Severe acute respiratory syndrome coronavirus 2 may cause liver injury via Na^(+)/H^(+) exchanger

The liver has many significant functions,such as detoxification,the urea cycle,gluconeogenesis,and protein synthesis.Systemic diseases,hypoxia,infections,drugs,and toxins can easily affect the liver,which is extremely sensitive to injury.Systemic infection of severe acute respiratory syndrome coronavirus 2 can cause liver damage.The primary regulator of intracellular pH in the liver is the Na+/H+exchanger(NHE).Physiologically,NHE protects hepatocytes from apoptosis by making the intracellular pH alkaline.Severe acute respiratory syndrome coronavirus 2 increases local angiotensin II levels by binding to angiotensinconverting enzyme 2.In severe cases of coronavirus disease 2019,high angiotensin II levels may cause NHE overstimulation and lipid accumulation in the liver.NHE overstimulation can lead to hepatocyte death.NHE overstimulation may trigger a cytokine storm by increasing proinflammatory cytokines in the liver.Since the release of proinflammatory cytokines such as interleukin-6 increases with NHE activation,the virus may indirectly cause an increase in fibrinogen and D-dimer levels.NHE overstimulation may cause thrombotic events and systemic damage by increasing fibrinogen levels and cytokine release.Also,NHE overstimulation causes an increase in the urea cycle while inhibiting vitamin D synthesis and gluconeogenesis in the liver.Increasing NHE3 activity leads to Na+loading,which impairs the containment and fluidity of bile acid.NHE overstimulation can change the gut microbiota composition by disrupting the structure and fluidity of bile acid,thus triggering systemic damage.Unlike other tissues,tumor necrosis factor-alpha and angiotensin II decrease NHE3 activity in the intestine.Thus,increased luminal Na+leads to diarrhea and cytokine release.Severe acute respiratory syndrome coronavirus 2-induced local and systemic damage can be improved by preventing virus-induced NHE overstimulation in the liver. 展开更多

目的 了解山东省2022-2023流感监测年度分离的A(H1N1)pdm09亚型流感病毒血凝素(hemagglutinin,HA)和神经氨酸酶(neuraminidase,NA)基因的变异特征,为流感的防控提供科学依据。方法 从流感监测网络实验室分离的流感毒株中按地市随机选取1... 目的 了解山东省2022-2023流感监测年度分离的A(H1N1)pdm09亚型流感病毒血凝素(hemagglutinin,HA)和神经氨酸酶(neuraminidase,NA)基因的变异特征,为流感的防控提供科学依据。方法 从流感监测网络实验室分离的流感毒株中按地市随机选取14株A(H1N1)pdm09亚型流感毒株,以WHO推荐的当季疫苗株为参考进行全基因组测序,并采用荧光法开展神经氨酸酶抑制(neuraminidase inhibition,NI)实验以评估药物敏感性。结果 山东省2022-2023年度分离的A(H1N1)pdm09流感病毒属于6B.1A分支中的5a.2a进化簇,核苷酸序列比对分析显示,HA和NA基因与2021-2023年度北半球疫苗株A/Victoria/2570/2019的亲缘关系较为接近,同源性分别为98.5%~98.7%和98.8%~99.1%。氨基酸序列分析显示,HA蛋白有20个位点发生了氨基酸序列变异,并发现1株病毒可能发生抗原漂移,有3株病毒发生了HA蛋白糖基化位点的缺失。NA酶相关重要位点未发生变异。NI实验显示,所测流感毒株均对抗流感病毒药物敏感。结论 所监测毒株与疫苗株整体同源性很高,但氨基酸存在一定程度变异,今后有必要持续开展流感病毒基因变异特征监测以了解流感流行的风险,以及评价基因变异对流感疫苗和治疗药物效果的影响。 展开更多

