The precise mechanism underlying the effects of anti-CD4 antibody and calcium ions(Ca^(2+)) in peanut allergy remains unknown.C3 H/HeJ mice sensitized with peanut protein extract(PPE)were injected with anti-CD4 antibodies for 4 weeks.Stimulation with PPE increased the specific immunoglobulin E(IgE),cytokine,histamine,and mMcp-1 levels,upregulated decorin(Dcn)expression,induced Ca^(2+) inflow in the spleen,and augmented the expression of the transcription factors GATA-3 and Foxp3,which resulted in Th2 and Treg cell activation.Notably,the Ca^(2+) levels were positively correlated with the histamine,interleukin(IL)-4,IL-5,and IL-13 levels,and negatively correlated with IL-10 levels.However,administration of anti-CD4 antibodies markedly alleviated allergic symptoms,activated T cells,and reduced Ca^(2+) inflow,cytokine,histamine,mMcp-1,and the IgHG3,CXCLI2,MMP2 and FABP4 gene.Our results indicated that anti-CD4 antibodies can ameliorate PPE-induced allergy,which is probably related to the suppression of Ca^(2+) inflow,and inhibiting histamine,cytokine and IgHG3,CXCL12,MMP2,and FABP4,thus exerting a protective effect against PPEsensitized food allergy. 展开更多

A significant decline of both Ia^+DECs and Thy-1^+DECs in the mice of the aged group was demonstrated anddifferent densities and different distribution patterns betweenIa^+DECs and Thy-1^+DECs were obserged. AIM: To determine whether study on the carcinogenic potential of reflux juice from patients with remote gastrectomy could clarify the inherent relationship between duodenal reflux and gastric stump cancer. METHODS: A ... AIM: To determine whether study on the carcinogenic potential of reflux juice from patients with remote gastrectomy could clarify the inherent relationship between duodenal reflux and gastric stump cancer. METHODS: A total of 37 reflux juice samples (13 Billroth I, 24 Billroth II) were employed in the present study. A two-stage transformation assay using BALB/c 3T3 cells was carried out to test the initiating or promoting activity of these samples. RESULTS: Two of 18 (11.1%) reflux samples exerted initiating activities, whereas 9/19 (47.4%) samples enhanced the MNNG-initiating cell transformation, suggesting the duodenal reflux juice might more frequently possess the tumor-promoter activity (P = 0.029). In addition, there was no difference in initiating activities of the samples irrespective of surgical procedures (P = 0.488), while Billroth II samples exhibited stronger tumor-promoter activity than Billroth I samples (P = 0.027). Furthermore, the promoter activities were well correlated with the histological changes of the stomas (r(s) = 0.625, P = 0.004), but neither their cytotoxicities nor initiating activities had this correlation (Probabilities were 0.523 and 0.085, respectively). CONCLUSION: The duodenal reflux juice from patients with remote postgastrectomy did have carcinogenic potential, and suggested that tumor-promoting activity should principally account for the high incidence of gastric cancer in gastrectomy patients. In contrast, it is difficult to explain the high stump-cancer incidence with the N-nitroso compounds theory-a popular theory for the intact stomach carcinogenesis, and it seemed to be justified to focus chemo-prevention of this cancer on the tumor-promoting potential of reflux juice. 展开更多

The results indicated that MS could enhance the proliferation of TIL,accelerate it to reach the exponential growth phase, and assist recombinant interleukin 2 (rIL-2) to enhance successively the percentage of CD3^+ positive cells, maintain the number of CD4^+ positive T cells, promote greatly the percentage of CD8^+ positive T cells among TILs, and reverse the CD4^+/CD8^+ ratio. Such cooperative effects rely on relative specificity of acupoints. It is suggested that MS is beneficial to the growth of TIL both in the aspects of proliferation and phenotypes. 展开更多

目的:研究基因转染后高表达吲哚胺2,3-双加氧酶(IDO)的树突状细胞(DC)对T细胞增殖的抑制作用,以探讨其在器官移植中排斥反应的作用。方法:采用逆转录-聚合酶链式反应(RT-PCR)从腹膜炎小鼠的小肠组织中获取IDO全长cDNA片段,将其克隆至真... 目的:研究基因转染后高表达吲哚胺2,3-双加氧酶(IDO)的树突状细胞(DC)对T细胞增殖的抑制作用,以探讨其在器官移植中排斥反应的作用。方法:采用逆转录-聚合酶链式反应(RT-PCR)从腹膜炎小鼠的小肠组织中获取IDO全长cDNA片段,将其克隆至真核表达质粒pTracer中,构建真核表达质粒IDO-pTracer,随后用LipofectamineTM2000介导转染小鼠脾脏DC,分为未转染组、空载体转染组、IDO基因转染组和IDO基因转染并添加1-甲基色氨酸(1-MT)组,以混合淋巴细胞反应的方法检测转染后DC对同种异系T细胞增殖的作用。结果:克隆的小鼠IDO基因全长cDNA大小约1.3kb,无碱基突变,以构建的重组真核表达质粒IDO-pTracer转染DC后,混合淋巴细胞反应结果显示IDO基因转染组的刺激指数(SI)低于未转染组、空载体转染组及IDO基因转染并添加1-MT组,差异均有统计学意义(P<0.05)。结论:成功构建了IDO基因的真核表达载体IDO-pTracer,且IDO基因转染DC具有抑制异系T细胞增殖的作用。 展开更多

