Acute kidney injury(AKI)is a significant global health issue with limited current treatment options.This study focused on the mechanism by which exosomes derived from bone marrow mesenchymal stem cells(BMSCs)promote renal tubule regeneration in AKI through the regulation of the PTEN signaling pathway by miR-21.BMSCs were isolated and characterized,and their exosomes were purified.In vitro,renal tubular epithelial cell injury models were established,and the co-culture of exosomes and cells demonstrated enhanced cell proliferation and reduced apoptosis.In vivo,AKI animal models showed improved renal function and histopathological changes after exosome treatment.miR-21 was found to be upregulated in exosomes and recipient cells,targeting PTEN and activating the PI3K/AKT pathway.The signaling network also interacted with other pathways related to renal tubule regeneration.The study highlights the potential of exosome therapy for AKI and provides insights into the underlying molecular mechanisms,although further research is needed to address remaining challenges and translate these findings into clinical applications. 展开更多

Acetaminophen is a drug used to treat many conditions as headache, muscle aches, arthritis, backache, toothache, and fever between others, but collateral effects of this drug are not well known yet. Here is tested its effect on proximal tubule epithelium. Acetaminophen (APAP) at doses of 200, 500, 1000 and 1500 mg/Kg i.p. caused cell damage and changes in F-actin distribution in the proximal tubule of male Wistar rats. After 48 hours of treatment, the proximal tubule epithelium showed tumefaction and necrosis. Dose of 200 mg/kg decreased the F-actin and was observed a structure in patches in the basal cytoplasm of epithelial cells of the proximal tubule. This effect was increased depending on the administered dose. Dose of 1000 mg/kg produced the highest histological damage and changes in the actin cytoskeleton. Results of this study suggested that nephrotoxic damage produced by high doses of APAP included breakdown of cytoskeleton in proximal tubule epithelium. 展开更多

[目的]探讨氧化应激对大鼠肾小管上皮细胞(NRK)生物学行为的影响及L -表没食子儿茶素没食子酸酯(L -EGCG)对NRK细胞氧化性损伤的保护作用.[方法]利用H2O2刺激离体培养的NRK细胞建立氧化损伤的模型,筛选H2O2对NRK细胞损伤研究的最适剂量与最佳时点.然后将培养细胞分EGCG正常对照组、H2O2组、不同浓度儿茶素组.MTT法检测细胞活力,流式细胞仪测定细胞凋亡,荧光探针JC-1测定线粒体膜电位.利用生物化学法检测各组NRK细胞培养上清中丙二醛(MDA)含量及乳酸脱氢酶(LDH)漏出量.[结果]①250 μmol/L H2O2作用6 h剂量组即可引起NRK细胞损伤;②L-EGCG能明显改善H2O2导致的细胞损伤,可使细胞存活率升高,LDH释放量降低,MDA生成减少,稳定了线粒体膜电位,减轻了细胞凋亡.[结论]氧化应激能导致大鼠肾小管上皮细胞活力下降,凋亡增加,L-EGCG可能是通过提高NRK细胞的抗氧化能力而提高其对H2O2损伤的保护及损伤修复能力. 展开更多

Acute kidney injury(AKI) is one of the frequent organ damage in severe COVID-19 patients. Previous studies have shown that human renal tubule cells could be the potential host cells targeted by SARS-CoV-2. Traditional cancer cell lines or immortalized cell lines are genetically and phenotypically different from host cells. Animal models are widely used, but often fail to reflect a physiological and pathogenic status because of species tropisms. There is an unmet need for normal human epithelial cells for disease modeling. In this study, we successfully established long term cultures of normal human kidney proximal tubule epithelial cells(KPTECs) in 2 D and 3 D culture systems using conditional reprogramming(CR) and organoids techniques.These cells had the ability to differentiate and repair DNA damage, and showed no transforming property. Importantly, the CR KPTECs maintained lineage function with expression of specific transporters(SLC34 A3 and cubilin). They also expressed angiotensin-converting enzyme 2(ACE2), a receptor for SARS-CoV and SARS-CoV-2. In contrast, cancer cell line did not express endogenous SLC34 A3, cubilin and ACE2. Very interestingly, ACE2 expression was around twofold higher in 3 D organoids culture compared to that in 2 D CR culture condition. Pseudovirion assays demonstrated that SARS-CoV spike(S) protein was able to enter CR cells with luciferase reporter. This integrated 2 D CR and 3 D organoid cultures provide a physiological ex vivo model to study kidney functions, innate immune response of kidney cells to viruses, and a novel platform for drug discovery and safety evaluation. 展开更多

