Exercise-induced modulation of miR-149-5p and MMP9 in LPS-triggered diabetic myoblast ER stress: licorice glycoside E as a potential therapeutic target

Background:This study explores the relationship between endoplasmic reticulum(ER)stress and diabetes,particularly focusing on the impact of physical exercise on ER stress mechanisms and identifying potential therapeutic drugs and targets for diabetes-related sepsis.The research also incorporates traditional physical therapy perspectives,emphasizing the genomic insights gained from exercise therapy in disease management and prevention.Methods:Gene analysis was conducted on the GSE168796 and GSE94717 datasets to identify ER stress-related genes.Gene interactions and immune cell correlations were mapped using GeneCard and STRING databases.A screening of 2,456 compounds from the TCMSP database was performed to identify potential therapeutic agents,with a focus on their docking potential.Techniques such as luciferase reporter gene assay and RNA interference were used to examine the interactions between microRNA-149-5p and MMP9.Results:The study identified 2,006 differentially expressed genes and 616 miRNAs.Key genes like MMP9,TNF-α,and IL1B were linked to an immunosuppressive state.Licorice glycoside E demonstrated high affinity for MMP9,suggesting its potential effectiveness in treating diabetes.The constructed miRNA network highlighted the regulatory roles of MMP9,IL1B,IFNG,and TNF-α.Experimental evidence confirmed the binding of microRNA-149-5p to MMP9,impacting apoptosis in diabetic cells.Conclusion:The findings highlight the regulatory role of microRNA-149-5p in managing MMP9,a crucial gene in diabetes pathophysiology.Licorice glycoside E emerges as a promising treatment option for diabetes,especially targeting MMP9 affected by ER stress.The study also underscores the significance of physical exercise in modulating ER stress pathways in diabetes management,bridging traditional physical therapy and modern scientific understanding.Our study has limitations.It focuses on the microRNA-149-5p-MMP9 network in sepsis,using cell-based methods without animal or clinical trials.Despite strong in vitro findings,in vivo studies are needed to confirm licorice glycoside E’s therapeutic potential and understand the microRNA-149-5p-MMP9 dynamics in real conditions. 展开更多

BACKGROUND MicroRNAs(miRNAs)regulate gene expression and play a critical role in cancer physiology.However,there is still a limited understanding of the function and regulatory mechanism of miRNAs in gastric cancer(GC).AIM To investigate the role and molecular mechanism of miRNA-145-5p(miR145-5p)in the progression of GC.METHODS Real-time polymerase chain reaction(RT-PCR)was used to detect miRNA expression in human GC tissues and cells.The ability of cancer cells to migrate and invade was assessed using wound-healing and transwell assays,respectively.Cell proliferation was measured using cell counting kit-8 and colony formation assays,and apoptosis was evaluated using flow cytometry.Expression of the epithelial-mesenchymal transition(EMT)-associated protein was determined by Western blot.Targets of miR-145-5p were predicated using bioinformatics analysis and verified using a dual-luciferase reporter system.Serpin family E member 1(SERPINE1)expression in GC tissues and cells was evaluated using RT-PCR and immunohistochemical staining.The correlation between SERPINE1 expression and overall patient survival was determined using Kaplan-Meier plot analysis.The association between SERPINE1 and GC progression was also tested.A rescue experiment of SERPINE1 overexpression was conducted to verify the relationship between this protein and miR-145-5p.The mechanism by which miR-145-5p influences GC progression was further explored by assessing tumor formation in nude mice.RESULTS GC tissues and cells had reduced miR-145-5p expression and SERPINE1 was identified as a direct target of this miRNA.Overexpression of miR-145-5p was associated with decreased GC cell proliferation,invasion,migration,and EMT,and these effects were reversed by forcing SERPINE1 expression.Kaplan-Meier plot analysis revealed that patients with higher SERPINE1 expression had a shorter survival rate than those with lower SERPINE1 expression.Nude mouse tumorigenesis experiments confirmed that miR-145-5p targets SERPINE1 to regulate extracellular signal-regulated kinase-1/2(ERK1/2).CONCLUSION This study found that miR-145-5p inhibits tumor progression and is expressed in lower amounts in patients with GC.MiR-145-5p was found to affect GC cell proliferation,migration,and invasion by negatively regulating SERPINE1 levels and controlling the ERK1/2 pathway. 展开更多