目的探讨长链非编码RNA(LncRNA)PVT1对变应性鼻炎(AR)进展的影响及其相关作用机制。方法24只BALB/c小鼠,其中6只作为对照组,余18只采用卵白蛋白和氢氧化铝建立AR模型并采用随机数字表法均分为模型组、AR+NC组(尾静脉注射空载体慢病毒)、... 目的探讨长链非编码RNA(LncRNA)PVT1对变应性鼻炎(AR)进展的影响及其相关作用机制。方法24只BALB/c小鼠,其中6只作为对照组,余18只采用卵白蛋白和氢氧化铝建立AR模型并采用随机数字表法均分为模型组、AR+NC组(尾静脉注射空载体慢病毒)、AR+shPVT1组(尾静脉注射LncRNA PVT1干扰载体慢病毒),每组6只。采用行为学评分评估各组小鼠过敏症状;HE染色观察各组小鼠鼻黏膜组织病理学改变;实时荧光定量PCR检测各组小鼠鼻黏膜组织中LncRNA PVT1、miR-149-5p和几丁质酶-3样蛋白1(CHI3L1)mRNA的表达;Western blot检测CHI3L1蛋白表达;ELISA分析各组小鼠血清IgE、白细胞介素(IL)-5、肿瘤坏死因子(TNF)-α水平。构建LncRNA PVT1和CHI3L1的野生型(WT)和突变型(MT)荧光素酶报告质粒,单独或同时与miR-149-5p mimic共转染293T细胞,采用双荧光素酶实验分析并计算各组细胞荧光素酶相对活性。结果与模型组相比,AR+shPVT1组小鼠行为学评分降低;血清中IgE、IL-5和TNF-α水平降低;鼻黏膜组织中LncRNA PVT1 mRNA表达水平降低,miR-149-5p mRNA表达水平升高,CHI3L1 mRNA及蛋白水平均降低,鼻黏膜组织炎症反应减轻。双荧光素酶实验证实LncRNA PVT1通过竞争性结合miR-149-5p,上调miR-149-5p靶基因CHI3L1的表达。结论在AR模型小鼠中异常高表达的LncRNA PVT1通过发挥竞争性内源RNA作用抑制miR-149-5p表达而上调CHI3L1的表达,引发小鼠鼻黏膜组织的炎性症状并促进过敏反应。 展开更多

目的:探讨内皮抑素重组腺相关病毒(rAAV-ES)对人宫颈癌HeLa细胞裸鼠移植瘤的生长抑制作用及其机制。方法:采用左腋皮下接种法,建立人宫颈癌裸鼠模型共60只。造模1周后,随机分为rAAV-ES实验组、空病毒组和磷酸盐缓冲液(PBS)对照组,每组2... 目的:探讨内皮抑素重组腺相关病毒(rAAV-ES)对人宫颈癌HeLa细胞裸鼠移植瘤的生长抑制作用及其机制。方法:采用左腋皮下接种法,建立人宫颈癌裸鼠模型共60只。造模1周后,随机分为rAAV-ES实验组、空病毒组和磷酸盐缓冲液(PBS)对照组,每组20只。观察裸鼠皮下移植瘤的生长情况,计算肿瘤体积。治疗3周后颈椎脱臼处死裸鼠,完整剥离肿瘤,称瘤质量并计算抑瘤率。HE染色观察肿瘤细胞形态,免疫组织化学技术检测各组肿瘤组织中Bcl-2、Bax和Caspase-3蛋白的表达情况。结果:成功建立宫颈癌裸鼠模型。实验组治疗后肿瘤生长速度较对照组和空病毒组明显减慢(P<0.01),抑瘤率为62.55%。Bcl-2在实验组的阳性率低于空病毒组和PBS对照组(均P<0.01);各组Bax阳性率比较差异无统计学意义(P>0.05);Caspase-3在实验组的阳性率高于空病毒组和PBS对照组(均P<0.01)。Bcl-2与Caspase-3的表达呈负相关(rs=-0.653,P<0.01)。结论:rAAV-ES可抑制宫颈癌移植瘤的生长,其机制可能与通过下调Bcl-2蛋白、激活下游的Caspase-3诱导细胞凋亡有关。 展开更多

