目的研究中药新药LW-AFC对代谢综合征(MS)的作用及其作用机制。方法高热量饲料喂养昆明小鼠6周,同时每天ig给予二甲双胍(阳性对照药)0.2g·kg-1和LW-AFC 0.2,0.8和3.2g·kg-1。实验结束时测量小鼠体质量和摄食量,检测空腹血清... 目的研究中药新药LW-AFC对代谢综合征(MS)的作用及其作用机制。方法高热量饲料喂养昆明小鼠6周,同时每天ig给予二甲双胍(阳性对照药)0.2g·kg-1和LW-AFC 0.2,0.8和3.2g·kg-1。实验结束时测量小鼠体质量和摄食量,检测空腹血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、C肽、血糖(FBG)和胰岛素(FINS)含量,并计算稳态模型胰岛素抵抗评价指数(HOMA-IR);测定小鼠内脏脂肪质量(VFM)、计算内脏脂肪系数(VFC),并检测血清瘦素、抵抗素、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和下丘脑神经肽Y(NPY)含量。TC和FBG含量用酶比色法检测,LDL-C和HDL-C含量用清除法检测,FINS和NPY含量用放射免疫分析法检测,C肽含量用均相酶联免疫法检测,瘦素、抵抗素、TNF-α和IL-6含量用液相芯片(luminex)法检测,并采用伊红染色法观察肝脏病理改变。结果与正常对照组比较,模型组小鼠腹型肥胖相关指标VFM和VFC、脂代谢相关指标TC和LDL-C和糖代谢相关指标FBG增高(P<0.01),肝细胞呈弥漫性小泡性脂变,日均摄食量增高(P<0.01),促食欲肽NPY、抑食欲激素瘦素和抵抗素增高(P<0.05)。与模型组比较,LW-AFC 0.2,0.8和3.2 g·kg-1可降低TC(P<0.05)和LDL-C(P<0.01)、升高HDL-C(P<0.05,P<0.01)等脂代谢相关指标水平,降低FBG,HOMA-IR和C肽(P<0.05,P<0.01)等糖代谢相关指标的水平,并可减轻肝脏小泡性脂变等病理损害,提示LW-AFC对模型小鼠糖脂代谢紊乱及肝脏病理损伤具有改善作用。LW-AFC 0.2,0.8和3.2g·kg-1可降低小鼠的日均摄食量和促食欲肽NPY水平(P<0.05),但可显著增高抑食欲激素瘦素的水平(P<0.01),提示LW-AFC对模型小鼠的食欲具有抑制作用。不同剂量的LW-AFC还可降低血清脂肪因子抵抗素和炎症细胞因子TNF-α和IL-6的水平(P<0.05,P<0.01);变量聚类分析结果表明,炎症因子IL-6与脂代谢关系密切,抑食欲激素瘦素与糖代谢关系密切,而抵抗素与炎症及食欲关系密切。结论 LW-AFC可调节血脂、降血糖、改善胰岛素敏感性、减轻肝脏病理损伤,从而改善代谢综合征。通过降低促食欲激素、升高抑食欲激素水平而抑制食欲以及降低炎症细胞因子分泌可能是其改善代谢综合征的部分作用机制。 展开更多

目的评价LW-AFC治疗糖尿病的作用,并探讨其作用特点。方法采用高热量饲料对Wistar大鼠饮食诱导5周,随后一次性ip给予链佐星(STZ)30 mg.kg-1制备糖尿病大鼠模型。7 d后ig给予LW-AFC 0.28,0.56和1.12 g.kg-1,每天1次,连续8周。在给药后2,... 目的评价LW-AFC治疗糖尿病的作用,并探讨其作用特点。方法采用高热量饲料对Wistar大鼠饮食诱导5周,随后一次性ip给予链佐星(STZ)30 mg.kg-1制备糖尿病大鼠模型。7 d后ig给予LW-AFC 0.28,0.56和1.12 g.kg-1,每天1次,连续8周。在给药后2,4,6和8周动态监测大鼠空腹血糖、胰岛素、胰岛素抵抗指数(HOMA-IR)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和甘油三酯(TG)水平,称量内脏脂肪质量(VFM)并计算内脏脂肪系数(VFC)。采用酶比色法检测血糖、TC和TG,采用清除法检测LDL-C,采用放射免疫分析法检测胰岛素。结果与正常对照组比较,模型组大鼠VFM和VFC显著升高(P<0.01),空腹血糖水平显著升高(P<0.01),胰岛素水平在2和6周明显升高(P<0.05),TC水平在4,6和8周明显升高,LDL-C水平在2,6和8周明显升高(P<0.05),TG水平在2周明显升高。与模型组比较,LW-AFC 0.56和1.12 g.kg-1给药8周可显著降低VFM至17.1±3.0和(16.0±3.6)g,可降低VFC至(4.5±0.6)%和(4.3±0.9)%(P<0.01);LW-AFC 1.12 g.kg-1给药4,6和8周可将空腹血糖水平由19.3±3.1,21.4±7.0和(17.0±4.7)mmol.L-1分别降低至14.2±4.0,11.8±4.9和(11.2±4.9)mmol.L-1;LW-AFC对胰岛素水平无明显影响。HOMA-IR在2~8周明显升高(P<0.01),HOMA-IR降低至17.3±4.8,17.4±6.7和4.1±2.4(P<0.05),LW-AFC 1.12 g.kg-1给药4,6和8周可明显降低TC至2.34±0.22,2.09±0.29和(2.16±0.22)mmol.L-1。LW-AFC 1.12 g.kg-1给药2周可将LDL-C降至(0.41±0.11)mmol.L-1;在6和8周降至0.50±0.10和(0.46±0.08)mmol.L-1。LW-AFC 0.56和1.12 g.kg-1给药2周TG降低至1.9±0.8和(1.8±0.8)mmol.L-1。结论 LW-AFC对糖尿病模型大鼠具有改善腹型肥胖、糖脂代谢紊乱和胰岛素抵抗的作用。 展开更多

