Ciliary neurotrophic factor has neuroprotective effects mediated through signal transducer and Janus kinase(JAK) 2/activator of transcription 3(STAT3) and phosphatidylinositol 3-kinase(PI3 K)/Akt signaling pathw...Ciliary neurotrophic factor has neuroprotective effects mediated through signal transducer and Janus kinase(JAK) 2/activator of transcription 3(STAT3) and phosphatidylinositol 3-kinase(PI3 K)/Akt signaling pathways.Whether ciliary neurotrophic factor is neuroprotective for glutamate-induced excitotoxicity of dorsal root ganglion neurons is poorly understood.In the present study,the in vitro neuroprotective effects of ciliary neurotrophic factor against glutamate-induced excitotoxicity were determined in a primary culture of dorsal root ganglion neurons from Wistar rat embryos at embryonic day 15.Whether the JAK2/STAT3 and PI3 K/Akt signaling pathways were related to the protective effects of ciliary neurotrophic factor was also determined.Glutamate exposure inhibited neurite outgrowth,cell viability,and growth-associated protein 43 expression and promoted apoptotic neuronal cell death,all of which were reversed by the administration of exogenous ciliary neurotrophic factor.Additionally,preincubation with either JAK2 inhibitor AG490 or PI3 K inhibitor LY294002 blocked the neuroprotective effect of ciliary neurotrophic factor.These data indicate that the two pathways JAK2/STAT3 and PI3 K/Akt play major roles in mediating the in vitro neuroprotective effects of ciliary neurotrophic factor on dorsal root ganglion neurons with glutamate-induced neurotoxicity.展开更多
Education is a complex system that has evolved over thousands of years to reach its current level.It has many objects and subjects.The education systems of the countries are very diverse.Almost every country has its o...Education is a complex system that has evolved over thousands of years to reach its current level.It has many objects and subjects.The education systems of the countries are very diverse.Almost every country has its own ranking approach,because there is no universally accepted scientific theory of education.The search for effective reform in education continues today,but any reform that is not based on scientific theory cannot solve the problem.There are many problems in the content and management of education.Knowledge assessment is also flawed.No country can build an ideal school.It can be considered that in the last hundred years,education has not developed conceptually in the desired direction.Thus,education aims to train strong personalities,not perfect(wise)people.Although individualistic education may seem beneficial locally,globally it divides humanity and prevents its sustainable and harmonious living.However,in societies made up of perfect people,in principle there will be no division,harmony will exist,because perfect people solve problems not by force,but by reason,prefer cooperation rather than conflict.This means protecting the planet.To make the world a gun-free society,the view of education must change conceptually.This article presents a new philosophical view of teaching knowledge and proposes a new model,criteria,and theory.展开更多

Methods: The standard medium was keratinocyte growth medium wit hout bovine pituitary extract (BPE), hydroco...To investigate the proliferative ef fect of keratinocyte growth factor (KGF 2) on human adult keratinocytes. Methods: The standard medium was keratinocyte growth medium wit hout bovine pituitary extract (BPE), hydrocortisone or epidermal growth factor ( EGF). Keratinocytes from a 48 year old subject were cultured and seeded on dis hes with standard medium of EGF in cell density of 2×10 4/32 mm 2. After 24 hours, the medium was replaced by the standard medium with 0, 4, 16, 125 and 50 0 ng/ml KGF 2, respectively. The standard medium with EGF was used as the posit ive control and the standard medium without EGF or KGF 2 was used as the negati ve controls. The growth of keratinocytes was monitored by 3 (4,5 dimethythiazo l 2 yl) 2,5 dipheyl tetrazolium bromide (MTT) assay and by photographs on day s 3, 5 and 7, respectively. Results: KGF 2 in concentrations of 4 500 ng/ml showed a sign ificant proliferative effect on days 5 and 7 as compared with that of the negati ve controls (P< 0.01 ). On day 3 the cells were prolifer ated to 1.5 2.5 fold, on day 5 to 3 5 fold and on day 7 to 3 12 fo ld in KGF 2 medium as that of the negative controls. The optimal response occur red when the concentration of KGF 2 was 125 ng/ml on day 7. Cell proliferation was also consistently higher in all KGF 2 concentrations as compared with that of the positive controls. Conclusions: KGF 2 has significant effects on the proliferatio n of adult keratinocytes, which are more effective than that of EGF. This study supports KGF 2 can improve the healing of chronic wounds in adults in clinic.展开更多

