In the first step of optimization, with Plackett-Burman design, ammoni...Response surface methodology (RSM) was used to optimize the fermentation medium for enhancing pyruvic acid production by Torulopsis glabrata TP19. In the first step of optimization, with Plackett-Burman design, ammonium sulfate, glucose and nicotinic acid were found to be the important factors affecting pyruvic acid production significantly. In the second step, a 23 full factorial central composite design and RSM were applied to determine the optimal concentration of each significant variable. A second-order polynomial was determined by the multiple regression analysis of the experimental data. The optimum values for the critical components were obtained as follows: ammonium sulfate 0.7498 (10.75 g/L), glucose 0.9383 (109.38 g/L) and nicotinic acid 0.3633 (7.86 mg/L) with a predicted value of maximum pyruvic acid production of 42.2 g/L. Under the optimal conditions, the practical pyruvic acid production was 42.4 g/L. The determination coefficient (R2) was 0.9483, which ensures adequate credibility of the model. By scaling up fermentation from flask to jar fermentor, we obtained promising results.展开更多

The overloading value for D301G is larger than that for D301R. The adsorption isotherm of pyruvic acid for both polymeric sorbents is greatly affected by the solution pH and MgSO4 concentration in the aqueous phase, and a high recovery efficiency of pyruvic acid from aqueous solution can be obtained at the solution pH around 2.展开更多
Background The study objective was to test the hypothesis that low crude protein(CP)diet with crystalline amino acids(CAA)supplementation improves Lys utilization efficiency for milk production and reduces protein tur...Background The study objective was to test the hypothesis that low crude protein(CP)diet with crystalline amino acids(CAA)supplementation improves Lys utilization efficiency for milk production and reduces protein turnover and muscle protein breakdown.Eighteen lactating multiparous Yorkshire sows were allotted to 1 of 2 isocaloric diets(10.80 MJ/kg net energy):control(CON;19.24%CP)and reduced CP with“optimal”AA profile(OPT;14.00%CP).Sow body weight and backfat were recorded on d 1 and 21 of lactation and piglets were weighed on d 1,14,18,and 21 of lactation.Between d 14 and 18,a subset of 9 sows(CON=4,OPT=5)was infused with a mixed solution of 3-[methyl-2H3]histidine(bolus injection)and[13C]bicarbonate(priming dose)first,then a constant 2-h[13C]bicarbonate infusion followed by a 6-h primed constant[1-13C]lysine infusion.Serial blood and milk sampling were performed to determine plasma and milk Lys enrichment,Lys oxidation rate,whole body protein turnover,and muscle protein breakdown.Results Over the 21-d lactation period,compared to CON,sows fed OPT had greater litter growth rate(P<0.05).Compared to CON,sows fed OPT had greater efficiency of Lys(P<0.05),Lys mammary flux(P<0.01)and whole-body protein turnover efficiency(P<0.05).Compared to CON,sows fed OPT tended to have lower whole body protein breakdown rate(P=0.069).Muscle protein breakdown rate did not differ between OPT and CON(P=0.197).Conclusion Feeding an improved AA balance diet increased efficiency of Lys and reduced whole-body protein turnover and protein breakdown.These results imply that the lower maternal N retention observed in lactating sows fed improved AA balance diets in previous studies may be a result of greater partitioning of AA towards milk rather than greater body protein breakdown.展开更多

We ana...In this study, we assess the effectiveness of utilizing a straight knee kick in the backstroke with regard to the overall time, max speed, and distance per stroke when using a straight knee butterfly technique. We analyzed freestyle kick of knee angles from 7-Olympic medalists, 1970's S-shaped pulling pattern stroke was used with a bending knee freestyle kick. The straight knee kick used in butterfly competitions resulted in improvements from 54.74 to 51.00 in Men's 100-meter butterfly and fi'om 23.43 to 23.02 in Men's 50-meter butterfly. Using a straight knee kick in backstroke competitions resulted in improvements from 1:01.74 to 1:00.55 in the 100-meter backstroke event. Increases in performance speed, max speed and distance per stroke among three elite level swimmers are highly correlated to duration of the kick resistance, and straight knee kick in backstroke. The straight knee kick also resulted in an increase in lactate acid.展开更多

