排毒消疹方联合西药治疗恶性肿瘤EGFRIs/PD-1相关性皮疹临床研究

目的:观察排毒消疹方联合西药治疗恶性肿瘤表皮生长因子受体通路抑制剂(EGFRIs)/程序性细胞死亡蛋白-1(PD-1)相关性皮疹热毒入营证的临床疗效。方法:选取52例接受EGFRIs/PD-1治疗后出现皮疹的恶性肿瘤热毒入营证患者,按随机数字表法分... 目的:观察排毒消疹方联合西药治疗恶性肿瘤表皮生长因子受体通路抑制剂(EGFRIs)/程序性细胞死亡蛋白-1(PD-1)相关性皮疹热毒入营证的临床疗效。方法:选取52例接受EGFRIs/PD-1治疗后出现皮疹的恶性肿瘤热毒入营证患者,按随机数字表法分为对照组与治疗组各26例。对照组给予枸地氯雷他定片治疗,治疗组在对照组基础上给予排毒消疹方治疗,2组均治疗14 d。比较2组临床疗效、症状总分、皮疹严重程度及生活质量。结果:治疗14 d,治疗组总有效率88.46%,高于对照组61.54%(P<0.05)。2组症状总分时间效应、组间效应及交互效应差异均有统计学意义(P<0.05)。2组治疗7 d的症状总分均较治疗前降低,2组治疗14 d的症状总分均较治疗前、治疗7 d降低,治疗组症状总分均低于同期对照组,差异均有统计学意义(P<0.05)。治疗14 d,2组皮疹严重程度分级均较治疗前改善,治疗组皮疹严重程度分级较对照组改善更明显,差异均有统计学意义(P<0.05)。治疗14 d,2组皮肤病生活质量指数(DLQI)评分均较治疗前降低,治疗组DLQI评分低于对照组,差异均有统计学意义(P<0.05)。结论:排毒消疹方联合西药治疗恶性肿瘤EGFRIs/PD-1相关性皮疹热毒入营证,可减轻患者的皮疹严重程度,提高其生活质量。 展开更多

To expand the evidence base to inform future public policy aimed at accident prevention, we investigated the impact of different categories of injury on average life expectancy in China. We used data from the National Death Cause Registration Information System and National Maternity and Children Health Surveillance databases, as well as 2010 population data from the National Bureau of Statistics. We then calculated the average life expectancy of the Chinese population, in addition to life expectancy after eliminating injury-related mortality. The average life expectancy of the Chinese population in 2010 was 74.93 years. After eliminating deaths due to injuries, the fourth leading cause of mortality in China, average life expectancy increased by 1.36 years. When this was broken down by population sub-groups, these gains were 1.76 and 0.79 years in men and women, 0.94 and 1.56 years in urban and rural residents, and 1.11, 1.30, and 1.67 years for residents in the Eastern, Central and Western regions respectively. After eliminating all categories of injury, the average life expectancy of the Chinese population was found to increase by 1.36 years. 目的 探讨仑伐替尼、程序性死亡受体-1(PD-1)抑制剂联合经导管动脉栓塞化疗(TACE)治疗不可切除肝细胞癌(uHCC)的临床疗效。方法 选取安徽省阜阳市阜南县人民医院2020年7月至2023年7月收治的uHCC患者106例,按治疗方案的不同分为对照组(32... 目的 探讨仑伐替尼、程序性死亡受体-1(PD-1)抑制剂联合经导管动脉栓塞化疗(TACE)治疗不可切除肝细胞癌(uHCC)的临床疗效。方法 选取安徽省阜阳市阜南县人民医院2020年7月至2023年7月收治的uHCC患者106例,按治疗方案的不同分为对照组(32例)、药物组(34例)和联合组(40例),分别予TACE、TACE+PD-1抑制剂、TACE+PD-1抑制剂+仑伐替尼方案治疗,其中PD-1抑制剂为信迪利单抗注射液、替雷利珠单抗注射液、注射用卡瑞利珠单抗中三选一。3组治疗周期均为6周。结果 联合组疾病控制率为90.00%,显著高于对照组的65.63%和药物组的70.59%(P <0.05)。联合组患者治疗后的丙氨酸氨基转移酶、天门冬氨酸氨基转移酶、总胆红素、甲胎蛋白、癌胚抗原、α-L-岩藻糖苷酶、糖类抗原199水平及生活质量核心量表评分均显著低于对照组及药物组(P <0.05)。3组不良反应发生率无显著差异(P> 0.05)。结论 仑伐替尼、PD-1抑制剂联合TACE治疗uHCC,可有效改善患者的肝功能,降低血清肿瘤标志物水平,提高患者预后的生活质量。 展开更多

Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) is a bacterial pathogen of tomato and of the model plants Arabidopsis and Nicotiana benthamiana (N. benthamiana). Like numerous Gram-negative bacterial pathogens of animals and plants, Pst DC3000 exploits the conserved type III secretion system (TTSS) to deliver multiple virulence effectors directly into the host cells. Type III effectors (T3Es) collectively participate in causing disease, by mechanisms that are not well clarity. Elucidating the virulence function of individual effector is fundamental for understanding bacterial infection of plants. Here, we focused on studying one of these effectors, HopAA1-1, and analyzed its potential function and subcellular localization in N. benthamiana. Using an Agrobacterium-mediated transient expression system, we found that HopAA1-1 can trigger domain-dependent cell death in N. benthamiana. The observation using confocal microscopy showed that the YFP-tagged HopAA1-1 localizes to diverse cellular components containing nucleus, cytoplasm and cell membrane, which was demonstrated through immunoblot analysis of membrane fractionation and nuclear separation. Enforced HopAA1-1 subcellular localization, by tagging with a nuclear localization sequence (NLS) or a nuclear export sequence (NES), shows that HopAA1-1-induced cell death in N. benthamiana is suppressed in the nucleus but enhanced in the cytoplasm. The development of aqueous battery with dual mechanisms is now arousing more and more interest.The dual mechanisms of Zn^(2+)(de)intercalation and I^(-)/I_(2)redox bring unexpected effects.Herein,differing from previous studies using Zn I_(2)additive,this work designs an aqueous Bi I_(3)-Zn battery with selfsupplied I^(-).Ex situ tests reveal the conversion of Bi I_(3)into Bi(discharge)and Bi OI(charge)at the 1st cycle and the dissolved I^(-)in electrolyte.The active I^(-)species enhances the specific capacity and discharge medium voltage of electrode as well as improves the generation of Zn dendrite and by-product.Furthermore,the porous hard carbon is introduced to enhance the electronic/ionic conductivity and adsorb iodine species,proven by experimental and theoretical studies.Accordingly,the well-designed Bi I_(3)-Zn battery delivers a high reversible capacity of 182 m A h g^(-1)at 0.2 A g^(-1),an excellent rate capability with 88 m A h g^(-1)at 10 A g^(-1),and an impressive cyclability with 63%capacity retention over 20 K cycles at 10 A g^(-1).An excellent electrochemical performance is obtained even at a high mass loading of 6 mg cm^(-2).Moreover,a flexible quasi-solid-state Bi I_(3)-Zn battery exhibits satisfactory battery performances.This work provides a new idea for designing high-performance aqueous battery with dual mechanisms. 展开更多