HPV E6/E7mRNA检测与P16/Ki-67免疫组化检测对宫颈CIN2级病变的筛查价值探究

目的:研究人乳头瘤病毒(Human papilloma virus,HPV)E6/E7mRNA与免疫组化抑癌基因P16/细胞增殖核抗原Ki-67在宫颈上皮内瘤样病变(Cervical intraepithelial neoplasia,CIN)2级病变筛查中的应用价值。方法:选取2020年2月至2023年2月期间... 目的:研究人乳头瘤病毒(Human papilloma virus,HPV)E6/E7mRNA与免疫组化抑癌基因P16/细胞增殖核抗原Ki-67在宫颈上皮内瘤样病变(Cervical intraepithelial neoplasia,CIN)2级病变筛查中的应用价值。方法:选取2020年2月至2023年2月期间本院收治的92例宫颈炎症及病变患者作为研究对象。根据病理检查结果,将CIN2级患者纳入研究组(n=45),宫颈CIN1级和宫颈炎患者纳入对照组(n=47)。对比两组HPV E6/E7mRNA、P16/Ki-67单独及联合检测阳性表达率差异,采用Logistic回归分析影响CIN2诊断的危险因素,并采用受试者工作曲线分析HPV E6/E7mRNA与P16/Ki-67单独及联合检测在宫颈CIN2级病变筛查的价值。结果:研究组HPV E6/E7mRNA、P16/Ki-67单独检测阳性率、联合检测阳性率(82.22%,77.78%,84.44%)高于对照组(63.83%,48.94%,53.19%)(P<0.05)。Logistic回归分析显示HPV E6/E7mRNA、P16/Ki-67是影响CIN2诊断的危险因素(P<0.05);HPV E6/E7mRNA、P16/Ki-67联合检测CIN2级病变的受试者工作特征曲线下面积为0.688(95%CI:0.579~0.798),优于HPV E6/E7mRNA、P16/Ki-67单独预测[0.592(95%CI:0.476~0.708);0.634(95%CI:0.519~0.748)]。联合检测的准确度、灵敏度、特异度(68.8%,84.4%,53.2%)均高于单独检测HPV E6/E7mRNA(59.2%,82.2%,36.2%)、P16/Ki-67(63.4%,77.8%,48.9%)。结论:HPV E6/E7mRNA与P16/Ki-67免疫组化检测在宫颈CIN2级病变筛查中具有一定的应用价值,可以显著提高筛查宫颈CIN2级病变的诊断效能。 展开更多

目的分析肿瘤组织的细胞周期素E(cyclin E)和p53、CD44s、CD44v6蛋白与早期乳腺癌的相关性及预后预测价值。方法选取2018年7月—2020年7月喀什地区第二人民医院收治的80例临床分期为Ⅰ~Ⅱ期的早期乳腺癌患者进行回顾性分析,将其分为恶性... 目的分析肿瘤组织的细胞周期素E(cyclin E)和p53、CD44s、CD44v6蛋白与早期乳腺癌的相关性及预后预测价值。方法选取2018年7月—2020年7月喀什地区第二人民医院收治的80例临床分期为Ⅰ~Ⅱ期的早期乳腺癌患者进行回顾性分析,将其分为恶性组;另选取同期喀什地区第二人民医院收治的80例通过病理学检验确诊为良性乳腺肿瘤患者,将其分为良性组。分别对两组患者留存的肿瘤组织进行免疫组化检测,检测cyclin E、p53、CD44s、CD44v6表达,并对比上述指标的阳性率,应用Spearman相关分析方法分析cyclin E、p53、CD44s、CD44v6与早期乳腺癌的相关性。所有患者均采取手术治疗,随后对80例早期乳腺癌患者进行3年随访,依照随访结果将患者分为2个亚组,即预后不良组(n=25)和预后良好组(n=55),对比两组患者一般临床特征及cyclin E、p53、CD44s、CD44v6阳性率,并应用Logistic回归分析cyclin E、p53、CD44s、CD44v6对早期乳腺癌预后的预测价值。结果恶性组患者cyclin E、p53、CD44s、CD44v6阳性率分别为42.50%、61.25%、77.50%、81.25%,明显高于良性组1.25%、5.00%、10.00%、12.50%(P<0.05);Spearman相关分析结果显示:cyclin E、p53、CD44s、CD44v6与早期乳腺癌呈正相关(P<0.05);预后良好组与预后不良组患者绝经情况、年龄、病理类型、治疗方式、三阴性乳腺癌、肿瘤大小、组织分化程度对比,差异无统计学意义(P>0.05),预后良好组与预后不良组患者临床分期、cyclin E、p53、CD44s、CD44v6阳性例数对比,差异有统计学意义(P<0.05);Logistic回归分析结果表明:cyclin E、p53、CD44s、CD44v6阳性表达为影响早期乳腺癌预后的独立影响因素(P<0.05)。结论cyclin E、p53、CD44s、CD44v6表达水平与乳腺癌的发生、发展具有明显关系,且上述指标呈阳性表达为早期乳腺癌预后不良的独立预测因素。 展开更多

