m^(6)A modification of lncRNA in middle ear cholesteatoma

Objective:Middle ear cholesteatoma is a non-tumorous condition that typically leads to hearing loss,bone destruction,and other severe complications.Despite surgery being the primary treatment,the recurrence rate remains high.Therefore,exploring the molecular mechanisms underlying cholesteatoma is crucial for discovering new therapeutic approaches.This study aims to explore the involvement of N6-methyladenosine(m^(6)A)methylation in long non-coding RNAs(lncRNAs)in the biological functions and related pathways of middle ear cholesteatoma.Methods:The m^(6)A modification patterns of lncRNA in middle ear cholesteatoma tissues(n=5)and normal post-auricular skin tissues(n=5)were analyzed using an lncRNA m^(6)A transcriptome microarray.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses were conducted to identify potential biological functions and signaling pathways involved in the pathogenesis of middle ear cholesteatoma.Methylated RNA immunoprecipitation(MeRIP)-PCR was used to validate the m^(6)A modifications in cholesteatoma and normal skin tissues.Results:Compared with normal skin tissues,1525 lncRNAs were differentially methylated in middle ear cholesteatoma tissues,with 1048 showing hypermethylation and 477 showing hypomethylation[fold change(FC)≥3 or<1/3,P<0.05].GO enrichment analysis indicated that hypermethylated lncRNAs were involved in protein phosphatase inhibitor activity,neuron-neuron synapse,and regulation ofα-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid(AMPA)receptor activity.Hypomethylated lncRNAs were associated with mRNA methyltransferase activity,secretory granule membrane,and mRNA methylation.KEGG analysis revealed that hypermethylated lncRNAs were mainly associated with 5 pathways:the Hedgehog signaling pathway,viral protein interaction with cytokines and cytokine receptors,mitogen-activated protein kinase(MAPK)signaling pathway,cytokine-cytokine receptor interaction,and adrenergic signaling in cardiomyocytes.Hypomethylated lncRNAs were mainly involved in 4 pathways:Renal cell carcinoma,tumor necrosis factor signaling pathway,transcriptional misregulation in cancer,and cytokine-cytokine receptor interaction.Additionally,MeRIP-PCR confirmed the changes in m^(6)A methylation levels in NR_033339,NR_122111,NR_130744,and NR_026800,consistent with microarray analysis.Real-time PCR also confirmed the significant upregulation of MAPK1 and NF-κB,key genes in the MAPK signaling pathway.Conclusion:This study reveals the m^(6)A modification patterns of lncRNAs in middle ear cholesteatoma,suggests a direction for further research into the role of lncRNA m^(6)A modification in the etiology of cholesteatoma.The findings provide potential therapeutic targets for the treatment of middle ear cholesteatoma. 展开更多

This study conducted a 16-week comparative experiment with 160 students from Jinan No.5 Middle School in Shandong Province as the experimental subjects.During the implementation of the experiment,the subjects were divided into two groups:an experimental group and a control group.The experimental group was subjected to physical exercise interventions using sports game teaching methods during physical education class,while the control group followed traditional physical education teaching methods.The results showed that the psychological health status of the students in the experimental group was significantly better than that of the control group after the experiment.The application of sports games helps to improve the psychological health of middle school students in the course of physical education,promoting a positive and forward direction in student psychology and is an effective means to enhance the psychological health level of middle school students. 展开更多

In middleschool English teaching,grammar instruction has always been an important and challenging aspect.Due to the strong influence of grammar explanation in Chinese,learning English grammar may be affected by negative transfer from their native language,which could pose certain obstacles to English acquisition.Self-efficacy,as a major emotional factor influencing learners'internal regulation strategies,has attracted wide attention from researchers both domestically and internationally for its impact on learning performance.High self-efficacy helps learners develop a more positive mindset towards learning,thereby contributing to improved learning performance.Therefore,this study focuses on investigating the relationship between grammatical self-efficacy and learning performance of English grammar among middle school students.This research contributes to a complementary understanding of grammatical self-efficacy among educators,enhancing their teaching skills,and effectively guiding students to learn English grammar with a more positive and healthy mindset,presenting a future grammar instruction in middle school English teaching. 展开更多

