‘Dianyinhua 3’is bred through the selection of a good single variant plant based on Hunan Lonicerae Flos variety‘Xianglei 1’as the basic material.The scions of the variant plant were grafted and propagated with wi... ‘Dianyinhua 3’is bred through the selection of a good single variant plant based on Hunan Lonicerae Flos variety‘Xianglei 1’as the basic material.The scions of the variant plant were grafted and propagated with wild Lonicera macranthoides Hand.-Mazz.as rootstocks.This variety belongs to L.macaroides,which has the characteristics of early flowering,large and numerous buds,concentrated flowering and abundant yield.The content of chlorogenic acid is as high as 6%,and the content of luteolin is 0.017%.It matures from late May to early June in Yunnan.The buds are rod-shaped,slightly curved,4.6-5.8 cm long,with a diameter of 6.47 mm at the top and 2.68 mm at the bottom,and the petals have a thickness of 0.33 mm.The thousand-flower bud weight was 15.8-18.2 g,and the average yield of dried flowers per plant was 1360-1680 g in the peak period,and the yield was 4887 kg/hm^2.It is suitable for the introduction and trial planting in Xiangyang Mountainous Area or rocky desertification mountainous area in Yunnan,with an altitude lower than 2200 m or in areas with similar climates such as Sichuan,Guizhou and Guangxi. 展开更多

Dietary supplementation of Lonicera macranthoides leaf powder improves amino acid profiles in serum and longissimus thoracis muscle of growing-finishing pigs 被引量:8

A 60-days feeding trial was conducted to determine the effect of dietary supplementation of Lonicera macranthoides leaf(LML) on growth performance, meat quality, and free amino acid profiles in growingfinishing pigs. L. macranthoides leaves were harvested, air-dried and milled to powder. A total of 288 crossbred barrows(Large White x Landrace) with an average initial BW of 54.0 ± 3.4 kg were randomly allocated to 3 treatment groups and fed either a basal diet or a diet supplemented with 0.5% or 1.0% LML powder. Results showed that diet supplemented with LML powder did not affect average daily feed intake(ADFI) and feed: gain ratio(P> 0.05) in growing-finishing pigs. Moreover, diet supplemented with LML powder showed no significant effects on carcass traits(P > 0.05) including dressing percentage,backfat thickness and loin muscle area, and meat quality traits(P > 0.05) including meat color, intramuscular fat and cooking loss. However, diet supplemented with LML powder significantly improved free amino acid profiles in serum and longissimus thoracis muscle in growing-finishing pigs. Most importantly, dietary LML powder increased total free amino acids content(P < 0.001) and essential free amino acid content(P < 0.001) in longissimus thoracis muscle. These results indicate that LML has the potential to improve the nutritional value of meat through improving free amino acid profiles. 展开更多

忍冬花中一种RG-I多糖的抗血管生成活性研究

目的本研究对忍冬花中均一多糖结构和抗新生血管生成活性进行研究,为忍冬的活性物质基础研究提供新的理论数据。方法水提醇沉并结合阴离子交换色谱和凝胶渗透色谱法获得忍冬均一多糖LF-02-2,并通过分子量检测、单糖组成分析、糖残基连... 目的本研究对忍冬花中均一多糖结构和抗新生血管生成活性进行研究,为忍冬的活性物质基础研究提供新的理论数据。方法水提醇沉并结合阴离子交换色谱和凝胶渗透色谱法获得忍冬均一多糖LF-02-2,并通过分子量检测、单糖组成分析、糖残基连接方式分析、部分酸水解、糖醛酸还原结合核磁共振数据推导出LF-02-2的结构。同时,运用人微血管内皮细胞管腔形成实验测定其体外抗新生血管活性。结果对LF-02-2的结构解析表明,均一多糖LF-02-2的重均分子量为74.1 kDa,其单糖组成由鼠李糖(Rha)、半乳糖(Gal)、半乳糖醛酸(GalA)和阿拉伯糖(Ara)组成,摩尔比分别为10.43∶14.94∶6.66∶67.97。其主链由1,4-连接的α-Galp A、1,2-连接的α-Rhap和1,2,4-连接的α-Rhap组成。分支由末端连接和1,4,6-连接的β-Galp,末端连接和1,5-连接的α-Araf连接,取代发生在1,2,4-连接的α-Rhap的O-4位。管腔形成实验结果表明LF-02-2能显著抑制人微血管内皮细胞(Human Microvascular Endothelial Cell,HMEC)的管腔形成。结论忍冬花中一种类RG-I型果胶多糖LF-02-2具有显著抗血管生成活性,具有开发为抗血管生成药物的潜力。 展开更多

