Topical Application of Cudrania tricuspidata Stem Extract Inhibits Atopic Dermatitis-Like Skin Lesions in an NC/Nga Mouse Model: An Experimental Animal Study

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by elevated immunoglobulin E (IgE), mast cell infiltration and skin lesions including pruritus, erythema and eczema. Cudrania tricuspidata extracts have been clinically administered for a long time in the East Asia including Korean and China as a home-remedy to diminish the inflammation of gastritis and hepatitis. To examine whether it works on AD or not, an AD-like animal model was experimented in this study. AD was induced by applying Dermatophagoides farinae (D. farinae) extract to the backs of 9-week old NC/Nga mice for 21 days. Following this, an ethanol extract of C. tricuspidata stems (EECT) was applied topically for 14 days to the sensitized skin, while distilled water was used as a control (EECT0 mice). Anti-AD effects of EECT were evaluated using scores for AD-like skin lesions, serum IgE levels and mast cell counts in the skin dermal layers to assess inflammation. Topically applied ethanol extract of Cudrania tricuspidata stems (EECT 7.5, 25 and 75 mg/mL) markedly reduced AD-like skin lesions after 4 days (by 30.1%, 31.4% and 38.5%, respectively) and also after 14 days (by 63.6%, 66.1% and 49.6%, respectively), while distilled water improved AD by 17.8% and 38.7%, respectively (p D. farinae extract (p = 0.003) and EECT attenuated the mast cell overproduction, and reduced mast cell degranulation markedly. Objective:To explore the effect of bacilli Galmette-Gurin(BCG)-polysaccharide nuceic acid on atopic dermatitis in mice and its mechanism.Methods:Forty NC/Nga mice were selected and randomly divided into Group A(model group),Group B(dexamethasone treatment group),Group C(BCG polysaccharide nucleic acid treatment group) and Group D(control group) with 10 mice in each group.Atopic dermatitis model were constructed by applying 2,4-dinitrochlorobenzene on the skin of the mice.Mice in Group D were treated with acetone solution(100 μ L) on the foot pad and abdomen after hair removal at the age of 7 weeks.then on ear skin at the age of 8-13 weeks.For mice in A,B and C groups,100 μL of acetone solution containing 2,4-dinitrochlorobenzene was applied to the foot pad and the abdomen at the age of 7 weeks,then on ear skins at the age of8 to 13 weeks.At the age of 7-13 weeks,mice in Group A and Group D were treated with 100 μL saline(i.p.);mice were given dexamethasone(0.1 mL/kg,i.p.) every other day for 7 weeks in Group B;mice were treated with BCG polysaccharide nucleic acid(0.5 mg/kg,i.p.) every other day for7 weeks in Group C.The ear thickness was measured every week and the scratching frequency was recorded 1 times for 10 min a week.The mice were sacrificed after the last administration of drugs,IgE,IL-4,IL-10,IL-I2 and IFN- γ in the plasma were detected using ELISA,and RT-PCR method was employed to detect the concentrations of IL-4,IL-10,IL-12 and IFN- γ proteins.After IIK staining,the lesion degree of inflammation in ear tissue was observed microscopically.Results:The ear thickness and scratching frequency of Group A were significantly higher than those in group B,C and D(P<0.05),and there was no significant difference between Group B and C(P>0.05);the concentrations of IgE,IL-4 and IL-10 in the plasma and the expression of IL-4,IL-10 mRNA in the spleen tissues of Group A,B and C were all significantly higher than those of Group D(P<0.05);the concentrations of plasma IL-12 and IFN- γ,and spleen protein expression of IL-12 and IFN- γ in Group C mice were significantly higher than those of Group A(P<0.05).Histological observation showed obvious ear tissue exudation,erythema,swelling,desquamation of skin,and scabbing in Group A.Histopathology of the skin lesion also showed hyperkeratosis,focal-parakeratosis,stratum spinosum hypertrophy,mild sponge-like edema,a large number of lymphocytes along with plasma cell infiltration in dermis,angiectasis and hyperemia in Group A,while degree of ear skin lesion in Group B and D mice was significandy lighter than that of Group A.Conclusions:BCC polysaccharide nucleic acid can significandy reduce the serum IgE concentrations,increase the expression of IL-12,IFN- γ protein,correct the imbalance of Th1/Th2 in atopic dermatitis mice,and has obvious inhibitory effect on atopic dermatitis in NC/Nga mice. 展开更多