中国人口普查死亡数据质量评估及2010-2020年死亡模式转变分析 被引量:3

人口的预期寿命和年龄死亡模式是评估人口健康水平、掌握人口发展规律的重要指标。人口普查为掌握年龄死亡模式的变动特点和人口预期寿命水平提供了基础数据,然而根据以往研究文献和对2020年人口普查数据的分析,人口普查所获得的死亡数... 人口的预期寿命和年龄死亡模式是评估人口健康水平、掌握人口发展规律的重要指标。人口普查为掌握年龄死亡模式的变动特点和人口预期寿命水平提供了基础数据,然而根据以往研究文献和对2020年人口普查数据的分析,人口普查所获得的死亡数据存在一定程度的漏报,需要对这一数据质量进行评估和修正。为此,本文根据不同年龄段的死亡特点对布拉斯-罗吉特模型进行适应性调整,使其符合观察年份的死亡模式,获得模型参数,进而实现对2010-2020年死亡数据的校正评估。结果显示2020年人口普查获得的死亡数据中低龄组男性漏报率高于女性,0-9岁男性平均漏报率为75.33%,女性为57.69%,男婴死亡漏报率为85.08%,女婴死亡漏报率为77.80%;70-90岁高龄组死亡漏报率男性平均为22.55%,女性为20.75%,90岁及以上的死亡漏报比较低;老年阶段男性漏报率普遍高于女性。2010年婴儿死亡概率为21.15‰,2020年为8.80‰;2010年出生预期寿命为74.82岁,2020年为78.21岁,10年间预期寿命增长了3.39岁。通过比较分析发现21世纪以来我国人口年龄死亡模式呈现“低龄降低,中龄平稳,中高龄先缓升后陡升,高龄略有回降”的变动态势。这表明进入老龄前期人口死亡概率降低带来一个虚弱累积的过程,达到一定年龄(70-80岁)后,老化速度就会加快虚弱累积的效应,带来更高年龄组失能或死亡概率的加速增长。 展开更多

In this paper, ultrasonic (20 kHz) fatigue tests were performed on specimens of a high-strength steel in very high cycle fatigue (VHCF) regime. Experimental results showed that for most tested specimens failed in a VHCF regime, a fatigue crack originated from the interior of specimen with a fish-eye pattern, which contained a fine granular area (FGA) centered by an inclusion as the crack origin. Then, a two-parameter model is proposed to predict the fatigue life of high-strength steels with fish-eye mode failure in a VHCF regime, which takes into account the inclusion size and the FGA size. The model was verified by the data of present experiments and those in the literature. Furthermore, an analytic formula was obtained for estimating the equivalent crack growth rate within the FGA. The results also indicated that the stress intensity factor range at the front of the FGA varies within a small range, which is irrespective of stress amplitude and fatigue life. 展开更多

BACKGROUND Anti-programmed death therapy has thrust immunotherapy into the spotlight.However,such therapy has a modest response in hepatocellular carcinoma(HCC).Epigenetic immunomodulation is a suggestive combinatorial therapy with immune checkpoint blockade.Non-coding ribonucleic acid(ncRNA)driven regulation is a major mechanism of epigenetic modulation.Given the wide range of ncRNAs that co-opt in programmed cell-death protein 1(PD-1)/programmed death ligand 1(PD-L1)regulation,and based on the literature,we hypothesized that miR-155-5p,miR-194-5p and long non-coding RNAs(lncRNAs)X-inactive specific transcript(XIST)and MALAT-1 are involved in a regulatory upstream pathway for PD-1/PD-L1.Recently,nutraceutical therapeutics in cancers have received increasing attention.Thus,it is interesting to study the impact of oleuropein on the respective study key players.AIM To explore potential upstream regulatory ncRNAs for the immune checkpoint PD-1/PD-L1.METHODS Bioinformatics tools including microrna.org and lnCeDB software were adopted to detect targeting of miR-155-5p,miR-194-5p and lncRNAs XIST and MALAT-1 to PD-L1 mRNA,respectively.In addition,Diana tool was used to predict targeting of both aforementioned miRNAs to lncRNAs XIST and MALAT-1.HCC and normal tissue samples were collected for scanning of PD-L1,XIST and MALAT-1 expression.To study the interaction among miR-155-5p,miR-194-5p,lncRNAs XIST and MALAT-1,as well as PD-L1 mRNA,a series of transfections of the Huh-7 cell line was carried out.RESULTS Bioinformatics software predicted that miR-155-5p and miR-194-5p can target PDL1,MALAT-1 and XIST.MALAT-1 and XIST were predicted to target PD-L1 mRNA.PD-L1 and XIST were significantly upregulated in 23 HCC biopsies compared to healthy controls;however,MALAT-1 was barely detected.MiR-194 induced expression elevated the expression of PD-L1,XIST and MALAT-1.However,overexpression of miR-155-5p induced the upregulation of PD-L1 and XIST,while it had a negative impact on MALAT-1 expression.Knockdown of XIST did have an impact on PD-L1 expression;however,following knockdown of the negative regulator of X-inactive specific transcript(TSIX),PD-L1 expression was elevated,and abolished MALAT-1 activity.Upon co-transfection of miR-194-5p with siMALAT-1,PD-L1 expression was elevated.Co-transfection of miR-194-5p with siXIST did not have an impact on PD-L1 expression.Upon co-transfection of miR-194 with siTSIX,PD-L1 expression was upregulated.Interestingly,the same PD-L1 expression pattern was observed following miR-155-5p cotransfections.Oleuropein treatment of Huh-7 cells reduced the expression profile of PD-L1,XIST,and miR-155-5p,upregulated the expression of miR-194-5p and had no significant impact on the MALAT-1 expression profile.CONCLUSION This study reported a novel finding revealing that opposing acting miRNAs in HCC,have the same impact on PD-1/PD-L1 immune checkpoint by sharing a common signaling pathway. 展开更多

