AIM: To identify the effect of Houttuynia cordata Thunb(HCT) on lipopolysaccharide(LPS)-induced microglial activation and investigate its possible molecular mechanisms. METHODS: The primary retinal microglial cells were cultured from the retinas of newborn Sprague-Dawley rats and exposed to LPS, and/or HCT with different concentrations. The survival ability of retinal microglia cells was tested by standard MTT method. BrdU cell proliferation assay was used to evaluate the proliferation of retinal microglia. Inflammatory factors in the culture supernatants, including TNF-α, iNOS and IL-1β, were measured using ELISA. Microglia cells’ migration was determined with Transwell migration assay. The total p38-MAPK and phosphorylation of p38-MAPK(p-p38-MAPK) were detected with Western blot.RESULTS: Primary retinal microglia in culture exposed to LPS to induce microglia activation. Pretreatment with HCT significantly inhibited the LPS-induced cell proliferation, but not the cell viability. LPS induced inflammatory reaction in microglia and cell migration. HCT significantly reduced LPS-stimulated release of pro-inflammatory factors and decreased the number of migrating cells substantially in a concentration-dependent manner. Moreover, the protein levels of p-p38 MAPK were identified as the up regulation and co-treatment with HCT obviously inhibited the upregulation of p-p38 MAPK, but had no effect on the levels of total p38-MAPK.CONCLUSION: The data suggest that HCT inhibits LPSinduced retinal microglial activation via suppression of the p-p38-MAPK. HCT may be used for the treatment of ocular diseases characterized by over-activated microglia. 展开更多

An in-depth mechanism in zonal activation of CO_(2)and H2molecular over dual-active sites has not been revealed yet.Here,Ni-Co-MgO was rationally constructed to elucidate the CO_(2)methanation mechanism. The abundant surface nickel and cobalt components as active sites led to strong Ni-Co interaction with charge transfer from nickel to cobalt.Notably,electron-enriched Coδ-species participated in efficient chemisorption and activation of CO_(2)to generate monodentate carbonate.Simultaneously, plentiful available Ni0sites facilitated H2dissociation,thus CO_(2)and H2were smoothly activated at zones of Coδ-species and Ni0,respectively.Detailed in situ DRIFTS,quasi situ XPS,TPSR,and DFT calculations substantiated a new formate evolution mechanism via monodentate carbonate instead of traditional bidentate carbonate based on synergistic catalysis of Coδ-species and Ni0.The zonal activation of CO_(2)and H2by tuning electron behaviors of double-center catalysts can boost heterogeneous catalytic hydrogenation performance. 展开更多

Background: Harvesting activities have proven effective in reducing stress in patients with dementia. At a psychiatric hospital, occupational therapists (OT) made patients with dementia perform vegetable harvesting activities as a part of their treatment. The patients became calmer and began to smile not only after but also before and during harvesting activity. Therefore, this study aimed to measure autonomic nervous system (ANS) responses over time in a female patient in her 80s with moderate Alzheimer’s disease (AD) who had experience in farming when performing harvesting activities. Additionally, this study aimed to consider factors associated with changes in ANS responses over time and to verify the physiological therapeutic effects of harvesting activities. Methods: An OT with more than 10 years of experience and a good relationship with the patient conducted one-on-one sessions with the patient. Harvesting activity was performed in a courtyard with trees. The patient harvested lettuce (three lettuce plants) grown in a planter while sitting on a chair. Additionally, the planter was set up on a desk. ANS responses were measured over time from before to after the activity for different events at six time periods. Changes in the patient’s ANS responses were assessed during each period. Results: With the involvement of environmental factors and OT during harvesting activities, parasympathetic nervous system (PNS) activity significantly increased. This may lead to improved mental stability. Conclusion: The findings of this study indicate that implementing harvesting activities may have a positive effect on the PNS in patients with AD with long-term agricultural experience. However, further studies with a larger sample size and multidimensional evaluations are needed. 展开更多

