Lipid peroxide (LPO) concentrations were increased slightly in the medium after incubation of macrophages with normal LDL (n-LDL), while decreased significantly in the media after incubation of macrophages with pox-LDL. In the three groups with pox-LDL, it could be found that there was a dose-dependent decrease of concentrations of LPO and total cholesterol (TCH) in the two RSM groups, and the decrease in the two RSM groups was much greater than in the group without RSM. RSM accelerated a more decrease of LPO than cholesterol contents in the media containing pox-LDL. The ultrastructural studies also showed that RSM induced the accumulation of lipid droplets in the cytoplasm of mouse peritoneal macrophages. The results suggested that RSM could accelerate the phagocytosis and degradation of pox-LDL by macrophages.展开更多

Changes in serum and hepatic lipid parameters were evaluated. Metabolites in serum, urine and liver samples were profiled using ultra-highperformance liquid chromatography/mass spectrometry followed by multivariate statistical analysis to identify potential biomarkers and metabolic pathways.RESULTS P. kingianum significantly inhibited the HFD-induced increase in total cholesterol and triglyceride in the liver and serum. P. kingianum also significantly regulated metabolites in the analyzed samples toward normal status. Nineteen, twenty-four and thirty-eight potential biomarkers were identified in serum, urine and liver samples, respectively. These biomarkers involved biosynthesis of phenylalanine, tyrosine, tryptophan, valine, leucine and isoleucine, along with metabolism of tryptophan, tyrosine, phenylalanine, starch, sucrose, glycerophospholipid, arachidonic acid, linoleic acid, nicotinate, nicotinamide and sphingolipid.CONCLUSION P. kingianum alleviates HFD-induced dyslipidemia by regulating many endogenous metabolites in serum, urine and liver samples. Collectively, our findings suggest that P. kingianum may be a promising lipid regulator to treat dyslipidemia and associated diseases.展开更多

The biological functions of these proteins have been reported to involve plant reproduction and biotic or abiotic stress response. With the completion of the barley genome sequence, a genome-wide analysis of nsLTPs in barley(Hordeum vulgare L.)(HvLTPs) will be helpful for understanding the function of nsLTPs in plants. We performed a genome-wide analysis of the nsLTP gene family in barley and identified 70 nsLTP genes,which can be divided into five types(1, 2, C, D, and G). Each type of nsLTPs shares similar exon and intron gene structures. Expression analysis showed that barley nsLTPs have diverse expression patterns, revealing their various roles. Our results shed light on the phylogenetic relationships and potential functions of barley nsLTPs and will be useful for future studies of barley development and molecular breeding.展开更多
AIM To observe the changes in erythrocyte membrane ATPases and plasma lipid peroxides (LPO) patients with in abdominal surgery under intravenous procaine-balanced anesthesia.METHODS By determining the ATPase activitie...AIM To observe the changes in erythrocyte membrane ATPases and plasma lipid peroxides (LPO) patients with in abdominal surgery under intravenous procaine-balanced anesthesia.METHODS By determining the ATPase activities of erythrocyte membrane, effects of upper abdominal surgery under intravenous procaine-balanced anesthesia on the function of erythrocytes were observed in 15 patients undergoing cholecystectomy and gastrectomy (5 males and 10 females, aged 45.9±10.20 years and weighed 60.60kg±11.93kg). All patients were free from severe renal, hepatic, pulmonary, cardiac, metabolic and endocrinological diseases and acute infection for at least 2 weeks before surgery. Patients receiving any drug known to affect carbohydrate metabolism prior to anesthesia were excluded from the study.RESULTS Erythrocyte membrane Na+, K+-ATPase, Mg2+-ATPase, Ca2+, Mg2+-ATPase activities were not significantly changed 60min-90min after incision as compared with 30min before anesthesia, but were decreased markedly 10min and 24 hours after completion of operation (P<0.01). Plasma lipid peroxides (LPO) were increased significantly 24 hours after surgery (P<0.01) following an initially marked but transient reduction. Plasma LPO changes were not correlated with erythrocyte membrane ATPase activities, r=-0.0396, -0.0097 and 0.4383, respectively (P>0.05).CONCLUSION Abdominal surgical trauma under intravenous procaine-balanced anesthesia may be associated with the decreased ATPase activities of erythrocyte membrane and increased LPO in plasma.展开更多

