This study measured platelet intracellular free calcium (PFCa 2+ ) of 1003 normal pregnancies in different trimester. The purpose of this study was to find the alteration of PFCa 2+ in normal pregnancy, to provide scientific basis for forecasting and preventing PIH (pregnancy induced hypertension). Preeclampsia is still a major cause of maternal death in pregnancy which needs efforts to prevent and early detection. Nitric oxide and Corticotrophin Releasing Hormone has an important role in the pathogenesis of preeclampsia to determine differences in concentrations of NO and CRH in normal pregnancy and preeclampsia, as well as to study the correlation both on the incidence of preeclampsia. This is a cross sectional study. Blood samples obtained from patients with preeclampsia (30 samples) and normal pregnancies as controls (30 samples) in accordance with inclusion and exclusion criteria. NO concentration was examined with essay protocol, and CRH concentrations with ELISA method. Then be calculated by statistical analysis: chi square, t test, Mann-Whitney, and Spearman Rank correlation coefficient. Based on statistical analysis, there was no significant difference (p > 0.05) from the characteristics of maternal age, pregnancy weeks, and parity in both study groups, so the two groups of homogeneous and comparable. The mean concentration of NO in preeclampsia was lower (3.780 μM) compared with normal pregnancy (13.360 μM). There was significant correlation between NO and CRH concentrations in normal pregnancy and preeclampsia (p < 0.001). The involvement of NO/c-GMP pathway is very possible in the pathogenesis of preeclampsia. 展开更多

There are increasing evidences for the presence of an endogenous digitalis-like substance (EDLS) which interacts with the digitalis receptor and acts against the digoxin serum response in human body. The plasma EDLS level increases in response to fluid or salt loading. EDLS may play a role in salt and blood pressure regulation. Being considered as a volume-expanding state, pregnancy may cause an increase in maternal EDLS. However, further studies to identify the origin of EDLS in normal pregnancy are needed. 展开更多

Piwi-interacting RNAs(piRNAs)is a novel class of non-coding RNAs.However,changes in piRNA expression profiles in recurrent spontaneous abortion(RSA)have not yet been investigated.The aim of this study was to identify differentially expressed piRNAs in deciduas of RSA patients.Decidua tissues were collected by curettage from recruited RSA patients and normal early pregnant(NEP)women with their informed consent.Small RNA sequencing was used to evaluate the differences in piRNA expression profiles between RSA and NEP.The present results demonstrated that the counts of total piRNA reads in RSA samples were increased compared with those in NEP samples(0.21%vs.0.11%).Differential expression analysis identified 29 upregulated piRNAs and 18 downregulated piRNAs in RSA samples.RT-qPCR further confirmed that the expression levels of uniq-109625,uniq-89328,uniq-50651 and uniq-4569 were decreased in 8 RSA tissues,compared with 13 NEP tissues.Otherwise,pi-22628 and uniq-173406 were increased in 8 RSA tissues.Based on GO term and KEGG pathway analysis,we speculate that these piRNAs regulate RSA by targeting extracellular matrix component pathway,cell adhesion pathway and focal adhesion pathway.PiRNAs may be involved in RSA pathogenesis by target genes function on adhesion and extracellular matrix component. 展开更多
