Chemical and physical mutagenesis has been used to increase genetic variability in crop plants. More than 430 new varieties have been derived as mutants of rice (Oryza sativa L.) via the application of different mutagenic agents. Chemical mutagens such as ethyl methane sulphonate (EMS), diepoxybutane-derived (DEB), sodium azide and irradiation (Gamma rays, X-rays and fast neutrons) have been widely used to induce a large number of functional variations in rice and others crops. Among chemical mutagens, the alkylating agent, ethyl methane sulfonate (EMS) is the most commonly used in plants as it causes a high frequency of nucleotide substitutions, as detected in different genomes. In this study, seeds of potential genotype of the popular variety, (Oryza sativa L. spp. Indica cv. MR219) were treated with EMS at concentrations of 0.25%, 0.50%, 0.75%, 1%, 1.25%, 1.5% and 2%. Sensitivity to EMS was determined by various measurements on the M1 generation. As concentration of applied EMS increased, will decrease in germination, seedling height, root length and emergence under field conditions was observed in M1 generation as compared to the non-treatment control. Plant height and root length also decreased with increases in EMS mutagenesis in an approximately linear fashion. The LD25 and LD50 values were observed based on growth reduction of seedlings after EMS treatment with 0.25% and 0.50% on the rice variety (Oryza sativa L. spp. Indica cv. MR219). 展开更多

Obtaining the Minimum Lethal Dose against <i>Fasciola hepatica in Vitro</i>Using Plant Extract Hexanes with Fasciolicide Activity and Toxicity Evaluation on CD1 Male Mice

Its importance lies in the economic losses it produces in the livestock industry. Its control is carried out by using a chemical fasciolicide showing resistance problems and environmental contamination. Looking for an alternative control for this disease the present study was aimed at determining the hexane anti-Fasciola hepatica in the in vitro effect of some plant extracts and the minimum lethal dose of the mentioned extracts. All selected plants were tested in vitro at concentrations of 500, 250, 125 and 50 mg/L):Achilleamillefolium (plumajillo), Artemisiaabsinthium (wormwood), Artemisia mexicana (estafiate), Castelatortuousa (chaparroamargo), Chenopodiumgraveolens (epazote de zorrillo), Gymnospermaglutinosum (popote) Justicia spicigera (muicle), Limpia critridora (cedron), Lippiagraveolens (oregano), Menthapiperita (Mint), Populus alba (alamo) and Thymusvulgaris (thyme). Subsequently proceeded to perform a toxicity study with these fractions in CD1 male mice 10-13 weeks of age, forming groups of 3-5 animals they were administered a single oral dose being (5 mg/kg, 50 mg/kg, 500 mg/kg, 2500 mg/kg and 5000 mg/kg) and were kept under observation 20 days, later were sacrificed and a kidney and liver histology was performed, finding the safety of the extracts. To perform the toxicity study with these fractions, groups of five CD1 male-mice were formed, they were treated with oral doses of 5, 50, 500, 2500 and 5000 mg/kg, administered with a cannule. All mice were kept under observation for 20 days. Finally they were sacrificed to perform histology of the kidney and liver in search of possible side effects. Results show that none of the extracts exhibited that fasciolocide activity for mice CD1 even at the highest dose thereforefinding the safety of the extracts. 展开更多

Somina (herbal preparation) prepared by Hamdard Laboratories (Waqf) Pakistan is a mixture of five different medicinal plants, widely prescribed for the treatment of mental illness. For acute toxicity, the Karber arithmetic method for the calculation of LD50 and Hodge and Sterner toxicity scale was used. In this study, different doses (10, 100, 285, 500, 1000, 5000 and 10,000 mg/kg) of the extract was administered orally to the different groups of rats and mice. Signs of toxicity and possible death of animals were monitored for 24 hrs to calculate the median lethal dose (LD50) of somina. At the end of the study, all the animals in all the dose groups were sacrificed and the internal organ-body was compared with values from the control group. The LD50 was found to be >10,000 mg/kg body weight upon oral administration in mice and rats as no mortality was observed after single dose administration. According to Hodge and Sterner toxicity scale, the obtained value of LD 50 > 10,000 mg/kg classified the Somina as Practically non-toxic herbal medicine. 展开更多