目的:探讨液基薄层细胞学检查(TCT)联合p16/Ki-67双染、HPV E6/E7 mRNA检测在宫颈鳞状上皮内病变中的诊断价值。方法:选择在郑州大学第一附属医院病理科行细胞学筛查并在1个月内行宫颈组织病理学检查的女性患者624例,年龄18~81岁,均进行... 目的:探讨液基薄层细胞学检查(TCT)联合p16/Ki-67双染、HPV E6/E7 mRNA检测在宫颈鳞状上皮内病变中的诊断价值。方法:选择在郑州大学第一附属医院病理科行细胞学筛查并在1个月内行宫颈组织病理学检查的女性患者624例,年龄18~81岁,均进行了TCT、p16/Ki-67双染、HPV E6/E7 mRNA检测。对比病理学结果,评价各项检测方法单独或联合的诊断价值。结果:病理组织学结果显示624例患者中阴性(慢性炎症)183例,阳性441例(低级别鳞状上皮内病变176例、高级别鳞状上皮内病变227例、鳞状细胞癌38例);TCT阳性376例,阴性248例;p16/Ki-67双染阳性317例,阴性307例;HPV E6/E7 mRNA阳性360例,阴性264例;TCT+p16/Ki-67双染阳性425例,阴性199例;TCT+HPV E6/E7 mRNA阳性434例,阴性190例;3项联合阳性367例,阴性257例。以病理组织学结果为金标准,TCT+p16/Ki-67、TCT+HPV E6/E7 mRNA和3项联合诊断宫颈鳞状上皮内病变的敏感度分别为87.30%、89.11%、77.32%,特异度分别为78.14%、77.60%、86.34%。结论:TCT联合p16/Ki-67双染、HPV E6/E7 mRNA检测可提高宫颈鳞状上皮内病变的诊断效能。 展开更多

