METHODS: The primary retinal microglial cells were cultured from the retinas of newborn Sprague-Dawley rats and exposed to LPS, and/or HCT with different concentrations. The survival ability of retinal microglia cells was tested by standard MTT method. BrdU cell proliferation assay was used to evaluate the proliferation of retinal microglia. Inflammatory factors in the culture supernatants, including TNF-α, iNOS and IL-1β, were measured using ELISA. Microglia cells’ migration was determined with Transwell migration assay. The total p38-MAPK and phosphorylation of p38-MAPK(p-p38-MAPK) were detected with Western blot.RESULTS: Primary retinal microglia in culture exposed to LPS to induce microglia activation. Pretreatment with HCT significantly inhibited the LPS-induced cell proliferation, but not the cell viability. LPS induced inflammatory reaction in microglia and cell migration. HCT significantly reduced LPS-stimulated release of pro-inflammatory factors and decreased the number of migrating cells substantially in a concentration-dependent manner. Moreover, the protein levels of p-p38 MAPK were identified as the up regulation and co-treatment with HCT obviously inhibited the upregulation of p-p38 MAPK, but had no effect on the levels of total p38-MAPK.CONCLUSION: The data suggest that HCT inhibits LPSinduced retinal microglial activation via suppression of the p-p38-MAPK. HCT may be used for the treatment of ocular diseases characterized by over-activated microglia.展开更多

At a psychiatric hospital, occupational therapists (OT) made patients with dementia perform vegetable harvesting activities as a part of their treatment. The patients became calmer and began to smile not only after but also before and during harvesting activity. Therefore, this study aimed to measure autonomic nervous system (ANS) responses over time in a female patient in her 80s with moderate Alzheimer’s disease (AD) who had experience in farming when performing harvesting activities. Additionally, this study aimed to consider factors associated with changes in ANS responses over time and to verify the physiological therapeutic effects of harvesting activities. Methods: An OT with more than 10 years of experience and a good relationship with the patient conducted one-on-one sessions with the patient. Harvesting activity was performed in a courtyard with trees. The patient harvested lettuce (three lettuce plants) grown in a planter while sitting on a chair. Additionally, the planter was set up on a desk. ANS responses were measured over time from before to after the activity for different events at six time periods. Changes in the patient’s ANS responses were assessed during each period. Results: With the involvement of environmental factors and OT during harvesting activities, parasympathetic nervous system (PNS) activity significantly increased. This may lead to improved mental stability. Conclusion: The findings of this study indicate that implementing harvesting activities may have a positive effect on the PNS in patients with AD with long-term agricultural experience. However, further studies with a larger sample size and multidimensional evaluations are needed.展开更多

We used functional MRI to investigate whether motor performance of the untrained limb could be improved using a serial reaction time task according to motor sequential learning of the trained limb, and whether these skill acquisitions led to changes in brain activation patterns. We recruited 20 right-handed healthy subjects, who were randomly allocated into training and control groups. The training group was trained in performance of a serial reaction time task using their non-dominant left hand, 40 minutes per day, for 10 days, over a period of 2 weeks. The control group did not receive training. Measurements of response time and percentile of response accuracy were performed twice during pre- and post-training, while brain functional MRI was scanned during performance of the serial reaction time task using the untrained right hand. In the training group, prominent changes in response time and percentile of response accuracy were observed in both the untrained right hand and the trained left hand between pre- and post-training. The control group showed no significant changes in the untrained hand between pre- and post-training. In the training group, the activated volume of the cortical areas related to motor function (i.e., primary motor cortex, premotor area, posterior parietal cortex) showed a gradual decrease, and enhanced cerebellar activation of the vermis and the newly activated ipsilateral dentate nucleus were observed during performance of the serial reaction time task using the untrained right hand, accompanied by the cross-motor learning effect. However, no significant changes were observed in the control group. Our findings indicate that motor skills learned over the 2-week training using the trained limb were transferred to the opposite homologous limb, and motor skill acquisition of the untrained limb led to changes in brain activation patterns in the cerebral cortex and cerebellum.展开更多