The conventional method was used and a series of novel linear and cyclic Schiff bases were obtained with or without catalyst. All the Schiff bases were fully characterized by CHN elemental analysis, FT-IR, 1H & 13C NMR, EI-MS and two of the Schiff bases were further characterized by X-ray crystallographic structure analysis. Compound 2 crystallizes in the triclinic space group P-1 and unit cell dimensions are: a = 4.1777(8), b = 5.9538(11), c = 13.458(3) &ARING;, α = 92.759(6), β = 90.813(6), γ = 100.040(6)°, R1 = 0.0439. Background:Under current dietary regimens,milk production by lactating sows is insufficient to sustain the maximal growth of their piglets.As precursors of glutamate and glutamine as well as substrates and activators ...Background:Under current dietary regimens,milk production by lactating sows is insufficient to sustain the maximal growth of their piglets.As precursors of glutamate and glutamine as well as substrates and activators of protein synthesis,branched-chain amino acids(BCAAs)have great potential for enhancing milk production by sows.Methods:Thirty multiparous sows were assigned randomly into one of three groups:control(a corn-and soybean meal-based diet),the basal diet+1.535%BCAAs;and the basal diet+3.07%BCAAs.The ratio(g/g)among the supplemental L-isoleucine,L-leucine and L-valine was 1.00:2.56:1.23.Diets were made isonitrogenous by the addition of appropriate amounts of L-alanine.Lactating sows had free access to drinking water and their respective diets.The number of live-born piglets was standardized to 9 per sow at d 0 of lactation(the day of parturition).On d 3,15 and 29 of lactation,body weights and milk consumption of piglets were measured,and blood samples were obtained from sows and piglets 2 h and 1 h after feeding and nursing,respectively.Results:Feed intake did not differ among the three groups of sows.Concentrations of asparagine,glutamate,glutamine,citrulline,arginine,proline,BCAAs,and many other amino acids were greater(P<0.05)in the plasma of BCAA-supplemented sows and their piglets than those in the control group.Compared with the control,dietary supplementation with 1.535%and 3.07%BCAAs increased(P<0.05)concentrations of free and protein-bound BCAAs,glutamate plus glutamine,aspartate plus asparagine,and many other amino acids in milk;milk production by 14%and 21%,respectively;daily weight gains of piglets by 19%and 28%,respectively,while reducing preweaning mortality rates by 50%and 70%,respectively.Conclusion:Dietary supplementation with up to 3.07%BCAAs enhanced milk production by lactating sows,and the growth and survival of their piglets.展开更多