目的旨在建立同时鉴别诊断H7亚型及其5种NA亚型(N2、N3、N4、N7和N9)AIV的检测方法。方法针对H7亚型HA基因、5种NA亚型NA基因和所有亚型AIV M基因的保守序列,分别设计了6对亚型特异性引物和1对AIV亚型通用检测引物,利用GeXP多基因表达... 目的旨在建立同时鉴别诊断H7亚型及其5种NA亚型(N2、N3、N4、N7和N9)AIV的检测方法。方法针对H7亚型HA基因、5种NA亚型NA基因和所有亚型AIV M基因的保守序列,分别设计了6对亚型特异性引物和1对AIV亚型通用检测引物,利用GeXP多基因表达和毛细管电泳分析技术,建立同时检测H7亚型和5种NA亚型AIV的GeXP高通量分型鉴别诊断方法,对其进行特异性、敏感性和临床样品检测。结果特异性结果显示该法单管可同时检测H7、N2、N3、N4、N7和N9亚型AIV,均能检出对应的亚型AIV,通用检测引物检出所有亚型AIV,与其它常见禽病病原体不存在交叉反应。敏感性结果显示该法对7种AIV目的基因(含H7、N2、N3、N4、N7、N9基因)浓度均为100拷贝/μL时,仍可同时检测出。该法对150份临床样品的检测结果与病毒分离鉴定一致。结论本研究建立的同时鉴别诊断H7亚型和5种亚型AIV GeXP高通量检测方法具有特异性强、敏感性高、快速和简便等优点,为防控AIV提供新型检测方法。 展开更多

Adsorption isotherms of carbon dioxide (CO2), methane (CH4), and nitrogen (N2) on Hβand sodium exchanged β-zeolite (Naβ) were volumetrically measured at 273 and 303 K. The results show that all isotherms were of Brunauer type I and well correlated with Langmuir-Freundlich model. After sodium ions exchange, the adsorption amounts of three adsorbates increased, while the increase magnitude of CO2 adsorption capacity was much higher than that of CH4 and N2. The selectivities of CO2 over CH4 and CO2 over N2 enhanced after sodium exchange. Also, the initial heat of adsorption data implied a stronger interaction of CO2 molecules with Na+ ions in Naβ . These results can be attributed to the larger electrostatic interaction of CO2 with extraframework cations in zeolites. However, Naβ showed a decrease in the selectivity of CH4 over N2, which can be ascribed to the moderate affinity of N2 with Naβ. The variation of isosteric heats of adsorption as a function of loading indicates that the adsorption of CO2 in Naβ presents an energetically heterogeneous profile. On the contrary, the adsorption of CH4 was found to be essentially homogeneous, which suggests the dispersion interaction between CH4 and lattice oxygen atoms, and such interaction does not depend on the exchangeable cations of zeolite. 展开更多

We determined whether La3+ enter human peripheral blood lymphocytes via Na+/Ca2+ exchanger (measured with fura-2). We first compared the sensitivity of fura-2 with La3+ and Ca2+, the result indicates that the sensitivity of fura-2 for La3+ is much greater than for Ca2+. La3+ forms a 1:1 La3+-fura-2 complex (apparent dissociation constant = 1.7x10(-12) mol/L, pH 7.05). Ouabain-pretreated cells, suspended in Na+-free medium, showed that La3+ can enter human lymphocytes via the Na-i(+)/Ca2+ (La3+)(o) exchanger and is found to be about 10(-12) mol/L in cells exposed to 0.4 mmol/L La3+. Otherwise, the higher concentration (0.1 mmol/L) blocks the Na-i(+)/Ca2+(La3+)(o) exchange-mediated influx of Ca2+, but the lower concentration (0.01 mmol/L) appears to increase Ca2+ entry. 展开更多

