目的:了解non-HDL-C和remnant cholesterol与肥胖儿童亚临床动脉粥样硬化(AS)的关系。方法:根据甘油三酯(TG)水平将65例肥胖儿童(肥胖组)分为I组(TG≥2.3 mmol/L)32例,II组(TG<2.3 mmol/L)33例,与肥胖组儿童性别年龄相匹配的正常体... 目的:了解non-HDL-C和remnant cholesterol与肥胖儿童亚临床动脉粥样硬化(AS)的关系。方法:根据甘油三酯(TG)水平将65例肥胖儿童(肥胖组)分为I组(TG≥2.3 mmol/L)32例,II组(TG<2.3 mmol/L)33例,与肥胖组儿童性别年龄相匹配的正常体质量儿童30例为III组(对照组)。比较3组儿童血脂指标,对肥胖组各项血脂指标与血浆致动脉粥样硬化指数(AIP)进行相关性分析,肥胖患儿体质指数、血脂指标与颈动脉内径、内膜-中层厚度(c-IMT)、血流参数进行相关性分析。结果:I组、II组的TG、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、non-HDL-C、remnant cholesterol、AIP均较对照组升高,高密度脂蛋白胆固醇(HDL-C)较对照组降低,且I组的remnant cholesterol、AIP均较II组升高。肥胖组儿童AIP与TG、non-HDL-C、remnant cholesterol呈正相关,与HDL-C呈负相关。37例行颈动脉超声检查的肥胖儿童的c-IMT较正常儿童的增厚。结论:non-HDL-C、remnant cholesterol在肥胖儿童亚临床AS中有重要作用,在肥胖儿童血脂异常的治疗中应关注non-HDL-C、remnant cholesterol的变化。 展开更多

Objective: To evaluate the clinical efficacy of Shenqi Fuzheng injection combined with gemcitabine plus cisplatin(GP) in the treatment of advanced non-small cell lung cancer (NSCLC). Methods: we performed a systematicsearch in the electronic databases such as Cochrane Library, Pubmed, Embase, Chinese Journal Full-text Database,Chinese Biomedical Literature Database, Chinese Science and Technology Periodical Full-text Database andWanfang Database up to 30 January 2017. Randomized controlled trials (RCT) of Shenqi Fuzheng Injectioncombined with GP chemotherapy in the treatment of advanced NSCLC were searched, and all the RCTs wereconducted on methodological quality assessment. Data extraction and data analysis were according to standards ofCochrane systematic review. Results: Eight trials were included including a total of 701 patients. Meta-analysisresults: Shenqi Fuzheng injection combined with GP chemotherapy could significantly improve the functionalstatus of patients with NSCLC (OR = 3.44, 95% CI [2.26, 5.25], P 〈 0.0001) and clinical treatment efficacy (OR =(OR = 0.31, 95%CI [0.20, 0.47], P 〈 0.0001. The rate of leukopenia (OR = .31, 95%CI [0.20,0.47], P 〈 0.0001),thrombocytopenia (OR = 0.58, 95%CI [0.37, 0.91], P = 0.020), hemoglobin decline ((OR = 0.31, 95%CI [0.16,0.59], P = 0.0004) and incidence of gastrointestinal reactions (OR = 0.58,P 〈 0.05) could be reduced. Conclusion:Shenqi Fuzheng injection combined with GP chemotherapy in the treatment of advanced NSCLC obtainedsignificantly clinical efficacy. The quality of the literature incorporated is low, the conclusion requires high-qualityresearch to further prove. 展开更多

It was found that SeB had the capacity to inhibit the metabolic growth of Escherichia coli. But in different sol-vent, the extent of inhibition is different. Removing the effect of solvent, we obtained the half-inhibition conce.ntra-tion(IC50) of SeB from the power-time curves. The value of IC50 of SeB was 1319μg ·mL-1. 展开更多

CYFRA 21-1 as an early predictor of first line chemotherapy response in advanced non small cell lung cancer

