The cytokine channel’s mechanism for self-regulation involves the application of antagonistic cytokines that are synthesized to connect to the receptors and release soluble cytokine receptors.The very first receptor antagonist of cytokine that was naturally present was interleukin-1 receptor antagonist(IL-1Ra).The IL-1Ra protein forms are disinfected from supernatants of cultured monocytes on stacked IgG.The family of IL-1 consists of IL-1α,IL-1βand IL-1Ra.Human monocytes regulate the production of IL-Ra.IL-Ra takes part in normal physiological functions by using specific antibodies,and acts as an anti-inflammatory agent.IL-Ra is synthesized in the tissues during the period of active disease and can be systematically measured and/or estimated.Maintenance of the levels of IL-Ra and IL-1 is the main factor for host resistance in patients during diseased conditions,as IL-Ra acts as an inherent regulator of various inflammatory responses.In this article,we focuse on how IL-Ra is synthesized and performs its functions once the inflammatory responses are activated. 展开更多

Non-invasive therapeutic methods have recently been used in medical sciences. Enzymes have shown high activity at very low concentrations in laboratories and pharmaceutical,enabling them to play crucial roles in different biological phenomena related to living organism, especially human medicine. Recently, using the therapeutic methods based on non-invasive approaches has been emphasized in medical society. Researchers have focused on producing medicines and tools reducing invasive procedures in medical.Collagenases are proteins which catalyze chemical processes and break the peptide bonds in collagen. Collagen may be generated more than the required amount or produced in unsuitable sites or may not degrade after a certain time. In such cases, using an injectable collagenase or its ointment can be helpful in collagen degradation. In both in vitro and in vivo tests, it has been revealed that collagenases have several therapeutic properties in wound healing, burns, nipple pain and some diseases including intervertebral disc herniation, keloid, cellulite, lipoma among others. This review describes the therapeutic application of collagenase in medical sciences and the process for its production using novel methods, paving the way for more effective and safe applications of collagenases. 展开更多

BACKGROUND: Hepatitis B virus (HBV) infection, a glo- bal public health problem, is the leading cause of cirrhosis and hepatocellular carcinoma (HCC) worldwide. There are more than 350 million HBV carriers in the world and up to one million die annually due to hepatitis B associated liv- er disease. So far no optimal treatment is available for pa- tients with chronic hepatitis B. In the paper we investigated the efficacy of intramuscular matrine in the treatment of chronic hepatitis B. METHODS: One hundred and twenty patients with chronic hepatitis B were randomly divided into matrine treatment group (n =60) and control group (n =60). The patients of the matrine group were given intramuscularly with matrine (an alkaloid extracted from a traditional Chinese herb Radix Sophorae Flavescentis by Guangzhou Ming Xing Pharmaceu cal Factory, Guangzhou, China) of 100 mg daily for 90 days in addition to conventional liver-protective drugs in- cluding glucurone, inosine, compound vitamin B and caryophyllin. The control group received conventional liv- er-protective drugs alone. Clinical manifestations and labo- ratory parameters including liver biochemistry and serum hepatitis B virus markers were monitored before and after treatment in the two groups. RESULTS: Significant differences were seen between the two groups in terms of improvement of clinical symptoms and signs, recovery of liver functions, and serum conver- sion from hepatitis Be antigen to HBe antibody and from positive to negative serum HBV DNA (P <0.05-0.01). The result of the matrine group was more marked than that of the control group. Serious side-effects were not observed except mild pain at the site of injection of matrine in a few patients. CONCLUSION: These results indicate that intramuscular matrine may be an economical, efficacious, safe drug for the treatment of chronic hepatitis B. 展开更多

Mesenchymal stem cells (MSCs) are non-hematopoietic stem cells with the capacity to differentiate into tissues of both mesenchymal and non-mesenchymal origin. MSCs can differentiate into osteoblastic, chondrogenic, and adipogenic lineages, although recent studies have demonstrated that MSCs are also able to differentiate into other lineages, including neuronal and cardiomyogenic lineages. Since their original isolation from the bone marrow, MSCs have been successfully harvested from many other tissues. Their ease of isolation and ex vivo expansion combined with their immunoprivileged nature has made these cells popular candidates for stem cell therapies. These cells have the potential to alter disease pathophysiology through many modalities including cytokine secretion, capacity to differentiate along various lineages, immune modulation and direct cell-cell interaction with diseased tissue. Here we first review basic features of MSC biology including MSC characteristics in culture, homing mechanisms, differentiation capabilities and immune modulation. We then highlight some in vivo and clinical evidence supporting the therapeutic roles of MSCs and their uses in orthopedic, autoimmune, and ischemic disorders. 展开更多