Objective Background and Objects: Naturally occurring temporal variability of action potentialduration (APD) in isolated myocytes has been noted. Most of the studies have beenfocusing on analyzes of the differences in ionic channels and currents among theepicardial-, mid-myocardial-(M) and endocardial myocytes, and the rate-dependent (adaptation) characteristics of APD. We have found that the change in APD during achange in frequency of stimulation mostly reflects a change in rate of repolarization at distinct membrane potential levels. We assumed that in the myocytes, there is balancing mechanism, which is constantly adjusting the various ionic currents accommodating to the changing conditions. This intrinsic ability of adaptation is important and may offer some of the consequences of the transmural heterogeneity in adaptation of APD. This adaptive behaviors maybe equally important in maintaining the normal electrophysiological properties and in induction of arrhythmia in a case of error in normal adaptation. Though most studies of Na +/Ca 2+ exchange (NCX) has been emphasized on its reverse activaty during pathyological condition. Our hypothesis is that reverse activaty of NCX also plays an important role in adjusting the repolarization of AP during a physiological condition. A mismatch between action potential (AP) repolarization and relaxation of the contraction can be caused by intracellular Ca 2+ transport abnormalities. Ca 2+ influx via reverse activation of NCX can load the sarcoplasmic recticulum (SR), which has arrhythmogenic effect.Methods We studied the single myocytes from the left ventricle of adult mongrel dogs. During the cell separation, collagenase was perfused through LAD by Langandorff system. We use the patch-clamp system to determinded AP in current clamp mode. Myocyte contraction was imaged by a video camera, shortening of unloaded myocytes was detected by a video edge motion detector, using changes in light intensity at the edges of the myocyte. Results From 60 consecutive recorded APs at a constant 1.0 Hz stimulation under steady state conditions we found there is a variance in the repolarization between 10mV and-40mV. We also found the variance in the APD during the rate adaptation range of repolarization. Fluctuation in the transient may contribute to the APD variability. To test thishypothesis we block the transient by intracellular dialysis with 10 mM EGTA(n=19), this caused a significant reduction in the coefficient variability (CV=SD/mean APD%) from 2.3± 0.8 to 1.3± 0.3 P< 0.01. During a rate change of the stimulation from 0.6 Hz to 1.0 Hz. The AP duration increased from 278±8 msec to 320±9 msec, Mean+SD, n=5, 50 APs, P< 0.05. contraction is accompanied by an after-contraction(A-CON). The relaxation of contraction precedes the repolarization of the AP. We assumed that the enhancement of repolarization and the production of after-contraction can be possibly induced by reverse mode of NCX. Reducing [Na +] o by substitution of 40mM Na + with Li + favors NCX activating the reverse mode, which significantly decreased the dome of the AP from 4.8± 0.3 to -10.6± 1.2mV, P< 0.05, and increased the APD from 330±13 to 368±14 msec. P< 0.05.Conclusions Intracellular calcium transient most likely contributes to the beat-to-beat variance of action potential duration in canine ventricular myocyte. And it attributes to the voltage-dependent switch of NCX mode. Calcium concentration is high inmyocytes during the repolarization, and high intracellular Ca 2+ activates NCX in such a manner, that it generates an inward (positive, depolarizing) current. This current works against the repolarization, it is prolonging it, with other words it increases the duration of the action potential. The magnitude of calcium concentration during repolarization is very much dependent on calcium transport in the SR. Cation exchange capacity (CEC) is an important characteristic of zeolites, especially when they are used as adsorbents in the aqueous system. However, no international standard method exists for the determination of CEC of zeolites. We determined CEC of Linde-type A and Na-P1 type zeolites at various pH (4 to 10) with a simple method, where Na+-saturated zeolites were prepared, and then various amounts of HCl were added. CEC was simply calculated by subtracting the amount of Na+ in the final supernatant from the content of Na+ of the Na+-saturated zeolites. CEC of the zeolites decreased with decreasing pH and with decreasing Na+ concentration of the final supernatant. The concentration of Na+ of the supernatant, CEC of the zeolites began to decrease at weakly alkaline or neutral pH, and that of the Linde-type A zeolite became about half at pH around 6. When CEC was plotted against pH-pNa;where pNa is negative logarithm of the activity of Na+;CEC of each zeolite was expressed by a curve. To determine the effects of HOE 694, a new and potent Na+- H+ exchanger blocker, on transient inward current (Iti) and Na+- Ca2+ exchange during hypoxia- reoxygenation in guinea pig cardiomyocytes. Methods. Cardiomyocytes were isolated from adult guinea pig ventricle. Experiment was performed in an experimental chamber that allowed the cells to be exposed to a sufficiently low O2 pressure. The cells were subjected to hypoxia and reoxygenation. The ionic currents were studied with patch clamp technique. Results. In the absence of HOE 694, hypoxia- reoxygenation induced Iti in 12 of 15 experiments; but in cardiomyocytes pretreated with HOE 694 (10~ 50μ mol/L), the incidence of Iti observed during reoxygenation was reduced to 5 of 11 experiments and 3 of 10 experiments, P Conclusions. Blockade of the Na+- H+ exchange by HOE 694 could reduce Ca2+ overload upon hypoxia- reoxygenation, and inhibition of Na+- H+ exchange may also indirectly decrease Na+- Ca2+ exchange activity during hypoxia. 展开更多