[ Result ] The daily feed intake, lactation yield and average daily gain of piglets in the experimental group were increased by 9.4%, 28.5% and 10.7%, respectively. The backfat of lactating sows was decreased less, and the same with the weaning-oestrus interval. The survival ratio of piglets was increased by 4.5%. Compared with the control group, the content of lactose and fat increased significantly, but other components almost did not change. Background:The study objective was to test the hypothesis that 1)lowering dietary crude protein(CP)increases dietary energetic efficiency and reduces metabolic heat associated with lactation,and 2)excessive dietary le...Background:The study objective was to test the hypothesis that 1)lowering dietary crude protein(CP)increases dietary energetic efficiency and reduces metabolic heat associated with lactation,and 2)excessive dietary leucine(Leu)supplementation in a low CP diet decreases dietary energetic efficiency and increases metabolic heat associated with lactation.Methods:Fifty-four lactating multiparous Yorkshire sows were allotted to 1 of 3 isocaloric diets(10.80 MJ/kg net energy):1)control(CON;18.75%CP),2)reduced CP with a near ideal or optimal AA profile(OPT;13.75%CP)and 3)diet OPT with excessive Leu(OPTLEU;14.25%CP).Sow body weight and backfat were recorded on day 1 and 21 of lactation and piglets were weighed on day 1,4,8,14,18,and 21 of lactation.Energy balance was measured on sows during early(day 4 to 8)and peak(day 14 to18)lactation,and milk was sampled on day 8 and 18.Results:Over 21-day lactation,sows fed OPT lost body weight and body lipid(P<0.05).In peak lactation,sows fed OPT had higher milk energy output(P<0.05)than CON.Sows fed OPTLEU tended(P=0.07)to have less milk energy output than OPT and did not differ from CON.Maternal energy retention was lower(P<0.05)in OPT and OPTLEU compared to CON sows,and did not differ between OPTLEU and OPT sows.Sows fed OPT had higher(P<0.05)apparent energy efficiency for milk production compared to CON.Heat production associated with lactation was lower(P<0.05)or tended to be lower(P=0.082),respectively,in OPT and OPTLEU compared to CON sows.Conclusion:The OPT diet,in peak lactation,improved dietary energy utilization for lactation due to less urinary energy and metabolic heat loss,and triggered dietary energy deposition into milk at the expense of maternal lipid mobilization.Leucine supplementation above requirement may reduce dietary energy utilization for lactation by decreasing the energy partitioning towards milk,partially explaining the effectiveness of OPT diet over CON diets.展开更多

A comprehensive review of recent studies showed that lipid supplementation increases average daily energy intake, which is partitioned for lactation as indicated by greater milk fat output and improved litter growth rate. Recent compelling findings showed that addition of particular lipids during lactation improved the subsequent reproductive outcome of sows. Such benefits were related to the level of dietary essential fatty acids(EFA, linoleic acid, C18:2n-6; and α-linolenic acid, C18:3n-3) during lactation. Lactation diets without supplemental EFA resulted in a pronounced negative balance(intake minus milk output) of linoleic(-25.49 g/d) and α-linolenic acid(-2.75 g/d); which compromised sow fertility(farrowing rate 〈 75 % and culling rates 〉 25 % of weaned sows). This phenomenon seems to be increasingly important with advancing sow age because of a progressive reduction of body EFA pool over successive lactations. The net effect of supplemental EFA during lactation was to create a positive EFA balance, which improved the subsequent reproduction of sows. Adequate linoleic acid intake improved the proportion of sows that farrowed in the subsequent cycle(Farrowing rate(%) = [(-1.5 × 10^-3× linoleic acid intake(g/d)^2) +(0.53 × linoleic acid intake(g/d)) +(45.2)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). In addition, increasing linoleic acid intake increased the number of pigs born in the subsequent cycle(total pigs born(n) = [(9.4 × 10^-5× linoleic acid intake(g/d)^2) +(0.04 × linoleic acid intake(g/d)) +(10.94)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). Supplemental α-linolenic acid resulted in a rapid return to estrus(sows bred: sows weaned = 94.2 %; wean-to-estrus interval = 4.0 d) and achieved a high retention of pregnancy(sows pregnant: sows bred = 98 %). Collectively, we conclude that a minimum dietary intake of10 g/d of α-linolenic acid, simultaneous with a minimum of 125 g/d of linoleic acid should be provided to ≥ 95 % of the sows; thereby, achieving a maximum sow reproductive efficiency through multiple mechanisms that include rapid return to estrus, high maintenance of pregnancy and large subsequent litter size in mature sows, that appear to be susceptible to EFA deficiency.展开更多