目的探讨LW-AFC对免疫功能低下小鼠是否有改善作用。方法昆明小鼠ip给予环磷酰胺(CTX)0.02g·kg-1,连续10d,制备免疫功能低下小鼠模型。六味地黄(LW)浓缩丸(1.4g·kg-1)和LW-AFC(0.2,0.4和0.8g·kg-1)组小鼠在每次给予CTX... 目的探讨LW-AFC对免疫功能低下小鼠是否有改善作用。方法昆明小鼠ip给予环磷酰胺(CTX)0.02g·kg-1,连续10d,制备免疫功能低下小鼠模型。六味地黄(LW)浓缩丸(1.4g·kg-1)和LW-AFC(0.2,0.4和0.8g·kg-1)组小鼠在每次给予CTX后ig给予相应药物,连续10d。监测小鼠体重,以及胸腺和脾脏重量变化,并计算胸腺和脾脏系数;[3H]TdR掺入法检测脾淋巴细胞增殖反应,MTT法检测脾脏自然杀伤(NK)细胞杀伤活性,流式细胞术检测脾淋巴细胞中CD4+CD8-和CD4-CD8+T细胞亚群百分率,并计算CD4+/CD8+T细胞亚群比值。结果与正常对照组比较,模型组小鼠胸腺和脾重量及其系数明显降低,体重增长显著缓慢;LW和LW-AFC对小鼠体重增长缓慢、胸腺重量及其系数降低无明显改善作用;LW-AFC0.2g·kg-1对CTX导致的脾重及系数降低有明显改善(P<0.05)。模型组小鼠脾淋巴细胞自发增殖反应、刀豆蛋白A(ConA)和脂多糖(LPS)诱导的脾细胞增殖反应与正常对照组比较均明显降低,[3H]TdR掺入值由正常对照组的6115±441,19432±1778和(23345±7296)cpm分别降低为2741±340,9210±1387和(3983±263)cpm;与模型组比较,LW-AFC0.8g·kg-1对脾淋巴细胞自发增殖反应有明显的改善作用;LW浓缩丸和LW-AFC对ConA诱导的脾细胞增殖反应均具有明显的促进作用,[3H]TdR掺入值分别为13996±5161,27550±2356,15427±1444和(27333±1701)cpm;对LPS诱导的脾细胞增殖降低无改善作用,甚至低于CTX模型组(P<0.01)。模型组小鼠NK细胞杀伤活性由正常对照组的(39.5±0.5)%降低为(37.0±1.0)%,LW和LW-AFC明显促进小鼠NK细胞杀伤活性,杀伤率分别为(40.9±0.6)%,(39.7±0.8)%,(42.4±0.5)%和(39.8±0.9)%。与正常对照组比较,模型组小鼠脾脏CD3+,CD4+CD8-和CD4-CD8+T细胞百分率明显增加,CD4+/CD8+比值无明显变化;LW浓缩丸和LW-AFC可使CTX导致的升高的CD3+,CD4+CD8-和CD4-CD8+T细胞百分率明显降低,并可明显降低CD4+/CD8+比值。结论 LW-AFC对CTX所致小鼠免疫功能低下具有一定的改善作用。 展开更多

