BACKGROUND Hepatocellular carcinoma(HCC)has been a pervasive malignancy throughout the world with elevated mortality.Efficient therapeutic targets are beneficial to treat and predict the disease.Currently,the exact molecular mechanisms leading to the progression of HCC are still unclear.Research has shown that the microRNA-142-3p level decreases in HCC,whereas bioinformatics analysis of the cancer genome atlas database shows the ASH1L expression increased among liver tumor tissues.In this paper,we will explore the effects and mechanisms of microRNA-142-3p and ASH1L affect the prognosis of HCC patients and HCC cell bioactivity,and the association between them.AIM To investigate the effects and mechanisms of microRNA-142-3p and ASH1L on the HCC cell bioactivity and prognosis of HCC patients.METHODS In this study,we grouped HCC patients according to their immunohistochemistry results of ASH1L with pathological tissues,and retrospectively analyzed the prognosis of HCC patients.Furthermore,explored the roles and mechanisms of microRNA-142-3p and ASH1L by cellular and animal experiments,which involved the following experimental methods:Immunohistochemical staining,western blot,quantitative real-time-polymerase chain reaction,flow cytometric analysis,tumor xenografts in nude mice,etc.The statistical methods involved in this study contained t-test,one-way analysis of variance,theχ^(2)test,the Kaplan-Meier approach and the log-rank test.RESULTS In this study,we found that HCC patients with high expression of ASH1L possess a more recurrence rate as well as a decreased overall survival rate.ASH1L promotes the tumorigenicity of HCC and microRNA-142-3p exhibits reduced expression in HCC tissues and interacts with ASH1L through targeting the ASH1L 3′untranslated region.Furthermore,microRNA-142-3p promotes apoptosis and inhibits proliferation,invasion,and migration of HCC cell lines in vitro via ASH1L.For the exploration mechanism,we found ASH1L may promote an immunosuppressive microenvironment in HCC and ASH1L affects the expression of the cell junction protein zonula occludens-1,which is potentially relevant to the immune system.CONCLUSION Loss function of microRNA-142-3p induces cancer progression and immune evasion through upregulation of ASH1L in HCC.Both microRNA-142-3p and ASH1L can feature as new biomarker for HCC in the future. 展开更多

[Result] The expression quality of the gene extracted from leaves, stem and earthnut increased and the expression quality reached the peak point 24,72 and 72 h after low temperature treatment respectively. 目的:探讨环状RNA(CircRNA)RTN4调节微小RNA(miR)-224-5p/髓磷脂转录因子1样(MYT1L)轴对胶质母细胞瘤(GM)细胞恶性生物学行为的影响。方法:以对数生长期的U251细胞进行设置分组:空白组、阴性对照(sh NC)组、沉默circRTN4 shRNA(sh circR... 目的:探讨环状RNA(CircRNA)RTN4调节微小RNA(miR)-224-5p/髓磷脂转录因子1样(MYT1L)轴对胶质母细胞瘤(GM)细胞恶性生物学行为的影响。方法:以对数生长期的U251细胞进行设置分组:空白组、阴性对照(sh NC)组、沉默circRTN4 shRNA(sh circRTN4)组、sh circRTN4+抑制剂对照(inhibitor NC)组、sh circRTN4+miR-224-5p抑制剂(miR-224-5p inhibitor)组。Transwell实验、流式细胞术、CCK-8、qRT-PCR、Western blot以及双荧光素酶分别检测细胞迁移与侵袭、凋亡、增殖、CircRTN4、miR-224-5p、MYT1L mRNA表达、增殖蛋白(ki-67)、凋亡蛋白(cleaved Caspase-3)、MYT1L蛋白表达以及miR-224-5p分别与CircRTN4、MYT1L靶向关系验证;qRT-PCR检测GM细胞系U118、U87、U251和LN229细胞及正常人星形胶质细胞系NHA细胞中CircRTN4、miR-224-5p、MYT1L mRNA表达水平。结果:U118、U87、U251和LN229细胞中CircRTN4、MYT1L mRNA表达较NHA细胞显著增加,miR-224-5p表达显著下降(P<0.05);miR-224-5p分别与CircRTN4、MYT1L的有靶向关系。与空白组、sh NC组相比,sh circRTN4组迁移、侵袭数、24h、48h A450值、ki-67、CircRTN4、MYT1L mRNA及蛋白表达显著降低,凋亡率、cleaved Caspase-3表达显著增加(P<0.05);与sh circRTN4+inhibitor NC组相比,sh circRTN4+miR-224-5p inhibitor组迁移、侵袭数、24h、48h A450值、ki-67、MYT1L mRNA及蛋白表达显著增加,凋亡率、cleaved Caspase-3表达显著下降(P<0.05),CircRTN4表达无统计学差异(P>0.05),体内实验证明干扰CircRTN4可显著抑制移植瘤裸鼠肿瘤质量(P<0.05)。结论:沉默CircRTN4调节miR-224-5p/MYT1L轴抑制GM细胞恶性生物学行为。 展开更多

