Lotus root polysaccharide inhibits the growth of ovarian cancer cells by blocking the cell cycle

Background:Lotus root polysaccharide is a natural antioxidant.As a new anticancer drug,it has anti-proliferation and pro-apoptotic effects in a variety of tumour cells,but its effect on ovarian cancer is not clear.In study,we attempted to elucidate the role and mechanism of lotus root polysaccharide in SKOV3 cells.Methods:In this study,the effect of lotus root polysaccharide on mRNA of SKVO3 cells was analyzed by RNA-seq,and verified by Western blot,flow cytometry,fluorescence detection and other techniques.Results:The results showed that lotus root polysaccharide could inhibit the proliferation of ovarian cancer cells.Then,a change in gene expression was found by RNA-seq.In the mRNA(differentially expressed mRNA)with these differences,significant changes in the cell cycle were found by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis.Subsequently,the proportion of cells in S phase decreases and G2/M phase increases,as seen with propidium iodide staining.Gene Set Enrichment Analysis showed inhibition of the cell cycle,and the gene and protein expression of CDK1,CCNA1 and CCNB1 were inhibited.Conclusion:Our results show that lotus root polysaccharide can inhibit the growth of SKOV3 cells in vitro by blocking the cell cycle at the G2/M phase,which reveals the potential of lotus root polysaccharide in the treatment of ovarian cancer. 展开更多

[Method] The Iotus root and Iemon were seIected as raw materials for preparing compound beverage. The Iotus root and Iemon were juiced respectiveIy. SubsequentIy, their juices were mixed together with a series of proportions so as to expIore a rational processing ratio of Iotus root and Iemon. [Result] The resuIts showed that the browning of Iotus root juice couId be effectiveIy prevented when the Iotus root juice was heated for 9 min and added with 0.8% citric acid. The optimized formuIa of compound beverage of Iotus root and Iemon was as foI ows: Iotus root juice to Iemon juice ratio, 5:1; addition amount of white granuIated sugar, 12%; addition amount of stabiIizer (CMC-Na), 0.15%. Af-ter moduIation, homogenization, steriIization and other processes, the compound beverage of Iotus root and Iemon was prepared. Bacterial cellulose/lotus root starch(BC/LRS) composites were prepared by cultivating Acetobacter xylinum in nutrient media containing gelatinized lotus root starch. Low concentrations of gelatinized LRS had increased BC production with the maximum value at 6.67 g/L when 5 g/L of LRS was added in the culture media and the composites had thicker and denser fibrils compared with those of BC with low concentrations of LRS(2.5 and 5 g/L). When the concentration of LRS was increased above 7.5 g/L, the morphology of the BC/LRS composites contained more fibril layers that were linked with LRS. The results from X-ray diffraction(XRD) demonstrated that there was no significant difference in structure between BC and BC/LRS composites except a slight increase in crystallinity for BC/LRS composites as the concentration of LRS was lifted up. The tensile tests were performed to display BC/LRS composites prepared with LRS concentration at 2.5 and 5 g/L in media had the tensile strength of 54 and 60 MPa, respectively, which indicated an improvement in mechanical property compared to the unmodified BC(45 MPa). Live/dead assay with chondrocytes seeded on BC/LRS composite revealed higher cell viability ranging from 85% to 90% than BC. Furthermore, cell morphology with typical spindle shape was observed on the surfaces of BC/LRS composite by confocal microscope. Through the overall results, it shows that this study has provided a guidance to prepare BC/LRS composites with better cell biocompatibility and higher mechanical strength than those of BC for the potential use in cartilage tissue engineering. 展开更多

A high incidence of bone defects and the limitation of autologous bone grafting require 3 D scaffolds for bone repair. Compared with synthetic materials, natural edible materials possess outstanding advantages in terms of biocompatibility, bioactivities and low manufacturing cost for bone tissue engineering. In this work, attracted by the natural porous/fabric structure, good biocompatibility and bioactivities of the lotus root, the lotus root-based scaffolds were fabricated and investigated their potential to serve as natural porous bone tissue engineering scaffolds. The results indicated that the lotus root-based scaffolds possess suitable natural microstructure, excellent biocompatibility and promising functions, such as antioxidant capacity and angiogenesis promotion. Remarkably, lotus root scaffolds showed encouraging possibility of bone tissue engineering while the mineralized lotus root could further improve the bone regeneration in vivo. All the results demonstrated the bone regeneration potential of lotus root-based scaffolds equipped with suitable natural architecture, excellent biocompatibility, specific bioactivities and low manufacturing cost. 展开更多