Objective: In an era of ever evolving, promising new therapies for advanced non small cell lung cancer (NSCLC), early predictors of response to therapy, are needed. We evaluated early variations in CYFRA 21-1 serum levels of patients with advanced NSCLC receiving first line chemotherapy and correlated the results with objective tumor response. Methods: 29 consecutive, previously untreated, patients of advanced non small cell lung cancer, with measurable disease on CT scan were evaluated. All patients were treated with conventional systemic chemotherapy, although the choice of chemotherapy was left to the discretion of the treating physicians. Serum samples were obtained immediately before the start of 1st and 2nd cycles of chemotherapy. CYFRA 21-1 was measured with an electrochemiluminescense immunoassay on an automatic analyzer (Elecsys 2000; Roche Diagnostics). Response was evaluated using Response evaluation criteria in solid tumors (RECIST) criteria. Results: 10 patients had partial response, 9 patients had stable disease and 9 had progressive disease. None of the patients had complete response. 21/29 (72%) patients had an elevated baseline value of CYFRA 21-1.62% patients (18/29) had a decrease in CYFRA 21-1 after 1 cycle of chemotherapy. The average reduction in the 2nd reading was irrespective of whether baseline value was normal or not. The average reduction was statistically significant (P = 0.002; 95% CI, from 0.8369 to 3.49464; t test). 8 out of 10 (80%) patients with partial response had a reduction in their 2nd reading of. CYFRA (P = 0.019; 95% CI, from 0.81965 to 7.20035; t test) which was significant. We also observed that 6/9 (66%) patients whose disease remains stable also had a decrease in their subsequent reading (P = 0.0106; 95% CI, from -0.44942 to 3.82720; t test), though it was not significant statistically. Although 5 out of 9 (55%) patients, who had an increase in their CYFRA 21-1 level, had progressive disease, but it was not statistically significant (P = 0.537; 95% CI, from -1.20021 to 2.13354; ttest). 14 out of 19 (73%) who either had partial response or had stable disease, had a reduction in their 2nd value of CYFRA 21-1 and was significant statistically (P = 0.004; 95% CI, from 0.74792 to 3.50208; t test). We also observed that except for 1 patient, all patients who had a decrease of 42% or more in their subsequent CYFRA 21-1 level, were those who had either responded to chemotherapy or had stable disease (P = 0.001), which was statistically significant. Conclusion: We can conclude that monitoring of serum marker CYFRA 21-1, early dudng first-line chemotherapy may be a useful prognostic tool for evaluation of early tumor response in patients with advanced NSCLC. 展开更多

Increasing evidence indicates that aberrant expressions of some microRNAs are associated with cancer progression.However,the roles and biological mechanisms of miRNA-16-5p in human non-small cell lung cancer(NSCLC)are not to be well studied.Here,we validated that the expression of miR-16-5p was decreased significantly in NSCLC samples and cell lines.The correlation between the clinicopathological features of NSCLC and the miR-16-5p expression showed that the expression of miR-16-5p in non-small cell lung cancer was linked with the advanced TNM stage,positive lymph node metastasis,with short overall survival(OS).Also,a negative correlation between miR-16-5p and Fermitin family member 2(FERMT2)was observed,implying there may be a potential link about their regulation.The hypothesis was further confirmed by in-silico analysis and dual-luciferase reporter assay.Moreover,we demonstrated that the transfections of miR-16-5p mimics could alter some biological characteristics of NSCLC cells remarkably accomplished by the expression variance of FERMT2 in vitro and in vivo assays.Summarily,this study demonstrated that miR-16-5p,as a tumor suppression factor in NSCLC by targeting FERMT2,could serve as one promising biomarker in the prediction for NSCLC patients. 展开更多