OBJECTIVE Tumor necrosis factor-α(TNF-α) plays a vital role in cognitive dysfunction caused by stress.In our previous study, Liuwei Dihuang-active fraction combination(LW-AFC) could attenuate the effects of mental stress and non-psychotic stress in mice. The aim of this study is to investigate the effects of LW-AFC on cognitive dysfunction caused by TNF-α in mice. METHODS 40 male BALB/c mice were divided into 4 groups according to their body weight,including control, TNF-α model, LW-AFC treatment and Etanercept(TNF-α antagonist) treatment groups. LW-AFC(1.6 or 3.2 g·kg-1 per day) were orally administrated for 7 consecutive days before TNF-α administration. Etanercept was injected subcutaneously at 30 mg·kg-1 the day before TNF-α administration. One hour before the behavioral test, TNF-αwere injected intraperitoneally at 0.2 mg·kg-1 to mice. RESULTS Compared with control group, the time of mice stayed in the target quadrant and the number of mice crossing the plate significantly decreased after TNF-α injection, suggested that the spatial learning and memory ability of the mice were impaired. LW-AFC administration could increase the time of mice stayed in the target quadrant and the number of mice crossing the plate significantly at 1.6 g·kg-1, indicated that LW-AFC could improve spatial learning and memory in TNF-α treated mice. CONCLUSION LW-AFC can improve spatial learning and memory impairment induced by TNF-α in mice, the further mechanism still need to be clarified. 展开更多

OBJECTIVE To investigate the effect of LW-AFC,a new formula of the main active components extracted from Liuwei Dihuang decoction,on treatment of Alzheimer′s disease in mouse models.METHODS The APP/PS1 transgenic mice and senescence-accelerated mouse prone 8(SAMP8)were administered orally by LW-AFC continuously for 3 months.The behavioral tests were conducted by the novel object recognition(recognition memory task),the Morris water maze(spatial memory test),the shuttle box and step-down tests(active and passive avoidance,associative learning task).The synaptic plasticity was evaluated by long-term potentiation(LTP)experiment in anesthetized mice.The cytokine profiles in serum were assayed by multiplex Luminex assay methods.The hormone levels associated with hypothalamo-pituitary-adrenal(HPA)and hypothalamo-pituitary-gonad(HPG)were measured by radioimmunoassay.The levels of Aβ1-40 and Aβ1-42 were assayed by AlphaLISA technique.RESULTS The administration of LW-AFC significantly improved the behavioral performances in novel object recognition tasks,spatial memory tasks,and associative learning tasks both in APP/PS1 transgenic mice and SAMP8 mice,and the inductions of LTP were also significantly facilitated.Meanwhile,the concentrations of Aβ1-40 and Aβ1-42 in cerebral cortex,hippocampus and serum were decreased in LW-AFC treated groups.The serum concentration of some proinflammatory cytokines,such as IL-1β,IL-2,IL-6,IL-17,IL-23,INF-γ,TNF-α,TNF-β,MCP-1 and MIP-1β were significantly decreased,while the anti-inflammatory cytokines,such as IL-4,IL-10,IL-5 and G-CSF were significantly increased.In addition,LW-AFC could also improved the abnormalities of the corticotropin releasing hormone(CRH)and gonadotropin-releasing hormone(GnRH)levels in hypothalamus,the adrenocorticotropic hormone(ACTH),luteinizing hormone(LH)and follicle stimulating hormone(FSH)levels in pituitary,the corticosterone and testosterone level in serum,respectively.CONCLUSION LW-AFC improves cognitive deficits and synaptic plasticity impairments in AD mouse models,and this may be due to,at least in part,the modulation of neuroimmunomodulation network by LW-AFC,suggesting that LW-AFC might be a promising therapy for AD. 展开更多

Effects of LW-AFC on anxiety-like behavior and immune function in corticosterone-induced mice