Background:Ischemic acute kidney injury(AKI)is a common syndrome associated with considerable mortality and healthcare costs.Up to now,the underlying pathogenesis of ischemic AKI remains incompletely understood,and specific strategies for early diagnosis and treatment of ischemic AKI are still lacking.Here,this study aimed to define the transcriptomic landscape of AKI patients through single-cell RNA sequencing(scRNA-seq)analysis in kidneys.Methods:In this study,scRNA-seq technology was applied to kidneys from two ischemic AKI patients,and three human public scRNA-seq datasets were collected as controls.Differentially expressed genes(DEGs)and cell clusters of kidneys were determined.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis,as well as the ligand-receptor interaction between cells,were performed.We also validated several DEGs expression in kidneys from human ischemic AKI and ischemia/reperfusion(I/R)injury induced AKI mice through immunohistochemistry staining.Results:15 distinct cell clusters were determined in kidney from subjects of ischemic AKI and control.The injured proximal tubules(PT)displayed a proapoptotic and proinflammatory phenotype.PT cells of ischemic AKI had up-regulation of novel pro-apoptotic genes including USP47,RASSF4,EBAG9,IER3,SASH1,SEPTIN7,and NUB1,which have not been reported in ischemic AKI previously.Several hub genes were validated in kidneys from human AKI and renal I/R injury mice,respectively.Furthermore,PT highly expressed DEGs enriched in endoplasmic reticulum stress,autophagy,and retinoic acid-inducible gene I(RIG-I)signaling.DEGs overexpressed in other tubular cells were primarily enriched in nucleotide-binding and oligomerization domain(NOD)-like receptor signaling,estrogen signaling,interleukin(IL)-12 signaling,and IL-17 signaling.Overexpressed genes in kidney-resident immune cells including macrophages,natural killer T(NKT)cells,monocytes,and dendritic cells were associated with leukocyte activation,chemotaxis,cell adhesion,and complement activation.In addition,the ligand-receptor interactions analysis revealed prominent communications between macrophages and monocytes with other cells in the process of ischemic AKI.Conclusion:Together,this study reveals distinct cell-specific transcriptomic atlas of kidney in ischemic AKI patients,altered signaling pathways,and potential cell-cell crosstalk in the development of AKI.These data reveal new insights into the pathogenesis and potential therapeutic strategies in ischemic AKI. 展开更多

Objective:Dopamine,via its receptors,plays a vital role in the maintenance of blood pressure by modulating renal sodium transport.However,the role of the D_(4)dopamine receptor(D_(4)receptor)in renal proximal tubules(PRTs)is still unclear.This study aimed to verify the hypothesis that activation of D_(4)receptor directly inhibits the activity of the Na+-K+-ATPase(NKA)in RPT cells.Methods:NKA activity,nitric oxide(NO)and cyclic guanosine monophosphate(cGMP)levels were measured in RPT cells treated with the D_(4)receptor agonist PD168077 and/or the D_(4)receptor antagonist L745870,the NO synthase inhibitor NG-nitro-L-arginine-methyl ester(L-NAME)or the soluble guanylyl cyclase inhibitor 1H-[1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one(ODQ).Total D_(4)receptor expression and its expression in the plasma membrane were investigated by immunoblotting in RPT cells from Wistar-Kyoto(WKY)rats and spontaneously hypertensive rats(SHRs).Results:Activation of D_(4)receptors with PD168077,inhibited NKA activity in RPT cells from WKY rats in a concentration-and time-dependent manner.The inhibitory effect of PD168077 on NKA activity was prevented by the addition of the D_(4)receptor antagonist L745870,which by itself had no effect.The NO synthase inhibitor L-NAME and the soluble guanylyl cyclase inhibitor ODQ,which by themselves had no effect on NKA activity,eliminated the inhibitory effect of PD168077 on NKA activity.Activation of D_(4)receptors also increased NO levels in the culture medium and cGMP levels in RPT cells.However,the inhibitory effect of D_(4)receptors on NKA activity was absent in RPT cells from SHRs,which could be related to decreased plasma membrane expression of D_(4)receptors in SHR RPT cells.Conclusions:Activation of D_(4)receptors directly inhibits NKA activity via the NO/cGMP signaling pathway in RPT cells from WKY rats but not SHRs.Aberrant regulation of NKA activity in RPT cells may be involved in the pathogenesis of hypertension. 展开更多