Water as a Standard Substance of a Logarithmic Poison Scale

The lethal dose LD<sub>50</sub> represents the most important experimental value for acute toxicity. The simple logarithmic calculation of -log<sub>10</sub> LD<sub>50</sub> = value leads to the possible poison power pLD. As with the pH or pK value, respectively, for acid or the scale of earthquake intensities the logarithm helps making large differences of orders of magnitude easier to understand since they are more comparable. The higher the pLD value, the higher is the power of poison. An increase of the pLD value by 1 stands for a tenfold increase in toxicity. The lethal acute dose for water, one of the most important and at the same time non-toxic substances of all, is about one tenth of the body weight. This leads to a possible pLD value for water of 1, an ideal starting value for a logarithmic poison scale. 展开更多

鳗鲡爱德华氏菌对美洲鳗鲡的致病性与宿主抗感染相关基因的表达

采用1.0×10^(7)~2.0×10^(8)cfu/mL的鳗鲡爱德华氏菌感染美洲鳗鲡以确定其半数致死量(LD_(50)),然后采用略低于LD_(50)的菌浓度(3.3×10^(6)cfu/尾)感染鳗鲡,并于感染后12、24、36、48、60、72 h等6个时间点检测鳗鲡肝脏... 采用1.0×10^(7)~2.0×10^(8)cfu/mL的鳗鲡爱德华氏菌感染美洲鳗鲡以确定其半数致死量(LD_(50)),然后采用略低于LD_(50)的菌浓度(3.3×10^(6)cfu/尾)感染鳗鲡,并于感染后12、24、36、48、60、72 h等6个时间点检测鳗鲡肝脏、肾脏和血液的载菌量,同时观察感染后36、60 h鳗鲡肝脏、肾脏和脾脏的病理变化。此外,采用qRT-PCR方法探究感染前和感染后12、36、60 h等4个时间点细菌外膜蛋白A(OmpA)和宿主抗鳗鲡爱德华氏菌感染的16个免疫相关基因表达。结果表明:感染鳗鲡爱德华氏菌后,美洲鳗鲡的LD_(50)为2.5×10^(5)cfu/g(5.0×10^(6)cfu/尾);美洲鳗鲡肾脏的载菌量于感染后36、48、60 h均显著高于在肝脏和血液中的载菌量,且感染后36 h菌浓度在肾脏和血液中均达到峰值,而在肝脏中的峰值则为感染后72 h。感染后36 h鳗鲡肝脏血管充血,肾小球萎缩且肾小管上皮细胞脱落,脾脏血管内形成血栓;感染后60 h肝血管出现血栓且部分肝细胞坏死,肾小管上皮细胞空泡变性并脱落入肾小管管腔,脾脏出现大量因细菌感染而导致的坏死性结节。与感染前相比,感染后12 h细菌OmpA基因在肝脏的表达量显著(P<0.05)升高;感染后OmpA基因在脾脏的表达量先下降后急剧上升,在肾脏的表达量则先上升后下降再上升。鳗鲡抗爱德华氏菌感染的5个免疫球蛋白相关的基因,在肝脏和肾脏不同时间点的表达量均无明显差异,而感染后早期在脾脏的表达量显著高于晚期;感染后3个肿瘤坏死因子相关基因在早期的表达量相对较高;此外,感染后12 h多个免疫相关基因表达量显著上升。 展开更多