Objective\ To test the effect of hypothermia on Na+/H+ exchange, activated by shrinkage and cytoplasmic acidosis. Method\ Amiloride-sensitive Na+ influx in guinea pig red cells was traced with isotope 22Na and intracellular Na+ concentration was measured by emission flame photometry. Result\ Amiloride-sensitive Na+ influx decreased linearly as a function of temperatures (about 37℃) in shrunken cells, but increased in acidified cells. The up-regulation of acid-induced Na+/H+ exchange by elevated temperature was enhanced by hypo-osmolarity. Less sensitivity of intracellular H+ site at 41℃ may be the mechanism for the inhibition of shrinkage-induced Na+/H+ exchange by elevated temperature. Heating-mediated explosive increase in the activity of acid-induced Na+/H+ exchange may be due to enhanced extracellular Na+ sensitivity and lower intracellular pH caused by acidic metabolites. Acid-induced Na+/H+ ewxchange contributes to cytoplasmic Na+ accumulation. Conclusion\ These two modes of Na+/H+ exchange with different response to elevated temperature may play different roles in the cellular pathogenesis of heatstroke. 展开更多

In order to reveal the reactivity of a functional group in an aromatic compound having two substituents inthe aromatic ring, the hydrogen-isotope exchange reaction (T-H exchange reaction) between tritiated water vapor(HTO vapor) and 4-amino-2-methylbenzenesulfonic acid (and 5-amino-2-methylphenol) were dynamically observedat 50 (and 70 ) in a gas ℃ ℃ -solid system. Consequently, the fact that the specific activity of the acid increased withtime was obtained, and the T-for-H exchange reaction occurred. By applying the A"-McKay plot method to the dataobserved, the rate constant of each functional group for the reaction was obtained. After the additive property of theHammett’s rule was applied to this work, the new substituent constants were obtained. From the above-mentioned,the following four items have been confirmed: (1) the reactivity of the functional groups can be dynamically analyzed,and the A"-McKay plot method is useful to analyze the reactivity; (2) the additive property of the Hammett’s rule isapplicable to quantitative comparison of the reactivity of the functional groups; (3) the reactivity of the functionalgroups can be simultaneously analyzed by using the A"-McKay plot method in the T-H exchange reaction; and (4) themethod used in this work is also useful for analyzing the reactivity of a certain material having some kinds of functional groups. 展开更多

采用熔融法制备了不同Na_(2)O含量的透明锂铝硅微晶玻璃,通过DSC、XRD、FESEM等测试方法研究了不同Na_(2)O含量对玻璃析晶及性能的影响。结果表明:Na_(2)O的引入能显著降低玻璃的转变温度和析晶温度,抑制LiAlSi_(4)O_(10)晶相的析出。但... 采用熔融法制备了不同Na_(2)O含量的透明锂铝硅微晶玻璃,通过DSC、XRD、FESEM等测试方法研究了不同Na_(2)O含量对玻璃析晶及性能的影响。结果表明:Na_(2)O的引入能显著降低玻璃的转变温度和析晶温度,抑制LiAlSi_(4)O_(10)晶相的析出。但Na_(2)O的引入促使微晶玻璃中析出Li_(2)Si_(2)O_(5)新相,并且随着Na_(2)O引入量的增加,Li_(2)Si_(2)O_(5)转变为主晶相。由于晶体尺寸均为纳米级,主晶相的转变对透过率影响较小,微晶玻璃的可见光透过率均高于85%。主晶相的转变有效增强了微晶玻璃的机械性能,其弯曲强度由300 MPa提升至331 MPa。Na_(2)O的引入有效增强了Na-K交换,Na_(2)O含量为4%(质量分数)的Li 2O-Al_(2)O_(3)-SiO_(2)微晶玻璃在410℃的KNO_(3)熔盐中交换6 h后,维氏硬度由7.108 GPa提升至7.403 GPa,弯曲强度由331 MPa提升至470 MPa。 展开更多