Investigation of NSSI loca... Background Non-suicidal self-injury (NSSI) behaviour is very common among adolescents. Its prevalence and behavioural characteristics may vary according to regional and cultural differences. Investigation of NSSI locations and diagnosis of adolescents with NSSI are relatively lacking in China. Aims The study objective was to determine the prevalence and features of NSSI among middle school students in Shanghai. Methods The participants were from grade 6 to 8 selected from three junior schools in Jing’an District. Consenting students completed the Ottawa Self-Injury Inventory to determine the prevalence and characteristics of NSSI. Those who indicated NSSI within the past month were administered the Mini-International Neuropsychiatric Interview for Children and Adolescents to assess for emotional disorders. Result The result shows 510 participants (21.7%;mean age 13.51 (0.97) years old;56.7% female) reported at least one instance of NSSI during the previous 12 months. NSSI was significantly more common in girls than boys (24.9% vs 18.5%;χ^2=14.03, p=0.00). Commonly reported reasons for NSSI were for internal and external emotion regulation (87.9%, 82.3%) and social influence (57.8%). Anxiety disorders were the most common (28.9%) disorder associated with NSSI. Conclusion The rate of NSSI of middle school students in Shanghai inner bound is similar to those reported in North American and European youth. It is essential that school mental health professionals are aware of how to manage NSSI within the school setting. 展开更多

目的评估450 nm蓝激光半导体治疗机6点位置定位法治疗中叶增生型良性前列腺增生(BPH)的疗效及安全性,以期推广该术式的临床应用。方法回顾性分析2023年3月—8月在泰安市中心医院迎胜院区泌尿肾病激光治疗中心行450 nm蓝激光半导体治疗机... 目的评估450 nm蓝激光半导体治疗机6点位置定位法治疗中叶增生型良性前列腺增生(BPH)的疗效及安全性,以期推广该术式的临床应用。方法回顾性分析2023年3月—8月在泰安市中心医院迎胜院区泌尿肾病激光治疗中心行450 nm蓝激光半导体治疗机6点位置定位法治疗20例中叶增生型BPH患者的临床资料,记录手术时间、术后膀胱冲洗时间、尿管留置时间、术后住院时间、手术并发症。比较患者术前及术后1个月最大尿流率(Qmax)、膀胱残余尿量(PVR)、生活质量评分(QoL)、国际前列腺症状评分(IPSS)变化情况。结果本组患者手术时间为(26.80±7.22)min,术后膀胱冲洗时间为(20.50±1.79)h,所有患者术后次日拔除导尿管,术后2 d顺利出院。与术前比较,患者术后1个月的Qmax显著升高[(7.40±1.05)mL/s vs.(19.60±1.76)mL/s]、PVR[(73.50±12.26)mL vs.(9.25±4.94)mL]、QoL[(4.55±1.19)分vs.(1.95±0.95)分]、IPSS[(26.55±1.88)分vs.(10.05±1.36)分]均显著降低,差异具有统计学意义(P<0.05)。术中及术后1个月随访期间无任何并发症发生。结论450 nm蓝激光半导体治疗机6点位置定位法治疗中叶增生型BPH是一种新型、安全、有效的手术方式,值得在临床推广应用。 展开更多