Plasma amino acids(AA)in sows are important precursors for milk protein synthesis in the mammary gland. In order to study the regulation of AA transported in sow mammary glands and possible underlying mechanisms, we measured the expression of genes coding for milk proteins, AA transporter expressions, and plasma AA concentrations in sows at three different physiological stages(D-17, D1 and D17 of lactation), and then further investigated the regulation of AA transport across the cell membrane by adaptive mechanisms using pig mammary epithelial cells(PMEC) as an in vitro model.PMEC were cultured in DMEM:F12 with 4 amino acid concentrations(0 × AA complex, 1 × AA complex, 5 × AA complex,and 25 × AA complex). Classes of AA complexes evaluated in this study included neutral AAs(L-Ala + L-Ser + L-Cys), acidic AAs(L-Asp, L-Glu) and neutral + basic AAs(L-Ala + L-Ser + L-Cys + L-Lys).Results: Our results indicated that m RNA expression of genes coding for milk protein(αs1-casein, αs2-casein,β-casein and κ-casein) increased significantly with the advance of physiological stage(P < 0.05), and plasma concentrations of most AAs including threonine, serine, glutamate, alanine, valine, cysteine, methionine, isoleucine and tyrosine were greater at D1 of lactation compared with D-17 and D17 of lactation(P < 0.05). Additionally, protein and gene expressions of AA transporters including excitatory AA transporter 3(EAAT3), alanine/serine/cysteine/threonine transporter(ASCT1) and sodium-coupled neutral AA transporter 1(SNAT2) were greater in lactating sow mammary glands compared with sow mammary glands in late pregnancy(P < 0.05). The m RNA expressions of SLC38 A2, SLC1 A1,SLC6 A14 increased significantly in the cell mediums supplemented with 5 × and 25 × of AA complexes compared with those cells cultured in DMEM/F12 cell medium(P < 0.05). The m RNA expressions of SLC38 A, SLC1 A4, and SLC6 A14 also increased in EBSS cell medium compared to DMEM/F12. However, only m RNA expression of SLC38 A decreased when AA complex was added into EBSS(P < 0.05).Conclusion: AA transportation was positively regulated in sow mammary glands with the advance of physiological stage from late pregnancy to peak of lactation and AA transporters in PMECs were adaptively regulated by changed AA concentrations.展开更多

In the second step, a 23 full factorial central composite design and RSM were applied to determine the optimal concentration of each significant variable. A second-order polynomial was determined by the multiple regression analysis of the experimental data. The optimum values for the critical components were obtained as follows: ammonium sulfate 0.7498 (10.75 g/L), glucose 0.9383 (109.38 g/L) and nicotinic acid 0.3633 (7.86 mg/L) with a predicted value of maximum pyruvic acid production of 42.2 g/L. Under the optimal conditions, the practical pyruvic acid production was 42.4 g/L. The determination coefficient (R2) was 0.9483, which ensures adequate credibility of the model. By scaling up fermentation from flask to jar fermentor, we obtained promising results.

文摘Uptakes of pyruvic acid for two types of commercially available weakly basic polymer sorbents, D301G and D301R, have been measured over a wide pH range and at various salinities of MgSO4. The results show that the overloading adsorption of pyruvic acid occurs on both weakly basic polymer sorbents, and the overloading models can predict the experimental data of uptake very well. The overloading value for D301G is larger than that for D301R. The adsorption isotherm of pyruvic acid for both polymeric sorbents is greatly affected by the solution pH and MgSO4 concentration in the aqueous phase, and a high recovery efficiency of pyruvic acid from aqueous solution can be obtained at the solution pH around 2.