目的 观察卡介菌多糖核酸(BCG-PSN)对二硝基氯苯(DNCB)诱导的Nc/Nga小鼠特应性皮炎样皮损的影响.方法 15只Nc/Nga小鼠随机分成3组,每组5只.第1周开始,对照组在小鼠足垫及除毛之后的腹部外涂丙酮并腹腔注射生理氯化钠溶液100μl,模... 目的 观察卡介菌多糖核酸(BCG-PSN)对二硝基氯苯(DNCB)诱导的Nc/Nga小鼠特应性皮炎样皮损的影响.方法 15只Nc/Nga小鼠随机分成3组,每组5只.第1周开始,对照组在小鼠足垫及除毛之后的腹部外涂丙酮并腹腔注射生理氯化钠溶液100μl,模型组外涂丙酮配置的5%DNCB并腹腔注射生理氯化钠溶液100μl,治疗组外涂丙酮配置的5%DNCB并腹腔注射100 mg/L的BCG/PSN 100μl,三组均为隔日给药1次,共7周.第2周开始,用丙酮配置的0.1%DNCB涂抹在小鼠双侧耳部和颈部皮肤,每周1次,共6周.通过小鼠耳朵厚度、组织病理学、免疫学指标评价皮炎损伤程度,观察BCG-PSN对小鼠的治疗作用.结果 外用DNCB可以引起Ne/Nga小鼠湿疹样皮炎的产生,模型组小鼠出现皮肤干燥、红斑、水肿、脱毛和糜烂,组织病理学显示表皮和真皮增厚、过度角化和炎症细胞浸润,小鼠血清IgE(174.72±12.64μg/L)和IL-4(91.49±6.32 ng/L)增高,与对照组IgE(17.32±3.56μg/L)和IL-4(83.95±6.63 ng/L)相比,P值均〈0.05;而治疗组小鼠血浆IL-12(122.10±4.64 ng/L)和IFN-γ(73.89±2.39 ng/L)增加,IgE(84.27±9.35 μg/L)下降,与模型组IL-12(20.14±6.15 ng/L)、IFN-γ/(51.53±3.45 ng/L)和IgE相比,差异均有统计学意义.结论 BCG-PSN可能通过促进IL-12和IFN-γ分泌,抑制IgE合成,发挥治疗Nc/Nga小鼠特应性皮炎样皮损的作用. 展开更多

背景:Nc/Nga小鼠皮损、病理和免疫学特征等均与人类特应性皮炎相一致,作为特应性皮炎动物模型其研究价值日益显现。目前国内尚无Nc/Nga小鼠的生理生化指标等相关数据报道。目的:观察特应性皮炎动物模型Nc/Nga小鼠的生长繁殖性能以及血... 背景:Nc/Nga小鼠皮损、病理和免疫学特征等均与人类特应性皮炎相一致,作为特应性皮炎动物模型其研究价值日益显现。目前国内尚无Nc/Nga小鼠的生理生化指标等相关数据报道。目的:观察特应性皮炎动物模型Nc/Nga小鼠的生长繁殖性能以及血液生理、生化指标。方法:统计Nc/Nga小鼠1-3胎的繁殖指标数据,包括每胎产仔数、离乳率、怀孕率及胎间隔。分别称取60只1-56 d Nc/Nga仔鼠的体质量,雌雄各半,绘制其生长表格。Nc/Nga鼠眼眶采血,测定其血常规和生化值。结果与结论:Nc/Nga小鼠平均交配分娩间隔为(25.8±3.1)d,平均产仔数为(7.5±2.5)只,平均离乳率为(97.2±1.2)%、平均怀孕率为(97.0±1.4)%,1-3胎间差异无显著性意义(P>0.05)。Nc/Nga小鼠的体质量随着日龄的增长而逐渐增加,断奶后一两周内增重最为迅速,6周后体质量雌雄间差异无显著性意义(P>0.05)。同一日龄的雌雄间血常规和生化值比较,雌性小鼠红细胞、血红蛋白明显高于雄性小鼠;雄性小鼠血小板计数明显高于雌性小鼠(P<0.05);雌性小鼠三酰甘油、白蛋白水平明显高于雄性小鼠(P<0.05)。提示性别和年龄都会影响Nc/Nga小鼠的血液生理和生化值指标。 展开更多

目的:观察清热除湿汤对2,4-二硝基氟苯(DNFB)诱导的NC/Nga小鼠特应性皮炎样模型的干预作用,并对其可能机制进行探讨。方法:48只NC/Nga小鼠随机分为正常对照组,模型组,阳性对照(波尼松龙)组,清热除湿高、中、低剂量组,每组8只。观察特应... 目的:观察清热除湿汤对2,4-二硝基氟苯(DNFB)诱导的NC/Nga小鼠特应性皮炎样模型的干预作用,并对其可能机制进行探讨。方法:48只NC/Nga小鼠随机分为正常对照组,模型组,阳性对照(波尼松龙)组,清热除湿高、中、低剂量组,每组8只。观察特应性皮炎小鼠模型皮损变化情况和行为学变化;HE与甲苯胺蓝染色观察皮损形态学变化;ELISA法检测血清中Ig E水平;流式细胞术检测脾脏Th1/Th2比例;RT-PCR法检测皮损中白细胞介素-4(IL-4)、干扰素γ(IFN-γ)m RNA相对表达量。结果:模型组小鼠出现搔抓增加(P<0.01),皮损红斑、水肿、糜烂、结痂并伴随血清Ig E升高(P<0.01)等AD样表现。清热除湿汤治疗可以缓解小鼠症状,包括搔抓次数减少(P<0.05),SCORAD评分降低(P<0.05),红斑、水肿、糜烂减轻,Ig E水平降低(P<0.05),组织病理学改善,肥大细胞数目减少(P<0.05),Th1/Th2比值升高(P<0.05)等。结论:清热除湿汤可以改善DNFB诱导的NC/Nga小鼠特应性皮炎样皮损,抑制肥大细胞的活化和调节Th1/Th2平衡均是其作用机制,可深入探索;高剂量组较优。 展开更多
