p < 0.001 and p = 0.004 for YBX-1 and IL-6 respectively) for predicting cancer metastasis. Conclusions: Serum YBX-1 and IL-6 are potential biomarkers of breast cancer patients with significant correlation with bad clinicopathological characteristics. Serum YBX-1 and IL-6 have superior sensitivity and specificity compared to CA15-3 and can serve as potential follow up and prognostic markers. 展开更多

目的 检测慢性牙髓炎患者龈沟液白介素-6 (IL-6)、基质金属蛋白酶-8 (MMP-8)、基质金属蛋白酶抑制剂-1(TIMP-1)水平,并分析其与患者牙髓活力的关系。方法 选取106例慢性牙龈炎患者(观察组)和106例健康志愿者(对照组)。取龈沟液测定IL-6... 目的 检测慢性牙髓炎患者龈沟液白介素-6 (IL-6)、基质金属蛋白酶-8 (MMP-8)、基质金属蛋白酶抑制剂-1(TIMP-1)水平,并分析其与患者牙髓活力的关系。方法 选取106例慢性牙龈炎患者(观察组)和106例健康志愿者(对照组)。取龈沟液测定IL-6、MMP-8、TIMP-1水平,比较两组差异;比较观察组不同临床特征患者龈沟液IL-6、MMP-8、TIMP-1水平。牙髓活力电测仪测定牙髓活力,比较不同牙髓活力患者龈沟液IL-6、MMP-8、TIMP-1水平,Pearson法分析观察组牙髓电活力(EPT)值与龈沟液IL-6、MMP-8、TIMP-1水平的相关性。结果 观察组龈沟液IL-6、MMP-8均高于对照组[(61.28±8.11) ng/L vs.(38.15±7.04) ng/L、(33.04±6.02) ng/mL vs.(20.05±4.18) ng/mL](P<0.05),TIMP-1水平低于对照组[(1.35±0.30) ng/mL vs.(2.04±0.41) ng/mL](P<0.05);观察组病程≥12个月、合并糖尿病患者龈沟液IL-6、MMP-8水平均高于病程<12个月、未合并糖尿病患者(P<0.05),TIMP-1水平均低于病程<12个月、未合并糖尿病患者(P<0.05);中度、重度疼痛患者龈沟液IL-6、MMP-8水平均高于轻度疼痛(P<0.05),且重度疼痛均高于中度疼痛(P<0.05),而TIMP-1水平变化趋势正相反;牙髓活力下降、无活力患者龈沟液IL-6、MMP-8水平均高于活力正常(P<0.05),且牙髓无活力均高于活力下降(P<0.05),而TIMP-1水平变化趋势正相反;观察组患牙EPT与龈沟液IL-6、MMP-8水平均呈正相关(r=0.715、0.689, P<0.05),与TIMP-1水平呈负相关(r=-0.673, P<0.05)。结论 慢性牙龈炎患者龈沟液IL-6、MMP-8水平均升高,TIMP-1水平下降,且与病程、疼痛程度、合并糖尿病、牙髓活力均有关。 展开更多