文摘Background The study objective was to test the hypothesis that low crude protein(CP)diet with crystalline amino acids(CAA)supplementation improves Lys utilization efficiency for milk production and reduces protein turnover and muscle protein breakdown.Eighteen lactating multiparous Yorkshire sows were allotted to 1 of 2 isocaloric diets(10.80 MJ/kg net energy):control(CON;19.24%CP)and reduced CP with“optimal”AA profile(OPT;14.00%CP).Sow body weight and backfat were recorded on d 1 and 21 of lactation and piglets were weighed on d 1,14,18,and 21 of lactation.Between d 14 and 18,a subset of 9 sows(CON=4,OPT=5)was infused with a mixed solution of 3-[methyl-2H3]histidine(bolus injection)and[13C]bicarbonate(priming dose)first,then a constant 2-h[13C]bicarbonate infusion followed by a 6-h primed constant[1-13C]lysine infusion.Serial blood and milk sampling were performed to determine plasma and milk Lys enrichment,Lys oxidation rate,whole body protein turnover,and muscle protein breakdown.Results Over the 21-d lactation period,compared to CON,sows fed OPT had greater litter growth rate(P<0.05).Compared to CON,sows fed OPT had greater efficiency of Lys(P<0.05),Lys mammary flux(P<0.01)and whole-body protein turnover efficiency(P<0.05).Compared to CON,sows fed OPT tended to have lower whole body protein breakdown rate(P=0.069).Muscle protein breakdown rate did not differ between OPT and CON(P=0.197).Conclusion Feeding an improved AA balance diet increased efficiency of Lys and reduced whole-body protein turnover and protein breakdown.These results imply that the lower maternal N retention observed in lactating sows fed improved AA balance diets in previous studies may be a result of greater partitioning of AA towards milk rather than greater body protein breakdown.

文摘In this study, we assess the effectiveness of utilizing a straight knee kick in the backstroke with regard to the overall time, max speed, and distance per stroke when using a straight knee butterfly technique. We analyzed freestyle kick of knee angles from 7-Olympic medalists, 1970's S-shaped pulling pattern stroke was used with a bending knee freestyle kick. The straight knee kick used in butterfly competitions resulted in improvements from 54.74 to 51.00 in Men's 100-meter butterfly and fi'om 23.43 to 23.02 in Men's 50-meter butterfly. Using a straight knee kick in backstroke competitions resulted in improvements from 1:01.74 to 1:00.55 in the 100-meter backstroke event. Increases in performance speed, max speed and distance per stroke among three elite level swimmers are highly correlated to duration of the kick resistance, and straight knee kick in backstroke. The straight knee kick also resulted in an increase in lactate acid.

文摘One‐step production of pyruvic acid through selective oxidation of glycerol was investigated using lead promoted platinum/activated carbon(Pb‐Pt/AC)catalysts under mild conditions.The results of N2physisorption,X‐ray diffraction,X‐ray photoelectron spectroscopy,and high‐resolution transmission electron microscopy revealed that the alloy phases of PtPb and PtxPb were favorable for pyruvic acid production from glycerol oxidation,whereas the Pb3(CO3)2(OH)2and surface Pb0species inhibited the glycerol conversion.The loading of Pb and the catalyst preparation method(including impregnation and deposition precipitation)affected the formation of different metal species.Pyruvic acid was obtained at a yield of18.4%on a5.0wt%Pb‐5.0wt%Pt/AC catalyst prepared by co‐deposition precipitation method and500°C argon treatment.
文摘Four Schiff bases, from pyruvic acid (1) with amines containing N and S donor atoms, thiocarbohydrazide (2, 61%), 2-methyl-3-thiosemicarbazide (3, 26%), S-benzyldithiocarbazate (4, 51%) and S-n-octyldithiocarbazate (5, 63%) have been successfully synthesized. The conventional method was used and a series of novel linear and cyclic Schiff bases were obtained with or without catalyst. All the Schiff bases were fully characterized by CHN elemental analysis, FT-IR, 1H & 13C NMR, EI-MS and two of the Schiff bases were further characterized by X-ray crystallographic structure analysis. Compound 2 crystallizes in the triclinic space group P-1 and unit cell dimensions are: a = 4.1777(8), b = 5.9538(11), c = 13.458(3) &ARING;, α = 92.759(6), β = 90.813(6), γ = 100.040(6)°, R1 = 0.0439. Compound 3 crystallizes in the orthorhombic space group P n a 2(1) and unit cell dimensions are: a = 5.5992(2), b = 11.3962(5), c = 10.6473(5), α = 92.759(6), β = 90.813(6), γ = 100.040(6)°, R1 = 0.0285. Compounds 2 and 3 were obtained as cyclic Schiff bases which are triazine derivatives.