目的:分析探讨淄博市周村区中小学生脊柱侧弯检出率及相关影响因素,为开展脊柱侧弯异常早期干预和防治提供依据。方法:2023年2—3月对淄博市周村区6 395名中小学生进行脊柱侧弯筛查和问卷调查。结果:6 395名中小学生脊柱侧弯检出率为12.... 目的:分析探讨淄博市周村区中小学生脊柱侧弯检出率及相关影响因素,为开展脊柱侧弯异常早期干预和防治提供依据。方法:2023年2—3月对淄博市周村区6 395名中小学生进行脊柱侧弯筛查和问卷调查。结果:6 395名中小学生脊柱侧弯检出率为12.38%,男、女生脊柱侧弯检出率比较,差异无统计学意义(P>0.05);不同年级学生脊柱侧弯检出率比较,差异有统计学意义(χ^(2)=140.24,P<0.01),学生脊柱侧弯检出率随年级增长呈逐渐增高的趋势。小学、初中、职业高中学生脊柱侧弯检出率比较,差异有统计学意义(χ^(2)=94.58,P<0.01)。脊柱侧弯异常程度以姿势不良、脊柱Ⅰ度弯曲为主,脊柱侧弯异常发生部位以胸段、胸腰段为主,随年级、学习阶段提高呈胸段—腰段—胸腰段异常率递增的趋势。实行年度脊柱侧弯筛查制度,采取脊柱侧弯综合防控干预措施后,脊柱侧弯异常率显著降低。结论:儿童青少年脊柱侧弯以非结构性脊柱侧弯为主,应严格执行未病先防、定期筛查、抓早抓小、已病早纠、协同管理原则,采取脊柱侧弯综合防控措施降低脊柱侧弯检出率,进一步提高儿童青少年的身心健康素质和卫生保健水平。 展开更多

The triptych (Smartphones, Video games, adolescents) has imposed itself on the collective consciousness in the form of questioning to the point of becoming a social phenomenon. It thus raises concerns about the uses that adolescents make of it and the effects on their academic performance, which we proposed to study among middle school students from the Dakar academy inspection. Where appropriate, we used mixed methods with the collection techniques of questionnaire survey, semi-structured interviews respectively with middle school students, adults (parents, supervisors, teachers, etc.), participant observation and literature review. Concretely, before accessibility to video games, the middle school students were more idle and well-behaved, watched a lot of TV, played with their brothers and sisters or did household chores. The majority of young people had good, very good or excellent conduct and their averages were fair, fairly good or good. With access to a diverse digital environment, middle school students have passionately turned to video games. As a result, their learning time, concentration and submission to parental injunctions have declined significantly. This situation negatively affected their academic performance and encouraged bad behavior. 展开更多

为考察学校氛围对3-6年级留守儿童焦虑的影响机制以及心理素质和自尊在其间的作用,采用学校氛围量表、小学生心理素质问卷、自尊量表和广泛性焦虑量表对安徽省3451名3-6年级留守儿童进行集体施测。结果发现:(1)学校氛围、心理素质与自... 为考察学校氛围对3-6年级留守儿童焦虑的影响机制以及心理素质和自尊在其间的作用,采用学校氛围量表、小学生心理素质问卷、自尊量表和广泛性焦虑量表对安徽省3451名3-6年级留守儿童进行集体施测。结果发现:(1)学校氛围、心理素质与自尊两两间显著正相关(r=0.39-0.54,p<0.001),学校氛围、心理素质、自尊分别与焦虑显著负相关(r=-0.17--0.16,p<0.001)。(2)学校氛围能有效预测3-6年级留守儿童心理素质(β=0.96,p<0.001)、自尊(β=0.06,p<0.001)和焦虑(β=-0.04,p<0.001)。(3)3-6年级留守儿童心理素质和自尊在学校氛围对焦虑的影响过程中起多重中介作用,其效应值占比总效应的12.5%。因此,营造良好的学校氛围,提高3-6年级留守儿童心理素质和自尊水平是减少3-6年级留守儿童焦虑的有效途径。 展开更多

To the middle school environment,combining education with scenery and environment is particularly important.The study described that during the landscape design of middle school,the planners should grasp the characteristics of school culture.To arrange the layout of school reasonably,designers should be familiar with the psychological and behavioral characteristics of middle school students with considering of the natural elements around the school.Landscape design needs to respect natural environment,taking cost savings,and the features of the school traffic into account.With the pursuit of natural,fresh,simple,and bright color style,landscape design in middle school should retain the feature of school history and the unique humanity environment.The whole planning is best to meet the public aesthetic,avoiding of blind pursuit of the novel,unique,and particular design style.Designers should also work hard at the design conception.With the help of modern language and materials,positive elements should be added into the school environment with the enlightenment of education,which highlight the appeal of school environment and the sense of the times. 展开更多