In this paper, we propose a novel game-theoretical solution to the multi-path routing problem in wireless ad hoc networks comprising selfish nodes with hidden information and actions. By incorporating a suitable traffic allocation policy, the proposed mechanism results in Nash equilibria where each node honestly reveals its true cost, and forwarding subgame perfect equilibrium in which each node does provide forwarding service with its declared service reliability. Based on the generalised second price auction, this mechanism effectively alleviates the over-payment of the well-known VCG mechanism. About 75% of all bladder cancer diagnosed are non-muscle invasive bladder cancer(NMIBC), recurring over 50% of them after transurethral resection of the bladder tumor. In order to prevent recurrences, adjuvant intravesical chemotherapy with mitomycin C and immunotherapy with bacillus Calmette-Gu-érin(BCG) is traditionally used. Unfortunately, many patients relapse after receiving these treatments and a significant proportion of them require surgery. After a one-to-three years BCG maintenance, the risk for progression at 5 years was 19.3% for T1G3 tumors. Many new treatment approaches are being investigated to increase the effectiveness of adjuvant intravesical therapy. One of the developing treatments for intermediate and high-risk NMIBC is the combination of intravesical chemotherapy and hyperthermia, called chemohyperthermia. This article provides a review of the mechanism of action, current status and indications, results and future perspectives. 展开更多

目的基于沉默信息调节因子1(SIRT1)调控的炎症和凋亡相关信号通路,探讨当飞利肝宁胶囊改善四氯化碳(CCl4)诱导的非酒精性脂肪性肝病(NAFLD)大鼠急性肝损伤的作用机制。方法雄性Wistar大鼠按体质量随机分为正常(N)组、NAFLD模型(HF)组、... 目的基于沉默信息调节因子1(SIRT1)调控的炎症和凋亡相关信号通路,探讨当飞利肝宁胶囊改善四氯化碳(CCl4)诱导的非酒精性脂肪性肝病(NAFLD)大鼠急性肝损伤的作用机制。方法雄性Wistar大鼠按体质量随机分为正常(N)组、NAFLD模型(HF)组、正常加CCl4(N-CCl4)组、NAFLD加CCl4(HF-CCl4)组和当飞利肝宁胶囊(DF)组。N和N-CCl4组大鼠给予普通饲料,HF、HF-CCl4组及DF组均给予高脂饲料,DF组同时予当飞利肝宁胶囊(0.3 g·kg^(-1))每天灌胃干预。8周后,除N和HF组外,其余组大鼠腹腔注射小剂量CCl4。48 h后取大鼠血清和肝组织。检测指标包括:①通过苏木精-伊红(HE)染色观察肝组织病理,检测血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST);②通过实时荧光定量逆转录聚合酶链式反应(RT-qPCR)法检测肝组织促炎因子白介素-1β(IL^(-1β))、单核细胞趋化蛋白1(MCP1)及肿瘤坏死因子-α(TNF-α)的mRNA水平,同时采用Western blot法检测肝组织组蛋白-赖氨酸N-甲基转移酶(SUV39H2)、SIRT1、乙酰化核因子-κB亚基p65(Acetyl-p65)、乙酰化肿瘤抑制蛋白p53(Acetyl-p53)、剪切型多聚ADP核糖多聚酶(Cleaved-PARP)及凋亡相关分子p53上调凋亡调节因子(PUMA)、Bcl-2相关蛋白(Bcl-xL)的蛋白表达水平。结果①HF组大鼠肝组织出现广泛肝细胞脂肪变性,HF-CCl4组除此之外还出现大量肝细胞气球样变性、炎症细胞浸润和凋亡小体,DF组大鼠肝组织脂肪变性、气球样变和炎症浸润程度均显著减轻。小剂量CCl4未导致肝组织异常改变。血清ALT、AST水平在各组大鼠中的变化趋势同肝组织病理改变,在HF组比N、N-CCl4组上调(P<0.05),在HF-CCl4组又较HF组增加(P<0.05),而在DF组上述转氨酶下调(P<0.05)。②肝组织SIRT1水平在HF组和HF-CCl4组大鼠低于N组(P<0.05),DF组SIRT1水平显著提高(P<0.05);SUV39H2表达水平变化则与SIRT1趋势相反(P<0.05)。大鼠肝组织Acetyl-p65水平及促炎因子IL^(-1β)、MCP1及TNF-αmRNA在HF组和HF-CCl4组较N组升高(P<0.05),而在DF组的表达较HF-CCl4组下调(P<0.05)。HF组和HF-CCl4组大鼠肝组织Acetyl-p53及PUMA、Cleaved-PARP高于N组(P<0.05),而Bcl-xL蛋白则相反(P<0.05);与HF组和HF-CCl4组比较,DF组逆转了Acetyl-p53、PUMA、Cleaved-PARP及Bcl-xL蛋白表达。结论当飞利肝宁胶囊可改善毒性物质诱导的NAFLD大鼠急性肝损伤,其机制可能与上调SIRT1抑制核因子-κB(NF-κB)和p53介导的炎症、凋亡信号通路有关。 展开更多