OBJECTIVE Chronic stress is one of the important factors in the development of many mental and neurological diseases,and cause damage to the central nervous system,affect animal emotions and damage the immune function of the body.The purpose of this study was to investigate the effects of LW-AFC which extracting from traditional Chinese medicine prescription Liuwei Dihuang decoctionon the anxiety-like behaviorand immune dysfunction abnormalities caused by chronic stress,and whether immune intervention affect the action of LW-AFC.METHODS Male BALB/c mice were subcutaneously injected with corticosterone(25 mg·kg^-1)for 28 d to establish a chronic stress model.Cyclophos⁃phamide(Cy,80 mg·kg^-1)was injected continuously for the initial three days,followed by once a week,LW-AFC(1.6 g·kg^-1)was given continuously for 28 d.Then investigate the emotion changes by open field and elevated plus maze tests,and detected the lymphocyte proliferation,lymphocyte subsets in peripheral blood,microglia and astrocyte expression,and inflammatory cytokines in peripheral blood and brain tissue.RESULTS The mice showed obvious depressive-like behaviorafter 28 d of continuous corticosterone injection.LW-AFC could significantly improve the anxietybehavior induced by corticosterone injection,but LW-AFC could not improve the anxietybehavior of mice by Cy intervention.The expression of glial cells in hippocampus of corticosterone-induced mice showed an upward trend,and the activation of microglia and astrocytes have significantly increase in corticosterone and Cy injected mice.LW-AFC significantly decreased the activation of microglia and astrocytes in corticosterone-induced mice with Cy intervention.This suggested that LW-AFC can reduce the damage of stress on the immune function of central nervous system under immunosuppres⁃sive state.Furthermore,LW-AFC could significantly up-regulate the proliferation of splenic lymphocyte stimulated by LPS and ConA,up-regulate the proportion of CD3+CD8+cells,reduce the proportion of CD4+/CD8+cells,decrease the secretion of inflammatory factors IL-6 and MCP-1 in plasma,and increase the level of anti-inflammatory factor IL-10 in plasma of mice induced by chronic corticosteroneinjection.While LW-AFC could promote the inflammatory factors TNF-α and IL-6in plasma,inhibit the secretion of anti-inflammatory factor IL-10 and inflammatory cytokine MCP-1 in hippocampus of corticosterone-induced mice with Cy intervention.CONCLUSION LW-AFC can improve anxiety-likebehavior induced by chronic stress,the Cy intervention affects the alleviation of anxiety-like behavior by LW-AFC as well as the regulation of immune function.The regulation of immune function might be the main way for LW-AFC to improve the function of central nervous system. 展开更多

OBJECTIVE To investigate the effect of LW-AFC,a new formula of the main active components extracted fromLiuwei Dihuang decoction,on treatment of Alzheimer disease(AD) in mouse models.METHODS After treatment LW-AFC,mice were cognitively evaluated in behavioral experiments.Neuron loss,amyloid-β(Αβ) deposition,and Αβ level were analyzed using Nissl staining,immunofluorescence,and an Alpha LISA assay,respectively.Multiplex bead analysis,a radioimmunoassay,immunochemiluminometry,and an ELISA were used to measure cytokine and hormone levels.Lymphocyte subsets were detected using flow cytometry.RESULTS LW-AFC ameliorated the cognitive impairment observed in APP/PS1 mice,including the impairment of object recognition memory,spatial learning and memory,and active and passive avoidance.In addition,LW-AFC alleviated the neuron loss in the hippocampus,suppressed Αβ deposition in the brain,and reduced the concentration of Aβ1-42 in the hippocampus and plasma of APP/PS1 mice.LW-AFC treatment also significantly decreased the secretion of corticotropin-releasing hormone and gonadotropin-releasing hormone in the hypothalamus,and adrenocorticotropic hormone,luteinizing hormone,and follicle-stimulating hormone in the pituitary.Moreover,LW-AFC increased CD8+CD28+T cells,and reduced CD4^+CD25^+Foxp3^+T cells in the spleen lymphocytes,down-regulated interleukin(IL)-1β,IL-2,IL-6,IL-23,granulocyte-macrophage colony stimulating factor,and tumor necrosis factor-α and-β,and up-regulated IL-4 and granulocyte colony stimulating factor in the plasma of APP/PS1 mice.CONCLUSION LW-AFC ameliorated the behavioral and pathological deterioration of APP/PS1 transgenic micevia the restoration of the NIM network to a greater extent than either memantineor donepezil,which supports the use of LW-AFC as a potential agent for AD therapy. 展开更多

LW-AFC and its active components ameliorate corticosterone-induced long-term potentiation impairment in mice

OBJECTIVE LW-AFC is extracted from the classical traditional Chinese medicinal prescription-Liuwei Dihuang Decoction.Previous studies have showed that LW-AFC could improve learning&memory ability in amny animal models.In this study,we focused on evaluating the effect of several main active components fromLW-AFC(B-B;loganin,LOG;morroniside,MOR;paeoniflorin,PF and stachyose,STA)on LTP.METHODS In vivo recording of LTP was used in this study to evaluate the effects of LW-AFC and it′s active components on coticorsterone(Cort)induced LTP impairment.RESULTS The results showed that LW-AFC could ameliorate Cort-induced LTP impairment.The effect of LW-AFC was abolished when the immune function was inhibited.Single administration(ig,ip,icv)of any of the components had no effect on Cort-induced LTP impairment.Consecutively intragastric administration or intraperitoneal injections(chronic administration)of B-B,LOG,MOR or PF for 7 d showed protective effect on Cort-induced LTP impairment.Intragastric administration of STA for 7 d protected LTP from impairment induced by Cort,while there was little improving effect when STA was administrated via intraperitoneal injection.In addition,when the intestinal microbiota was disrupted by applying the antibiotic cocktail,STA showed little protective effect against Cort.CONCLUSION In conclusion,LW-AFC and it′s components showed positive effects against cort induced LTP impairment,it seems that all displayed protective effects via indirectly,immune modulation might be the common pathway for all components;the exact pathways are different in each component,B-B,LOG,MOR and PF could be absorbed into the bloods tream and then modulate the peripheral immune function,while STA could not be absorbed and modulates the immune function via modulating intestinal microbiota.Further studies are needed to invesgate the underlying mechanisms and the synergetic effects of all components. 展开更多