Cross-training is a phenomenon related to motor learning, where motor performance of the untrained limb shows improvement in strength and skill execution following unilateral training of the homologous contralateral limb. We used functional MRI to investigate whether motor performance of the untrained limb could be improved using a serial reaction time task according to motor sequential learning of the trained limb, and whether these skill acquisitions led to changes in brain activation patterns. We recruited 20 right-handed healthy subjects, who were randomly allocated into training and control groups. The training group was trained in performance of a serial reaction time task using their non-dominant left hand, 40 minutes per day, for 10 days, over a period of 2 weeks. The control group did not receive training. Measurements of response time and percentile of response accuracy were performed twice during pre- and post-training, while brain functional MRI was scanned during performance of the serial reaction time task using the untrained right hand. In the training group, prominent changes in response time and percentile of response accuracy were observed in both the untrained right hand and the trained left hand between pre- and post-training. The control group showed no significant changes in the untrained hand between pre- and post-training. In the training group, the activated volume of the cortical areas related to motor function (i.e., primary motor cortex, premotor area, posterior parietal cortex) showed a gradual decrease, and enhanced cerebellar activation of the vermis and the newly activated ipsilateral dentate nucleus were observed during performance of the serial reaction time task using the untrained right hand, accompanied by the cross-motor learning effect. However, no significant changes were observed in the control group. Our findings indicate that motor skills learned over the 2-week training using the trained limb were transferred to the opposite homologous limb, and motor skill acquisition of the untrained limb led to changes in brain activation patterns in the cerebral cortex and cerebellum. 展开更多

This study examined the influence of semantic and phonological priming on L2 speech planning,as well as the difference between native and non-native speakers of English in terms of lemma activation. Two potential explanations for the contrast between the performance of L2 speakers and native controls were considered. The first of which was that L2 speakers’ phonological forms are activated before selection of syntactic frame occurred, whereas the reverse is true for native speakers. The second explanation posits that the organisation of the speech production procedure is fundamentally similar in native and L2 speakers, and the disparity in performance arises from difference in the levels of activation of stored items. The results of the present experiment suggest that lemma selection is indeed what drives syntactic frame selection. However, lemmas in L2 speakers can be primed through a chain of connections demonstrated as:L2 phonological form →L1 phonological form → L1 lemma. 展开更多

The polysaccharides in yam bulbils were extracted, and their antioxidant activity and hypoglycemic effect in diabetic mice were discussed. The results showed that the antioxidant activity of polysaccharides in yam bulbils was signifi- cantly enhanced with the increase of concentration; they showed a strong scaveng- ing ability against DPPH. and .OH, and the scavenging ability was dose dependent to some extent; the scavenging rates reached 91.15% and 89.06% respectively when the dose reached 4.0 mg/ml; the polysaccharides in yam bulbils significantly educed the blood glucose in model rice, and the hypoglycemic effect of large-dose polysaccharides was more obvious. The polysaccharides in yam bulbils has good antioxidant activity and hypoglycemic effect, which provides a new source for devel- opment of safe and natural food antioxidants and blood sugar-lowering agents. 展开更多

This study focuses on drawing a hydrothermal synthesis process map for Co3O4 nanoparticles with various morphologies and investigating the effects of Co3O4 nanocatalyst morphology on CO oxidation.A series of cobalt-hydroxide-carbonate nanoparticles with various morphologies(i.e.,nanorods,nanosheets,and nanocubes) were successfully synthesized, and Co3O4 nanoparticles were obtained by thermal decomposition of the cobalt-hydroxide-carbonate precursors. The results suggest that the cobalt source is a key factor for controlling the morphology of cobalt-hydroxide-carbonate at relatively low hydrothermal temperatures(≤ 140℃).Nanorods can be synthesized in CoCl2 solution, while Co(NO3)2 solution promotes the formation of nanosheets.Further increasing the synthesis temperature(higher than 140 ℃) results in the formation of nanocubes in either Co(NO3)2 or CoCl2 solution. The reaction time only affects the size of the obtained nanoparticles. The presence of CTAB could improve the uniformity and dispersion of particles.Co3O4 nanosheets showed much higher catalytic activity for CO oxidation than nanorods and nanocubes because it has more abundant Co^(3+) on the surface, much higher reducibility, and better oxygen desorption capacity. 展开更多