BACKGROUND Gastric cancer is one of the most common malignant tumors in the world,and its occurrence and development involve complex biological processes.Iron death,as a new cell death mode,has attracted wide attentio...BACKGROUND Gastric cancer is one of the most common malignant tumors in the world,and its occurrence and development involve complex biological processes.Iron death,as a new cell death mode,has attracted wide attention in recent years.However,the regulatory mechanism of iron death in gastric cancer and its effect on lipid peroxidation metabolism remain unclear.AIM To explore the role of iron death in the development of gastric cancer,reveal its relationship with lipid peroxidation,and provide a new theoretical basis for revealing the molecular mechanism of the occurrence and development of gastric cancer.METHODS The process of iron death in gastric cancer cells was simulated by cell culture model,and the occurrence of iron death was detected by fluorescence microscopy and flow cytometry.The changes of gene expression related to iron death and lipid peroxidation metabolism were analyzed by high-throughput sequencing technology.In addition,a mouse model of gastric cancer was established,and the role of iron death in vivo was studied by histology and immunohistochemistry,and the level of lipid peroxidation was detected.These methods comprehensively and deeply reveal the regulatory mechanism of iron death on lipid peroxidation metabolism in the occurrence and development of gastric cancer.RESULTS Iron death was significantly activated in gastric cancer cells,and at the same time,associated lipid peroxidation levels increased significantly.Through high-throughput sequencing analysis,it was found that iron death regulated the expression of several genes related to lipid metabolism.In vivo experiments demonstrated that increased iron death in gastric cancer mice was accompanied by a significant increase in lipid peroxidation.CONCLUSION This study confirmed the important role of iron death in regulating lipid peroxidation metabolism in the occurrence and development of gastric cancer.The activation of iron death significantly increased lipid peroxidation levels,revealing its regulatory mechanism inside the cell.展开更多
Marine microalga Isochrysis galbana is an important feed species with a high nutritional value.Different from other uni-cellular algae,its cell contains two chloroplasts which are the major sites for lipid synthesis.H...Marine microalga Isochrysis galbana is an important feed species with a high nutritional value.Different from other uni-cellular algae,its cell contains two chloroplasts which are the major sites for lipid synthesis.Here,we optimized a chloroplast isola-tion approach suitable for the isolation of I.galbana chloroplasts and determined the purity and integrity of the isolated chloroplasts through microscopic observations and enzyme activity assay.The chloroplast lipids were analyzed with a ultrahigh-performance li-quid chromatography-Q Exactive Orbitrap-mass spectrometry.This newly developed isolation approach is simple and reliable to isolate chloroplasts with high integrity and purity.The average yield of intact chloroplasts was 15.3%±0.1%.Glycolipids and acyl-glycerols were the main chloroplast lipids.Glycolipids accounted for 56.6%of chloroplast lipid.Digalactosyldiacylglycerol(DGDG),monogalactosyldiacylglycerol(MGDG)and sulfoquinovosyldiacylglycerol(SQDG)were the main glyceroglycolipids.The fatty acyl R1/R2 were mostly 18:4/16:1,18:3/16:1 and 18:4/18:5 in DGDGs,14:0/18:4,18:4/18:5,18:4/18:4 and 18:3/18:4 in MGDGs and 16:0/14:0,16:0/18:3,and 18:4/18:3 in SQDGs.In addition,diacylglycerol(DAG)was the most abundant acylglycerols;the content of 22:6/18:4-DAG was the highest.There was a little amount of glycosphingolipid(GSL)in chloroplast.Digalactosylmonoglyceride(DGMG),monogalactosylmonoglyceride(MGMG),sulfoquinovosylmonoacylglycerol(SQMG),monoglyceride(MAG),phospholi-pids(PLs),ceramide(Cer)and betaine lipids were nearly undetectable in chloroplast.The fatty acid proportions of DGDGs,MGDGs,SQDGs,DAGs,triglycerides(TAGs)and GSLs were either higher or lower than or similar to those of whole-cell.Collectively,our isolation approach is applicable to many aspects of chloroplast biology,and may offer a reference for the isolation of chloroplasts from other marine microalgae.展开更多
Nervonic acid plays an important role in nutrition and function of the human body.Malania oleifera,Acer truncatum and Xanthoceras sorbifolium are China’s unique woody plant rich in nervonic acid in seed oil.This stud...Nervonic acid plays an important role in nutrition and function of the human body.Malania oleifera,Acer truncatum and Xanthoceras sorbifolium are China’s unique woody plant rich in nervonic acid in seed oil.This study aims to investigate the lipid composition of these 3 special resources.Their fatty acids were detected by gas chromatography coupled with flame detector(GC-FID).Triglycerides(TAGs)and phospholipids(PLs)were detected by shotgun-mass spectrometry(shotgun-MS).Results showed that M.oleifera oil presented the highest level of nervonic acid(46.20±0.22%)among the 3 oils.Seeds oil of A.truncatum and X.sorbifolium had 3.53±0.20%and 1.83±0.21%nervonic acid respectively.53 species of TAGs and 15 species of PLs were identified in M.oleifera oil,with PLs content of 499.94±22.34μg/g.In A.truncatum oil,PL and TAG species were twice more than those in M.oleifera oil,and its’content of PLs was 76.27±3.21μg/g.In X.sorbifolium oil,75 TAGs and 34 PLs were detected,with the lowest PLs at 23.84±0.17μg/g.The results demonstrated that these 3 vegetable oils have great potential to become nervonic acid supplements for human health.展开更多

Despite extensive research, a cure for Alzheimer's disease has not yet been found. Oxidative stress mediates excessive oxidative responses, and its involvement in Alzheimer's disease pathogenesis as a primary or secondary pathological event is widely accepted. As a member of the selenium-containing antioxidant enzyme family, glutathione peroxidase 4 reduces esterified phospholipid hydroperoxides to maintain cellular redox homeostasis. With the discovery of ferroptosis, the central role of glutathione peroxidase 4 in anti-lipid peroxidation in several diseases, including Alzheimer's disease, has received widespread attention. Increasing evidence suggests that glutathione peroxidase 4 expression is inhibited in the Alzheimer's disease brain, resulting in oxidative stress, inflammation, ferroptosis, and apoptosis, which are closely associated with pathological damage in Alzheimer's disease. Several therapeutic approaches, such as small molecule drugs, natural plant products, and non-pharmacological treatments, ameliorate pathological damage and cognitive function in Alzheimer's disease by promoting glutathione peroxidase 4 expression and enhancing glutathione peroxidase 4 activity. Therefore, glutathione peroxidase 4 upregulation may be a promising strategy for the treatment of Alzheimer's disease. This review provides an overview of the gene structure, biological functions, and regulatory mechanisms of glutathione peroxidase 4, a discussion on the important role of glutathione peroxidase 4 in pathological events closely related to Alzheimer's disease, and a summary of the advances in small-molecule drugs, natural plant products, and non-pharmacological therapies targeting glutathione peroxidase 4 for the treatment of Alzheimer's disease. Most prior studies on this subject used animal models, and relevant clinical studies are lacking. Future clinical trials are required to validate the therapeutic effects of strategies targeting glutathione peroxidase 4 in the treatment of Alzheimer's disease.展开更多