Based on the theory of continuum damage mechanics,a bi-variable damage mechanics model is developed,which,according to thermodynamics,is accessible to derivation of damage driving force,damage evolution equation and damage evolution criteria. Furthermore,damage evolution equations of time rate are established by the generalized Drucker's postulate. The damage evolution equation of cycle rate is obtained by integrating the time damage evolution equations,and the fatigue life prediction method for smooth specimens under repeated loading with constant strain amplitude is constructed. Likewise,for notched specimens under the repeated loading with constant strain amplitude,the fatigue life prediction method is obtained on the ground of the theory of conservative integral in damage mechanics. Thus,the material parameters in the damage evolution equation can be obtained by reference to the fatigue test results of standard specimens with stress concentration factor equal to 1,2 and 3. 展开更多

On the contrary, recently it has been described that RecA promotes cellular apoptotic-like death (ALD), a pathway of programmed cellular death (PCD). In fact, RecA has been dubbed as the major apoptotic executor in E. coli. From these studies, RecA emerges as a prototypical Gin/Gan protein that despite of their intrinsic vital and lethal anfi-funcionality becomes in a WISE factor: a Worker to Integrate Survival and Evolution in E. coli evolving populations living in community. Here, I provide a review of recent experimental and conceptual advances trying to understand these RecA’s antagonistic roles in appearance contradictory under a unified biological vision. 展开更多

目的探究程序性死亡因子-1(PD-1)单抗联合安罗替尼治疗晚期肺癌的病灶局部控制率及对患者营养状况和生活质量的影响。方法选择2019年1月至2021年12月于我院接受治疗的91例晚期肺癌患者为研究对象,按照随机数字表法结合患者意愿的方式将... 目的探究程序性死亡因子-1(PD-1)单抗联合安罗替尼治疗晚期肺癌的病灶局部控制率及对患者营养状况和生活质量的影响。方法选择2019年1月至2021年12月于我院接受治疗的91例晚期肺癌患者为研究对象,按照随机数字表法结合患者意愿的方式将其区分为研究组(51例,接受PD-1联合安罗替尼治疗)和对照组(40例,接受安罗替尼治疗),2组患者均连续治疗4个周期,对比2组患者治疗后病灶局部控制率差异,随访比较2组患者治疗中营养状况及生活质量的变化,并统计不良反应发生情况。结果研究组患者的客观有效率(ORR)和疾病控制率(DCR)分别为66.7%(34/41)和80.4%(41/51),明显高于对照组患者的45.0%(18/40)和60.0%(24/40),差异具有统计学意义(P<0.05);治疗前2组患者的营养指标、生活质量评分组间差异均无统计学意义(P>0.05),治疗后研究组患者BMI指数、血清总蛋白及白蛋白水平高于对照组(P<0.05),FACT-L量表各维度及总分高于对照组(P<0.05);统计2组患者各类不良反应发生率组间差异无统计学意义(45.1%和45.0%,P>0.05)。结论对晚期肺癌患者开展PD-1单抗联合安罗替尼治疗效果肯定,有助于提高患者病灶局部控制率,同时还可以改善患者的营养指标与生活质量,具有一定的推广应用价值。 展开更多