目的比较p16/Ki-67双染、HPV E6/E7 mRNA联合检测与单独检测对宫颈癌前病变的检出效能。方法收集2021—2022年接受宫颈癌前病变筛查的患者112例,液基薄层细胞检测(TCT)宫颈细胞的病变情况,并经病理学检查确诊。根据宫颈HPV E6/E7 mRNA... 目的比较p16/Ki-67双染、HPV E6/E7 mRNA联合检测与单独检测对宫颈癌前病变的检出效能。方法收集2021—2022年接受宫颈癌前病变筛查的患者112例,液基薄层细胞检测(TCT)宫颈细胞的病变情况,并经病理学检查确诊。根据宫颈HPV E6/E7 mRNA检测试剂盒检测宫颈脱落细胞的HPV E6/E7 mRNA水平,利用CINtec PLUS细胞学试剂盒检测宫颈细胞p16/Ki-67双染情况。分析p16/Ki-67双染、HPV E6/E7 mRNA单独或联合检测对宫颈癌前病变检出效能的影响。结果112例宫颈癌前病变筛查患者的病理学结果显示,低度鳞状上皮内病变(LSIL)33例,高度鳞状上皮内病变(HSIL)79例。HPV E6/E7 mRNA阳性72例(64.3%),阴性40例(35.7%);p16/Ki-67双染阳性83例(74.1%),阴性29例(25.9%)。不同癌前病变组间HPV E6/E7 mRNA表达和p16/Ki-67双染间差别均有统计学意义(P<0.05)。不同年龄段癌前病变组间差别有统计学意义(P<0.05),HPV E6/E7 mRNA表达和p16/Ki-67双染间差别无统计学意义(P>0.05)。HPV E6/E7 mRNA联合p16/Ki-67双染检测宫颈癌前病变的敏感度和准确性均高于两者单独检测,阴性预测值高于HPV E6/E7 mRNA单独检测(P<0.05)。两者联合检测HSIL的敏感度、特异度和阳性预测值均高于HPV E6/E7 mRNA单独检测,准确性和阴性预测值均高于两者单独检测。另外,p16/Ki-67双染检测HSIL的敏感度、准确性和阴性预测值均高于HPV E6/E7 mRNA单独检测(P<0.05)。结论HPV E6/E7 mRNA和p16/Ki-67双染联合检测在宫颈癌前病变检出中具有临床价值。 展开更多

目的:研究β-D-葡聚糖在宫颈癌细胞中的作用及可能机制。方法:CCK-8法检测β-D-葡聚糖对宫颈癌细胞CaSki、HeLa和SiHa细胞增殖的影响,伤口愈合试验检测细胞迁移情况,流式细胞术检测细胞周期和细胞凋亡情况。采用实时荧光定量聚合酶链式... 目的:研究β-D-葡聚糖在宫颈癌细胞中的作用及可能机制。方法:CCK-8法检测β-D-葡聚糖对宫颈癌细胞CaSki、HeLa和SiHa细胞增殖的影响,伤口愈合试验检测细胞迁移情况,流式细胞术检测细胞周期和细胞凋亡情况。采用实时荧光定量聚合酶链式反应(qRT-PCR)检测细胞中HPV16 E6/E7、HPV18 E6/E7、p53和p21 mRNA表达水平,Western blot检测N-钙黏蛋白、E-钙黏蛋白、p53、波形蛋白、p21蛋白表达水平。以宫颈癌CaSki细胞为研究对象,在雌性BALB/c小鼠皮下注射成瘤,验证β-D-葡聚糖在体内对肿瘤生长的抑制作用。结果:β-D-葡聚糖对不同宫颈癌细胞增殖的影响不同,对CaSki细胞增殖有显著的时间依赖性抑制作用,对HeLa细胞增殖无显著影响,对SiHa细胞作用72h后表现为促进增殖作用(P<0.05)。β-D-葡聚糖对CaSki、HeLa和SiHa细胞的迁移都有抑制作用(P<0.05)。β-D-葡聚糖对CaSki、SiHa的细胞周期有调节作用并能促进其凋亡(P<0.05),但对HeLa细胞无显著影响。与对照组相比,β-D-葡聚糖处理组CaSki和HeLa细胞中HPV16 E6 mRNA表达水平降低,p53和p21 mRNA表达水平在3种细胞中均升高(P<0.05)。与对照组相比,β-D-葡聚糖处理组3种细胞波形蛋白表达量降低,E-钙黏蛋白、p53和p21表达量增加(P<0.05)。结论:β-D-葡聚糖抑制宫颈癌细胞中HPV16 E6表达并上调p53和p21表达,抑制HPV16阳性宫颈癌细胞的增殖、迁移,调节细胞周期,促进细胞凋亡,并在体内发挥抗肿瘤作用。 展开更多