微小RNA-378调控小鼠急性肝衰竭作用机制的研究

目的探究微小RNA-378(miR-378)调控小鼠急性肝衰竭的作用机制。方法将36只雄性巴比塞小鼠以随机数字表法均分为实验组和对照组。实验组腹腔注射D氨基半乳糖(D-GalN)800 mg/kg联合脂多糖10μg/kg诱导急性肝衰竭,对照组为腹腔内注射等量... 目的探究微小RNA-378(miR-378)调控小鼠急性肝衰竭的作用机制。方法将36只雄性巴比塞小鼠以随机数字表法均分为实验组和对照组。实验组腹腔注射D氨基半乳糖(D-GalN)800 mg/kg联合脂多糖10μg/kg诱导急性肝衰竭,对照组为腹腔内注射等量的生理盐水。两组在诱导后0、1、3、5、7、9 h处死小鼠取静脉血和肝组织,每个时间点有3只实验鼠。检测两组在各时间点血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平,酶联免疫吸附测定(ELISA)检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平,逆转录聚合酶链反应(RT-PCR)测定肝组织中的胱天蛋白酶-3(Caspase-3)、TNF-α及IL-6 mRNA表达量,探针法确定肝组织中miR-378表达量,蛋白质印迹法(Westernblotting)检测Caspase-3蛋白表达量。结果实验组ALT/AST水平逐渐升高且在诱导后7 h达到峰值,对照组ALT/AST未见明显变化。相较对照组,实验组在诱导后1 h和7 h TNF-α/IL-6血清浓度及mRNA水平明显升高;实验组在诱导后5 h[(0.0038±0.0003)比(0.0054±0.0003),P<0.05]和7 h[(0.0014±0.0005)比(0.0054±0.0003),P<0.05]miR-378表达量明显降低,Caspase-3 mRNA[5 h(0.0052±0.0003),7 h(0.0070±0.0006)比(0.0033±0.0001),均P<0.05]和蛋白[5 h(0.867±0.012),7 h(1.062±0.039)比(0.577±0.035),均P<0.05]表达量升高。结论在小鼠D-GalN联合脂多糖诱导的急性肝衰竭模型中,miR-378表达量的下调促进了Caspase-3的活化,增强了肝细胞凋亡活动,进而调控急性肝衰竭的发展。miR-378可作为治疗急性肝衰竭新靶点。 展开更多

目的①探索非霍奇金淋巴瘤(NHL)动物模型建模方法;②探讨吲哚胺2,3双加氧酶(IDO)及色氨酰tRNA合成酶(TTS)参与的色氨酸代谢紊乱与NHL细胞增殖及NHL免疫耐受的相关性。方法将近交系BALB/c小鼠随机分为对照组(4只)、肿瘤组(8只)和治疗组(8... 目的①探索非霍奇金淋巴瘤(NHL)动物模型建模方法;②探讨吲哚胺2,3双加氧酶(IDO)及色氨酰tRNA合成酶(TTS)参与的色氨酸代谢紊乱与NHL细胞增殖及NHL免疫耐受的相关性。方法将近交系BALB/c小鼠随机分为对照组(4只)、肿瘤组(8只)和治疗组(8只),以鼠源性NHL细胞株A20皮下注射(5×106个/只)建立NHL动物模型,瘤旁及瘤内注射IDO抑制剂1-甲基色氨酸(1-MT),并采用免疫组化SP法和Western blot技术检测肿瘤组织和淋巴结中TTS的表达水平。结果①BALB/c小鼠皮下接种A20细胞建立淋巴瘤动物模型,成瘤率高,小鼠死亡率低,伴有明显的淋巴结和脾脏转移;②免疫组化示肿瘤组织及转移淋巴结TTS表达均升高(P<0.05);③Western blot结果显示,与正常对照比较,肿瘤组及治疗组小鼠肿瘤组织TTS表达明显升高(P<0.001);与肿瘤组相比,治疗组TTS表达水平显著下降(P<0.01),肿瘤转移淋巴结TTS表达无变化(P>0.05)。结论 IDO诱导的低色氨酸微环境中,TTS表达上调是肿瘤细胞对抗IDO诱导的色氨酸耗竭、维持自身代谢过程的重要机制之一。1-MT对色氨酸代谢活性及表达水平的调控有望成为辅助治疗恶性肿瘤的新方向。 展开更多