Methods: According to a random number table, 24 Tats w...Objective: To explore the protective effect of sodium tanshinone ⅡA sulfonate (STS) on small intestine injury in rats with sepsis and its possible mechanism. Methods: According to a random number table, 24 Tats were randomly divided into 3 groups: sham operation group (sham group), sepsis model group (model group) and STS treatment group (STS group), with 8 Tats in each group. A rat model of sepsis was induced by cecal ligation and puncture (CLP) for 5 h. STS (1 mg/kg) was slowly injected through the right external jugular vein after CLP. The histopathologic changes in the intestine tissue were observed under a light microscope, and the intestinal epithelial cell apoptosis was evaluated by terminal deoxynucleoddyl transferase (TdT)-mediated dUTP nick-end labeling (TUNEL) method. The expressions of Bcl-2, Bax and nuclear factor κB (NF- κ B) p65 in the intestinal tissue was determined by Western blot. The levels of tumor necrosis factor α (TNF-α) and interleukin 6 (IL-6) in the intestinal tissue were determined using enzyme-linked immuno-sorbent assay (ELISA). Results: Obvious injuries were observed in the intestinal tissue in the CLP group compared with the sham group. The expression of NF- K B p65 and the levels of TNF- α and IL-6 were up-regulated after CLP, the apoptosis of intestinal epithelial cells was increased after CLP, and the ratio of Bcl-2 to Bax was decreased. STS post- treatment could attenuate the injury on the intestinal tissue induced by CLP, decrease the apoptosis of intestinal epithelial cells and the levels of NF- κ B p65, TNF-α and IL-6, and increase the ratio of Bcl-2 to Bax. Conclusion: STS can protect the small intestine in rats with sepsis, and the mechanism may be associated with the inhibition of intestinal epithelial apoptosis and the reduction of activation of inflammatory cytokines.展开更多

In this paper, the impact of damping on the their stat...High speed VCSELs are important optical devices in short-reach optical communication links and interconnects because of their low cost and high modulation speeds. In this paper, the impact of damping on the their static and dynamic characteristics is analyzed and demonstrated. Through the shallow corrosion of the top layer DBR, the VCSELs with different damping is designed and fabricated. With the increase of the surface etch depth from 0 to ~55 nm for 9 μm oxide-aperture VCSEL, the K factor related with the damping is reduced from0.31 to 0.23 ns;. When the etch depth of the VCSEL with 9 μm oxide-aperture is decreased to ~25 nm,output power is increased from 4.03 to 4.70 mW and small signal modulation bandwidth is also increased from15.46 to 16.37 GHz. It shows that there is a tradeoff between damping and differential gain for improving modulation speed.展开更多
Soil erodibility(K factor)mapping has been accomplished mainly by soil map-linked or geo-statistical interpolation.However,the resulting maps usually have coarse spatial resolution at a regional scale.The objectives o...Soil erodibility(K factor)mapping has been accomplished mainly by soil map-linked or geo-statistical interpolation.However,the resulting maps usually have coarse spatial resolution at a regional scale.The objectives of this study were a)to map the K factors using a set of environmental variables and random forest(RF)model,and b)to identify the important environmental variables in the predictive mapping on a regional scale.We collected 101 surface soil samples across southeast China in the summer of 2019.For each sample,we measured the particle size distribution and organic matter content,and calculated the K factors using the nomograph equation.The hyperparameters of RF were optimized through 5-fold cross validation(m_(ay)=2,n_(tree)=500,p=63),and a digital map with 250 m resolution was generated for the K factor.The lower and upper limits of a 90% prediction interval were also pro-duced for uncertainty analysis.It was found that the important environmental variables for the K factor prediction were relief,climate,land surface temperature and vegetation indexes.Since the existing K factor map has an average polygonal area of 6.8 km^(2),our approach dramatically improves the spatial resolution of the K factor to 0.0625 km^(2).The new method captures more distinct differences in spatial details,and the spatial distribution of the K factor derived from RF prediction followed a similar pattern with kriging interpolation.This suggests the presented approach in this study is effective for mapping the K factor with limited sampling data.展开更多

Soil erosion is one of the main environmental problems in the Mediterranean area.This problem is becoming even more important especially in Italy,in the Apennines,where severe erosive processes occur due to the action...Soil erosion is one of the main environmental problems in the Mediterranean area.This problem is becoming even more important especially in Italy,in the Apennines,where severe erosive processes occur due to the action of concentrated running water.The erodibility(K-Factor)of a soil,estimated using the Revised Universal Soil Loss Equation(RUSLE),is a measure of its susceptibility to erosion and depends on several soil properties such as organic matter,texture and permeability and structure.To assess the spatial variability of soil properties and soil erodibility in hilly agricultural areas and to investigate the relationships between soil features and landscape morphodynamics,a detailed study in Molise region(southern Italy),in a small drainange basin located along its hilly Adriatic flank,was carried out.In this catchment,63 topsoil samples(A horizons)were collected and 10 soil profiles,forming a catena crossing 3 land units,were sampled.The calculated K-Factors ranges between 0.012 and 0.048 t ha h ha-1 MJ-1 mm-1 indicating a complex spatial distribution,due to the several local pedological and geomorphological factors affecting soil erodibility.The results give clear evidence about the relationships among soil characteristics,soil erodibility and landscape morpho-dynamics(land units).Comparing the soil loss rates estimated for the study area with those reported in literature,a good correspondence can be observed only for the more stable land unit,not characterized by intense erosive processes.The proposed methodology is suitable to highlight areas characterized by similar morphodynamics features,and comparable soil erodibility,for a more effective spatialization of K factor.展开更多