多纹钱蝶鱼源海豚链球菌的分离鉴定及灭活疫苗的免疫效果

为探究养殖的多纹钱蝶鱼(Selenotoca multifasciata)病害频发的原因,建立有效的病害防控方法,对暴发于海南养殖场的多纹钱蝶鱼病害开展调查并进行灭活疫苗的研制及免疫保护效果评估。分离到2株病原菌LSSM和DFSM,经形态学和16S rDNA序列... 为探究养殖的多纹钱蝶鱼(Selenotoca multifasciata)病害频发的原因,建立有效的病害防控方法,对暴发于海南养殖场的多纹钱蝶鱼病害开展调查并进行灭活疫苗的研制及免疫保护效果评估。分离到2株病原菌LSSM和DFSM,经形态学和16S rDNA序列分析,将病原鉴定为海豚链球菌(Streptococcus iniae)。将这2株菌与实验室收集保藏的2株鱼源海豚链球菌(菌株GXTO和GXAS)进行了血清型鉴定和耐药性、致病性分析,制备了灭活疫苗并评价了疫苗的免疫保护效果。研究表明:4株海豚链球菌鉴定为血清Ⅰ型。所有菌株均对青霉素类、头孢类、四环素等23种抗生素高度敏感、对丁胺卡那耐药,其中海南株LSSM、DFSM对庆大霉素和卡那霉素抗生素耐药,广西株GXTO和GXAS对庆大霉素和卡那霉素敏感。DFSM、GXTO和LSSM对多纹钱蝶鱼的半数致死量(LD_(50))分别为7.20×10^(2)、2.72×10^(3)和3.40×10^(6) CFU/尾,GXAS未显示毒力。分别制备3株毒株(DFSM、GXTO和LSSM)的灭活疫苗,通过腹腔注射途径免疫多纹钱蝶鱼,各组免疫鱼在同源菌株攻毒后,获得的免疫保护率分别为84.6%~85.7%、69.2%~71.4%和33.3%~40.0%;此外,DFSM免疫鱼在异源毒株LSSM和GXTO攻毒后,获得的交叉免疫保护率分别为86.7%和100%。本研究确定了海豚链球菌是海南养殖的多纹钱蝶鱼病害频发的病原,使用DFSM株制备的海豚链球菌灭活疫苗对多纹钱蝶鱼有良好的免疫保护效果。 展开更多

In order to prevent severe pollution by de-icing salt on greenery along urban roads, a half lethal dose (LD_50)for a plant population was confirmed through stress simulation of chloride de-icing salt on Euonymus japonicus, with an ianalysis of physiological changes, statistics on mortality rate on plant populations and mathematic modeling during a 30- day subacute toxicity test. The results indicate that a significant positive correlation in the early stages and a significant negative correlation in the later stages were observed between the amount of chlorophyll a and b in plants and a cumulative dose of de-icing salt. The amounts of free proline in plants and the dose of de-icing salt were positively correlated Over the entire period. No significant correlation in the initial stage, but a significant negative correlation in later stages was observed between the soluble protein and the dose of de-icing salt. LDs0 of this chloride agent on E. japonicus is 5 kg.(L·m2)-1 over 30 days. 展开更多

Dose-response studies in arthropod research usually involve observing and collecting successive information at different times on the same group of insects exposed to different concentrations of stimulus. When the same measure is collected repeatedly over time, the data become correlated and Probit Analysis technique which is the standard method in analyzing bioassay experiments data cannot be used. Lethal time is estimated when the speed of kill is of interest since mortality varies over time. We evaluate a complementary approach, repeated measures logistic regression using Generalized Estimating Equations (GEE), for lethal time determination in mosquito dose response. Mortality data from anopheles larva exposed to 3 botanical extracts (B,C,E) at 4 concentration levels: 500 mg/ml, 250 mg/ml, 50 mg/ml and 12.5 mg/ml were used. The result shows the estimated LT50 values with concentration 500 mg/ml being the most virulent chemical for extract B (LT50 = 10.3 hrs), C (LT50 = 7.2 hrs) and E (LT50 = 10.3 hrs). The least virulent chemical was concentration 12.5 mg/ml for extract B (LT50 = 52.1 hrs), C (LT50 = 70.7 hrs) and E (LT50 = 55.0 hrs). We conclude that repeated measures of logistic regression via GEE can be used as a tool to estimate LT50 more effectively in repeated measures of arthropod data. 展开更多

Predictive formulas expressing relationship between dose rate and survival time in total body irradiation in mice

The Gompertz model is the long-time well-known mathematical model of exponential expression among mortality models in the literature that are used to describe mortality and survival data of a population. The death rate of the “probacent” model developed by the author based on animal experiments, clinical applications and mathematical reasoning was applied to predict age-specific death rates in the US elderly population, 2001, and to express a relationship among dose rate, duration of exposure and mortality probability in total body irradiation in humans. The results of both studies revealed a remarkable agreement between “probacent”-formula-predicted and published-reported values of death rates in the US elderly population or mortality probabilities in total body irradiation in humans (p - value > 0.995 in χ2 test in each study). In this study, both the Gompertz and “probacent” models are applied to the Sacher’s comprehensive experimental data on survival times of mice daily exposed to various doses of total body irradiation until death occurs with an assumption that each of both models is applicable to the data. The purpose of this study is to construct general formulas expressing relationship between dose rate and survival time in total body irradiation in mice. In addition, it is attempted to test which model better fits the reported data. The results of the comparative study revealed that the “probacent” model not only fit the Sacher’s reported data but also remarkably better fit the reported data than the Gompertz model. The “probacent” model might be hopefully helpful in research in human tolerance to low dose rates for long durations of exposure in total body irradiation, and further in research in a variety of biomedical phenomena. 展开更多