本研究旨在通过构建非洲猪瘟病毒(ASFV)MGF360-13L基因原核表达系统表达13L蛋白,并制备其鼠源多克隆抗体。利用生物信息学方法,对ASFV MGF360-13L基因进行序列比对,分析其同源性、构建遗传进化树;将非洲猪瘟病毒MGF360-13L基因密码子优... 本研究旨在通过构建非洲猪瘟病毒(ASFV)MGF360-13L基因原核表达系统表达13L蛋白,并制备其鼠源多克隆抗体。利用生物信息学方法,对ASFV MGF360-13L基因进行序列比对,分析其同源性、构建遗传进化树;将非洲猪瘟病毒MGF360-13L基因密码子优化后进行合成,连接至PET32a载体构建重组质粒pET32a-13L,转化至E.coliBL21(DE3)感受态细胞,经IPTG诱导表达获得目的蛋白,采用镍柱纯化法进行蛋白纯化。将纯化后的蛋白乳化后免疫8周龄BALB/c雌鼠制备多克隆抗体,间接ELISA和Western blot检测抗体特异性。SDS-PAGE结果显示,重组蛋白分子量大小为58.2 kDa,主要以包涵体形式存在;Western blot结果显示免疫猪阳性血清可特异性识别该蛋白,具有良好的反应性,表明该重组蛋白获得正确表达。利用该蛋白免疫小鼠制备多克隆抗体进行Western blot,结果显示其能与MGF360-13L重组蛋白发生特异性反应。间接ELISA测定抗体效价高达1∶256000。本研究成功制备非洲猪瘟病毒MGF360-13L重组蛋白,以其为免疫原制备的多克隆抗体具备较高的特异性和反应性,为进一步阐述MGF360-13L蛋白的生物学功能和研制非洲猪瘟新型疫苗奠定了基础。 展开更多

多次加压条件下L-Met和MWCNT促进剂对CO_(2)水合物生成的影响

自工业革命以来,随着化石燃料被广泛应用,温室气体的排放不断增加,导致气候变化,引发国内外广泛关注。由于温室气体主要为CO_(2),因此近年新出现的废气中CO_(2)捕获、分离技术被广泛研究。其中,水合物法是一种新兴技术,其在应用中的突... 自工业革命以来,随着化石燃料被广泛应用,温室气体的排放不断增加,导致气候变化,引发国内外广泛关注。由于温室气体主要为CO_(2),因此近年新出现的废气中CO_(2)捕获、分离技术被广泛研究。其中,水合物法是一种新兴技术,其在应用中的突出问题是如何获得较快水合物生成速率与较高生成量。CO_(2)水合物快速生成需要高压气体条件,伴随水合物不断生成,系统内气体压力会持续下降,因此多次对反应加压使其恢复至初始压力是生成反应中常用方法。本研究选取L-蛋氨酸(L-Met)和多壁碳纳米管(MWCNT)两种典型动力学促进剂,采用多次加压至反应体系初始压力的方法,比较了此反应条件下不同促进剂对CO_(2)水合物生成反应动力学促进效果。实验结果表明,当反应体系初始压力反复恢复时,MWCNT和L-Met体系均能够显著减少水合物的诱导成核时间,而1.1g/L L-Met能够显著促进二氧化碳水合物的生成,水合物生成量是纯水体系的5倍;L-Met体系水合物生成速率优于MWCNT体系,1.1g/L L-Met气体转化率最高,转化率为70.3%,是纯水体系的6倍。最终实验结果表明,L-Met作为CO_(2)水合物动力学促进剂优于MWCNT,在水合物法捕集、分离烟道废气CO_(2)技术的规模化应用中,可用作一种高效、可靠、环保的动力学促进剂。 展开更多

