In this report, we describe the isolation and characterization of microsatellite loci for forest musk deer (Moschus berezovskii) through an improved enrichment protocol. Five new markers were isolated from the genomic DNA of forest musk deer and showed high polymorphism with 4 - 13 alleles in 24 sampled individuals from the population of Jinfeng Mountain, Sichuan Province, China. The observed and expected heterozygesities were from 0.429 - 0.957 and 0.587 - 0.902, respectively. The average polymorphism information content (PIC) value in these five loci was 0. 730. This suggests that the five microsatellite loci are a valuable tool for further studies about forest musk deer. 展开更多

In order to solve the problem that the musk deer concentrated feed were easy to get rancidity in spring, summer, autumn and freezing in winter, high quality and palatability pellet feed of musk deer was researched and developed to ensure the quality of musk deer's feed. Twenty-eight male musk deer (3 - 10 years old) (Moschus berezovskii) with normal body condition in the same culture conditions were chosen and were randomly divided into experimental group (fed pellets group) and control group (original feeding diet group), the results show that musk deer was able to adapt to the pellet feed generally in 13 days and the duration varied with the palatability of pellet feed. During the trial period, the difference of the body weight, the average musk yield of producing musk deer and the producing musk incidence between the experimental group and the control group was not significant, the incidence of disease and mortality of experimental group deer was significantly lower than the control group. 展开更多

气候背景下林麝适宜生境的最大熵模型(MaxEnt)研究 被引量:4

基于公开发表的林麝(Moschus berezovskii)在中国范围内地理分布数据和生境气候数据,利用刀切法提取影响林麝存在概率的关键气象因子,并运用MaxEnt模型与ArcGIS软件分析不同情景下林麝在中国的适生范围。结果表明,最暖季降水量、最干季... 基于公开发表的林麝(Moschus berezovskii)在中国范围内地理分布数据和生境气候数据,利用刀切法提取影响林麝存在概率的关键气象因子,并运用MaxEnt模型与ArcGIS软件分析不同情景下林麝在中国的适生范围。结果表明,最暖季降水量、最干季均温、最湿季降水量、年均温、季节性温差、最湿季均温、最暖季均温、最干季降水量8个关键气候因子对林麝的分布有重要影响;利用受试者工作特征曲线检验林麝生境范围预测模型,得出模型预测结果达到优秀水平(AUC=0.993)。当前气候情景下,林麝生境适宜区主要分布在腾冲-漠河线以南,适宜生境面积为4.13×106km2,占中国国土面积的43%;RCP2.6、RCP4.5和RCP8.5三种未来气候情景下,至2050s(2040—2059年)林麝高、中、低适生面积均有所减少,其中低适生面积减幅最大(达到50%);2080s(2070—2089年)较2050s,RCP2.6和RCP4.5情景下林麝高、中、低适生面积有所增加,RCP8.5情景下则有所减少。以平原、丘陵地貌为主的林麝适生区东南区域,对未来气候条件的变化负面响应较明显,而以高山为主的西南地区则能够较好地应对未来气候条件变化,所以建议以西南地区为核心建立林麝保护区,并严格控制人员进入保护区,以达到更好地保护野生林麝的目的。 展开更多

圈养林麝粪样类固醇激素水平与麝香分泌的关系 被引量:6

基于非损伤取样和放射免疫法(RIA),于2018年9月1日—10月15日期间对四川马尔康林麝繁育场的36头雄性圈养林麝(Moschus berezovskii)的粪样类固醇激素水平进行了检测,结合麝香分泌数据采集,探究了林麝的粪样睾酮和皮质醇水平与其麝香分... 基于非损伤取样和放射免疫法(RIA),于2018年9月1日—10月15日期间对四川马尔康林麝繁育场的36头雄性圈养林麝(Moschus berezovskii)的粪样类固醇激素水平进行了检测,结合麝香分泌数据采集,探究了林麝的粪样睾酮和皮质醇水平与其麝香分泌的关系,结果表明:四川马尔康圈养林麝的麝香分泌(12.53±0.76) g (n=36)与粪样睾酮水平(106.03±12.34) ng/g (n=36)呈显著正相关(r=0.436,P<0.01),泌香较多雄麝(16.50±0.61) g (n=17)的睾酮水平(141.51±21.86) ng/g (n=17)显著地高于泌香较少(8.98±0.59) g (n=19)雄麝(74.27±7.93) ng/g (n=19)(t=-3.252,df=34,P<0.01),方程y=0.021x+10.270(R2=0.120,P<0.05)可较好地拟合二者间关系。麝香分泌与皮质醇浓度(47.49±1.53) ng/g (n=36)相关不显著(r=0.078,P>0.05),泌香较多雄麝的皮质醇浓度(48.98±2.28) ng/g (n=17)略高于泌香较少雄麝(46.17±2.14) ng/g (n=19),差异不显著(P>0.05),方程y=0.038x+10.730(R2=0.023,P>0.05)可近似拟合皮质醇与泌香量间的关系。此外,雄麝睾酮水平对泌香量有显著影响(P<0.05),睾酮水平较高林麝泌香量(14.97±1.17) g (n=12)显著高于睾酮水平较低个体的泌香量(11.32±0.90) g (n=24)(F=4.79,P<0.05),而睾酮、皮质醇和年龄对麝香分泌的综合效应及变量间的交互作用均不显著(P>0.05)。研究确定了圈养林麝粪样睾酮和皮质醇水平与麝香分泌量间的关系,可用于麝类驯养实践的麝香产量预测及品系选育。 展开更多