Traditional PID controllers are no longer suitable formagnetic-bearing-supported high-speed flywheels with significant gyroscopic effects. Becausegyroscopic effects greatly influence the stability of the flywheel rotor, especially at highrotational speeds. Velocity cross feedback and displacement cross feedback are used to overcomeharmful effects of nutation and precession modes, and stabilize the rotor at high rotational speeds.Theoretical analysis is given to show their effects. A control platform based on RTLinut and a PCis built to control the active magnetic bearing (AMB) system, and relevant results are reported.Using velocity cross feedback and displacement cross feedback in a closed loop control system, theflywheel successfully runs at over 20000 r/min. 展开更多

The in vitro and in vivo effects of corn peptides(CPs) prepared from corn gluten meal by proteolysis with an alkaline protease and fractions of CPs from Sephadex G-15 and G-10 columns on activities of alcohol dehydroge-nase(ADH) were studied. The results show that CPs and fraction 3 of CPs from Sephadex G-10 column enhance in vitro ADH activity. Furthermore, the in vitro accelerating effect of the fraction 3 of CPs on ADH activity was superior to that of glutathione, which was also found even in the presence of ADH inhibitor, such as pyrazole. In the in vivo experiments, the animals were fed with different dosages of CPs and with a dose of Chinese distilled spirit orally, and sacrificed for the measurement of ADH activity. In vivo experimental results indicate that CPS enhanced hepatic ADH activities. To test the safety of CPs as health food, 30 d feeding test was performed. No obvious toxic effects were detected in treated Wistar rats. 展开更多

BACKGROUND Study shows that signal transducer and activator of transcription 3(STAT3) can increase the Warburg effect by stimulating hexokinase 2 in breast cancer and upregulate lactate dehydrogenase A and pyruvate dehydrogenase kinase 1 in myeloma. STAT3 and pyruvate kinase M2(PKM2) can also be activated and enhance the Warburg effect in hepatocellular carcinoma. Precancerous lesions are critical to human and rodent hepatocarcinogenesis. However, the underlying molecular mechanism for the development of liver precancerous lesions remains unknown. We hypothesized that STAT3 promotes the Warburg effect possibly by upregulating p-PKM2 in liver precancerous lesions in rats.AIM To investigate the mechanism of the Warburg effect in liver precancerous lesions in rats.METHODS A model of liver precancerous lesions was established by a modified Solt-Farber method. The liver pathological changes were observed by HE staining and immunohistochemistry. The transformation of WB-F344 cells induced with Nmethyl-N'-nitro-N-nitrosoguanidine and hydrogen peroxide was evaluated by the soft agar assay and aneuploidy. The levels of glucose and lactate in the tissue and culture medium were detected with a spectrophotometer. The protein levels of glutathione S-transferase-π, proliferating cell nuclear antigen(PCNA), STAT3,and PKM2 were examined by Western blot and immunofluorescence.RESULTS We found that the Warburg effect was increased in liver precancerous lesions in rats. PKM2 and p-STAT3 were upregulated in activated oval cells in liverprecancerous lesions in rats. The Warburg effect, p-PKM2, and p-STAT3 expression were also increased in transformed WB-F344 cells. STAT3 activation promoted the clonal formation rate, aneuploidy, alpha-fetoprotein expression,PCNA expression, G1/S phase transition, the Warburg effect, PKM2 phosphorylation, and nuclear translocation in transformed WB-F344 cells. Moreover, the Warburg effect was inhibited by stattic, a specific inhibitor of STAT3, and further reduced in transformed WB-F344 cells after the intervention for PKM2.CONCLUSION The Warburg effect is initiated in liver precancerous lesions in rats. STAT3 activation promotes the Warburg effect by enhancing the phosphorylation of PKM2 in transformed WB-F344 cells. 展开更多