白粉病是威胁全球小麦生产的一种严重病害,从小麦野生近缘种发掘成株抗白粉病基因资源对培育持久抗性品种十分重要。本研究从硬粒小麦-簇毛麦01I139(V#6)双二倍体AABBVV-3与普通小麦NAU0686杂交、回交的后代中鉴定到一个小麦-簇毛麦2V#6... 白粉病是威胁全球小麦生产的一种严重病害,从小麦野生近缘种发掘成株抗白粉病基因资源对培育持久抗性品种十分重要。本研究从硬粒小麦-簇毛麦01I139(V#6)双二倍体AABBVV-3与普通小麦NAU0686杂交、回交的后代中鉴定到一个小麦-簇毛麦2V#6(2D)异代换系11-2V-1,利用11-2V-1在染色体2V和2D之间产生重组,将11-2V-1与高感白粉病普通小麦NAU0686杂交,F_(1)自交。利用GISH/FISH和分子标记对F_(2)世代131个单株进行分子细胞学鉴定,获得了小麦-簇毛麦T2DS·2V#6L易位系11-2V-2、2V#6S端体系11-2V-3和2V#6L端体系11-2V-4新种质。白粉病抗性鉴定发现,所有F_(2)单株苗期均高感小麦白粉菌生理小种E09,但成株期含有2V#6和2V#6L染色体臂的所有单株均表现高抗(IT=1~2),而仅含有2V#6S染色体臂或无外源染色体的单株均高感白粉病(IT=7~9),表明簇毛麦01I139的2V#6L染色体臂上携带成株抗白粉病基因,该基因可能与小麦-簇毛麦T2BS·2V#5L易位系携带的Pm62是等位基因。遗传效应分析表明,T2DS·2V#6L易位染色体在小麦背景传递正常,对主要农艺性状没有显著的负向影响,对穗长、每穗粒数和单株粒重还表现出一定程度的正向效应。因此,本研究创制的成株抗白粉病T2DS·2V#6L易位系11-2V-2为小麦抗白粉病育种提供了新抗源,也为后续簇毛麦2VL上优异基因遗传定位和图位克隆奠定了基础。 展开更多

Merr.) is a major crop that provides plant-origin protein and oil for humans and livestock. Al- though the soybean vegetative tissues and seeds provide a major source of high-quality protein, they suffer from low con- centration of an essential sulfur-containing amino acid, methionine, which significantly limits their nutritional quality. The level of methionine is mainly controlled by the first unique enzyme of methionine synthesis, cystathione y-synthase (CGS). Aiming to elevate methionine level in vegetative tissues and seeds, we constitutively over-expressed a feedback-insensitive Arabidopsis CGS (AtD-CGS) in soybean cultivars, Zigongdongdou (ZD) and Jilinxiaoli 1 (JX). The levels of soluble methionine increased remarkably in leaves of transgenic soybeans compared to wild-type plants (6.6- and 7.3-fold in two transgenic ZD lines, and 3.7-fold in one transgenic JX line). Furthermore, the total methionine contents were significantly increased in seeds of the transgenic ZD lines (1.5- to 4.8-fold increase) and the transgenic JX lines (1.3- to 2.3-fold increase) than in the wild type. The protein contents of the transgenic soybean seeds were significantly elevated compared to the wild type, suggesting that the scarcity of methionine in soybeans may limit protein accumulation in soybean seeds. The increased protein content did not alter the profile of major storage proteins in the seeds. Generally, this study provides a promising strategy to increase the levels of methionine and protein in soybean through the breeding programs. 展开更多

血清3-NT UCH-L1对帕金森病并发认知功能障碍的诊断效能分析

目的探讨血清3-硝基酪氨酸(3-NT)、泛素C末端水解酶L1(UCH-L1)水平与帕金森病患者并发认知功能障碍的关系及其诊断价值。方法纳入驻马店市中心医院2020-07—2023-07诊治的104例帕金森病患者作为研究对象,根据蒙特利尔认知功能评估量表(M... 目的探讨血清3-硝基酪氨酸(3-NT)、泛素C末端水解酶L1(UCH-L1)水平与帕金森病患者并发认知功能障碍的关系及其诊断价值。方法纳入驻马店市中心医院2020-07—2023-07诊治的104例帕金森病患者作为研究对象,根据蒙特利尔认知功能评估量表(MoCA)评分将患者分为认知功能正常组48例和认知功能障碍组56例。采用酶联免疫吸附法检测血清3-NT、UCH-L1水平,采用Pearson法分析帕金森病认知功能障碍患者血清3-NT、UCH-L1水平与MoCA评分的相关性,采用受试者工作特征(ROC)曲线分析血清3-NT、UCH-L1水平对帕金森病患者认知功能障碍的诊断价值,采用Logistic多因素回归分析帕金森病患者认知功能障碍的影响因素。结果认知功能障碍组血清3-NT[(7.26±2.04)μg/L比(4.58±1.40)μg/L]、UCH-L1[(0.94±0.21)μg/L比(0.62±0.16)μg/L]水平及H-Y分期中/晚期患者比例(78.57%比16.67%)高于认知功能正常组(P<0.05)。认知功能障碍组语言[(4.93±0.52)分比(5.23±0.49)分]、命名[(2.11±0.35)分比(2.46±0.34)分]、注意力[(4.80±0.67)分比(5.31±0.45)分]、抽象思维[(1.21±0.26)分比(1.63±0.20)分]、视空间及执行能力[(2.89±0.39)分比(3.54±0.30)分]、定向力[(5.11±0.48)分比(5.50±0.31)分]、延迟记忆[(2.88±0.36)分比(4.02±0.41)分]、MoCA总分[(23.93±2.05)分比(27.69±1.08)分]低于认知功能正常组(P<0.05)。帕金森病认知功能障碍患者血清3-NT、UCH-L1水平与语言、命名、注意力、抽象思维、视空间及执行能力、定向力、延迟记忆、MoCA总分均呈负相关(P<0.05)。3-NT、UCH-L1单独及其联合诊断帕金森病患者认知功能障碍的AUC分别为0.869、0.852、0.951。3-NT、UCH-L1、H-Y分期均是帕金森病患者并发认知功能障碍的独立危险因素(P<0.05)。结论血清3-NT、UCH-L1水平可用于评估帕金森病患者认知功能障碍的发生。 展开更多