为研究不同H5亚型高致病性禽流感病毒(H5 HPAIV)的HA和NA匹配度对流感假病毒组装和功能的影响,本研究将5株H5 HPAIV的HA和NA基因节段分别克隆于pCMV/R载体,构建3个HA和5个NA重组质粒,两两随机组合后将HA、NA质粒与pCMV/RΔ8.9质粒、p HR... 为研究不同H5亚型高致病性禽流感病毒(H5 HPAIV)的HA和NA匹配度对流感假病毒组装和功能的影响,本研究将5株H5 HPAIV的HA和NA基因节段分别克隆于pCMV/R载体,构建3个HA和5个NA重组质粒,两两随机组合后将HA、NA质粒与pCMV/RΔ8.9质粒、p HR-CMV-Luc质粒构成4质粒系统,利用磷酸钙法将4个质粒转染HEK293T细胞,检测各细胞培养上清液血凝效价,结果显示:有3个上清液无血凝效价,其余12个上清液的血凝效价在100 HAU/m L~400 HAU/m L,表明获得12株具有血凝效价的流感假病毒。超速离心后获得浓缩假病毒,通过western blot检测假病毒HA蛋白的表达,结果显示各假病毒均在75 ku、50 ku和25 ku有特异性条带。利用各假病毒分别感染MDCK.1细胞60 h后裂解细胞,采用荧光素酶试验检测假病毒的相对荧光素酶活性(RLA),结果显示各假病毒RLA在5.0×10~1~4.5×10~5不等。上述结果首次表明不同H5 HPAIV的HA和NA相互匹配会对假病毒的血凝效价和相对荧光酶活性(RLA)造成影响。本研究为后续流感病毒灭活疫苗制备及抗病毒药物的研发奠定基础。 展开更多

为了解浙江省H9N2亚型禽流感病毒(AIV)血凝素(HA)和神经氨酸酶(NA)基因的分子特征和遗传变异情况,本试验对2012—2021年分离到的14株H9N2亚型AIV分离株进行HA和NA基因序列测定和分析。结果显示:14株分离株的HA和NA基因均属于Y280分支,H... 为了解浙江省H9N2亚型禽流感病毒(AIV)血凝素(HA)和神经氨酸酶(NA)基因的分子特征和遗传变异情况,本试验对2012—2021年分离到的14株H9N2亚型AIV分离株进行HA和NA基因序列测定和分析。结果显示:14株分离株的HA和NA基因均属于Y280分支,HA核苷酸同源性为91.5%~99.4%,NA核苷酸同源性为89.0%~98.7%,其与疫苗株CK/SS/94、CK/F/98、CK/6/96和CK/1/00的HA核苷酸同源性为86.0%~91.1%。14株分离株HA蛋白裂解位点氨基酸组成均为RSSR↓GLF;受体结合位点主要表现为K141N、A142T、T155N、H183N和V190T突变,其中第226位和第228位全部为L和G,可与α-2,6唾液酸受体结合,具有感染人的特性;蛋白潜在糖基化位点均为6个,主要变异表现在第200~202位1个位点缺失和第295~297位1个位点增加;抗原相关位点主要表现为G82E、S137D、D145G、N159G、A160D、T192R和N193D突变,此外,有5株出现D160N突变。14株分离株NA蛋白存在6个潜在糖基化位点,其中1株发生N86H突变,缺失1个潜在糖基化位点;均出现第61~63位颈部氨基酸缺失。结果表明,浙江省H9N2亚型AIV流行毒株不断变异,与疫苗株存在抗原性差异,需加强病毒变异监控和新疫苗研发。 展开更多