It was found that selenium content in the rat tissues decreased remarkably af-ter injury, which in turn resulted in serious reduction of glutathione peroxidasc activity and significantincrease of lipid peroxides in the scrum, crythrocytcs and liver. However the muscular tissue showedno significant changes. These facts imply that after burn injury, the body is in a state of selenium deficiency, the lossof selenium might be responsible for the reduction of anti - peroxidation ability of glutathioneperoxidase, and conscqucntly there is an increase of lipid peroxides in the tissues. Only the musculartissue is insensitive to lipid peroxidation. It is believed that the reduction of anti-peroxidation abilityof glutathione peroxidasc after bum injury might be one of the main causes to intensify, the injury re-suiting from free radicals.展开更多
The level of lipid peroxides (LPO) and copper-zinc containing superoxide dismutase (SOD-Ⅰ) in the gastric mucosa and serum was determined in 141 samples from 25 patients after con-ventional subtotal gastrectomy (11 c...The level of lipid peroxides (LPO) and copper-zinc containing superoxide dismutase (SOD-Ⅰ) in the gastric mucosa and serum was determined in 141 samples from 25 patients after con-ventional subtotal gastrectomy (11 cases of Billroth Ⅰ and 14 cases of Billroth Ⅱ) and 11 patientsafter pylorus and antroseromuscular flap preserving gastrectomy (PAFPG).Those of 11 normal subjects were examined likewise to serve as control.It was found that:(1) The average LPO levelwas much higher and the average SOD-Ⅰ level much lower in the gastric mucosa of the patientsafter conventional subtotal gastrectomy especially the Billroth Ⅱ type than in that of those afterPAFPG.(2) In 36 specimens of stump mucosa,the average LPO level was significantly higher inthe tissue around the anastomotic ring than in that of the body of the stump;no marked differ-ence of SOD-Ⅰ level between the 2 was revealed.(3) The LPO value in the stump mucosa waspositively correlated to and the SOD-Ⅰ value negatively correlated to the pH value of gastric juice.These rindings suggest that the reation of oxygen free radicals in the stump mucosa may be influ-enced by the intragastric pH or by the type of digestive continuity reconstruction,and that thereaction of oxygen free radicals especially the lipid peroxidation may play a role in the pathogenesisof the lesions in the anastomotic stoma.展开更多

AIM:To clarify this controversy and to provide evidence for application of lipid lowering agents in treatment of diabetic retinopathy(DR).METHODS:We searched the databases of Pub Med,Embase and Cochrane Library Ce...AIM:To clarify this controversy and to provide evidence for application of lipid lowering agents in treatment of diabetic retinopathy(DR).METHODS:We searched the databases of Pub Med,Embase and Cochrane Library Central Register of Controlled Trials(CENTRAL)and abstracts from main annual meetings up to January 1,2017.Google scholar and Clinical Trials.gov were also searched for unpublished relevant studies.We included randomized controlled trials(RCTs)that studied lipid-lowering agents in type 1 or type 2 diabetes in this Meta-analysis.The primary endpoint was the progression of DR,and the secondary endpoints included vision loss,development of diabetic macular edema(DME)and aggravation of hard exudates.The pooled odds ratios(OR)with corresponding 95%confidence intervals(95%CIs)were calculated.RESULTS:After systemic and manual literature search by two independent investigators,we included 8 RCTs from 7 published articles with 13 454 participants in this Meta-analysis.The results revealed that lipid-lowering drugs were associated with reduced risk in DR progression[OR=0.77(95%CI:0.62,0.96),P=0.02].Lipid-lowering agents might have protective effect on DME compared to placebo,although the difference was not statistically significant[OR=0.60(95%CI:0.34,1.08),P=0.09].However,no significant differences in the worsening of vision acuity[OR=0.96(95%CI:0.81,1.14),P=0.64]and hard exudates[OR=0.50(95%CI:0.15,1.74),P=0.28]were found between the lipidlowering drugs and the placebo groups.CONCLUSION:In DR patients,lipid-lowering agents show a protective effect on DR progression and might be associated with reduced risk in the development of DME.However,lipid-lowering agents have no effects on vision loss and hard exudates aggravation.Further clinical trials in larger scale are required to confirm the conclusion of this study and thus justify the use of intensive control lipids with anti-lipid agents at the early stages of DR.展开更多