目的运用超高压液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术结合网络药理学技术探究参葛颗粒治疗非酒精性脂肪性肝病的活性成分及作用机制。方法通过UPLC-Q-TOF-MS/MS技术鉴定参葛颗粒中的化学成分,结合中药系统药理学数据... 目的运用超高压液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术结合网络药理学技术探究参葛颗粒治疗非酒精性脂肪性肝病的活性成分及作用机制。方法通过UPLC-Q-TOF-MS/MS技术鉴定参葛颗粒中的化学成分,结合中药系统药理学数据库(TCMSP)、PubChem数据库以及SwissTargetPrediction数据库筛选参葛颗粒的活性成分和靶点,在GeneCards等数据库检索非酒精性脂肪性肝病的疾病靶点,在Venny 2.1.0在线作图平台绘制药物-疾病韦恩图,将交集靶点导入String数据库,再利用Cytoscape 3.10.0软件进行可视化处理,同时基于交集靶点通过DAVID数据库进行GO和KEGG通路富集。利用Cytoscape 3.10.0软件构建核心靶点网络图以及疾病-通路-靶点-成分-药物网络图。结果质谱解析化合物75个,筛选得到药物活性成分29个,药物靶点499个,疾病靶点4776个,共同靶点353个,关键基因10个,分别为TP53、CASP3、HIF1A、JUN、BCL2L1、SRC、MTOR、HRAS、TNF、ALB,关键活性成分8个,分别为丹酚酸B、葛根素、Puerol B、芒柄花苷、葛苷A、丹参素、大豆苷、染料木素苷。结论参葛颗粒治疗非酒精性脂肪性肝病具有多成分、多靶点、多通路的特点,主要通过减少肝脏脂肪堆积、调节内分泌减缓炎症以及细胞凋亡,从而发挥治疗非酒精性脂肪肝病的作用。 展开更多

目的研究17β-雌二醇(17β-E2)对子宫内膜异位症(内异症)患者在位子宫内膜间质细胞胞外信号调节激酶1/2(ERK1/2)信号转导通路活化的影响,探讨ERK1/2信号转导通路在介导雌激素促进内异症发生发展中的作用。方法体外分离培养内异症... 目的研究17β-雌二醇(17β-E2)对子宫内膜异位症(内异症)患者在位子宫内膜间质细胞胞外信号调节激酶1/2(ERK1/2)信号转导通路活化的影响,探讨ERK1/2信号转导通路在介导雌激素促进内异症发生发展中的作用。方法体外分离培养内异症患者在位子宫内膜间质细胞。用1×10^-6mol/L 17β-E2处理子宫内膜间质细胞不同时间(0~120 min),然后用MTT法和免疫印迹法(Western blot)检测子宫内膜间质细胞的活性和ERK1/2的活化情况,检测出子宫内膜间质细胞活性最强的时间点。用Western blot和MTT法分别检测不同浓度ERK1/2抑制剂PD98059对1×10^-6mol/L 17β-E2作用20 min后子宫内膜间质细胞活性及ERK1/2活性的变化。结果 1×10^-6mol/L的17β-E2作用20 min时,子宫内膜间质细胞的活性最强,ERK1/2活化在17-βE2作用10 min后出现,在20 min时达高峰;随着PD98059作用浓度的增加,ERK1/2活化水平逐渐下降,浓度为50μmol/L时已被完全阻断;随着PD98059作用浓度的增加,子宫内膜间质细胞活性逐渐下降,在50μmol/L和100μmol/L时,活性最低,但此时仍高于空白对照组。结论 17β-E2增强子宫内膜间质细胞的活性可能与17β-E2通过非转录机制,迅速激活子宫内膜间质细胞的ERK1/2信号转导通路有关。 展开更多