目的探究健脾祛邪汤治疗幽门螺杆菌(Hp)感染合并慢性萎缩性胃炎(CAG)的临床疗效及对白细胞介素(IL)-1β、IL-6、IL-8水平的影响。方法选取2019年3月至2021年3月天津市西青医院收治的90例Hp感染合并CAG患者为研究对象,按照治疗方法不同... 目的探究健脾祛邪汤治疗幽门螺杆菌(Hp)感染合并慢性萎缩性胃炎(CAG)的临床疗效及对白细胞介素(IL)-1β、IL-6、IL-8水平的影响。方法选取2019年3月至2021年3月天津市西青医院收治的90例Hp感染合并CAG患者为研究对象,按照治疗方法不同分为对照组(45例)和观察组(45例)。两组患者入院后均进行健康宣教,规范饮食习惯、作息时间以及生活方式。对照组采用常规西药治疗,口服奥美拉唑钠肠溶胶囊(每次20 mg、每日1次)、阿莫西林胶囊(每次0.5 g、每日2次)、克拉霉素片(每次0.5 g、每日2次);观察组在对照组基础上采用健脾祛邪汤治疗,用水煎服150 ml,每日2次。两组均持续治疗4周。比较两组患者治疗前后的炎症因子(IL-1β、IL-6、IL-8)水平、病理组织学(萎缩、肠化生、上皮内瘤变)积分、中医证候(神疲懒言、食少呆纳、食后饱胀、胃脘隐痛)积分以及临床疗效。结果治疗后,两组患者IL-1β、IL-6、IL-8水平均低于治疗前(P<0.05),且观察组低于对照组[(61.6±6.6)ng/L比(69.5±6.8)ng/L、(19.9±2.9)ng/L比(29.6±3.6)ng/L、(7.8±1.1)ng/L比(9.9±1.8)ng/L](P<0.01)。治疗后,两组患者萎缩、肠化生、上皮内瘤变评分均低于治疗前(P<0.05),且观察组低于对照组[(2.93±0.29)分比(3.56±0.39)分、(2.45±0.25)分比(3.01±0.31)分、(2.51±0.25)分比(3.09±0.31)分](P<0.01)。治疗后,两组患者神疲懒言、食少呆纳、食后饱胀、胃脘隐痛均低于治疗前(P<0.05),且观察组低于对照组[(0.55±0.05)分比(0.75±0.08)分、(0.64±0.07)分比(0.89±0.09)分、(0.61±0.06)分比(0.81±0.09)分、(0.73±0.07)分比(1.38±0.39)分](P<0.01)。观察组患者Hp感染率低于对照组[33.33%(15/45)比60.00%(27/45)](χ^(2)=4.252,P=0.039),总有效率高于对照组[95.56%(43/45)比80.00%(36/45)](χ^(2)=5.075,P=0.024)。结论健胃祛邪汤可有效抑制Hp感染合并CAG患者的炎症,改善萎缩、肠化生、上皮内瘤变情况,减轻中医证候症状,提高临床疗效。 展开更多

目的:观察金氏鼻渊方治疗鼻窦炎肺气虚寒证的临床疗效及对鼻分泌物白细胞介素(IL)-6、IL-8、肿瘤坏死因子(TNF)-α、转化生长因子(TGF)-β1水平的影响。方法:选取2021年9月~2023年6月苏州市中西医结合医院耳鼻咽喉科门诊接诊的慢性鼻窦... 目的:观察金氏鼻渊方治疗鼻窦炎肺气虚寒证的临床疗效及对鼻分泌物白细胞介素(IL)-6、IL-8、肿瘤坏死因子(TNF)-α、转化生长因子(TGF)-β1水平的影响。方法:选取2021年9月~2023年6月苏州市中西医结合医院耳鼻咽喉科门诊接诊的慢性鼻窦炎病例共计100例,根据随机数字法分为对照组与观察组各50例。对照组患者予常规药物治疗;观察组患者予金氏鼻渊方服用。两组均连续观察1个月。比较两组患者主观病情评分、鼻窦CT评分与鼻内镜检测评分、治疗效果以及鼻分泌物IL-6、IL-8、TNF-α、TGF-β1水平。结果:治疗后,两组病例视觉模拟量表(VAS)、Lund-Mackay、Lund-Kennedy评分显著减少,且观察组减少更显著(P<0.05);1个月疗程后,观察组病例的总有效率为94.00%,显著高于对照组(78.00%)(P<0.05);1个月疗程后,两组患者鼻分泌物IL-6、IL-8、TNF-α、TGF-β1水平明显减少,且观察组减少更显著(P<0.05)。结论:金氏鼻渊方治疗鼻窦炎肺气虚寒证的临床疗效显著,能改善患者的临床症状,抑制炎症反应。 展开更多

目的研究脐血白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素8(IL-8)、白细胞介素10(IL-10)、肿瘤坏死因子-α(TNF-α)与母体、胎儿炎症反应及早产预后的相关性。方法选取2015年1月至2016年1月在我院妇产科就诊的早产孕妇及早... 目的研究脐血白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素8(IL-8)、白细胞介素10(IL-10)、肿瘤坏死因子-α(TNF-α)与母体、胎儿炎症反应及早产预后的相关性。方法选取2015年1月至2016年1月在我院妇产科就诊的早产孕妇及早产儿各93例作为研究对象。比较母体炎症反应不同时期的脐血细胞因子水平,胎儿炎症反应不同分型的脐血细胞因子水平,不同早产预后的脐血细胞因子水平,利用Spearman相关性分析法分析脐血细胞因子与母体或胎儿炎症反应及早产预后的相关性。结果母体炎症反应≥2期者的IL-1β、IL-6、IL-8及TNF-α的水平明显高于无炎症反应与1期者,IL-10水平明显低于无炎症反应与1期者,而1期的IL-1β、IL-6、IL-8及TNF-α的水平明显高于无炎症反应者,IL-10明显低于无炎症反应者,差异均有统计学意义(P<0.05);胎儿2型炎症反应的IL-1β、IL-6、IL-8及TNF-α的水平明显高于无炎症反应与1型者,IL-10水平明显低于无炎症反应与1型者,而1型的IL-1β、IL-6、IL-8及TNF-α的水平明显高于无炎症反应者,IL-10水平明显低于无炎症反应者,差异均有统计学意义(P<0.05);预后不良的早产儿脐血IL-1β、IL-6、IL-8及TNF-α水平均明显高于预后良好者,而IL-10水平明显低于预后良好者,差异均有统计学意义(P<0.05);根据Spearman法评价相关性可知,IL-1β、IL-6、IL-8及TNF-α分别与母体或胎儿炎症反应及早产预后均呈正相关(P<0.05),而IL-10与母体或胎儿炎症反应以及早产预后呈负相关(P<0.05)。结论脐血IL-1β、IL-6、IL-8及TNF-α水平分别与母体或胎儿炎症反应及早产预后呈正相关,而IL-10与母体或胎儿炎症反应及早产预后呈负相关。 展开更多