OBJECTIVE To investigate the effects of LW-AFC,a new formula derived fromLiuwei Dihuang decoction,on gut microbiota and the behavior of learning and memory of SAMP8 mice,a mouse model of Alzheimer Disease(AD),and identify the specific intestinal microbiota correlating with cognitive ability.METHODS Morris-water maze test,novel object recognition test and shuttle-box test were conducted to observe the ability of learning and memory.16S rRNA amplicon sequencing(Illumina,San Diego,CA,USA)was employed to investigate gut microbiota.RESULTS The treatment of LW-AFC improved cognitive impairments of SAMP8 mice,including spatial learning and memory ability,active avoidance response,and object recognition memory capability.Our data indicated that there were significantly 8 increased and 12 decreased operational taxonomic units(OTUs)in the gut microbiota of SAMP8 mice compared with senescence accelerated mouse resistant 1(SAMR1) strains,the control of SAMP8 mice.The treatment of LW-AFC altered 22(16 increased and 6 decreased)OTUs in SAMP8 mice and among them,15 OTUs could be reversed by LW-AFC treatment resulting in a microbial composition similar to that of SAMR1 mice.We further showed that there were7(3 negative and 4 positive correlation)OTUs significantly correlated with all the three types of cognitive abilities,at the order level,including Bacteroidales,Clostridiales,Desulfovibrionales,CW040,and two unclassified orders.LW-AFC had influences on bacterial taxa correlated with the abilities of learning and memory in SAMP8 mice and restored them to SAMR1 mice.CONCLUSION The effects of LW-AFC on improving cognitive impairments of SAMP8 mice might be via modulating intestinal microbiome and LW-AFC could be used as a potential anti-AD agent. 展开更多

目的肿瘤坏死因子α(TNF-α)是精神性应激和非精神性应激(如感染、疾病)致情绪障碍的主要参与因素。本实验室前期研究表明,六味地黄汤活性成分群复方LW-AFC可以改善急性及慢性应激小鼠情绪障碍,并且降低海马和皮质TNF-α水平,提示调节T... 目的肿瘤坏死因子α(TNF-α)是精神性应激和非精神性应激(如感染、疾病)致情绪障碍的主要参与因素。本实验室前期研究表明,六味地黄汤活性成分群复方LW-AFC可以改善急性及慢性应激小鼠情绪障碍,并且降低海马和皮质TNF-α水平,提示调节TNF-α表达或功能可能是LW-AFC发挥抗应激作用的重要环节。本研究拟以TNF-α致情绪障碍小鼠为模型,观察LW-AFC的作用,为进一步研究TNF-α在LW-AFC抗应激中的作用及机制奠定基础。方法将40只BALB/c小鼠按体质量、自主活动距离随机分成5组:对照组、TNF-α处理组(ip)、LW-AFC给药组、伊那西普(TNF-α拮抗剂)处理组(ip)和伊那西普对照组(ip)(每组8只)。高架十字迷宫(EPM)实验前,LW-AFC 1.6 g·kg^-1连续灌胃给药14 d,伊那西普2.5 mg·kg^-1连续腹腔注射3 d,其他各组给予等量去离子水,EPM实验前1 h,TNF-α处理组、LW-AFC给药组和伊那西普(TNF-α拮抗剂)处理组腹腔注射TNF-α0.2 mg·kg^-1。结果TNF-α处理组小鼠表现出明显焦虑样行为,表现为小鼠在封闭臂时间明显增加(P<0.01),开放臂时间明显减少(P<0.01);伊那西普对照组对EPM实验小鼠无影响。LW-AFC可以改善TNF-α致模型小鼠焦虑样行为,表现为小鼠在开放臂时间延长(P<0.01),在封闭臂时间明显减少(P<0.01);伊那西普预处理与LW-AFC预防给药结果类似,有改善小鼠焦虑样行为的作用。结论六味地黄汤活性成分群复方LW-AFC可以改善TNF-α所致小鼠焦虑样情绪,提示这可能是LW-AFC对应激小鼠具有保护作用的基础之一,为进一步作用机制研究奠定了基础。 展开更多