In the past decades, iron based magnetic nanomaterials have attracted much attention in the biomedicine field due to their superior magnetic properties and great potential in biomedical applications. In particular, magnetic iron oxide nanoparticles(MIONPs) have been playing a crucial role in the biomedicine field because of their diagnostic and therapeutic functions. Meanwhile, MIONPs are benign, low toxic, biocompatible, and biodegradable, so they are the only inorganic magnetic nanomaterials approved by the U.S. Food and Drug Administration(FDA) for clinical use at present. In this review, we mainly introduce the progress in the preparation of iron based magnetic nanomaterials for biomedical applications, including pure iron nanoparticles, iron-based alloy nanoparticles, and MIONPs, with a focus on MIONPs. 目的 本研究旨在评估参苓白术散结合穴位贴敷疗法在腹泻型肠易激综合征(irritable bowel syndrome with diarrhea,IBS-D)治疗中的临床效果。方法 选取2022年5月-2023年8月期间就诊于安徽中医药大学第二附属医院脾胃病科门诊与住院部的... 目的 本研究旨在评估参苓白术散结合穴位贴敷疗法在腹泻型肠易激综合征(irritable bowel syndrome with diarrhea,IBS-D)治疗中的临床效果。方法 选取2022年5月-2023年8月期间就诊于安徽中医药大学第二附属医院脾胃病科门诊与住院部的肠易激综合征腹泻型的患者88例,采用随机数字表法分组为对照组与研究组,各44例。在控制日常饮食与适量运动的前提下,对照组采用盐酸洛哌丁胺治疗,而研究组则应用参苓白术散与穴位贴敷的联合疗法。经过为期28 d的治疗周期,对2组腹泻型肠易激综合征患者的治疗效果进行全面评估。结果 研究组的临床治疗效果在总有效率上明显优于对照组,差异具有统计学意义(P<0.05),其腹胀、排便频率、性状及腹痛程度的改善情况均优于对照组(P<0.05)。经过治疗周期后,2组的血清5-羟色胺(5-HT)以及神经肽Y(NPY)指标均呈现下降趋势,差异具有统计学意义(P<0.05)。在不良反应方面,2组无统计学差异(P>0.05)。对2组中治疗有效的患者进行随访,研究组复发率4.76%(2/42)低于对照组20.00%(7/35)(P<0.05)。结论 参苓白术散联合穴位贴敷治疗腹泻型肠易激综合征具有显著疗效,在改善临床症状和实验室指标方面具有明显优势,且复发率低。 展开更多

引导编辑(Prime editing,PE)系统是一种全新的、革命性的基因组编辑策略。该系统由引导编辑器(Prime editor)组成,包括nCas9(H840A)与逆转录酶(Reverse transcriptase,RT)的融合蛋白;以及包含PBS(Primer binding site)序列和RT模板(RT t... 引导编辑(Prime editing,PE)系统是一种全新的、革命性的基因组编辑策略。该系统由引导编辑器(Prime editor)组成,包括nCas9(H840A)与逆转录酶(Reverse transcriptase,RT)的融合蛋白;以及包含PBS(Primer binding site)序列和RT模板(RT template,RTT)序列的pegRNA(Prime editing guide RNA)两大部分。PE系统可以在双链不断裂的情况下实现所有12种类型的碱基替换及小片段DNA增删,是精准编辑的全新范式。自2019年开发至今不到4年时间,PE系统作为一种通用的技术平台,已广泛应用于医疗、农业等各个领域,产生了一大批新种质资源、基因治疗药物等优秀应用案例。PE作为目前最灵活、最具发展前景的基因组精准编辑新手段,仍旧存在效率偏低、大片段操纵能力不足、系统组分设计复杂(如pegRNA)、安全性未全面评估等问题,仍需要深入研究。本文详细介绍了PE系统的技术原理及限制因素,全面总结了PE系统自开发以来的优化策略及在动植物系统、医疗领域的应用现状,并对PE的发展前景进行了展望。 展开更多