目的观察西黄胶囊对非哺乳期乳腺炎大鼠模型创面愈合及白细胞介素-6(interleukin-6,IL-6)/非受体型酪氨酸蛋白激酶2(Janus kinase 2,JAK2)/信号转导及转录激活蛋白3(signal transducer and activator of transcription 3,STAT3)信号通... 目的观察西黄胶囊对非哺乳期乳腺炎大鼠模型创面愈合及白细胞介素-6(interleukin-6,IL-6)/非受体型酪氨酸蛋白激酶2(Janus kinase 2,JAK2)/信号转导及转录激活蛋白3(signal transducer and activator of transcription 3,STAT3)信号通路的影响。方法复制非哺乳期乳腺炎大鼠模型,随机分为模型对照组、阳性药物组、中药治疗组、联合用药组,每组10只,另选取10只同批次SPF级健康SD大鼠作为假手术组。干预后观察并评估各组大鼠的创面愈合情况及乳腺炎症指数,HE染色法观察各组大鼠乳腺组织的病理改变;免疫组织化学法检测各组大鼠乳腺组织B淋巴细胞瘤-2基因关联X蛋白(B-cell lymphoma-2-associated X,BAX)、B淋巴细胞瘤-2基因(B-cell lymphoma-2,Bcl-2)的平均光密度值;Western blot法及RT-PCR法检测各组大鼠乳腺组织IL-6、JAK2、STAT3、人胱天蛋白酶9(Caspase-9)蛋白及mRNA的表达水平。结果与假手术组大鼠比较,模型对照组大鼠的创面愈合率明显降低(P<0.05),而乳腺炎症指数,乳腺组织BAX、Bcl-2平均光密度值,IL-6、JAK2、STAT3、Caspase-9蛋白及mRNA的表达水平显著升高(P<0.05)。HE染色可见模型对照组大鼠乳腺腺叶存在明显的肉芽肿变性,周围充斥大量炎症细胞,阳性药物组、中药治疗组、联合用药组大鼠的病理情况存在不同程度的缓解,炎性细胞浸润及肉芽肿组织明显减少,“空泡样”坏死存在不同程度的愈合。与模型对照组比较,阳性药物组、中药治疗组、联合用药组的创面愈合率、Bcl-2光密度值均明显升高(P<0.05);其中联合用药组的创面愈合率、Bcl-2光密度值明显高于阳性药物组及中药治疗组(P<0.05);阳性药物组的创面愈合率高于中药治疗组(P<0.05),Bcl-2光密度值与中药治疗组比较差异无统计学意义(P>0.05)。与模型对照组比较,阳性药物组、中药治疗组、联合用药组的BAX光密度值、乳腺炎症指数均明显下降(P<0.05);其中联合用药组的BAX光密度值、乳腺炎症指数明显低于阳性药物组及中药治疗组(P<0.05);阳性药物组的乳腺炎症指数低于中药治疗组(P<0.05),BAX光密度值与中药治疗组比较差异无统计学意义(P>0.05)。与模型对照组比较,阳性药物组、中药治疗组、联合用药组的IL-6、JAK2、STAT3、Caspase-9蛋白及mRNA的表达水平均明显降低(P<0.05);其中联合用药组上述蛋白及mRNA表达水平明显低于阳性药物组及中药治疗组(P<0.05),而阳性药物组的上述蛋白及mRNA表达水平均明显高于中药治疗组(P<0.05)。结论西黄胶囊能有效促进非哺乳期乳腺炎大鼠模型的乳腺组织创面愈合,抑制乳腺组织细胞凋亡及炎症反应,其作用机制可能与西黄胶囊抑制IL-6/JAK2/STAT3信号通路相关。 展开更多