The effect of L-arginine on the progression of chronic renal scarring in remnant kidney

OBJECTIVE: To investigate the effect of L-arginine (L-arg) on early compensatory renal growth (CRG), tubulointerstitial accumulation of extracellular matrix (ECM), long term survival rate and renal scarring in rats with 5/6 nephrectomy (SNx). METHODS: The experiment included four groups of rats (n = 5 each group): (1) Sham group, (2) SNx group, (3) SNx + L-arg group, and (4) Sham + L-arg group (L-arg 1% in drinking water). Parameters related with CRG and early tubulointerstitial expression of ECM and alpha smooth muscle actin (alphaSMA) were evaluated by immunohistochemistry at day 30. The survival rate and the extent of renal scarring in the rats were observed at day 120. 目的比较观察甲基胍、1。甲脲乙醇酸酐对人肾小管上皮细胞的毒性作用。方法HK.2细胞作为研究对象,分为三组培养:正常对照(A)组,甲基胍(B)组、1-甲脲乙醇酸酐(C)组,用MTr比色试验法检测细胞增殖,NAG酶释放检测对细胞的毒性... 目的比较观察甲基胍、1。甲脲乙醇酸酐对人肾小管上皮细胞的毒性作用。方法HK.2细胞作为研究对象,分为三组培养:正常对照(A)组,甲基胍(B)组、1-甲脲乙醇酸酐(C)组,用MTr比色试验法检测细胞增殖,NAG酶释放检测对细胞的毒性,Hoechst染色及AnnexinV—FITC/PI流式细胞术检测凋亡。结果HK-2细胞加入甲基胍、1-甲脲乙醇酸酐后,较正常对照组吸光度值显著下降(0.188±0.011,0.176±0.010VS0.545±0.021,F=1557.74,P〈0.01),NAG酶浓度显著上升(20.488±0.473,22.225±0.565VS5.1254-0.198,F:3848.22,P〈0.01)。Hoechst33258染色显示凋亡细胞呈现核固缩,染色加深,并出现凋亡小体。流式细胞术显示B组、C组HK-2细胞凋亡率显著高于A组(18.23±1.1581,20.22±1.1433VS2.4734-0.321,F=526.06,P〈0.01)。C组吸光度值显著低于B组(0.176±0.010VS0.1884-0.011,t=2.26,P〈0.05),NAG酶浓度显著高于B组(22.225±0.565VS20.488±0.473,t=-6.67.P〈0.01),凋亡率显著高于B组(20.224-1.1433VS18.23±1.1581,t=-2.762,P〈0.05)。结论1.甲脲乙醇酸酐对肾小管上皮细胞的毒性作用强于甲基胍。 展开更多

目的研究微小RNA(miR)-29a在急性肾损伤患者血清中的表达情况,探究miR-29a是否通过调控膦酸酯酶张力蛋白同源物(PTEN)表达影响肾小管细胞凋亡。方法收集113例急性肾损伤(AKI)患者以及110例健康对照者血清,通过实时荧光定量PCR检测血清中... 目的研究微小RNA(miR)-29a在急性肾损伤患者血清中的表达情况,探究miR-29a是否通过调控膦酸酯酶张力蛋白同源物(PTEN)表达影响肾小管细胞凋亡。方法收集113例急性肾损伤(AKI)患者以及110例健康对照者血清,通过实时荧光定量PCR检测血清中miR-29a的表达情况。体外培养人肾小管上皮细胞(HK-2),通过转染miR-29a NC和miR-29a mimic到HK-2细胞中,用TGF-β1诱导HK-2细胞凋亡,采用流式细胞仪检测细胞凋亡,用生物信息学预测miR-29a的潜在靶基因,荧光素酶报告基因实验验证miR-29a与PTEN的互补配对关系,Western blot检测潜在靶基因PTEN的表达情况。再通过转染过表达PTEN的质粒,用流式细胞仪检测HK-2细胞的凋亡情况。结果与健康对照者血清相比,AKI患者血清中的miR-29a表达水平降低(P<0.05);流式细胞仪结果显示转染miR-29a mimic后,HK-2细胞的凋亡明显少于转染miR-29a NC组(P<0.05);荧光素酶报告基因实验结果显示,miR-29a与PTEN具有互补配对关系;转染过表达PTEN后,肾小管上皮细胞的凋亡率增加(P<0.05)。结论在AKI患者血清中,miR-29a表达水平降低,过表达miR-29a通过抑制PTEN蛋白的表达从而抑制肾小管上皮细胞凋亡。 展开更多