背景:血小板细胞外囊泡是循环中最丰富的囊泡,富含丰富的生物活性分子、遗传物质及蛋白质等信息分子,参与细胞通讯和物质交换,不仅具有良好的促凝活性,还具有促进组织修复和再生的巨大潜力,在再生医学具有广泛的应用前景。目的:从血小... 背景:血小板细胞外囊泡是循环中最丰富的囊泡,富含丰富的生物活性分子、遗传物质及蛋白质等信息分子,参与细胞通讯和物质交换,不仅具有良好的促凝活性,还具有促进组织修复和再生的巨大潜力,在再生医学具有广泛的应用前景。目的:从血小板细胞外囊泡的分泌机制、在再生医学领域的应用及临床转化的限制因素进行阐述,为血小板细胞外囊泡的临床转化提供一些理论支撑。方法:应用计算机检索2005年1月至2023年8月PubMed数据库与血小板细胞外囊泡有关的文章,英文检索词为“platelet-derived,plateletrich plasma,extracellular vesicles,isolated,microvesicles exosomes,applications”,最后纳入符合主题标准的文献62篇进行综述分析。结果与结论:(1)活化的血小板细胞外囊泡可产生血小板微囊泡和血小板外泌体两种类型的囊泡,前者的分泌可能与肌动蛋白细胞骨架的不对称性相关,后者的分泌可能与H^(+)-ATP酶的调节相关。(2)血小板细胞外囊泡是血小板浓缩物和血小板本身的潜在效应物,可能通过促进血管生成、影响细胞行为、促凝与止血以及发挥炎症作用等几方面影响组织再生。(3)血小板细胞外囊泡在组织损伤、肌肉再生、软骨再生、骨关节炎等再生医学领域已有临床前报道,作为伤口愈合的潜在治疗方法已有临床试验数据,但血小板细胞外囊泡的分离方法、样本来源以及激活剂的种类等因素限制了血小板细胞外囊泡在再生医学领域的临床转化。(4)未来血小板细胞外囊泡可能成为再生医学中替代富血小板血浆的无细胞疗法,其临床转化还需要积极寻找鉴别血小板细胞外囊泡和血小板外泌体的特异性标志物,在激活剂刺激血小板细胞外囊泡产生的机制以及血小板细胞外囊泡的最佳收集方式、最佳储存方法、保质期限、临床应用的推荐剂量和最佳临床适应证还需要继续深入研究。 展开更多

目的:探讨经口腔前庭入路腔镜甲状腺切除术(TOETVA)在甲状腺乳头状癌(PTC)治疗中的应用价值。方法:选取PTC患者82例,按照手术方式不同分为TOETVA组(37例)和开放组(45例)。比较两组患者围手术期指标和临床病理学指标。比较两组患者术前... 目的:探讨经口腔前庭入路腔镜甲状腺切除术(TOETVA)在甲状腺乳头状癌(PTC)治疗中的应用价值。方法:选取PTC患者82例,按照手术方式不同分为TOETVA组(37例)和开放组(45例)。比较两组患者围手术期指标和临床病理学指标。比较两组患者术前及术后6个月甲状腺功能[血钙、甲状旁腺素(PTH)和甲状腺球蛋白抗体(TgAb)]。比较两组患者术后6个月对切口瘢痕美观满意度[温哥华瘢痕评估量表(VSS)和患者瘢痕评价量表(PSAS)]以及术后并发症发生情况。结果:与开放组比较,TOETVA组患者手术时间增加,术后24 h颈部VAS评分降低,住院时间缩短(均P<0.05)。两组患者单/双侧甲状腺清扫、阳性淋巴结数目和淋巴结转移阳性率比较差异无统计学意义(均P>0.05)。与同组术前比较,,两组术后6个月血钙、PTH和TgAb水平降低(均P<0.05);血钙、PTH和TgAb水平两组间比较差异无统计学意义(均P>0.05)。TOETVA组患者VAA和PSAS评分低于开放组(均P<0.05)。术后6个月内,两组患者并发症总发生率比较差异无统计学意义(P>0.05)。结论:TOETVA在治疗PTC时与传统开放手术具有同样的有效性及安全性,但在瘢痕美观方面具有明显优势。 展开更多

The potential roles of the intestinal microbiome play in human health have gained considerable attention. The imbalance of gut microbial community has been looked to multiple chronic diseases. Cardiovascular diseases (CVDs) are leading causes of morbidity worldwide and are influenced by genetic and environmental factors. Recent advances have provided scientific evidence that CVD may also be attributed to gut microbiome. In this review, we highlight the complex interplay between microbes, their metabolites, and the potential influence on the generation and development of CVDs. The therapeutic potential of using intestinal microbiomes to treat CVD is also discussed. It is quite possible that gut microbes may be used for clinical treatments of CVD in the near future. 展开更多

BAT对于小型哺乳动物和人类婴儿的体温维持具有重要作用.棕色脂肪细胞内含有很多小脂滴和丰富的血管、神经和线粒体.棕色脂肪的线粒体通过去偶联反应把ATP转化成热量.近年来的研究结果表明,增加棕色脂肪的数量或者提高棕色脂肪的活性可以抑制高脂饮食诱导的体重增加,改善胰岛素抵抗,治疗肥胖.这说明棕色脂肪很可能是防治肥胖以及相关疾病的强有力的靶器官.本文综述了近年来有关棕色脂肪在改善代谢功能、激活棕色脂肪和米色脂肪的相关因子以及其临床运用等方面的研究进展. 展开更多