文摘Density functional theory BLYP (using Becke's and Lee-Yang-Pars's correlation functionals), ab initio Hartree-Fock (HF) and hybrid DFT/HF B3LYP calculations were carried out to study the structure and vibrational spectra of pyruvic acid. The scaled B3LYP/6-31G* frequencies correspond well with available experimental assignment of the functional vibrational modes and the mean absolut devation is only 12.3cm^(-1).

文摘In this paper a new chemical oscillating reaction in the pyruvic acid-BrO-H2SO4 - [CuL](ClO4)2 system, where L is 5, 7, 12, 14-tetraethyl-7, 14-dimethyl-1, 4, 8, 11 -tetraazacyclotetradeca-4, 11-diene, is reported. The features of the oscillations are studied in detail. The effects of Ag+,Hg2+,CCl4, Vc, H2O2, acrylonitrile, and temperature on the oscillation system are also discussed.

文摘It is important to study the solvent effect on keto-enol tautomerism that has applications in many areas of chemical engineering.In this work,we use a multiscale reaction density functional theory(Rx DFT)to study the keto-enol tautomerism and isomerization of pyruvic acid.The results show that both effects of solvation and water assistance could reduce the reaction barriers.The water molecule participates the reaction as a catalyst to accept/give the protons with forming a hexagonal ring in the transition state.As a result of this temporary and intermediate hexagonal ring,the solute configuration undergoes a small variation during the reaction,giving a diminished contribution to the intrinsic reaction free energy.The solvent distribution shows a local ordering behavior near the solute that also reduces the contribution of solvation effect to the reaction barrier.Water assistance plays a major role in both pre-reaction and postreaction process.In terms of the driving force for the reaction,the effects of both solvation and water assistance are important.

文摘In this study,we assess the effectiveness of utilizing a downward breaststroke kick with regard to the overall time,max speed and velocity per stroke when using a downward breaststroke kick technique.We analyzed breaststroke kick of knee angles from 8 elite world class swimmers.The downward breaststroke kick used in breaststroke competitions resulted in improvements average of 100 yard or 100 meter breaststroke from 1:05.10 s to 1:01.97 s.Increases in performance max speed and velocity per stroke among 9 elite world class swimmers are highly correlated to duration of the kick aerodynamic buoyant farce in breaststroke.The downward breaststroke kick also resulted in an increase in lactate acid.
文摘Altered blood chemistry, acid-base and electrolyte are suggested determinants of sleep disturbance, with frequent arousal at high altitude even in well and long-trained altitude marathon runners. 文摘Background:Under current dietary regimens,milk production by lactating sows is insufficient to sustain the maximal growth of their piglets.As precursors of glutamate and glutamine as well as substrates and activators of protein synthesis,branched-chain amino acids(BCAAs)have great potential for enhancing milk production by sows.Methods:Thirty multiparous sows were assigned randomly into one of three groups:control(a corn-and soybean meal-based diet),the basal diet+1.535%BCAAs;and the basal diet+3.07%BCAAs.The ratio(g/g)among the supplemental L-isoleucine,L-leucine and L-valine was 1.00:2.56:1.23.Diets were made isonitrogenous by the addition of appropriate amounts of L-alanine.Lactating sows had free access to drinking water and their respective diets.The number of live-born piglets was standardized to 9 per sow at d 0 of lactation(the day of parturition).On d 3,15 and 29 of lactation,body weights and milk consumption of piglets were measured,and blood samples were obtained from sows and piglets 2 h and 1 h after feeding and nursing,respectively.Results:Feed intake did not differ among the three groups of sows.Concentrations of asparagine,glutamate,glutamine,citrulline,arginine,proline,BCAAs,and many other amino acids were greater(P<0.05)in the plasma of BCAA-supplemented sows and their piglets than those in the control group.Compared with the control,dietary supplementation with 1.535%and 3.07%BCAAs increased(P<0.05)concentrations of free and protein-bound BCAAs,glutamate plus glutamine,aspartate plus asparagine,and many other amino acids in milk;milk production by 14%and 21%,respectively;daily weight gains of piglets by 19%and 28%,respectively,while reducing preweaning mortality rates by 50%and 70%,respectively.Conclusion:Dietary supplementation with up to 3.07%BCAAs enhanced milk production by lactating sows,and the growth and survival of their piglets.