HLMs (1 mg/ml) were exposed to each of the above flavonoids at 0, 5, 10, 15, 20 or 25 μM and incubated for the respective times as previously stated. Our hypothesis was that HLMs exposed to the flavonoids for the respective exposure times can decrease LP and increase GSH in HLMs to better cope with the oxidative stress. </span><span style="font-family:Verdana;">The results of our studies indicate that each of the flavonoids significantly (p < 0.01) decreased LP compared to their respective controls. The highest decrease in LP was observed for K followed by Q and G. Significant increases (p < 0.01) in GSH were observed for the flavonoid doses tested with the highest</span><span style="font-family:Verdana;"> levels observed for Q for the 24-hr. incubation. Blood lipid indicators: total cholesterol, triglycerides, high density lipoprotein cholesterol, low density lipoprotein cholesterol, apolipoprotein A, apolipoprotein B;coagulation indicators: activated partial thromboplastin time, plasma prothrombin time, fibrinogen, thrombin time indicators, using analysis of variance method for different types of osteonecrosis of the femoral head and blood lipids, coagulation indicators Search for relevance. Results: 1. Comparison of three groups of blood lipid indicators: total cholesterol and low density lipoprotein cholesterol in the alcohol group were higher than those in the trauma group (P<0.05);total cholesterol, triglycerides, and low density lipoprotein in the alcohol group The three indexes of cholesterol were higher than those of the hormone group (P<0.05). There was no significant difference in the serum lipid indexes of the hormone group and the trauma group (P>0.05). 2. Comparison of three groups of coagulation indicators: hormone The fibrinogen index of the group was higher than that of the trauma group (P<0.05). There was no significant difference in the coagulation index between the alcohol group and the trauma group (P>0.05). The coagulation index of the hormone group and the alcohol group was compared. The differences were not statistically significant (P>0.05). Conclusions: 1. Non-traumatic osteonecrosis of the femoral head (alcoholic, hormonal) patients are characterized by markedly elevated blood lipids (total cholesterol, low density lipoprotein cholesterol) and coagulation (fibrinogen) indicators;2. Methods: A total of 129 sexually mature female SD rats were divided into three gro...Objective: To analyze the characteristics and possible mechanism of lipid metabolism in pregnant rats with intestinal flora imbalance. Methods: A total of 129 sexually mature female SD rats were divided into three groups: non-pregnant group (untreated healthy rats), healthy pregnant group (natural insemination pregnant rats), and pregnant microflora disorder group (pregnant rats were given mixed antibiotics by gavage to build the modeling), with 43 rats in each group. The contents of TG, LDL, HDL and TC were detected by automatic biochemical analyzer, and the contents of SCD1, PGC-1 alpha, PEPCK, ApoE and MTTP genes were detected by fluorescence quantitative PCR technology. Regression analysis was used to explore the comprehensive influence of each gene on total cholesterol expression in rats. Principal component analysis was used to explore the internal mechanism of lipid metabolism in pregnant rats with intestinal flora disorder. Results: The contents of TG, TC, LDL and HDL were compared among the three groups of rats and the differences were statistically significant (P<0.05) . The expression levels of related genes (SCD1, PGC-1, PEPCK, ApoE, MTTP) in the three groups were statistically significant (P<0.05) . SCD1 content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (0.92±0.12) μg/mL, (1.20±0.15)μg/mL, and (1.53±0.20) μg/mL, respectively. PGC-1 alpha content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (1.34±0.21) μg/mL, (0.93±0.12) micron /mL, and (0.41±0.08) μg/mL, respectively. PEPCK content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (0.48±0.06) μg/mL, (0.35±0.09)μg/mL, and (0.22±0.05) μg/mL, and the differences were statistically significant (P<0.05) . Multivariate linear regression analysis showed that the influence of gene content on The effect of each gene content on TC content was in order from large to small: SCD1 (OR=4.572) , PGC-1 (OR=3.387) , PEPCK (OR=3.935) , ApoE (OR=3.597) , MTTP (OR=3.096) . The principal component analysis showed that three principal components could be extracted from five related genes of lipid metabolism in pregnant rats with intestinal dysbiosis: SCD1/PEPCK pathway (contribution rate: 36.28%) , PGC-1 /ApoE pathway (contribution rate: 30.42%) , and MTTP pathway (contribution rate: 15.37%) . Conclusion: After pregnancy, blood lipids in rats are significantly increased while the imbalance of intestinal flora will lead to decreased blood lipids. The disorder of lipid metabolism in pregnant rats with intestinal flora imbalance is mainly related to the disorder of gene expression, which further affects the functions of SCD1/PEPCK, PGC-1 /ApoE and MTTP pathways.展开更多
With indica ( Oryza sativa L.) hybrid Shanyou 63 as control, the hybrid rice varieties including Peiai 64S/E32, Peiai 64S/9311, X07S/Zihui 100, Guangyou 881 and japonica 9516 were used to study changes of chlorophyll ...With indica ( Oryza sativa L.) hybrid Shanyou 63 as control, the hybrid rice varieties including Peiai 64S/E32, Peiai 64S/9311, X07S/Zihui 100, Guangyou 881 and japonica 9516 were used to study changes of chlorophyll content, photosynthetic response to light intensity and temperature, chlorophyll fluorescence characteristics and membrane lipid peroxidation in their flag leaves at the late stage of development under natural conditions in Nanjing. The results were as follows:. primary photochemical efficiency of PS II ( F-v / F-m), quantum yield of linear electron transport of PS II (phi(PSII)), electron transfer rate (ETR) in these rice varieties decreased with their decrease of chlorophyll content during this period. This kind of impediment to energy conversion induced the transfer of excessive energy to the reducing side of PS I, hence the accumulation of O-2(radical anion) and peroxidation of membrane lipid, and resulting in the accumulation of malondialdehyde (MDA), that is the destroys of photosynthetic pigments and membranes and the consequent, premature senescence. This phenomenon is variable conspicuously in different rice varieties. Under natural condition in Nanjing, F-v/F-m, phi(PSII), ETR and quenching coefficient ( qP) in japonica 9516 tolerant to photooxidation decreased less and the conversion capacity of light energy was stable, premature senescence was unlikely, and consequently the seed-setting rate was higher. While F-v/F-m, phi(PSII), ETR and photochemical qP in Shanyou 63 sensitive to photooxidation decreased more and therefore premature senescence was easy to happen, thus the seed-setting rate and yield were all reduced. The tolerance to photooxidation and premature senescence in other hybrids derived from typical two line or three line crossing laid in the middle. From the rice breeding for super-high-yield, on the basis of the good plant-type of current rice, considering both hybrid vigor and the prevention premature senescence, it would be a notable strategy to use japonica maternal line or maternal. lines with some japonica genotype as the sterile lines in rice breeding.展开更多

[ Method] Responses of membrane lipid peroxidation and endogenous hormones of soybean seedlings to UV-B radiation and rare earth were studied through hydroponics in laboratory. [ Result] The results showed that under irradiation of UV-B( T1-0.15 W/m^2 and T2-0.45 W/m^2), chlorophyll and indole-3-acetic acid(IAA) contents firstly decreased during the stress phase (1 -5 d) and then increased during the restoration phase (6 -9 d) while contents of malonadialdehyde(MDA) and abscisic acid(ABA) gradually increased during the imposition of UV-B radiation (1 -5 d) and subsequently decreased during recovery from UV-B stress (6 -9 d) . With adding of La (III) with the concentration of 20 mg · L^-1 , the decline/dse trend of chlorophyll, IAA, MDA and ABA contents was slowed down during the stress period while the rise/decline speed was accelerated during the recovery period. [ Conclusion] It suggests that the regulation of La ( III ) on membrane lipid peroxidation and endogenous hormones could increase chlorophyll and IAA contents, improve the metabolism of reactive oxygen species ( ROS), inhibit membrane lipid peroxidation, decrease the accumulation amount of ABA and alleviate injury of UV-B radiation to soybean seedlings. 文摘Mouse peritoneal macrophages were incubated in DMEM with pox-LDL and Rradlx Salviae Miltiorrhizae (RSM) to investigate the effects of RSM on the internalization of peroxidized low density lipoprotein (pox-LDL) by using lipid analysis and electron microscopy. Lipid peroxide (LPO) concentrations were increased slightly in the medium after incubation of macrophages with normal LDL (n-LDL), while decreased significantly in the media after incubation of macrophages with pox-LDL. In the three groups with pox-LDL, it could be found that there was a dose-dependent decrease of concentrations of LPO and total cholesterol (TCH) in the two RSM groups, and the decrease in the two RSM groups was much greater than in the group without RSM. RSM accelerated a more decrease of LPO than cholesterol contents in the media containing pox-LDL. The ultrastructural studies also showed that RSM induced the accumulation of lipid droplets in the cytoplasm of mouse peritoneal macrophages. The results suggested that RSM could accelerate the phagocytosis and degradation of pox-LDL by macrophages.
文摘As a highly aggressive tumor,the pathophysiological mechanism of primary liver cancer has attracted much attention.In recent years,factors such as ferroptosis regulation,lipid peroxidation and metabolic abnormalities have emerged in the study of liver cancer,providing a new perspective for understanding the development of liver cancer.Ferroptosis regulation,lipid peroxidation and metabolic abnormalities play important roles in the occurrence and development of liver cancer.The regulation of ferroptosis is involved in apoptosis and necrosis,affecting cell survival and death.Lipid peroxidation promotes oxidative damage and promotes the invasion of liver cancer cells.Metabolic abnormalities,especially the disorders of glucose and lipid metabolism,directly affect the proliferation and growth of liver cancer cells.Studies of ferroptosis regulation and lipid peroxidation may help to discover new therapeutic targets and improve therapeutic outcomes.The understanding of metabolic abnormalities can provide new ideas for the prevention of liver cancer,and reduce the risk of disease by adjusting the metabolic process.This review focuses on the key roles of ferroptosis regulation,lipid peroxidation and metabolic abnormalities in this process.