The results show that CPs and fraction 3 of CPs from Sephadex G-10 column enhance in vitro ADH activity. Furthermore, the in vitro accelerating effect of the fraction 3 of CPs on ADH activity was superior to that of glutathione, which was also found even in the presence of ADH inhibitor, such as pyrazole. In the in vivo experiments, the animals were fed with different dosages of CPs and with a dose of Chinese distilled spirit orally, and sacrificed for the measurement of ADH activity. In vivo experimental results indicate that CPS enhanced hepatic ADH activities. To test the safety of CPs as health food, 30 d feeding test was performed. They are commercially valuable and prized as a food and folk medicine in Asia. Nutritionally, sea cucumbers have an impressive profile of valuable nutrients such as vitamins, minerals and amino acids. A number of unique biological and pharmacological activities/properties, including anticancer, anticoagulant/antithrombotic, antimicrobial, antioxidant, antihyperlipidemic, antihyperglycemic, anti-inflammatory, antihypertension and radioprotective, have been ascribed to various compounds isolated from sea cucumbers. The therapeutic properties and medicinal benefits of sea cucumbers can be linked to the presence of a wide array ofbioactives, especially triterpene glycosides, acid mucopolysaccharide, sphingoid bases, glycolipids, fucosylated chondroitin sulfate, polysaccharides, phospholipids, cerebrosides, phosphatidylcholines, and other extracts and hydrolysates. This review highlights the valuable bioactive components as well as the multiple therapeutic properties of sea cucumbers with a view to exploring their potential uses as functional foods and a natural source of new multifunctional drugs.展开更多

STAT3 and pyruvate kinase M2(PKM2) can also be activated and enhance the Warburg effect in hepatocellular carcinoma. Precancerous lesions are critical to human and rodent hepatocarcinogenesis. However, the underlying molecular mechanism for the development of liver precancerous lesions remains unknown. We hypothesized that STAT3 promotes the Warburg effect possibly by upregulating p-PKM2 in liver precancerous lesions in rats.AIM To investigate the mechanism of the Warburg effect in liver precancerous lesions in rats.METHODS A model of liver precancerous lesions was established by a modified Solt-Farber method. The liver pathological changes were observed by HE staining and immunohistochemistry. The transformation of WB-F344 cells induced with Nmethyl-N'-nitro-N-nitrosoguanidine and hydrogen peroxide was evaluated by the soft agar assay and aneuploidy. The levels of glucose and lactate in the tissue and culture medium were detected with a spectrophotometer. The protein levels of glutathione S-transferase-π, proliferating cell nuclear antigen(PCNA), STAT3,and PKM2 were examined by Western blot and immunofluorescence.RESULTS We found that the Warburg effect was increased in liver precancerous lesions in rats. PKM2 and p-STAT3 were upregulated in activated oval cells in liverprecancerous lesions in rats. The Warburg effect, p-PKM2, and p-STAT3 expression were also increased in transformed WB-F344 cells. STAT3 activation promoted the clonal formation rate, aneuploidy, alpha-fetoprotein expression,PCNA expression, G1/S phase transition, the Warburg effect, PKM2 phosphorylation, and nuclear translocation in transformed WB-F344 cells. Moreover, the Warburg effect was inhibited by stattic, a specific inhibitor of STAT3, and further reduced in transformed WB-F344 cells after the intervention for PKM2.CONCLUSION The Warburg effect is initiated in liver precancerous lesions in rats. STAT3 activation promotes the Warburg effect by enhancing the phosphorylation of PKM2 in transformed WB-F344 cells.展开更多

Temperature and illumination are two major factors affecting seedling quality during nursery propagation. Suboptimal temperature or light of nurseries generally result in leggy or weak seedlings and great economic loss. However, production of healthy seedlings is challenging due to the lack of knowledge in systemic management of nursery environments. In this study, we have established simulation models to predict how temperature and illumination coordinately influence the growth of tomato and cabbage seedlings. Specifically, correlation between seedling quality characteristics(root-shoot ratio, G value(growth function: defined as ratio of whole plant dry weight to days of seedling), healthy indexes) and TEP(thermal effectiveness and photosynthetically active radiation) were explored to establish the models, which were validated with independent test data. Our results suggested that the curve of healthy index 1(HI1) and TEP fitted well with high coefficient of determination(R2) in both species, indicating that the model is highly reliable. The HI1 simulation models for tomato and cabbage are HI1=0.0009e0.0308TEP-0.0015 and HI1= 0.0003e0.0671TEP-0.0003, respectively, which can be used for predicting vigors of tomato and cabbage seedlings grown under different temperature and light conditions.展开更多