Selenium nanoparticles(SeNPs)have been demonstrated potential for use in diseases associated with oxidative stress.Functionalized SeNPs with lower toxicity and higher biocompatibility could bring better therapeutic activity and clinical application value.Herein,this work was conducted to investigate the protective effect of Pleurotus tuber-regium polysaccharide-protein complex funtionnalized SeNPs(PTR-SeNPs)against acetaminophen(APAP)-induced oxidative injure in HepG2 cells and C57BL/6J mouse liver.Further elucidation of the underlying molecular mechanism,in particular their modulation of Nrf2 signaling pathway was also performed.The results showed that PTR-SeNPs could significantly ameliorate APAP-induced oxidative injury as evidenced by a range of biochemical analysis,histopathological examination and immunoblotting study.PTR-SeNPs could hosphorylate and activate PKCδ,depress Keap1,and increase nuclear accumulation of Nrf2,resulting in upregulation of GCLC,GCLM,HO-1 and NQO-1 expression.Besides,PTR-SeNPs suppressed the biotransformation of APAP to generate intracellular ROS through CYP 2E1 inhibition,restoring the mitochondrial morphology.Furthermore,the protective effect of PTR-SeNPs against APAP induced hepatotoxicity was weakened as Nrf2 was depleted in vivo,indicating the pivotal role of Nrf2 signaling pathway in PTR-SeNPs mediated hepatoprotective efficacy.Being a potential hepatic protectant,PTR-SeNPs could serve as a new source of selenium supplement for health-promoting and biomedical applications. 展开更多

目的:评价p16/Ki-67双染联合HPV E6/E7 mRNA检测在宫颈病变中的筛查价值。方法:检索CNKI、万方、PubMed数据库,收集自建库至2022年5月发表的有关p16/Ki-67双染和HPV E6/E7 mRNA联合检测进行宫颈病变筛查的文献。根据纳入和排除标准进行... 目的:评价p16/Ki-67双染联合HPV E6/E7 mRNA检测在宫颈病变中的筛查价值。方法:检索CNKI、万方、PubMed数据库,收集自建库至2022年5月发表的有关p16/Ki-67双染和HPV E6/E7 mRNA联合检测进行宫颈病变筛查的文献。根据纳入和排除标准进行文献筛选,应用Stata12.0进行诊断性试验的Meta分析。结果:共纳入文献7篇,p16/Ki-67双染与HPV E6/E7 mRNA联合检测在诊断宫颈病变的合并灵敏度为0.964(95%CI 0.923~0.984),合并特异度为0.825(95%CI 0.788~0.858),合并的诊断比值比为127.623(95%CI 62.062~262.444)。联合检测的灵敏度优于单项检测,诊断比值比高于HPVE6/E7 mRNA检测,差异有统计学意义(P<0.05)。结论:在宫颈病变的诊断中,p16/Ki-67双染与HPV E6/E7 mRNA联合检测相对于单项检测灵敏度高,诊断比值比较好,特异度相当,具有较好的诊断价值,可以应用在宫颈病变筛查中。 展开更多

METHODS:Subjects of the current study included 50 cases of chronic hepatitis C(CHC) without cirrhosis,30 cases of CHC with liver cirrhosis(LC) ,and 30 cases of hepatitis C-related hepatocellular carcinoma(HCC) admitted to the Department of Hepato-Gastroenterology,Theodor Bilharz Research Institute(TBRI) ,Giza,Egypt.Fifteen wedge liver biopsies,taken during laparoscopic cholecystectomy,were also included as normal controls.Laboratory investigations including urine and stool analysis,liver function tests and prothrombin concentration;serologic markers for viral hepatitis and ultrasonography were done for all cases of the study together with immunohistochemical analysis using primary antibodies against Cyclin D1,Cyclin E,p21,p27 and Rb/p105 proteins. RESULTS:Normal wedge liver biopsies didn't express Cyclin E or Rb/p105 immunostaining but show positive staining for Cyclin D1,p21 and p27.Cyclin D1 expressed nuclear staining that was sequentially increased from CHC to LC(P<0.01) to HCC(P<0.001) cases;meanwhile,Cyclin E revealed nuclear positivity only in the case of HCCs patients that was directly correlated to Rb/p105 immuno-reactivity.The expression of p21 and p27 was significantly increased in CHC and LC cases compared to normal controls and HCCs with no significant difference between well-and poorlydifferentiated tumors.p21 showed only a nuclear pattern of staining,while,p27 presented with either cytoplasmic and/or nuclear reactivity in all studied cases.Correlation analysis revealed a direct relation between Cyclin D1 and p21 in CHC cases(P<0.001) ,between Cyclin D1 and Cyclin E in HCCs(P<0.01);however,an inverserelationship was detected between Cyclin D1 and p21 or p27(P<0.001) and between p21 and Rb/p105(P<0.05) in HCCs. CONCLUSION:Upregulation of Cyclin D1 in CHC plays a vital role in the development and differentiation of HCC;while,Cyclin E may be a useful marker for monitoring tumor behavior.p21 and p27 can be used as predictive markers for HCC.Furthermore,higher expression of Rb/p105 as well as inverse relation with p21 and histologic grades suggests its important role in hepatic carcinogenesis. 展开更多