目的神经炎症参是阿尔茨海默病(AD)的发生发展的重要因素之一,可抑制海马突触可塑性。六味地黄苷糖(LW-AFC)是从经典名方六味地黄汤中提取活性组分所构成的活性成分群,包括糖苷、寡糖和多糖。本研究主要考察LW-AFC对神经炎症所致突触可... 目的神经炎症参是阿尔茨海默病(AD)的发生发展的重要因素之一,可抑制海马突触可塑性。六味地黄苷糖(LW-AFC)是从经典名方六味地黄汤中提取活性组分所构成的活性成分群,包括糖苷、寡糖和多糖。本研究主要考察LW-AFC对神经炎症所致突触可塑性损伤的作用,并对其所含单体免疫活性进行初步评价。方法灌胃给予雄性BALB/c小鼠LW-AFC两周,在体记录海马-齿状回长时程增强变化,检测外周血淋巴细胞亚群、外周血和脑组织中炎性细胞因子,免疫荧光染色观察小胶质细胞激活水平,单次腹腔注射脂多糖(LPS)(250μg·kg-1,ip)建立炎症模型,观察LW-AFC的作用。在体外,对LW-AFC中部分单体对正常BV-2细胞及LPS刺激BV-2所致TNF-α分泌水平进行检测。结果腹腔注射LPS 4 h后小鼠的兴奋性突触后电位降低,即长时程增强减弱,预防给予LW-AFC可显著改善炎症所导致小鼠神经突触可塑性的降低,可显著减轻海马中小胶质细胞及星形胶质细胞的激活水平。LW-AFC可显著上调炎症模型小鼠脾脏淋巴细胞中CD19+细胞的比例,降低CD3+CD4+细胞比例;同时,LW-AFC能明显升高外周血中CD3+CD4+和CD3+CD25+细胞比例。LW-AFC可显著降低海马中IL-1β,IL-6,G-CSF和MCP-1,皮质中MCP-1以及外周血中TNF-α,IL-6,IL-12和INF-γ炎性细胞因子的水平。体外研究表明,LW-AFC中单体芍药苷、马钱苷酸、莫诺苷和水苏糖对正常BV-2细胞TNF-α的分泌有降低作用,而100μmol·L-1的芍药苷、马钱苷酸、莫诺苷以及10μmol·L-1的甘露三糖和CA-4-3对LPS刺激的BV-2细胞TNF-α的分泌有促进作用。结论LW-AFC对LPS所致突触可塑性损伤的改善作用以及对胶质细胞过度激活的减轻与LW-AFC对中枢神经和外周免疫系统炎症因子的降低及调节外周B细胞、辅助T细胞和调节性T细胞比例相关。 展开更多

OBJECTIVE To explore the effect and mechanisms of LW-AFC,a new formula derived fromLiuwei Dihuang decoction,on chronic unpredictable mild stress(CUMS)-induced mood and cogni.tion impairment in mice.METHODS C57 BL/6 J mice were randomly placed into seven groups(n=10):normal control group,CUMS group,Fluoxetine(10 mg·kg^(-1),once per day) group,Liuwei Dihuang decoction group(LW,10 g·kg^(-1),once per day),and LW-AFC(0.8 g·kg^(-1),1.6 g·kg^(-1),3.2 g·kg^(-1),once per day) group.The stressed group was given CUMS for 4 weeks to set up a chronic multiple-stressed model.LW and LW-AFC was oral administered a week prior to CUMS and until the end of the study(a total of 35 d),while fluoxetine was administrated orally for 4 weeks.The anxiety behavior was analyzed using the open field test(OFT) and elevated plus maze test(EPM).The depression behavior was ana.lyzed using the sucrose preference test(SPT) and forced swimming test(FST).Spatial cognition was evaluated using Morris water maze(MWM) test and working memory was evaluated using new object recognition test(NORT).RESULTS CUMS for 28 d increased depressive-and anxiety-like behaviors.LW-AFC(1.6 g·kg^(-1)) significantly increased the numbers of entries into the open arm and time in the open arm of CUMS mice(P<0.05).LW-AFC(3.2 g·kg^(-1)) increased sucrose consumption and de.creased the immobility time of FST(P<0.01) of CUMS mice.The MWM test showed that spatial learning andmemory in CUMS mice were remarkably affected relative to controls,whereas LW-AFC(3.2 g·kg^(-1)) im.proves cognitive functions(P<0.05).CONCLUSION The mood and theability of learning and memory of thestressed group can be affected after exposure to CUS.Oral administration of LW-AFC significant.ly improved CUMS-induced impairments of mood and cognition in mice. 展开更多