目的:观察不同灸法对实验性RA模型关节滑膜液中炎症因子IL-6、IL-8、MCP-1表达的影响,探索不同灸法治疗实验性RA的作用机制。方法:日本大耳兔40只,体重(2.5±0.25)kg,雌雄各半,分为空白对照组、RA模型组、直接灸治疗组、隔姜灸治疗... 目的:观察不同灸法对实验性RA模型关节滑膜液中炎症因子IL-6、IL-8、MCP-1表达的影响,探索不同灸法治疗实验性RA的作用机制。方法:日本大耳兔40只,体重(2.5±0.25)kg,雌雄各半,分为空白对照组、RA模型组、直接灸治疗组、隔姜灸治疗组、温和灸治疗组,每组8只日本大耳兔。采用福氏完全佐剂塑造实验性RA模型,于造模后第7天开始,分别用直接灸、隔姜灸、温和灸3种不同方法施灸日本大耳兔"肾俞"、"足三里"穴各5壮,两侧交替进行,每日1次。6天为1个疗程,共3个疗程。结果:直接灸、隔姜灸、温和灸均能明显降低RA模型动物关节滑膜液中IL-6、IL-8、MCP-1含量(P<0.05,0.01),其中尤以隔姜灸作用最为明显。结论:不同灸法治疗实验性RA均有明显的抗炎效应,且以隔姜灸作用最为显著;不同灸法抑制IL-6、IL-8、MCP-1炎症因子表达可能是其发挥抗炎效应的重要机制之一。 展开更多

采用双抗体夹心酶联免疫吸附试验(ELISA)测定40例儿童支气管哮喘发作期、20例缓解期血清白细胞介素1(IL-1)、白细胞介素6(IL-6)、白细胞介素8(IL-8)水平。并设20例健康儿童作正常对照。结果:发作期IL-1(11.90pg/ml±2.84pg/ml)、IL-... 采用双抗体夹心酶联免疫吸附试验(ELISA)测定40例儿童支气管哮喘发作期、20例缓解期血清白细胞介素1(IL-1)、白细胞介素6(IL-6)、白细胞介素8(IL-8)水平。并设20例健康儿童作正常对照。结果:发作期IL-1(11.90pg/ml±2.84pg/ml)、IL-6(0.75ng/ml±0.30ng/ml)、IL-8(0.08ng/ml±0.05ng/ml)水平明显高于缓解期(8.41pg/ml±0.05pg/ml、0.49ng/ml±0.08ng/ml、0.02ng/ml±0.01ng/ml)及对照组(8.40pg/ml±0.32pg/ml、0.40ng/ml±0.09ng/ml、0.02ng/ml±0.02ng/ml),经统计学处理有显著差异(P<0.01)。且血清IL-1水平与IL-6、IL-8比较呈正相关(γ=0.753,γ=0.795,P<0.01)。提示IL-1与IL-6、IL-8一样参与了哮喘的病理过程,哮喘缓解期仍存在气道炎症反应。细胞因子网络失衡可能是哮喘发病的分子生物学基础。可将血清IL-1、IL-6、IL-8水平升高作为支气管哮喘发作的指标之一。 展开更多