Seedling quality is a prerequisite for successful field performance and therefore influences crop yields. Temperature and illumination are two major factors affecting seedling quality during nursery propagation. Suboptimal temperature or light of nurseries generally result in leggy or weak seedlings and great economic loss. However, production of healthy seedlings is challenging due to the lack of knowledge in systemic management of nursery environments. In this study, we have established simulation models to predict how temperature and illumination coordinately influence the growth of tomato and cabbage seedlings. Specifically, correlation between seedling quality characteristics(root-shoot ratio, G value(growth function: defined as ratio of whole plant dry weight to days of seedling), healthy indexes) and TEP(thermal effectiveness and photosynthetically active radiation) were explored to establish the models, which were validated with independent test data. Our results suggested that the curve of healthy index 1(HI1) and TEP fitted well with high coefficient of determination(R2) in both species, indicating that the model is highly reliable. The HI1 simulation models for tomato and cabbage are HI1=0.0009e0.0308TEP-0.0015 and HI1= 0.0003e0.0671TEP-0.0003, respectively, which can be used for predicting vigors of tomato and cabbage seedlings grown under different temperature and light conditions. 展开更多

Three large-scale episodes of volcanic activity occurred during the Tertiary in the Xihu Depression, located in the East China Sea. Intermediate-felsic magmas intruded along faults and the associated hydrothermal fluids resulted in the hydrothermal alteration of the clastic country rock. To better describe reservoir characteristics, reservoir samples were subjected to the following investigations: thin section examination, scanning electron microscope-energy dispersive spectrometer analysis(SEM–EDS), fluid inclusion homogenization temperature tests, vitrinite reflectance measurements, and X-ray di raction. The results of this study provide evidence of the following hydrothermal alteration phenomena: brittle fracturing, clastic particle alteration, precipitation of unique hydrothermal minerals(celestite, zircon, apatite, barite, and cerous phosphate). The presence of abnormally high temperatures is indicated by fluid inclusion analysis, the precipitation of high-temperature authigenic minerals such as quartz, illite alteration, and anomalous vitrinite reflectance. Two aspects related to hydrothermal effects on reservoir properties have been investigated in this study:(1) Deep magmatic hydrothermal fluids carry large amounts of dissolved carbon dioxide and sulfur dioxide gas. These fluids percolate into the country rocks along fault zones, resulting in dissolution within the sandstone reservoirs and the development of significant secondary porosity.(2) Magma intrusions increase the temperature of the surrounding rocks and accelerate the thermal evolution of hydrocarbon source rocks. This results in the release of large amounts of organic acids and carbon dioxide, leading the dissolution of the aluminosilicate minerals and volcanic fragments in the reservoirs, and the generation of significant secondary porosity. 展开更多

Searching alternative feed antibiotics is always a study hotspot in the field of animal production. In this study, the antipathogenic activities and probiotics stimulatory effects of 30 kinds of herbs were screened through Oxford cup method and Hungate roll method. 15 herbs showed significantly antibacterial activities(P〈0.05) against Escherichia coli, Salmonella enteritidis, Salmonella typhimurium and Staphylococcus aureus. Seven herbs showed greatly stimulatory promoting effects on Lactobacillus acidophilus and Bifidobacterium longum. Finally, five herbs were combined for the developed formulation with selective antibacterial properties and they were Mume Fructus(35%), Isatidis Folium(25%), Moslae Herba(20%), Chrysanthemi Indici Flos(13%) and Bupleuri Radix(7%). The herbal formulation showed significantly antibacterial abilities against four pathogens and stimulatory promoting abilities on two probiotics in vitro and the equivalent activities in broiler chickens in vivo against E. coli and L. acidophilus. The toxicity study showed it had no toxicity, which indicated that it would be a kind of preferred candidate for an alternative antibiotic in future animal production. 展开更多