OBJECTIVE Alzheimer disease(AD),the most common cause of dementia among older people,could not be prevented,halted,or reversed up till now.A large body of pharmacological study has revealed that Liuwei Dihuang(LW) possesses potential therapeutic effects on AD.LW-AFC is key fractions fromLW.In the present study,we investigated the effect of LW-AFC on AD mouse models.METHODS PrP-hAβPPswe/PS1ΔE9(APP/PS1) mice and senescence-accelerated mouse prone 8 strain(SAMP8),classic AD animal models,were employed.After the treatment of LW-AFC,mice were cognitively evaluated in behavioral experiments.Neuron loss,amyloid-β(Αβ)deposition,and Αβ level were analyzed using Nissl staining,immunofluorescence,and an AlphaLISA assay,respectively.Multiplex bead analysis,a radioimmunoassay,immunochemiluminometry,and an ELISA were used to measure cytokine and hormone levels.Lymphocyte subsets were detected using fl ow cytometry.RESULTS LW-AFC ameliorated the cognitive impairment observed in APP/PS1 and SAMP8 mice,including the impairment of object recognition memory,spatial learning and memory,and active and passive avoidance.In addition,LW-AFC alleviated the neuron loss in the hippocampus,suppressed amyloid-β(Αβ) deposition in the brain,and reduced the concentration of Aβ1-42 in the hippo.campus and plasma of APP/PS1 mice.LW-AFC treatment also significantly restored the imbalance of hypothalamic-pituitary-adrenal(HPA) and hypothalamic-pituitary-gonadal(HPG) axis,enhanced the proliferation of splenocytes,corrected the disorder of lymphocyte subsets,and regulated the abnormal production of cytokine in APP/PS1 and SAMP8 mice.Effects of LW-AFC on pharmacodynamics and neuroendocrine immunomodulation network in APP/PS1 and SAMP8 mice were better than meman.tine and donepezil.CONCLUSION LW-AFC ameliorated the behavioral and pathological deterioration of AD mouse models via the restoration of the NIM network,which supports the use of LW-AFC as a potential agent for AD therapy. 展开更多

OBJECTIVE Cranial radiotherapy is clinically used in the treatment of brain tumors;however,the consequent cognitive and emotional dysfunctions seriously impair the life quality of patients.LW-AFC,an active fraction combination extracted from classical traditional Chinese medicine prescription Liuwei Dihuang decoction,can improve cognitive and emotional dysfunctions in many animal models;however,the protective effect of LW-AFC on cranial irradiation-induced cognitive and emotional dysfunctions has not been reported.Recent studies indicate that impairment of adult hippocampal neurogenesis(AHN)and alterations of the neurogenic microenvironment in the hippocampus constitute critical factors in cognitive and emotional dysfunctions following cranial irradiation.Here,our research further investigated the potential protective effects and mechanisms of LW-AFC on cranial irradiation-induced cognitive and emotional dysfunctions in mice.METHODS LW-AFC(1.6 g·kg^(-1))was intragastrically administered to mice for 14 d before cranial irradiation(7 Gyγ-ray).AHN was examined by quantifying the number of proliferative neural stem cells and immature neurons in the dorsal and ventral hippocampus.The contextual fear conditioning test,open field test,and tail suspension test were used to assess cognitive and emotional functions in mice.To detect the change of the neurogenic microenvironment,colorimetry and multiplex bead analysis were performed to measure the level of oxidative stress,neurotrophic and growth factors,and inflammation in the hippocampus.RESULTS LW-AFC exerted beneficial effects on the contextual fear memory,anxiety behavior,and depression behavior in irradiated mice.Moreover,LW-AFC increased the number of proliferative neural stem cells and immature neurons in the dorsal hippocampus,displaying a regional specificity of neurogenic response.For the neurogenic microenvironment,LW-AFC significantly increased the contents of superoxide dismutase,glutathione peroxidase,glutathione,and catalase and decreased the content of malondialdehyde in the hippocampus of irradiated mice,accompanied by the increase in brain-derived neurotrophic factor,insulin-like growth factor-1,and interleukin-4 content.Together,LW-AFC improved cognitive and emotional dysfunctions,promoted AHN preferentially in the dorsal hippocampus,and ameliorated disturbance in the neurogenic microenvironment in irradiated mice.CONCLUSION LW-AFC ameliorates cranial irradiation-induced cognitive and emotional dysfunctions,and the underlying mechanisms are mediated by promoting AHN in the dorsal hippocampus and improving the neurogenic microenvironment.LW-AFC might be a promising therapeutic agent to treat cognitive and emotional dysfunctions in patients receiving cranial radiotherapy. 展开更多