目的 探讨TGF-β1诱导大鼠NRK-52E细胞转分化模型中miR-23b对PINK1/Parkin通路的靶向调节机制,并阐明清肾颗粒含药血清对NRK-52E细胞转分化的干预机理。方法 采用超高效液相色谱(UPLC)指纹图谱法对清肾颗粒进行全指纹图谱分析。构建TGF-... 目的 探讨TGF-β1诱导大鼠NRK-52E细胞转分化模型中miR-23b对PINK1/Parkin通路的靶向调节机制,并阐明清肾颗粒含药血清对NRK-52E细胞转分化的干预机理。方法 采用超高效液相色谱(UPLC)指纹图谱法对清肾颗粒进行全指纹图谱分析。构建TGF-β1诱导大鼠NRK-52E细胞转分化模型,转染siRNA后分为模拟物空载对照组、miR-23b-5p模拟物组、抑制剂空载对照组、miR-23b-5p抑制剂组,观察miR-23b-5p对PINK1表达量的影响。再将NRK-52E细胞分组为正常组、TGF-β1组、清肾颗粒组、miR-23b-mimic-NC组、miR-23b-mimic组、miR-23b-mimic+清肾颗粒组,Western blot法检测NRK-52E细胞中Pink1、Parkin、LC3Ⅱ、Beclin-1、P62、α-SMA蛋白表达,RT-qPCR法检测NRK-52E细胞中miR-23b-5p、Pink1、Parkin、Beclin-1、α-SMA mRNA的表达,双荧光素酶报告基因实验检测miR-23b-5p与PINK1的靶向关系。结果 UPLC指纹图谱法鉴定出清肾颗粒中11个活性成分。miR-23b-5p过表达后,PINK1 mRNA表达量也显著增加(P<0.05);而miR-23b-5p表达沉默后,PINK1 mRNA表达量也显著减少(P<0.05)。双荧光素酶报告显示,Rno-miR-23b-5p能显著下调Rno-PINK1-WT荧光素酶活性(P<0.05),但未能下调突变Rno-PINK1-mut荧光素酶活性(P>0.05)。清肾颗粒含药血清干预实验发现,TGF-β1组的miR-23b-5p、Pink1、Parkin、Beclin-1、LC3Ⅱ表达及LC3Ⅱ/Ⅰ比值均明显低于正常组,P62和α-SMA表达明显高于正常组(P<0.05)。清肾颗粒组和miR-23b-mimic组的miR-23b-5p、Pink1、Parkin、Beclin-1、LC3Ⅱ表达及LC3Ⅱ/Ⅰ比值均明显高于TGF-β1组,P62和α-SMA表达明显低于TGF-β1组(P<0.05)。miR-23b-mimic+清肾颗粒组的表现更优于miR-23b-mimic组(P<0.05)。结论 清肾颗粒能够上调NRK-52E细胞内miR-23b-5p表达,并通过增强PINK1/Parkin通路介导的线粒体自噬活性,抑制NRK-52E细胞转分化进程。 展开更多