目的:探讨2型糖尿病(DM)患者血清分泌型卷曲蛋白-4(SFRP-4)、壳多糖3样蛋白1(CHI3L1)水平与糖尿病视网膜病变(DR)的相关性。方法:前瞻性研究。选取2018-10/2023-10本院收治的DR患者103例为DR组,其中DR早期39例,DR中期42例,DR晚期22例;... 目的:探讨2型糖尿病(DM)患者血清分泌型卷曲蛋白-4(SFRP-4)、壳多糖3样蛋白1(CHI3L1)水平与糖尿病视网膜病变(DR)的相关性。方法:前瞻性研究。选取2018-10/2023-10本院收治的DR患者103例为DR组,其中DR早期39例,DR中期42例,DR晚期22例;选取单纯2型糖尿病患者98例为DM组,同期选取健康体检者101例为对照组。收集各组受试者基线资料及临床指标。采用酶联免疫吸附(ELISA)法检测血清SFRP-4、CHI3L1水平。结果:DR组患者血清SFRP-4、CHI3L1水平及TG、LDL-C、HbA1C、FPG、HOMA-IR均高于DM组和对照组(均P<0.05);DM组患者血清SFRP-4、CHI3L1水平及TG、LDL-C、HbA1C、FPG、HOMA-IR均高于对照组(均P<0.05)。DR晚期患者病程、TG、LDL-C、HbA1C、FPG、HOMA-IR、SFRP-4、CHI3L1均高于DR中期、DR早期(均P<0.05)。DR患者血清SFRP-4、CHI3L1水平与病程、TG、LDL-C、HbA1C、FPG、HOMA-IR、DR分期均呈正相关(均P<0.05)。血清SFRP-4、CHI3L1及联合诊断DR的曲线下面积(AUC)分别为0.809、0.801、0.898。血清SFRP-4、CHI3L1水平是影响DR发生的危险因素(P<0.05)。结论:血清SFRP-4、CHI3L1水平与2型糖尿病患者DR关系密切,二者水平升高提示患者发生DR的风险较高。 展开更多