文摘AIM To identify the effects and mechanism of action of Polygonatum kingianum(P. kingianum) on dyslipidemia in rats using an integrated untargeted metabolomic method.METHODS A rat model of dyslipidemia was induced with a high-fat diet(HFD) and rats were given P. kingianum [4 g/(kg·d)] intragastrically for 14 wk. Changes in serum and hepatic lipid parameters were evaluated. Metabolites in serum, urine and liver samples were profiled using ultra-highperformance liquid chromatography/mass spectrometry followed by multivariate statistical analysis to identify potential biomarkers and metabolic pathways.RESULTS P. kingianum significantly inhibited the HFD-induced increase in total cholesterol and triglyceride in the liver and serum. P. kingianum also significantly regulated metabolites in the analyzed samples toward normal status. Nineteen, twenty-four and thirty-eight potential biomarkers were identified in serum, urine and liver samples, respectively. These biomarkers involved biosynthesis of phenylalanine, tyrosine, tryptophan, valine, leucine and isoleucine, along with metabolism of tryptophan, tyrosine, phenylalanine, starch, sucrose, glycerophospholipid, arachidonic acid, linoleic acid, nicotinate, nicotinamide and sphingolipid.CONCLUSION P. kingianum alleviates HFD-induced dyslipidemia by regulating many endogenous metabolites in serum, urine and liver samples. Collectively, our findings suggest that P. kingianum may be a promising lipid regulator to treat dyslipidemia and associated diseases.

文摘Non-specific lipid transfer proteins(nsLTPs) are small, basic proteins that are characterized by an eight-cysteine motif. The biological functions of these proteins have been reported to involve plant reproduction and biotic or abiotic stress response. With the completion of the barley genome sequence, a genome-wide analysis of nsLTPs in barley(Hordeum vulgare L.)(HvLTPs) will be helpful for understanding the function of nsLTPs in plants. We performed a genome-wide analysis of the nsLTP gene family in barley and identified 70 nsLTP genes,which can be divided into five types(1, 2, C, D, and G). Each type of nsLTPs shares similar exon and intron gene structures. Expression analysis showed that barley nsLTPs have diverse expression patterns, revealing their various roles. Our results shed light on the phylogenetic relationships and potential functions of barley nsLTPs and will be useful for future studies of barley development and molecular breeding.
文摘AIM To observe the changes in erythrocyte membrane ATPases and plasma lipid peroxides (LPO) patients with in abdominal surgery under intravenous procaine-balanced anesthesia.METHODS By determining the ATPase activities of erythrocyte membrane, effects of upper abdominal surgery under intravenous procaine-balanced anesthesia on the function of erythrocytes were observed in 15 patients undergoing cholecystectomy and gastrectomy (5 males and 10 females, aged 45.9±10.20 years and weighed 60.60kg±11.93kg). All patients were free from severe renal, hepatic, pulmonary, cardiac, metabolic and endocrinological diseases and acute infection for at least 2 weeks before surgery. Patients receiving any drug known to affect carbohydrate metabolism prior to anesthesia were excluded from the study.RESULTS Erythrocyte membrane Na+, K+-ATPase, Mg2+-ATPase, Ca2+, Mg2+-ATPase activities were not significantly changed 60min-90min after incision as compared with 30min before anesthesia, but were decreased markedly 10min and 24 hours after completion of operation (P<0.01). Plasma lipid peroxides (LPO) were increased significantly 24 hours after surgery (P<0.01) following an initially marked but transient reduction. Plasma LPO changes were not correlated with erythrocyte membrane ATPase activities, r=-0.0396, -0.0097 and 0.4383, respectively (P>0.05).CONCLUSION Abdominal surgical trauma under intravenous procaine-balanced anesthesia may be associated with the decreased ATPase activities of erythrocyte membrane and increased LPO in plasma.

文摘Ferroptosis is a novel form of cell death driven by iron-dependent lipid peroxidation and it is implicated in various diseases,such as liver disease,acute kidney injury,cardiovascular disease,neurodegenerative disease and cancer.Lipid-based reactive oxygen species(ROS),particularly lipid hydroperoxides in the cellular membrane can lead to membrane disruption and cell death mediated by ferroptosis.There are three necessary stages involving in the process of lipid peroxidation regulation in ferroptosis,including the synthesis of membrane phospholipids,initiation of lipid peroxidation and clearance of lipid peroxides.In this review,we summarized the molecular modulation mechanisms of lipid peroxidation in ferroptosis from the above three stages,as well as various ferroptosis modulators targeting lipid peroxidation,including commonly used products,natural bioactive compounds and selenocompounds.Collectively,these findings suggest the vital role of lipid peroxidation in ferroptosis,and targeting lipid peroxidation in ferroptosis is potential to treat ferroptosis-associated diseases.

文摘BACKGROUND Gastric cancer is one of the most common malignant tumors in the world,and its occurrence and development involve complex biological processes.Iron death,as a new cell death mode,has attracted wide attention in recent years.However,the regulatory mechanism of iron death in gastric cancer and its effect on lipid peroxidation metabolism remain unclear.AIM To explore the role of iron death in the development of gastric cancer,reveal its relationship with lipid peroxidation,and provide a new theoretical basis for revealing the molecular mechanism of the occurrence and development of gastric cancer.METHODS The process of iron death in gastric cancer cells was simulated by cell culture model,and the occurrence of iron death was detected by fluorescence microscopy and flow cytometry.The changes of gene expression related to iron death and lipid peroxidation metabolism were analyzed by high-throughput sequencing technology.In addition,a mouse model of gastric cancer was established,and the role of iron death in vivo was studied by histology and immunohistochemistry,and the level of lipid peroxidation was detected.These methods comprehensively and deeply reveal the regulatory mechanism of iron death on lipid peroxidation metabolism in the occurrence and development of gastric cancer.RESULTS Iron death was significantly activated in gastric cancer cells,and at the same time,associated lipid peroxidation levels increased significantly.Through high-throughput sequencing analysis,it was found that iron death regulated the expression of several genes related to lipid metabolism.In vivo experiments demonstrated that increased iron death in gastric cancer mice was accompanied by a significant increase in lipid peroxidation.CONCLUSION This study confirmed the important role of iron death in regulating lipid peroxidation metabolism in the occurrence and development of gastric cancer.The activation of iron death significantly increased lipid peroxidation levels,revealing its regulatory mechanism inside the cell.

