背景与目的:全基因组相关联研究发现了染色体10q11区域的单核苷酸多态性(single nucleotide polymorphism,SNP)位点rs10993994与前列腺癌的遗传易感性相关,本研究探讨rs10993994影响前列腺癌发病风险的可能机制,即研究rs10993994对前列... 背景与目的:全基因组相关联研究发现了染色体10q11区域的单核苷酸多态性(single nucleotide polymorphism,SNP)位点rs10993994与前列腺癌的遗传易感性相关,本研究探讨rs10993994影响前列腺癌发病风险的可能机制,即研究rs10993994对前列腺癌细胞微精液蛋白β(microseminoprotein beta,MSMB)基因转录活性的影响。方法:化学合成MSMB基因的启动子序列,由于位于序列中间的SNP位点rs10993994有2种等位基因(T/C)可能,合成2条启动子MSMB promoter-T和MSMB promoter-C。将2条启动子通过酶切的方法导入荧光素酶报告基因载体(pGL3-basic vectors)中,筛选阳性克隆后,将携带启动子的质粒转染入前列腺癌细胞PC-3和LNCaP中,最后通过荧光检测仪检测重组质粒中启动子的转录活性。结果:成功合成2条启动子序列:MSMB promoter-T和MSMB promoter-C;分别将启动子序列与荧光素酶报告基因载体重组,形成重组质粒pGL3-MSMB promoter-T和pGL3-MSMB promoter-C。在前列腺癌细胞PC-3中,重组质粒MSMB promoter-C组的相对荧光度为2.27±0.39,显著高于重组质粒MSMB promoter-T组(0.57±0.13),差异有统计学意义(P<0.05);在前列腺癌细胞LNCaP中,重组质粒MSMB promoter-C组的相对荧光度为1.70±0.32,显著高于重组质粒MSMB promoter-T组(0.37±0.09),差异有统计学意义(P<0.05)。结论:重组质粒pGL3-MSMB promoter-C的转录活性强于重组质粒pGL3-MSMB promoter-T的转录活性;rs10993994可以影响到MSMB基因启动子的启动转录活性,rs10993994位点上等位基因胞嘧啶(C)较胸腺嘧啶(T)更能促使MSMB基因转录。 展开更多

微精原蛋白β(MSMB)对动物的繁殖功能具有重要的调控作用。采集成年健康母牦牛的卵巢组织,通过RT-PCR技术克隆牦牛的MSMB基因,并进行了生物信息学分析。结果表明:牦牛MSMB基因CDS区长117 bp,编码38个氨基酸;MSMB蛋白分子式C194H308N50O5... 微精原蛋白β(MSMB)对动物的繁殖功能具有重要的调控作用。采集成年健康母牦牛的卵巢组织,通过RT-PCR技术克隆牦牛的MSMB基因,并进行了生物信息学分析。结果表明:牦牛MSMB基因CDS区长117 bp,编码38个氨基酸;MSMB蛋白分子式C194H308N50O59S4,理论等电点7.69,分子质量4.4 ku,脂肪系数66.58,不稳定系数21.23,属于稳定蛋白质,半衰期30 h,不具有跨膜结构和信号肽,是亲水性蛋白;与黄牛MSMB基因对比发现,牦牛MSMB基因存在1处碱基突变;牦牛与黄牛和绵羊CDS区核苷酸同源性均在80%以上,说明MSMB基因在进化中保守;MEGA 6.0和Clustal X软件分析结果表明,牦牛与黄牛首先聚为一小类,其次与绵羊、小鼠和大鼠聚为一类,最终与猪、鸡和鱼聚为一类,符合物种进化规律,进化树与同源性比对结果相符;二级结构α螺旋、延长链、β-转角和无规卷曲,分别占13.16%、44.74%、13.16%、28.95%,二级结构与三级结构相符;预测MSMB蛋白其结构域是PSP94超家族成员。该实验结果为进一步研究牦牛MSMB基因功能提供了一定理论依据。 展开更多