动物社群的社会等级可对个体的资源分配、社群稳定及个体的行为对策产生重要影响,圈养动物的类固醇激素水平与饲养管理有关,可反映动物的社群紧张水平。深入理解圈养动物的社会等级与其类固醇激素水平的关系是进行濒危动物迁地保育和成... 动物社群的社会等级可对个体的资源分配、社群稳定及个体的行为对策产生重要影响,圈养动物的类固醇激素水平与饲养管理有关,可反映动物的社群紧张水平。深入理解圈养动物的社会等级与其类固醇激素水平的关系是进行濒危动物迁地保育和成功驯养的基础。2018年6月15日—8月15日,用焦点取样法对四川马尔康林麝繁育场的25头林麝进行了冲突行为取样及社会等级计算,采用放射免疫分析法检测了同期粪样的类固醇激素水平,分析了林麝社会等级与粪样皮质醇、睾酮及雌二醇水平的关系。结果表明:低等级雌麝的雌二醇水平(289.037±59.710 pg·g^(-1),n=11)显著高于高等级雌麝(45.670±27.283pg·g^(-1),n=6)(P<0.05),低等级雄麝的睾酮水平(3.863±1.538 ng·g^(-1),n=3)和高等级雄麝(8.017±1.295 ng·g^(-1),n=5)无显著差异(P>0.05);低等级雄麝(37.891±7.564 ng·g^(-1),n=3)和雌麝(37.262±1.544 ng·g^(-1),n=11)的皮质醇水平与高等级雄麝(29.947±2.441 ng·g^(-1),n=5)及雌麝(37.478±4.628 ng·g^(-1),n=6)间的差异不显著(P>0.05)。不区分性别,低等级林麝的皮质醇水平(37.397±1.826 ng·g^(-1),n=14)和高等级个体(34.055±2.886ng·g^(-1),n=11)间无显著差异(P>0.05)。研究结果表明,圈养林麝雌体的社会等级与其粪样雌二醇水平呈负相关,低等级雌麝的粪样雌二醇水平显著高于高等级雌麝。在麝类驯养实践中,可监测雌麝的社会等级和粪样雌二醇水平变化,预测雌麝的行为健康及繁殖成效。 展开更多

Lys R-type transcriptional regulators are involved in the regulation of numerous cellular metabolic processes in Klebsiella pneumoniae,leading to severe infection.Earlier,we found a novel Lys R family gene,named kp05372,in a strain of K.pneumoniae(designated GPKP)isolated from forest musk deer.To study the function of this gene in relation to the biological characteristics of GPKP,we used the suicide plasmid and conjugative transfer methods to construct deletion mutant strain GPKP-Δkp05372;moreover,we also constructed the GPKP-Δkp05372+complemented strain.The role of this gene was determined by comparing the following characteristics of three strains:growth curves,biofilm formation,drug resistance,stress resistance,median lethal dose(LD50),organ colonization ability,and the histopathology of GPKP.Real-time polymerase chain reaction(RT-PCR)was used to test the expression level of seven genes upstream of kp05372.There was no significant difference in the growth rates when comparing the three bacterial strains,and no significant difference was recorded at different osmotic pressures,temperatures,salt contents,or hydrogen peroxide concentrations.The GPKP-Δkp05372 mutant formed a weak biofilm,and the other two strains formed medium biofilm.The drug resistance of the GPKP-Δkp05372 mutant toward cephalothin,cotrimoxazole,and polymyxin B was changed.The acid tolerance of the deletion strain was stronger than that of the other two strains.The LD50 values of the wild-type and complemented strains were 174-fold and 77-fold higher than that of the GPKP-Δkp05372 mutant,respectively.The colonization ability of the GPKP-Δkp05372 mutant in the heart,liver,spleen,kidney,and intestine was the weakest.The three strains caused different histopathological changes in the liver and lungs.In the GPKP-Δkp05372 mutant,the relative expression levels of kp05374 and kp05379 were increased to 1.32-fold and 1.42-fold,respectively,while the level of kp05378 was decreased by 42%.Overall,the deletion of kp05372 gene leads to changes in the following:drug resistance and acid tolerance;decreases in virulence,biofilm formation,and colonization ability of GPKP;and regulation of the upstream region of adjacent genes. 展开更多