These examples range from the practical aspect to almost esoteric considerations but at the end, everything converges towards a unity of theory and computation presented in the form of modern fractal logic and transfinite quantum field theory in a Cantorian spacetime. Certain pseudogenes may regulate their protein-coding cousins by competing for miRNAs and play an active biological role in cancer. However, few studies have focused on the association of genetic variations in pseudogenes with cancer prognosis. We selected six potentially functional single nucleotide polymorphisms (SNPs) in cancerrelated pseudogenes, and performed a case-only study to assess the association between those SNPs and the prognosis of hepatocellular carcinoma (HCC) in 331 HBV-positive HCC patients without surgical treatment. Log-rank test and Cox proportional hazard models were used for survival analysis. We found that the A allele of rs9909601 in E2F3P1 was significantly associated with a better prognosis compared with the G allele [adjusted hazard ratio (HR) = 0.69, 95% confidence interval (CI) = 0.56-0.86, P = 0.001]. Additionally, this protective effect was more predominant for patients without chemotherapy and transcatheter hepatic arterial chemoembolization (TACE) treatment. Interestingly, we also detected a statistically significant multiplicative interaction between genotypes of rs9909601 and chemotherapy or TACE status on HCC survival (P for multiplicative interaction 〈 0.001). These findings indicate that rs9909601 in the pseudogene E2F3P1 may be a genetic marker for HCC prognosis in Chinese. 展开更多

AIM:To explore the regulatory mechanism of nuclear paraspeckle assembly transcript 1(NEAT1)in the pathogenesis of posterior capsule opacification(PCO).METHODS:Quantitative reverse transcription polymerase chain reaction(RT-q PCR)was executed to analyze NEAT1 and micro RNA(miR)-26a-5p expression in transforming growth factor-beta 2(TGF-β2)-disposed lens epithelial cells(LECs).The proliferation,cell cycle progression,apoptosis,and migration of TGF-β2-disposed LECs were evaluated.The relationship between NEAT1 or fanconi anemia(FA)complementation group E(FANCE)and miR-26a-5p was verified by dual-luciferase reporter assay.RESULTS:TGF-β2 induced NEAT1 expression in LECs.NEAT1 inhibition accelerated apoptosis,cell cycle arrest,decreased proliferation,epithelial-mesenchymal transition(EMT),and migration of TGF-β2-disposed LECs.NEAT1 sponged miR-26a-5p to further regulate FANCE expression.Rescue experiments presented that miR-26a-5p downregulation overturned NEAT1 silencing-mediated impacts on TGF-β2-disposed LEC biological behaviors.Additionally,FANCE overexpression reversed miR-26a-5p mimic-mediated impacts on TGF-β2-disposed LEC biological behaviors.CONCLUSION:TGF-β2-induced NEAT1 facilitates LEC proliferation,migration,and EMT by upregulating FANCE via sequestering miR-26a-5p. 展开更多

目的:探讨p16/Ki-67双染检测、HPVE6/E7mRNA检测在宫颈病变诊断中的价值。方法:选择2021年4月至2023年2月我院收治的605例高危型HPV阳性患者为研究对象,所有患者入院后均进行阴道镜检查与病理活检,取患者细胞学样本分别进行p16/Ki-67双... 目的:探讨p16/Ki-67双染检测、HPVE6/E7mRNA检测在宫颈病变诊断中的价值。方法:选择2021年4月至2023年2月我院收治的605例高危型HPV阳性患者为研究对象,所有患者入院后均进行阴道镜检查与病理活检,取患者细胞学样本分别进行p16/Ki-67双染检测、HPVE6/E7mRNA检测。比较两种方法单独检查与联合检测对宫颈病变阳性检出率的差异;比较三种检查方法对宫颈病变诊断效能的差异。结果:联合检测检出CIN1~3阳性率均高于p16/Ki-67双标记、HPVE6/E7mRNA检测(P<0.05);三种检测方式的浸润癌阳性检出率比较无明显差异(P>0.05)。Kappa检验分析结果显示,p16/Ki-67双标记与HPVE6/E7mRNA检查结果与病理诊断结果具有较好的一致性(Kappa值=0.719,P<0.05);联合检测结果与病理诊断结果具有高度的一致性(Kappa值=0.894,P<0.05)。联合检测对宫颈病变的诊断灵敏度、特异性与准确率高于p16/Ki-67双标记与HPVE6/E7mRNA检查(P<0.05)。结论:p16Ki-67双染与HPVE6/E7mRNA联合检测可准确检出宫颈病变程度,对宫颈病变疾病类型的鉴别可提供充足可靠的数据支持,联合检查的诊断效能较好,对患者后续相关治疗开展有积极指导意义。 展开更多