灰毡毛忍冬(Lonicera macranthoides)为中药山银花的一种基原植物,在中国南方广泛分布,具有很高的药用价值和经济价值。本研究利用MISA工具对灰毡毛忍冬转录组74057条Unigenes中的SSR位点进行了分析,其中15587条Unigene共计含有SSR位点2... 灰毡毛忍冬(Lonicera macranthoides)为中药山银花的一种基原植物,在中国南方广泛分布,具有很高的药用价值和经济价值。本研究利用MISA工具对灰毡毛忍冬转录组74057条Unigenes中的SSR位点进行了分析,其中15587条Unigene共计含有SSR位点20161个,SSR发生频率为21.05%。灰毡毛忍冬转录组SSR位点的种类从单核苷酸至六核苷酸重复均有分布,主要重复类型是二核苷酸(9704个)和三核苷酸(5847个),分别占SSR总数的48.13%和29.00%,主要重复单元为AG/CT(6086)、AT/AT(3071)和A/T(2826),分别占SSR总数的30.19%、15.23%和14.02%。SSR位点的重复次数分布最多的是6和5次,分别有4897和3531个,分别占总SSR数的24.29%和17.51%。利用Primer 6.0软件设计了17611对SSR引物,随机选出30对引物进行扩增验证,其中14对扩增条带表现出多态性,进而利用这14对引物对6个灰毡毛忍冬品种开展了遗传多样性分析。本研究结果将为灰毡毛忍冬物种鉴定、遗传多样性分析和分子标记辅助育种等提供科学依据。 展开更多

绿原酸是灰毡毛忍冬生长发育过程中产生的重要的次级代谢产物,而CCoAOMT是绿原酸合成过程中的关键基因。为进一步揭示灰毡毛忍冬LmCCoAOMT基因的功能,本研究利用RACE技术克隆LmCCoAOMT全长基因,通过生物信息学进行分析,并在大肠杆菌中... 绿原酸是灰毡毛忍冬生长发育过程中产生的重要的次级代谢产物,而CCoAOMT是绿原酸合成过程中的关键基因。为进一步揭示灰毡毛忍冬LmCCoAOMT基因的功能,本研究利用RACE技术克隆LmCCoAOMT全长基因,通过生物信息学进行分析,并在大肠杆菌中表达该蛋白。此外,通过RT-qPCR和HPLC的方法研究CCoAOMT基因表达量和绿原酸的含量之间的相关性。结果显示LmCCoAOMT基因全长1 096 bp(GenBank:MN103348),ORF 735 bp,编码245个氨基酸。系统进化树分析发现LmCCoAOMT与藤本植物牵牛花的亲缘关系最近。生物信息学分析表明LmCCoAOMT的氨基酸序列不具有跨膜结构,其蛋白三级结构主要以α-螺旋和无规卷曲的形式存在。在大肠杆菌BL21 (DE3)中表达结果显示,其分子量为31.36 kD。RT-qPCR及HPLC结果表明,CCoAOMT基因在灰毡毛忍冬和金银花早期花中表达量最高,在末期表达量最低,且表达量与绿原酸含量有相关性。本研究为深入研究LmCCoAOMT基因功能及阐明灰毡毛忍冬绿原酸合成的分子机制提供了科学依据。 展开更多