目的:探讨卵巢癌(OC)患者血清活性氧调节剂-1(Romo-1)、分泌型卷曲蛋白4(sFRP-4)、线粒体核糖体蛋白L15(MRPL15)水平及与淋巴结转移及预后生存关系。方法:回顾性收集本院2019年7月-2021年3月住院治疗的132例OC患者为OC组,良性病变患者... 目的:探讨卵巢癌(OC)患者血清活性氧调节剂-1(Romo-1)、分泌型卷曲蛋白4(sFRP-4)、线粒体核糖体蛋白L15(MRPL15)水平及与淋巴结转移及预后生存关系。方法:回顾性收集本院2019年7月-2021年3月住院治疗的132例OC患者为OC组,良性病变患者为病变组,体检健康者132例对照组。对OC患者随访3年分为死亡组(n=82)和生存组(n=50)。检测各组血清Romo-1、sFRP-4、MRPL15水平;Cox回归分析影响OC患者预后的相关因素;运用受试者工作特征(ROC)曲线分析血清Romo-1、sFRP-4、MRPL15对OC患者预后生存的预测价值。结果:对照组、病变组、OC组血清Romo-1、MRPL15水平依次升高,血清sFRP-4水平依次降低;OC组淋巴结转移患者血清Romo-1、MRPL15水平高于未转移患者,血清sFRP-4水平低于未转移患者(均P<0.05)。死亡组FIGO分期Ⅲ期~Ⅳ期占比(86.6%)、淋巴结转移占比(74.4%)、低分化程度占比(58.5%)、血清Romo-1(3.81±0.67 ng/ml)、MRPL15(13.26±2.28 ng/ml)水平均高于生存组(48.0%、48.0%、28.0%、2.92±0.58 ng/ml、10.52±1.73 ng/ml),而血清sFRP-4水平(1.75±0.42 ng/ml)低于生存组(2.36±0.49 ng/ml)(均P<0.05)。血清Romo-1、sFRP-4、MRPL15、淋巴结转移、FIGO分期高、分化程度低为影响OC患者预后生存的相关因素(均P<0.05)。血清Romo-1、sFRP-4、MRPL15联合预测OC患者预后生存的曲线下面积为0.957,优于各自单独指标预测(P<0.05)。结论:OC患者血清Romo-1、MRPL15水平异常升高,血清sFRP-4水平异常降低,且与患者淋巴结转移及预后有关,3者联合检测对患者预后生存有较高的预测价值。 展开更多

A total of 2268 1-day ol... Studies on the relative bioavailability (RBV) of DL-Methionine (DL-Met) to L-Methionine (L-Met) have produced variable results. An experiment was conducted to determine the RBV of DL to L-Met. A total of 2268 1-day old male chicken were housed in 54 floor pens (42 bird/pen). There were 9 treatments (6 repetitions) including the basal diet (BD). The BD was deficient in Met content with 0.27, 0.26 and 0.25 in the starter, grower and finisher periods respectively. Four levels of experimental diets for each DL-Met and L-Met were created by supplementing 0.05%, 0.10%, 0.15% and 0.20% of DL- or L-Met to the BD. The feeding program consisted of starter (0-14 d, 21% CP and 2900 kcal ME/kg), grower (15 - 28 d, 20% CP and 3000 kcal ME/kg) and finisher period (29 - 37 d, 18.5% CP and 3050 kcal ME/kg). Chickens and feed were weighed at the end of each age period. Regression coefficients of a common plateau asymptotic regression were used to calculate RBV. Birds responded to gradual increase in Met levels, BW, FCR and ADG were significantly (P 0.05) higher in treatment groups as compared to control. Through the study period (37 d), the RBVs of DL-Met for BW and FCR were 89 and 77 respectively. 展开更多

An important factor in the emergence and progre sion of osteosarcoma(OS)is the dysregulated expression of microRNAs(miRNAs).Transcription factor 7-like 1(TCF7LI),a member of the T cell factor/lymphoid enhancer factor(TCF/LEF)transcription factor family,interacts with the Wnt signaling pathway regulator β-catenin and acts as a DNA-specific binding protein.This study sought to elucidate the impact of the interaction between miR 3293p and TCF7L1 on.the growth and apoptosis of OS and analyze the regulatory expression relationship between miRNA and mRNA in osteosarcoma cells using a variety of approaches.MiR329-3p was significantly downregulated,while TCF7L1 was considerably up-regulated in all examined OS cell lines.Additionally,a clinical comparison study was performed using the TCGA database.Subsequently,the regulatory relationship between miR-329-3p and TCF7L1 on the proliferation and apoptosis of OS cells was verified through in vitro and in vivo experiments.When miR 329-3p was transfected into the OS cell line,the expression of TCF7L1 decreased,the proliferation of OS cells was inhibited,the cytoskeleton disintegrated,and the nucleus condensed to fom apoptotic bodies.The expression of proteins that indicate apoptosis increased simultaneously.The cell cycle was arrested in the G0/G1 phase,and the G1/S transition was blocked.The introduction of miR 3293p also inhibited downstream Cyclin D1 of the Wnt pathway.Xenograf experiments indicated that the overexpression of miR-329-3p signi ficanly inhibited the growth of OS xenografts in nude mice,and the expression of TCF7L1 and C-Myc in tumor tssues decreased.MiR 329-3p was significantly reduced in OS cells and played a suppressive role in tumorigenesis and proliferation by targeting TCF7L1 both in vitro and in vivo.Osteosarcoma cell cycle arrest and pathway inhibition were observed upon the regulation of TCF7LI by miR 3293p.Summarizing these results,it can be inferred that miR.3293p exerts anticancer efects in osteosarcoma by inhibiting TCF7L1. 展开更多