目的 探讨血凝素样氧化低密度脂蛋白受体-1(LOX-1)在氧化低密度脂蛋白(oxLDL)刺激肾小管上皮细胞摄取脂质中的作用.方法 体外培养NRK52E,加入不同浓度(0,25,50,100μg/ml)的ox-LDL及预先与LOX-1阻滞剂多聚肌苷酸(polyinosonic ac... 目的 探讨血凝素样氧化低密度脂蛋白受体-1(LOX-1)在氧化低密度脂蛋白(oxLDL)刺激肾小管上皮细胞摄取脂质中的作用.方法 体外培养NRK52E,加入不同浓度(0,25,50,100μg/ml)的ox-LDL及预先与LOX-1阻滞剂多聚肌苷酸(polyinosonic acid,poly I)和爱兰苔胶(carrageenan)作用2 h,再加入50μg/ml的ox-LDL,24 h后用Realtime-PCR测定LOX-1 mRNA的表达水平,Western blot测定LOX-1蛋白表达,用油红O法测定细胞内脂质聚集情况.结果 在25~100μ/ml范围内随着ox-LDL浓度的增加,LOX-1 mRNA和蛋白的表达增加,分别为0 μg/ml的2.13、10.14、20.81倍和2.53、12.18、21.45倍,各组间比较差异均有统计学意义(P〈0.05);而随着LOX-1 mRNA和蛋白的表达增加细胞内脂质聚集也增加,分别为0μg/ml的3.2,6.4和12.5倍(P〈0.05);polyI或carrageenan使50μg/ml的ox-LDL诱导的LOX-1 mRNA和蛋白的表达分别下降(48%,47%)和(72%,65%),同时细胞内脂质聚集减少41%和49%,LOX-1与细胞内脂质呈正相关(r=0.97,P〈0.05).结论 ox-LDL诱导NRK52E细胞表达LOX-1,又通过LOX-1促进脂质在细胞内聚集从而损伤细胞,这种诱导作用可被LOX-1的阻断剂部分阻断. 展开更多

目的 了解体外高糖刺激对肾小管上皮细胞(HK-2)脂联素受体(adipoR)表达的影响.方法 将HK-2细胞在含10%胎牛血清的低糖DMEM培养液中培养至贴壁且80%融合时,无血清培养24 h,改用含葡萄糖1、2,4、6、8 mg/ml的培养液继续培养48 h,RT-PC... 目的 了解体外高糖刺激对肾小管上皮细胞(HK-2)脂联素受体(adipoR)表达的影响.方法 将HK-2细胞在含10%胎牛血清的低糖DMEM培养液中培养至贴壁且80%融合时,无血清培养24 h,改用含葡萄糖1、2,4、6、8 mg/ml的培养液继续培养48 h,RT-PCR及westernblot检测ad-ipoR的表达.将HK-2细胞分别在含10%胎牛血清的高糖(4 mg/ml)、低糖(1 ms/ml)DMEM培养液中培养0、12、24、48、72、96 h后提取总RNA,RT-PCR检测adipoR mRNA的表达.结果 adipoR1/R2基因在HK-2细胞上均有表达,且adipoR1(0.63±0.12)基因表达量是adipoR2(0.16±0.03)的3.9倍,差异有统计学意义(P〈0.01).各不同浓度葡萄糖及作用时间对HK-2细胞adipoR的基因表达无明显影响(P〉0.05).westernblot检测到HK-2细胞上adipoR1的蛋白表达,其表达量不受葡萄糖浓度的影响(P〉0.05).结论 adipoR1/R2基因在HK-2细胞上均有表达,且以adipoR1为主,提示adipoR1在肾脏疾病中具有更重要的意义.HK-2细胞adipoR的表达不受高糖的影响. 展开更多