Objective: Prostate secretory protein of 94 amino acids(PSP94) is a target gene of the EZH2 transcriptional repressor and is often downregulated in prostate cancer;however, its prognostic value is disputed.Methods: Immunohistochemical analysis of a tissue microarray of 12, 432 prostate cancer specimens was performed to evaluate PSP94 expression. Correlation of PSP94 expression with tumor phenotype, patient prognosis, TMPRSS2:ERG fusion status, EZH2 expression and PTEN deletion was studied.Results: PSP94 expression was increased in benign prostatic hyperplasia;however, it was downregulated in 48% and negative in42% of the 9, 881 interpretable prostate cancer specimens. The loss of PSP94 expression was inversely correlated to EZH2 expression(P < 0.0001) and largely unrelated to the ERG status, but strongly correlated with high Gleason grade, advanced tumor stage, and nodal metastasis(P <0.0001 each). The fraction of PSP94-negative cancer specimens increased from 40% in pT2 to 52%in pT3 b-pT4(P < 0.0001) and from 40% in Gleason 3+3 = 6 to 46% in Gleason 4+3 = 7 and 60% in Gleason ≥4+4 = 8(P <0.0001). Loss of PSP94 was linked to early prostate-specific antigen recurrence, but with little absolute effect(P < 0.0001).However, it provided additional prognostic impact in cancer specimens with PTEN deletion. Loss of PSP94 deteriorated prognosis of cancer patients with PTEN deletion by more than 10%(P < 0.0001). The combination of PTEN deletion and PSP94 loss provided independent prognostic information that was observed in several subgroups defined by classical and quantitative Gleason grade.Conclusions: The results of our study suggest that combined PSP94/PTEN analysis can be potentially used in the clinical prognosis of prostate cancer. 展开更多

目的探讨整合素α6(ITGA6)基因(rsl2621278,G)、染色体8q24区(rs10086908,T)和β-微精浆蛋白(MSMB)基因(rs10993994,T)与北京市居民中前列腺癌(PCa)的关联,了解PCa患者基因型和表型的关系。方法收集112例PCa患者临床、... 目的探讨整合素α6(ITGA6)基因(rsl2621278,G)、染色体8q24区(rs10086908,T)和β-微精浆蛋白(MSMB)基因(rs10993994,T)与北京市居民中前列腺癌(PCa)的关联,了解PCa患者基因型和表型的关系。方法收集112例PCa患者临床、遗传、膳食习惯、嗜好等表型,对PCa患者和91名正常对照者的ITGA6基因(rs12621278,G)、染色体8q24区(rs10086908,T)和MSMB基因(rs10993994,T)进行比较,并进行病例组的基因型-表型分析。结果2组间相比,MSMB基因rs10993994,T风险等位基因频率差异有统计学意义(病例组56.4%,对照组46.2%;P=0.001,OR:1.97,95%CI为1.28~3.04)。8q24区的rs10086908,T(病例组83.5%,对照组79.2%)和ITGA6基因的rs12621278,G(病例组27.2%,对照组27.0%)组间差异无统计学意义(P〉0.05)。数量性状分析发现ITGA6风险基因型(G/G)的患者病程为(1.40±0.55)年,显著短于A/G型的(4.38±3.10)年和A/A型的(2.37±1.84)年(P=0.003)。结论MSMB基因变异和PCa易感性之间存在相关件.根示MSMB甚丙可能与PCa右芒麟. 展开更多

FK228 is an FDA-approved anticancer drug naturally produced by Chromobacterium violaceum No.968 up to 19 mg/L in a pilot industry-scale batch fermentation.Here we report a genomics-guided discovery of Burkholderia thailandensis MSMB43 as a new and significantly better source of FK228.The genome of B.thailandensis MSMB43 was found to contain a functional biosynthetic gene cluster highly homologous to that of FK228 in C.violaceum No.968,and the bacterium indeed produces authentic FK228.By simple fermentation in shaking flasks in a preferred M8 medium,B.thailandensis MSMB43 produced FK228 up to 67.7 mg/L;by fedbatch fermentation in a 20-L fermentor in M8 medium,B.thailandensis MSMB43 produced FK228 up to 115.9 mg/L,which is 95 fold higher than that of C.violaceum No.968 under the same laboratory fermentation conditions.RT-PCR analysis indicated that the high FK228 yield of B.thailandensis MSMB43 was due to high expression of biosynthetic genes,represented by Bth_depA,during the fermentation process.Further genetic manipulation resulted in a recombinant strain,B.thailandensis MSMB43/pBMTL3-tdpR,which harbors a broad host-range vector expressing the thailandepsin biosynthetic pathway regulatory gene tdpR.This engineered strain produced up to 168.5 mg/L of FK228 in fed-batch fermentation in a 20-L fermentor in M8 medium.Therefore,the wild-type B.thailandensis MSMB43 or its engineered derivative could potentially be a good starting point for an industrial process to improve FK228 production for its expanding use in therapy. 展开更多