文摘Marine microalga Isochrysis galbana is an important feed species with a high nutritional value.Different from other uni-cellular algae,its cell contains two chloroplasts which are the major sites for lipid synthesis.Here,we optimized a chloroplast isola-tion approach suitable for the isolation of I.galbana chloroplasts and determined the purity and integrity of the isolated chloroplasts through microscopic observations and enzyme activity assay.The chloroplast lipids were analyzed with a ultrahigh-performance li-quid chromatography-Q Exactive Orbitrap-mass spectrometry.This newly developed isolation approach is simple and reliable to isolate chloroplasts with high integrity and purity.The average yield of intact chloroplasts was 15.3%±0.1%.Glycolipids and acyl-glycerols were the main chloroplast lipids.Glycolipids accounted for 56.6%of chloroplast lipid.Digalactosyldiacylglycerol(DGDG),monogalactosyldiacylglycerol(MGDG)and sulfoquinovosyldiacylglycerol(SQDG)were the main glyceroglycolipids.The fatty acyl R1/R2 were mostly 18:4/16:1,18:3/16:1 and 18:4/18:5 in DGDGs,14:0/18:4,18:4/18:5,18:4/18:4 and 18:3/18:4 in MGDGs and 16:0/14:0,16:0/18:3,and 18:4/18:3 in SQDGs.In addition,diacylglycerol(DAG)was the most abundant acylglycerols;the content of 22:6/18:4-DAG was the highest.There was a little amount of glycosphingolipid(GSL)in chloroplast.Digalactosylmonoglyceride(DGMG),monogalactosylmonoglyceride(MGMG),sulfoquinovosylmonoacylglycerol(SQMG),monoglyceride(MAG),phospholi-pids(PLs),ceramide(Cer)and betaine lipids were nearly undetectable in chloroplast.The fatty acid proportions of DGDGs,MGDGs,SQDGs,DAGs,triglycerides(TAGs)and GSLs were either higher or lower than or similar to those of whole-cell.Collectively,our isolation approach is applicable to many aspects of chloroplast biology,and may offer a reference for the isolation of chloroplasts from other marine microalgae.

文摘Nervonic acid plays an important role in nutrition and function of the human body.Malania oleifera,Acer truncatum and Xanthoceras sorbifolium are China’s unique woody plant rich in nervonic acid in seed oil.This study aims to investigate the lipid composition of these 3 special resources.Their fatty acids were detected by gas chromatography coupled with flame detector(GC-FID).Triglycerides(TAGs)and phospholipids(PLs)were detected by shotgun-mass spectrometry(shotgun-MS).Results showed that M.oleifera oil presented the highest level of nervonic acid(46.20±0.22%)among the 3 oils.Seeds oil of A.truncatum and X.sorbifolium had 3.53±0.20%and 1.83±0.21%nervonic acid respectively.53 species of TAGs and 15 species of PLs were identified in M.oleifera oil,with PLs content of 499.94±22.34μg/g.In A.truncatum oil,PL and TAG species were twice more than those in M.oleifera oil,and its’content of PLs was 76.27±3.21μg/g.In X.sorbifolium oil,75 TAGs and 34 PLs were detected,with the lowest PLs at 23.84±0.17μg/g.The results demonstrated that these 3 vegetable oils have great potential to become nervonic acid supplements for human health.

文摘Alzheimer's disease is an age-related neurodegenerative disorder with a complex and incompletely understood pathogenesis. Despite extensive research, a cure for Alzheimer's disease has not yet been found. Oxidative stress mediates excessive oxidative responses, and its involvement in Alzheimer's disease pathogenesis as a primary or secondary pathological event is widely accepted. As a member of the selenium-containing antioxidant enzyme family, glutathione peroxidase 4 reduces esterified phospholipid hydroperoxides to maintain cellular redox homeostasis. With the discovery of ferroptosis, the central role of glutathione peroxidase 4 in anti-lipid peroxidation in several diseases, including Alzheimer's disease, has received widespread attention. Increasing evidence suggests that glutathione peroxidase 4 expression is inhibited in the Alzheimer's disease brain, resulting in oxidative stress, inflammation, ferroptosis, and apoptosis, which are closely associated with pathological damage in Alzheimer's disease. Several therapeutic approaches, such as small molecule drugs, natural plant products, and non-pharmacological treatments, ameliorate pathological damage and cognitive function in Alzheimer's disease by promoting glutathione peroxidase 4 expression and enhancing glutathione peroxidase 4 activity. Therefore, glutathione peroxidase 4 upregulation may be a promising strategy for the treatment of Alzheimer's disease. This review provides an overview of the gene structure, biological functions, and regulatory mechanisms of glutathione peroxidase 4, a discussion on the important role of glutathione peroxidase 4 in pathological events closely related to Alzheimer's disease, and a summary of the advances in small-molecule drugs, natural plant products, and non-pharmacological therapies targeting glutathione peroxidase 4 for the treatment of Alzheimer's disease. Most prior studies on this subject used animal models, and relevant clinical studies are lacking. Future clinical trials are required to validate the therapeutic effects of strategies targeting glutathione peroxidase 4 in the treatment of Alzheimer's disease.
文摘The changes of sclenium metabolism, glutathione peroxidase activity and lipid peroxidescontent in the tissues of rats suffering from 30% TBSA full thickness scalding were observed in thefirst 7 days after injury. It was found that selenium content in the rat tissues decreased remarkably af-ter injury, which in turn resulted in serious reduction of glutathione peroxidasc activity and significantincrease of lipid peroxides in the scrum, crythrocytcs and liver. However the muscular tissue showedno significant changes. These facts imply that after burn injury, the body is in a state of selenium deficiency, the lossof selenium might be responsible for the reduction of anti - peroxidation ability of glutathioneperoxidase, and conscqucntly there is an increase of lipid peroxides in the tissues. Only the musculartissue is insensitive to lipid peroxidation. It is believed that the reduction of anti-peroxidation abilityof glutathione peroxidasc after bum injury might be one of the main causes to intensify, the injury re-suiting from free radicals.
文摘The level of lipid peroxides (LPO) and copper-zinc containing superoxide dismutase (SOD-Ⅰ) in the gastric mucosa and serum was determined in 141 samples from 25 patients after con-ventional subtotal gastrectomy (11 cases of Billroth Ⅰ and 14 cases of Billroth Ⅱ) and 11 patientsafter pylorus and antroseromuscular flap preserving gastrectomy (PAFPG).Those of 11 normal subjects were examined likewise to serve as control.It was found that:(1) The average LPO levelwas much higher and the average SOD-Ⅰ level much lower in the gastric mucosa of the patientsafter conventional subtotal gastrectomy especially the Billroth Ⅱ type than in that of those afterPAFPG.(2) In 36 specimens of stump mucosa,the average LPO level was significantly higher inthe tissue around the anastomotic ring than in that of the body of the stump;no marked differ-ence of SOD-Ⅰ level between the 2 was revealed.(3) The LPO value in the stump mucosa waspositively correlated to and the SOD-Ⅰ value negatively correlated to the pH value of gastric juice.These rindings suggest that the reation of oxygen free radicals in the stump mucosa may be influ-enced by the intragastric pH or by the type of digestive continuity reconstruction,and that thereaction of oxygen free radicals especially the lipid peroxidation may play a role in the pathogenesisof the lesions in the anastomotic stoma.