哈维氏弧菌对泥蚶的毒性及半致死浓度研究

目的探讨不同剂量的柑橘类多甲氧基黄酮对小鼠的急性毒性。方法分别给予氯化钠溶液和多甲氧基黄酮灌胃给药后,观察小鼠的活动、饮食、饮水、体质量、死亡等情况,用加权回归法(weighted regression method,Bliss)计算半数致死量(median l... 目的探讨不同剂量的柑橘类多甲氧基黄酮对小鼠的急性毒性。方法分别给予氯化钠溶液和多甲氧基黄酮灌胃给药后,观察小鼠的活动、饮食、饮水、体质量、死亡等情况,用加权回归法(weighted regression method,Bliss)计算半数致死量(median lethal dose,LD_(50))及95%置信区间。结果多甲氧基黄酮对小鼠的毒性实验结果表明多甲氧基黄酮对存活小鼠饮食、饮水及体质量无明显影响。通过Bliss法计算得多甲氧基黄酮的LD_(50)为3.561 g/kg,表现出一定的毒性,LD_(50)的95%置信区间为2.258~6.451 g/kg;回归方程为Y(Probit)=-1.034+1.874 lgD。结论多甲氧基黄酮具有一定的毒性,且脏器指数与病理观察显示毒性主要表现在血液血管方面,主要作用在肝、脾和肾脏,其毒性与给药剂量相关,在一定剂量范围内可安全使用。 展开更多

【目的】为探讨^(60)Co-γ射线对北美红花七叶树种子的诱变效应并确定其辐射半致死剂量。【方法】以北美红花七叶树种子为试材,采用不同辐照剂量(0、100、200、300、400 Gy)^(60)Co-γ进行辐射处理,分析辐射对北美红花七叶树种子出苗及... 【目的】为探讨^(60)Co-γ射线对北美红花七叶树种子的诱变效应并确定其辐射半致死剂量。【方法】以北美红花七叶树种子为试材,采用不同辐照剂量(0、100、200、300、400 Gy)^(60)Co-γ进行辐射处理,分析辐射对北美红花七叶树种子出苗及幼苗生长的影响。【结果】辐照剂量与种子相对出苗率为极显著负相关,种子辐射半致死剂量为217.76 Gy。辐射后幼苗出现叶型改变、叶面扭曲、叶边缘变异等,300 Gy剂量下叶型和叶边缘变异比例最高。辐射处理的北美红花七叶树生长性状与对照差异不显著。随着^(60)Co-γ射线辐照剂量的增加,抗氧化酶(SOD、POD、CAT)活性,叶绿素及类胡萝卜素含量均呈现先升高后下降的趋势,抗氧化酶活性在300 Gy最高。相关性分析得出,辐照剂量与复叶宽、小叶宽、可溶性蛋白、相对电导率存在显著相关性。【结论】综合种子半致死剂量和生长、生理指标,确定北美红花七叶树种子适宜的辐照剂量为200~300 Gy。可为七叶树属植物辐射育种提供科学参考。 展开更多