目的:探讨甘氨酸对急性坏死性胰腺炎鼠血和胰腺组织中TNF-α、IL-1β、IL-6、IL-8和IL-10的影响。方法:Wistar大鼠150只,随机分为3组:假手术组(SO)、急性坏死性胰腺炎组(ANP)和急性坏死性胰腺炎+甘氨酸处理组(ANP+Gly),以牛磺胆酸钠复... 目的:探讨甘氨酸对急性坏死性胰腺炎鼠血和胰腺组织中TNF-α、IL-1β、IL-6、IL-8和IL-10的影响。方法:Wistar大鼠150只,随机分为3组:假手术组(SO)、急性坏死性胰腺炎组(ANP)和急性坏死性胰腺炎+甘氨酸处理组(ANP+Gly),以牛磺胆酸钠复制急性坏死性胰腺炎大鼠模型,ANP+Gly组大鼠胰管内给予牛磺胆酸钠前10min静脉给予甘氨酸1g/kg。观察血和胰腺组织中ET、TNF-α、IL-1β、IL-6、IL-8和IL-10含量变化,同时观察各组大鼠胰腺组织的病理变化、24h病死率及平均生存时间。结果:在0h、2h、4h、8h和12h的血和胰腺组织中ET、TNF-α、IL-1β、IL-6、IL-8和IL-10水平,ANP组和ANP+Gly组均显著高于SO组(除0h组及ANP组的IL-1012h外)(P<0·01/P<0·05),且在ANP和ANP+Gly组ET、TNF-α、IL-1β、IL-6和IL-8随病程进展而升高(P<0·05),在ANP组IL-10水平随病程进展降低(P<0·05),而在ANP+Gly组IL-10水平随病程进展增高(P<0·05)。在相同时段除ET在ANP组和ANP+Gly组无显著差异外(P>0·05),除0h组外,TNF-α、IL-1β、IL-6和IL-8水平在ANP组显著高于ANP+Gly组(P<0·01),而IL-10水平(除0h和2h组无差异外)ANP+Gly组显著高于ANP组(P<0·05/0·01)。虽然ANP组和ANP+Gly组大鼠胰腺组织的病理变化无显著差异,但ANP+Gly组大鼠24h病死率显著低于ANP组(P<0·05),平均生存时间显著延长于ANP组(P<0·05)。结论:ET、TNF-α、IL-1β、IL-6、IL-8和IL-10参与了大鼠ANP的病理过程,甘氨酸可减少促炎细胞因子TNF-α、IL-1β、IL-6和IL-8的产生,上调抗炎细胞因子IL-10的负调控作用,有助于减轻ANP时炎性细胞因子瀑布样级联反应,降低大鼠24h病死率及延长平均生存时间。 展开更多

目的:观察地奥司明(DOSM)对肾缺血/再灌注(I/R)大鼠血清和肾组织中TNF-α、IL-1β、IL-6、IL-8和IL-10水平的影响,并探讨其对肾脏的保护机制。方法:180只SD大鼠随机分成3组:假手术组(sham)、肾缺血/再灌注模型组(I/R)和地奥司明+肾缺血... 目的:观察地奥司明(DOSM)对肾缺血/再灌注(I/R)大鼠血清和肾组织中TNF-α、IL-1β、IL-6、IL-8和IL-10水平的影响,并探讨其对肾脏的保护机制。方法:180只SD大鼠随机分成3组:假手术组(sham)、肾缺血/再灌注模型组(I/R)和地奥司明+肾缺血/再灌注模型组(DOSM+I/R)。采用ELISA方法测定血清和肾脏组织中TNF-α、IL-1β、IL-6、IL-8和IL-10的水平,同时检测血肌酐(Cr)和尿素氮(BUN)水平。结果:在1 h、3 h、6 h、12 h、24 h和48 h不同时点的血清和肾组织中TNF-α、IL-1β、IL-6、IL-8和IL-10水平在I/R和DOSM+I/R组均显著高于sham组(P<0.01或P<0.05),且在I/R和DOSM+I/R组TNF-α、IL-1β、IL-6和IL-8随病程进展而升高,在I/R组IL-10水平随病程进展降低,而在DOSM+I/R组IL-10水平随病程进展而升高,TNF-α、IL-1β、IL-6和IL-8水平在I/R组显著高于DOSM+I/R组(P<0.05或P<0.01,除血清TNF-α1 h组外),而IL-10水平在DOMS+I/R组显著高于I/R组(P<0.01或P<0.05)。血Cr和BUN水平在DOSM+I/R组显著低于I/R组(P<0.05或P<0.01)。结论:DOSM可减少肾I/R病理过程中促炎性细胞因子TNF-α、IL-1β、IL-6和IL-8的产生,促进抗炎细胞因子IL-10的产生,有助于减轻I/R时炎性细胞因子瀑布样级联反应,发挥抗炎作用,从而减轻缺血再灌注损伤,保护肾功能。 展开更多