目的:探讨p16 INK4A和HPV16 E7/HPV18 E6在宫颈细胞学诊断为非典型性鳞状细胞不明确意义型(ASCUS)中的表达及筛查潜在宫颈病变的价值。方法:对150例ASCUS患者行阴道镜检查并取活检,同时对该150例患者的TCT标本进行免疫细胞化学染色检测p... 目的:探讨p16 INK4A和HPV16 E7/HPV18 E6在宫颈细胞学诊断为非典型性鳞状细胞不明确意义型(ASCUS)中的表达及筛查潜在宫颈病变的价值。方法:对150例ASCUS患者行阴道镜检查并取活检,同时对该150例患者的TCT标本进行免疫细胞化学染色检测p16INK4A的表达和RT-PCR法检测其中HPV16型E7蛋白和18型E6蛋白(HPV16 E7/HPV18 E6)mRNA的表达。结果:p16INK4A和HPV16 E7/HPV18 E6 mRNA在ASCUS中表达的阳性率分别为37.33%和46.67%,随着病理级别的增加,p16INK4A和HPV16E7/HPV18 E6 mRNA表达的阳性率也随之增加;p16INK4A和HPV16 E7/HPV18 E6 mRNA筛查ASCUS中宫颈病变的灵敏度、特异度、阳性预测值和阴性预测值分别为0.88、0.95、0.91、0.93和0.81、0.75、0.67、0.86,在p16INK4A和HPV16 E7/HPV18 E6 mRNA阳性的样本中,宫颈病变发生率分别为91.07%和67.14%,均明显高于阴性样本中的发病率7.45%和13.75%(P<0.001);ASCUS中宫颈病变样本中p16INK4A和HPV16 E7/HPV18 E6 mRNA呈高表达,且具有较高的一致性(κ=0.6475)。结论:p16INK4A和HPV16 E7/HPV18 E6 mRNA在ASCUS中病理诊断为宫颈病变的样本中均呈高表达,对筛查潜在的宫颈病变具有重要意义,其中p16INK4A的筛查效能优于HPV16E7/HPV18E6mRNA;p16INK4A能间接反映HPV的转录活性,在ASCUS的分流中有重要意义,且可视性的p16INK4A免疫染色比HPV检测更直观。 展开更多

The high impact properties of copolymer ICP2 may attribute tothe relatively higher ethylene content and homogeneous ethylene unit distribution. The Prognostic Value of Pathological and Molecular Margins Marked by p53 and eIF4E in Laryngeal Carcinoma

Objective: To study the prognostic value of the pathological margin and molecular margin marked by eIF4E and P53 protein in laryngeal carcinoma. Methods: The prognostic value of pathological and molecular margin was studied in 253 cases and 67 cases respectively, the latter were pathological negative margin chosen from the former. Immunohistochemisty was used to detect the expression of eIF4E and p53 proteins. Results: The rate of pathological, p53 and eIF4E positive margins was 20.2%, 19.4% and 32.8% respectively. The recurrent rate of those with positive margins was higher than that of negative margins, which including pathological margin (70.6% vs 35.1%, P =0.0000), p53 margin (69.2% vs 33.3%, P =0.018) and eIF4E margin (63.6% vs 28.9%, P =0.018); The survival rate of those with negative margins was higher than those with positive margins, including pathological margin (the 5-year cumulative survival rate was 37.52% and 64.37% respectively, P =0.0023), p53 margin (the 5-year cumulative survival rate was 24.62% and 75.69% respectively, P =0.0012) and eIF4E margin (the 5-year cumulative survival rate was 43.31% and 77.52% respectively, P =0.0006). Conclusion: The prognosis of those with both pathological and molecular positive margins was worse than that of the negative margins; Both the eIF4E and p53 were useful markers to pick out the poor prognostic patients from those with pathological negative margin, and the former seemed to be more potential. 展开更多