硼酸钠对小鼠的急性毒性实验研究

目的探讨硼酸钠单次经口给药对小鼠的急性毒性。方法根据预实验结果将C3H/HeN小鼠分为2.00、2.40、2.88、3.46、4.15、4.98和5.97 mg/g(Bliss法)7个硼酸钠实验组,对照组给予蒸馏水。灌胃均为最大给药容积,1 d内经口给药1次。观察14 d,... 目的探讨硼酸钠单次经口给药对小鼠的急性毒性。方法根据预实验结果将C3H/HeN小鼠分为2.00、2.40、2.88、3.46、4.15、4.98和5.97 mg/g(Bliss法)7个硼酸钠实验组,对照组给予蒸馏水。灌胃均为最大给药容积,1 d内经口给药1次。观察14 d,记录小鼠的临床状态,死亡率和体重变化,并进行器官指标评价和病理检查。结果实验组小鼠表现出蜷卧少动、气短、毛发松散和粪便不成形,部分小鼠在1 h内死亡。解剖可见胃肠胀气,其他脏器未见异常。组织病理学结果显示脑、心、脾未见异常变化;肝细胞水肿、中央静脉扩张、瘀血;肺泡上皮结构正常,肺泡间隔毛细血管扩张外渗,淋巴细胞浸润,少量红细胞从肺泡腔漏出;肾小球及肾小管间质瘀血。硼酸钠雄性和雌性小鼠的LD50分别为3.794和3.646 mg/g。结论硼酸钠属于低毒化合物;急性硼酸钠中毒对小鼠肝脏、肺脏和肾脏有明显的损伤和毒性作用。 展开更多

叠氮化钠对燕麦种子萌发及幼苗生长的影响

【目的】明确不同NaN_(3)浓度及处理时间对燕麦种子萌发及幼苗生长的影响。【方法】筛选半致死剂量,以2个燕麦品种爱沃和青永久709子为材料,设置不同NaN_(3)浓度(0、5、10、15、20 mmol/L)和处理时间(1、2、3 h),测定种子萌发活力及幼... 【目的】明确不同NaN_(3)浓度及处理时间对燕麦种子萌发及幼苗生长的影响。【方法】筛选半致死剂量,以2个燕麦品种爱沃和青永久709子为材料,设置不同NaN_(3)浓度(0、5、10、15、20 mmol/L)和处理时间(1、2、3 h),测定种子萌发活力及幼苗生长指标。【结果】NaN_(3)浓度是影响燕麦种子萌发及幼苗生长的最主要因子。燕麦种子的相对发芽率、相对发芽势及相对发芽指数随NaN_(3)浓度的升高而显著下降,当NaN_(3)浓度为10 mmol/L、处理时间为1 h时,青永久709的相对发芽率为51.08%、相对致死率为47.92%;同一浓度下处理2 h时,爱沃的相对发芽率为51.47%、相对致死率为48.53%。NaN_(3)浓度超过10 mmol/L,两品种的相对致死率均大于60%,最高浓度(20 mmol/L)下种子完全丧失发芽能力。NaN_(3)处理还显著抑制了幼苗生长,同一条件下青永久709的出苗率、根长和苗高受到抑制的程度重于爱沃。【结论】青永久709和爱沃的半致死剂量分别为10 mmol/L处理1 h和10 mmol/L处理2 h,青永久709对NaN_(3)处理比爱沃更敏感。 展开更多

目的探究抗高血压1类新药脯氨酸丹参素冰片酯的急性毒性,评价其安全性。方法24只小鼠,雌雄各半,随机分为溶剂对照组和给药组,根据脯氨酸丹参素冰片酯溶液的最大浓度(150 mg·mL^(-1)),单次灌胃,连续观察7 d,记录小鼠毒性反应、体重... 目的探究抗高血压1类新药脯氨酸丹参素冰片酯的急性毒性,评价其安全性。方法24只小鼠,雌雄各半,随机分为溶剂对照组和给药组,根据脯氨酸丹参素冰片酯溶液的最大浓度(150 mg·mL^(-1)),单次灌胃,连续观察7 d,记录小鼠毒性反应、体重及饮食饮水量,7 d后小鼠处死,观察主要脏器变化,测定心、肝、肾、肺脏等主要脏器指数,HE染色观察脏器病理性改变。结果实验过程中,脯氨酸丹参素冰片酯给药后小鼠均未出现死亡。与溶剂对照组相比,脯氨酸丹参素冰片酯给药后7 d内,小鼠活动、体重、饮食饮水量均无显著性差异。给药组肝、肾、肺脏指数与对照组相比无显著差异(P>0.05)。与溶剂对照组小鼠相比,雄性小鼠给药组心脏指数无显著差异(P>0.05),而雌性小鼠给药组的心脏指数具有显著降低(P<0.05),且心肌细胞排列紊乱。结论测得脯氨酸丹参素冰片酯对小鼠灌胃的最大耐受量为3000 mg·kg^(-1)。脯氨酸丹参素冰片酯毒性小,安全性较好。 展开更多