目的观察六味地黄苷糖(LW-AFC)对睡眠剥夺小鼠所致焦虑抑郁样行为的作用及其机制。方法雄性C57小鼠依据小鼠体质量和自主活动性随机分为正常对照组、睡眠剥夺模型组、LW-AFC 0.8 g/kg、1.6 g/kg和3.2 g/kg组。应用72 h多平台水环境法建... 目的观察六味地黄苷糖(LW-AFC)对睡眠剥夺小鼠所致焦虑抑郁样行为的作用及其机制。方法雄性C57小鼠依据小鼠体质量和自主活动性随机分为正常对照组、睡眠剥夺模型组、LW-AFC 0.8 g/kg、1.6 g/kg和3.2 g/kg组。应用72 h多平台水环境法建立失眠动物模型。给药组预防给药7 d后进行睡眠剥夺72 h,同时给予LW-AFC至建模后第14天。采用旷场实验评价小鼠的焦虑样行为,采用悬尾和强迫游泳实验评价小鼠的抑郁样行为,应用液相芯片分析技术检测小鼠海马组织中促炎因子IL-1β、IL-6、TNF-α以及抗炎因子IL-4的含量。采用Pearson相关性分析LW-AFC对睡眠剥夺小鼠细胞因子的影响与焦虑抑郁样行为的相关性。采用Western印迹法检测小鼠海马星形胶质细胞激活标志物胶质原纤维酸性蛋白(GFAP)的变化。结果与正常对照组相比,睡眠剥夺模型组小鼠在旷场中央的探索时间显著缩短(P<0.05),悬尾和强迫游泳的不动时间显著延长(P<0.05),小鼠海马组织中GFAP表达显著升高(P<0.01),IL-1β、IL-6和TNF-α含量均显著增加(P<0.01),IL-4含量显著降低(P<0.01);与睡眠剥夺模型组相比,LW-AFC 1.6和3.2 g/kg组小鼠旷场实验的中央探索时间显著延长(P<0.05),GFAP表达量显著降低(P<0.05)。LW-AFC 3个剂量组小鼠悬尾和强迫游泳的不动时间显著缩短(P<0.05,P<0.01),海马组织中IL-1β和IL-6含量均显著降低(P<0.05,P<0.01);LW-AFC 1.6和3.2 g/kg组小鼠海马组织中TNF-α含量均显著降低(P<0.01),IL-4含量显著增加(P<0.05,P<0.01)。Pearson相关性分析结果提示,LW-AFC改善小鼠的焦虑抑郁样行为与促炎细胞因子(IL-1β、TNF-α)密切相关(P<0.05,P<0.01)。结论 LW-AFC能改善睡眠剥夺导致的小鼠焦虑和抑郁样行为改变,其机制可能与其调节细胞因子的失衡以及抑制星形胶质细胞活化有关。 展开更多

OBJECTIVE To explore the effect and mechanisms of LW-AFC,a new formula derived fromLiu Wei Di Huang Tang,on irradiation-induced reduction of mice adult hippocampal neurogenesis.METHODS C57 BL/6 J mice were randomly divided into seven groups(n=10):control group,LW-AFC group(1.6 g·kg^(-1)),Liu Wei Di Huang Tang(LW) group(10 g·kg^(-1)),brain derived neurotrophic factor(BDNF) group,irradiation group,irradiation + LW group,and irradiation + LW-AFC group.Reduction of mice adult hippocampal neurogenesis was induced by cranial irradiation.LW-AFC was administered by oral gavage for 30 d after cranial irradiation treatment.Immunofluorescence and Nissl′s staining were performed for histological morphology assessment.Bromodeoxyuridine(BrdU) staining was used in the detection of proliferation cells.The peripheral blood and hippocampal homogenate were collected to measure the content of tumor necrosis factor-α(TNF-α),interferon-gamma(INF-γ),interleukin-1β(IL-1β),IL-4 and IL-10.The hippocampal homogenate was used for Western blot to detect the BDNFTrkB signal pathway,including extracellular regulated protein kinases1/2(ERK1/2) and BDNF target protein.Morris water maze and new object recognition test were performed to examine the cognitive function of mice.The mice forced swimming and tail suspension test were used to assess alteration in depressive behavior.Long term potentiation was used to examine the synaptic plasticity change of mice.RESULTS Adult hippocampal neurogenesis was significantly reduced after irradiation of 20 Gray dose(10 Gray per day,total 2 d).LW-AFC treatment increased the BrdU number of irradiated mice(P<0.05).In Morris water maze test,LW-AFC group showed decreased escape latency in the learning period(P<0.05),while increased the number of crossing the platform in the memory period.LW-AFC can also reduce the immobility time of mice in the tail suspension test(P<0.01).CONCLU.SION LW-AFC modulates adult neurogenesis to ameliorate cognitive impairment and reduce depres.sive behavior in radiation injury mice. 展开更多
