[ Method] Effect of C/N ratio on DHA production by Schizochytrium sp. JN-3 was explored. The industrial fermentation potential of Schizochytrium sp. JN-3 was investigated by pilot-scale fermentation in a 200 L fermentation tank. [ Result] Results of fermentation in a 7 L fermentation tank showed that the maximum biomass and total lipid content were 34.3 and 20.1 g/L, respectively, when C/N ratio was 10 ; the maximum DHA yield was 46.9% when C/N ratio was 20. When pilot-scale fermentation was conducted in a 200 L fermentation tank, the biomass was 30. 2 g/L, total lipid content was 18.4 g/L, accounting for 63.2% of the dry weight of cells; DHA content was 7.4 g/L, accounting for 40. 2% of the total lipid content. Biobutanol is an advanced biofuel that can be produced from excess lignocellulose via acetone-butanol-ethanol(ABE)fermentation.Although significant technological progress has been made in this field,attempts at larges...Biobutanol is an advanced biofuel that can be produced from excess lignocellulose via acetone-butanol-ethanol(ABE)fermentation.Although significant technological progress has been made in this field,attempts at largescale lignocellulosic ABE production remain scarce.In this study,1m^(3)scale ABE fermentation was investigated using high inhibitor tolerance Clostridium acetobutylicum ABE-P1201 and steam-exploded corn stover hydrolysate(SECSH).Before expanding the fermentation scale,the detoxification process for SECSH was simplified by process engineering.Results revealed that appropriate pH management during the fed-batch cultivation could largely decrease the inhibition of the toxic components in undetoxified SECSH to the solventogenesis phase of the ABE-P1201 strains,avoiding“acid crash”.Therefore,after naturalizing the pH by Ca(OH)_(2),the undetoxified SECSH,without removal of the solid components,reached 17.68±1.30 g/L of ABE production with 0.34±0.01 g/g of yield in 1 L scale bioreactor.Based on this strategy,the fermentation scale gradually expanded from laboratory-scale apparatus to pilot-scale bioreactors.Finally,17.05±1.20 g/L of ABE titer and 0.32±0.01 g/g of ABE yield were realized in 1m3 bioreactor,corresponding to approximately 145 kg of ABE production from 1 t of dry corn stover.The pilot-scale ABE fermentation demonstrated excellent stability during repeated operations.This study provided a simplified ABE fermentation strategy and verified the feasibility of the pilot process,providing tremendous significance and a solid foundation for the future industrialization of second-generation ABE plants.展开更多

The aim is to simulate the large-scale conditions, characterized by long mixing times introducing local gradients into the bioreactor, in laboratory scale bioreactor. We have expanded the current physiological knowledge about the consequences of such cultivation conditions from well-known mixed-acid fermentation to amino acid metabolism. Several interesting deviations were observed for amino acids derived from pyruvate when a STR-PFR scale-down simulator and a regular STR fed-batch cultivation were compared. In this article we show that the cultivation of Escherichia coli K-12 substrain W3110 in a scale-down two-compartment bioreactor has consequences on the free amino acid pools. Repeated occurrence of high glucose zones affects the biosynthesis of amino acids derived from pyruvate. This is interesting since the pyruvate based amino acids are precursors of the branched chain amino acid pathway, which is further disturbed by an unbalanced regulation caused by a frameshift mutation in the ilvG gene of E. coli K-12 strains. As a consequence the formation of non-typical amino acid norvaline was detected in the two compartment bioreactor cultivation.展开更多
Sweet potato shochu is a traditional Japanese spirit produced mainly in the South Kyushu area in Japan. The amount of stillage reaches approximately 8 x 105 tons per year. Wastewater mainly containing stillage from th...Sweet potato shochu is a traditional Japanese spirit produced mainly in the South Kyushu area in Japan. The amount of stillage reaches approximately 8 x 105 tons per year. Wastewater mainly containing stillage from the production of sweet potato-shochu was treated thermophilically in a fullscale treatment plant using fixed-bed reactors (8 reactors ×283 m3). Following the addition of Ni2+ and Co2+, the reactors have been stably operated for six years at a high chemical oxygen demand (COD) loading rate of 14 kg/(m3.day). Analysis of coenzyme content and microbial communities indicated that similar microbial communities were present in the liquid phase and on the fiber carders installed in reactors. Bacteria in the phyla Firmieutes as well as Bacteroidetes were dominant bacteria, and Methanosarcina thermophila as well as Methanothermobacter crinale were dominant methanogens in the reactors. This study reveals that stillage from sweet potato-shochu production can be treated effectively in a full-scale fixed-bed reactor under thermophilic conditions with the help of Ni2~ and Co2+. The high diversity of bacterial community and the coexistence of both aceticlastic and hydrogenotrophic methanogens contributed to the excellent fermentation performance.展开更多