文摘AIM:To clarify this controversy and to provide evidence for application of lipid lowering agents in treatment of diabetic retinopathy(DR).METHODS:We searched the databases of Pub Med,Embase and Cochrane Library Central Register of Controlled Trials(CENTRAL)and abstracts from main annual meetings up to January 1,2017.Google scholar and Clinical Trials.gov were also searched for unpublished relevant studies.We included randomized controlled trials(RCTs)that studied lipid-lowering agents in type 1 or type 2 diabetes in this Meta-analysis.The primary endpoint was the progression of DR,and the secondary endpoints included vision loss,development of diabetic macular edema(DME)and aggravation of hard exudates.The pooled odds ratios(OR)with corresponding 95%confidence intervals(95%CIs)were calculated.RESULTS:After systemic and manual literature search by two independent investigators,we included 8 RCTs from 7 published articles with 13 454 participants in this Meta-analysis.The results revealed that lipid-lowering drugs were associated with reduced risk in DR progression[OR=0.77(95%CI:0.62,0.96),P=0.02].Lipid-lowering agents might have protective effect on DME compared to placebo,although the difference was not statistically significant[OR=0.60(95%CI:0.34,1.08),P=0.09].However,no significant differences in the worsening of vision acuity[OR=0.96(95%CI:0.81,1.14),P=0.64]and hard exudates[OR=0.50(95%CI:0.15,1.74),P=0.28]were found between the lipidlowering drugs and the placebo groups.CONCLUSION:In DR patients,lipid-lowering agents show a protective effect on DR progression and might be associated with reduced risk in the development of DME.However,lipid-lowering agents have no effects on vision loss and hard exudates aggravation.Further clinical trials in larger scale are required to confirm the conclusion of this study and thus justify the use of intensive control lipids with anti-lipid agents at the early stages of DR.
文摘<span style="font-family:Verdana;">The effects of each of the flavonoids;genistein (G), quercetin (Q) and</span><span style="font-family:""><span style="font-family:Verdana;"> kaempferol (K) at several doses on lipid peroxides (LP) and reduced glutathione (GSH) in pooled human liver microsomes (HLMs) were investigated following the oxidative damage for 4, 6, 18 and 24 hr. HLMs (1 mg/ml) were exposed to each of the above flavonoids at 0, 5, 10, 15, 20 or 25 μM and incubated for the respective times as previously stated. Our hypothesis was that HLMs exposed to the flavonoids for the respective exposure times can decrease LP and increase GSH in HLMs to better cope with the oxidative stress. </span><span style="font-family:Verdana;">The results of our studies indicate that each of the flavonoids significantly (p < 0.01) decreased LP compared to their respective controls. The highest decrease in LP was observed for K followed by Q and G. Significant increases (p < 0.01) in GSH were observed for the flavonoid doses tested with the highest</span><span style="font-family:Verdana;"> levels observed for Q for the 24-hr. incubation. The findings suggest that the flavonoids modulate oxidative stress in HLMs by decreasing LP and such decreases in LPs may be due to the increasing and or the replenished levels of GSH in the said cells to better cope with the oxidative stress.</span></span>

文摘Objective: To explore the correlation between different types of osteonecrosis of the femoral head and blood lipids and coagulation indexes, in order to provide relevant references for the pathogenesis of osteonecrosis of the femoral head. Methods: 308 cases of ONFH were divided into alcohol group (165 cases), hormone group (113 cases), trauma group (30 cases), and basic data of patients were collected. Blood lipid indicators: total cholesterol, triglycerides, high density lipoprotein cholesterol, low density lipoprotein cholesterol, apolipoprotein A, apolipoprotein B;coagulation indicators: activated partial thromboplastin time, plasma prothrombin time, fibrinogen, thrombin time indicators, using analysis of variance method for different types of osteonecrosis of the femoral head and blood lipids, coagulation indicators Search for relevance. Results: 1. Comparison of three groups of blood lipid indicators: total cholesterol and low density lipoprotein cholesterol in the alcohol group were higher than those in the trauma group (P<0.05);total cholesterol, triglycerides, and low density lipoprotein in the alcohol group The three indexes of cholesterol were higher than those of the hormone group (P<0.05). There was no significant difference in the serum lipid indexes of the hormone group and the trauma group (P>0.05). 2. Comparison of three groups of coagulation indicators: hormone The fibrinogen index of the group was higher than that of the trauma group (P<0.05). There was no significant difference in the coagulation index between the alcohol group and the trauma group (P>0.05). The coagulation index of the hormone group and the alcohol group was compared. The differences were not statistically significant (P>0.05). Conclusions: 1. Non-traumatic osteonecrosis of the femoral head (alcoholic, hormonal) patients are characterized by markedly elevated blood lipids (total cholesterol, low density lipoprotein cholesterol) and coagulation (fibrinogen) indicators;2. Intravascular coagulation and lipid metabolism disorders may be the pathological basis of non-traumatic Osteonecrosis of the femoral head.