文摘[ Objective] To study the effects of gamma-aminobutyric acid (GABA) on performance of lactating sows during heat stress. [ Metbod] A total of 14 sows at the same parity and with close expected date of childbirth and similar body we:,ght were randomly divided into control group and experimental group. They were fed a common basal diet and a GABA (300 mg/kg) supplementary diet, respectively. The trial lasted for 21 d. [ Result ] The daily feed intake, lactation yield and average daily gain of piglets in the experimental group were increased by 9.4%, 28.5% and 10.7%, respectively. The backfat of lactating sows was decreased less, and the same with the weaning-oestrus interval. The survival ratio of piglets was increased by 4.5%. Compared with the control group, the content of lactose and fat increased significantly, but other components almost did not change. [Conclusien] Supplementing GABA in diet can improve the performance of lactating sows and promote the growth of piglets effectively.

文摘Background:The study objective was to test the hypothesis that 1)lowering dietary crude protein(CP)increases dietary energetic efficiency and reduces metabolic heat associated with lactation,and 2)excessive dietary leucine(Leu)supplementation in a low CP diet decreases dietary energetic efficiency and increases metabolic heat associated with lactation.Methods:Fifty-four lactating multiparous Yorkshire sows were allotted to 1 of 3 isocaloric diets(10.80 MJ/kg net energy):1)control(CON;18.75%CP),2)reduced CP with a near ideal or optimal AA profile(OPT;13.75%CP)and 3)diet OPT with excessive Leu(OPTLEU;14.25%CP).Sow body weight and backfat were recorded on day 1 and 21 of lactation and piglets were weighed on day 1,4,8,14,18,and 21 of lactation.Energy balance was measured on sows during early(day 4 to 8)and peak(day 14 to18)lactation,and milk was sampled on day 8 and 18.Results:Over 21-day lactation,sows fed OPT lost body weight and body lipid(P<0.05).In peak lactation,sows fed OPT had higher milk energy output(P<0.05)than CON.Sows fed OPTLEU tended(P=0.07)to have less milk energy output than OPT and did not differ from CON.Maternal energy retention was lower(P<0.05)in OPT and OPTLEU compared to CON sows,and did not differ between OPTLEU and OPT sows.Sows fed OPT had higher(P<0.05)apparent energy efficiency for milk production compared to CON.Heat production associated with lactation was lower(P<0.05)or tended to be lower(P=0.082),respectively,in OPT and OPTLEU compared to CON sows.Conclusion:The OPT diet,in peak lactation,improved dietary energy utilization for lactation due to less urinary energy and metabolic heat loss,and triggered dietary energy deposition into milk at the expense of maternal lipid mobilization.Leucine supplementation above requirement may reduce dietary energy utilization for lactation by decreasing the energy partitioning towards milk,partially explaining the effectiveness of OPT diet over CON diets.