Intermediate-felsic magmas intruded along faults and the associated hydrothermal fluids resulted in the hydrothermal alteration of the clastic country rock. To better describe reservoir characteristics, reservoir samples were subjected to the following investigations: thin section examination, scanning electron microscope-energy dispersive spectrometer analysis(SEM–EDS), fluid inclusion homogenization temperature tests, vitrinite reflectance measurements, and X-ray di raction. The results of this study provide evidence of the following hydrothermal alteration phenomena: brittle fracturing, clastic particle alteration, precipitation of unique hydrothermal minerals(celestite, zircon, apatite, barite, and cerous phosphate). The presence of abnormally high temperatures is indicated by fluid inclusion analysis, the precipitation of high-temperature authigenic minerals such as quartz, illite alteration, and anomalous vitrinite reflectance. Two aspects related to hydrothermal effects on reservoir properties have been investigated in this study:(1) Deep magmatic hydrothermal fluids carry large amounts of dissolved carbon dioxide and sulfur dioxide gas. These fluids percolate into the country rocks along fault zones, resulting in dissolution within the sandstone reservoirs and the development of significant secondary porosity.(2) Magma intrusions increase the temperature of the surrounding rocks and accelerate the thermal evolution of hydrocarbon source rocks. This results in the release of large amounts of organic acids and carbon dioxide, leading the dissolution of the aluminosilicate minerals and volcanic fragments in the reservoirs, and the generation of significant secondary porosity.展开更多

In this study, the antipathogenic activities and probiotics stimulatory effects of 30 kinds of herbs were screened through Oxford cup method and Hungate roll method. 15 herbs showed significantly antibacterial activities(P〈0.05) against Escherichia coli, Salmonella enteritidis, Salmonella typhimurium and Staphylococcus aureus. Seven herbs showed greatly stimulatory promoting effects on Lactobacillus acidophilus and Bifidobacterium longum. Finally, five herbs were combined for the developed formulation with selective antibacterial properties and they were Mume Fructus(35%), Isatidis Folium(25%), Moslae Herba(20%), Chrysanthemi Indici Flos(13%) and Bupleuri Radix(7%). The herbal formulation showed significantly antibacterial abilities against four pathogens and stimulatory promoting abilities on two probiotics in vitro and the equivalent activities in broiler chickens in vivo against E. coli and L. acidophilus. The toxicity study showed it had no toxicity, which indicated that it would be a kind of preferred candidate for an alternative antibiotic in future animal production.展开更多

HCT may be used for the treatment of ocular diseases characterized by over-activated microglia.

This review highlights the valuable bioactive components as well as the multiple therapeutic properties of sea cucumbers with a view to exploring their potential uses as functional foods and a natural source of new multifunctional drugs.

文摘BACKGROUND Study shows that signal transducer and activator of transcription 3(STAT3) can increase the Warburg effect by stimulating hexokinase 2 in breast cancer and upregulate lactate dehydrogenase A and pyruvate dehydrogenase kinase 1 in myeloma. STAT3 activation promotes the Warburg effect by enhancing the phosphorylation of PKM2 in transformed WB-F344 cells.

文摘Seedling quality is a prerequisite for successful field performance and therefore influences crop yields. The HI1 simulation models for tomato and cabbage are HI1=0.0009e0.0308TEP-0.0015 and HI1= 0.0003e0.0671TEP-0.0003, respectively, which can be used for predicting vigors of tomato and cabbage seedlings grown under different temperature and light conditions.

文摘Three large-scale episodes of volcanic activity occurred during the Tertiary in the Xihu Depression, located in the East China Sea. This results in the release of large amounts of organic acids and carbon dioxide, leading the dissolution of the aluminosilicate minerals and volcanic fragments in the reservoirs, and the generation of significant secondary porosity.

文摘Searching alternative feed antibiotics is always a study hotspot in the field of animal production. The toxicity study showed it had no toxicity, which indicated that it would be a kind of preferred candidate for an alternative antibiotic in future animal production.