文摘Objective: To analyze the characteristics and possible mechanism of lipid metabolism in pregnant rats with intestinal flora imbalance. Methods: A total of 129 sexually mature female SD rats were divided into three groups: non-pregnant group (untreated healthy rats), healthy pregnant group (natural insemination pregnant rats), and pregnant microflora disorder group (pregnant rats were given mixed antibiotics by gavage to build the modeling), with 43 rats in each group. The contents of TG, LDL, HDL and TC were detected by automatic biochemical analyzer, and the contents of SCD1, PGC-1 alpha, PEPCK, ApoE and MTTP genes were detected by fluorescence quantitative PCR technology. Regression analysis was used to explore the comprehensive influence of each gene on total cholesterol expression in rats. Principal component analysis was used to explore the internal mechanism of lipid metabolism in pregnant rats with intestinal flora disorder. Results: The contents of TG, TC, LDL and HDL were compared among the three groups of rats and the differences were statistically significant (P<0.05) . The expression levels of related genes (SCD1, PGC-1, PEPCK, ApoE, MTTP) in the three groups were statistically significant (P<0.05) . SCD1 content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (0.92±0.12) μg/mL, (1.20±0.15)μg/mL, and (1.53±0.20) μg/mL, respectively. PGC-1 alpha content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (1.34±0.21) μg/mL, (0.93±0.12) micron /mL, and (0.41±0.08) μg/mL, respectively. PEPCK content in the non-pregnant group, healthy pregnancy group, and disordered pregnancy group was (0.48±0.06) μg/mL, (0.35±0.09)μg/mL, and (0.22±0.05) μg/mL, and the differences were statistically significant (P<0.05) . Multivariate linear regression analysis showed that the influence of gene content on The effect of each gene content on TC content was in order from large to small: SCD1 (OR=4.572) , PGC-1 (OR=3.387) , PEPCK (OR=3.935) , ApoE (OR=3.597) , MTTP (OR=3.096) . The principal component analysis showed that three principal components could be extracted from five related genes of lipid metabolism in pregnant rats with intestinal dysbiosis: SCD1/PEPCK pathway (contribution rate: 36.28%) , PGC-1 /ApoE pathway (contribution rate: 30.42%) , and MTTP pathway (contribution rate: 15.37%) . Conclusion: After pregnancy, blood lipids in rats are significantly increased while the imbalance of intestinal flora will lead to decreased blood lipids. The disorder of lipid metabolism in pregnant rats with intestinal flora imbalance is mainly related to the disorder of gene expression, which further affects the functions of SCD1/PEPCK, PGC-1 /ApoE and MTTP pathways.
文摘With indica ( Oryza sativa L.) hybrid Shanyou 63 as control, the hybrid rice varieties including Peiai 64S/E32, Peiai 64S/9311, X07S/Zihui 100, Guangyou 881 and japonica 9516 were used to study changes of chlorophyll content, photosynthetic response to light intensity and temperature, chlorophyll fluorescence characteristics and membrane lipid peroxidation in their flag leaves at the late stage of development under natural conditions in Nanjing. The results were as follows:. primary photochemical efficiency of PS II ( F-v / F-m), quantum yield of linear electron transport of PS II (phi(PSII)), electron transfer rate (ETR) in these rice varieties decreased with their decrease of chlorophyll content during this period. This kind of impediment to energy conversion induced the transfer of excessive energy to the reducing side of PS I, hence the accumulation of O-2(radical anion) and peroxidation of membrane lipid, and resulting in the accumulation of malondialdehyde (MDA), that is the destroys of photosynthetic pigments and membranes and the consequent, premature senescence. This phenomenon is variable conspicuously in different rice varieties. Under natural condition in Nanjing, F-v/F-m, phi(PSII), ETR and quenching coefficient ( qP) in japonica 9516 tolerant to photooxidation decreased less and the conversion capacity of light energy was stable, premature senescence was unlikely, and consequently the seed-setting rate was higher. While F-v/F-m, phi(PSII), ETR and photochemical qP in Shanyou 63 sensitive to photooxidation decreased more and therefore premature senescence was easy to happen, thus the seed-setting rate and yield were all reduced. The tolerance to photooxidation and premature senescence in other hybrids derived from typical two line or three line crossing laid in the middle. From the rice breeding for super-high-yield, on the basis of the good plant-type of current rice, considering both hybrid vigor and the prevention premature senescence, it would be a notable strategy to use japonica maternal line or maternal. lines with some japonica genotype as the sterile lines in rice breeding.

文摘[ Objective] The aim was to provide strategies for development of rare earth and control of environmental pollution. [ Method] Responses of membrane lipid peroxidation and endogenous hormones of soybean seedlings to UV-B radiation and rare earth were studied through hydroponics in laboratory. [ Result] The results showed that under irradiation of UV-B( T1-0.15 W/m^2 and T2-0.45 W/m^2), chlorophyll and indole-3-acetic acid(IAA) contents firstly decreased during the stress phase (1 -5 d) and then increased during the restoration phase (6 -9 d) while contents of malonadialdehyde(MDA) and abscisic acid(ABA) gradually increased during the imposition of UV-B radiation (1 -5 d) and subsequently decreased during recovery from UV-B stress (6 -9 d) . With adding of La (III) with the concentration of 20 mg · L^-1 , the decline/dse trend of chlorophyll, IAA, MDA and ABA contents was slowed down during the stress period while the rise/decline speed was accelerated during the recovery period. [ Conclusion] It suggests that the regulation of La ( III ) on membrane lipid peroxidation and endogenous hormones could increase chlorophyll and IAA contents, improve the metabolism of reactive oxygen species ( ROS), inhibit membrane lipid peroxidation, decrease the accumulation amount of ABA and alleviate injury of UV-B radiation to soybean seedlings. Further, the protective potential of La ( III ) was better under low UV-B radiation than under high one.