文摘Dietary lipid supplementation benefits the prolific and high-producing modern lactating sow. A comprehensive review of recent studies showed that lipid supplementation increases average daily energy intake, which is partitioned for lactation as indicated by greater milk fat output and improved litter growth rate. Recent compelling findings showed that addition of particular lipids during lactation improved the subsequent reproductive outcome of sows. Such benefits were related to the level of dietary essential fatty acids(EFA, linoleic acid, C18:2n-6; and α-linolenic acid, C18:3n-3) during lactation. Lactation diets without supplemental EFA resulted in a pronounced negative balance(intake minus milk output) of linoleic(-25.49 g/d) and α-linolenic acid(-2.75 g/d); which compromised sow fertility(farrowing rate 〈 75 % and culling rates 〉 25 % of weaned sows). This phenomenon seems to be increasingly important with advancing sow age because of a progressive reduction of body EFA pool over successive lactations. The net effect of supplemental EFA during lactation was to create a positive EFA balance, which improved the subsequent reproduction of sows. Adequate linoleic acid intake improved the proportion of sows that farrowed in the subsequent cycle(Farrowing rate(%) = [(-1.5 × 10^-3× linoleic acid intake(g/d)^2) +(0.53 × linoleic acid intake(g/d)) +(45.2)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). In addition, increasing linoleic acid intake increased the number of pigs born in the subsequent cycle(total pigs born(n) = [(9.4 × 10^-5× linoleic acid intake(g/d)^2) +(0.04 × linoleic acid intake(g/d)) +(10.94)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). Supplemental α-linolenic acid resulted in a rapid return to estrus(sows bred: sows weaned = 94.2 %; wean-to-estrus interval = 4.0 d) and achieved a high retention of pregnancy(sows pregnant: sows bred = 98 %). Collectively, we conclude that a minimum dietary intake of10 g/d of α-linolenic acid, simultaneous with a minimum of 125 g/d of linoleic acid should be provided to ≥ 95 % of the sows; thereby, achieving a maximum sow reproductive efficiency through multiple mechanisms that include rapid return to estrus, high maintenance of pregnancy and large subsequent litter size in mature sows, that appear to be susceptible to EFA deficiency.

文摘Background: Milk protein is crucial for milk quality in sows and health of newborn piglets. Plasma amino acids(AA)in sows are important precursors for milk protein synthesis in the mammary gland. In order to study the regulation of AA transported in sow mammary glands and possible underlying mechanisms, we measured the expression of genes coding for milk proteins, AA transporter expressions, and plasma AA concentrations in sows at three different physiological stages(D-17, D1 and D17 of lactation), and then further investigated the regulation of AA transport across the cell membrane by adaptive mechanisms using pig mammary epithelial cells(PMEC) as an in vitro model.PMEC were cultured in DMEM:F12 with 4 amino acid concentrations(0 × AA complex, 1 × AA complex, 5 × AA complex,and 25 × AA complex). Classes of AA complexes evaluated in this study included neutral AAs(L-Ala + L-Ser + L-Cys), acidic AAs(L-Asp, L-Glu) and neutral + basic AAs(L-Ala + L-Ser + L-Cys + L-Lys).Results: Our results indicated that m RNA expression of genes coding for milk protein(αs1-casein, αs2-casein,β-casein and κ-casein) increased significantly with the advance of physiological stage(P < 0.05), and plasma concentrations of most AAs including threonine, serine, glutamate, alanine, valine, cysteine, methionine, isoleucine and tyrosine were greater at D1 of lactation compared with D-17 and D17 of lactation(P < 0.05). Additionally, protein and gene expressions of AA transporters including excitatory AA transporter 3(EAAT3), alanine/serine/cysteine/threonine transporter(ASCT1) and sodium-coupled neutral AA transporter 1(SNAT2) were greater in lactating sow mammary glands compared with sow mammary glands in late pregnancy(P < 0.05). The m RNA expressions of SLC38 A2, SLC1 A1,SLC6 A14 increased significantly in the cell mediums supplemented with 5 × and 25 × of AA complexes compared with those cells cultured in DMEM/F12 cell medium(P < 0.05). The m RNA expressions of SLC38 A, SLC1 A4, and SLC6 A14 also increased in EBSS cell medium compared to DMEM/F12. However, only m RNA expression of SLC38 A decreased when AA complex was added into EBSS(P < 0.05).Conclusion: AA transportation was positively regulated in sow mammary glands with the advance of physiological stage from late pregnancy to peak of lactation and AA transporters in PMECs were adaptively regulated by changed AA concentrations.
